Description : Crop nutrient management is a critical component of productivity and quality. Commercial horticulture has relied almost exclusively on inorganic nutrient sources to satisfy plant nutritional requirements. Compost tea represents a sustainable alternative technology but its use has not been extensively researched. A factorial experiment was conducted subjecting lettuce plants to four increasing concentrations of compost tea; 0, 5, 10 and 20 % (m/v) applied at either 100 or 200 cm3 plant-1 day-1, for 30 days. At the end of the trial, measurements were made of leaf area (LA), yield, shoot and root dry mass (DM), root to shoot ratio, and nitrogen (N) and phosphorus (P) contents of shoots and roots. Compost tea concentration significantly (P < 0.05) affected all growth indices showing a similar pattern. Increasing concentration up to 10 % (m/v), positively improved growth and yield above the control. Increasing further to 20 % (m/v) reduced LA, yield and dry mass. For these variables, applying compost tea at 200 cm3 plant-1 day-1 resulted in a better response. Increasing concentration of compost tea showed a directly proportional relationship to shoot and root N content. However, the effect was non-significant for P. Yield and shoot DM were affected by the combination of compost tea concentration and application rate. Similar patterns of increasing yield were seen for increasing concentration within both rates however, the differences were significant for all concentrations at the higher application rate. Application of 200 cm3, 5 % compost tea resulted in the greatest yield, shoot and root DM. In addition to root DM, root surface area was significantly correlated with yield and shoot DM. Compost tea used at appropriate concentrations improved root growth and lettuce productivity.

STUDY QUESTION: Do freezing and in vitro culture procedures enhance the expression of proteins involved in apoptotic or autophagic pathways in murine pre-pubertal testicular tissue? SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. We present an integrated study of geochemistry, petrofabrics and seismic properties of strongly sheared eclogites from the Chinese Continental Scientific Drilling (CCSD) project in the Sulu ultrahigh-pressure (UHP) metamorphic terrane, eastern China. First, geochemical data characterize diverse protoliths of the studied eclogites. The positive Eu- and Sr-anomalies, negative Nb anomaly and flat portion of heavy rare earth elements in coarse-grained rutile eclogites (samples B270 and B295) suggest a cumulate origin in the continental crust, whereas the negative Nb anomaly and enrichment of light rare earth elements in retrograde eclogites (samples B504, B15 and B19) imply an origin of continental basalts or island arc basalts. Second, P-wave velocities (V-p) of three typical eclogite samples were measured under confining pressures up to 500 MPa and temperatures to 700 degrees C. At 500 MPa and room temperature, the mean V-p reaches 8.50-8.53 km/s in samples B270 and B295 but drops to 7.86 km/s in sample B504, and the P-wave anisotropy changes from 1.7-2.7% to 5.5%, respectively. The pressure and temperature derivatives of V-p are larger in the retrograde eclogite than in fresh ones. Third, the electron backscatter diffraction (EBSD) measurements of the eclogites reveal random crystal preferred orientation (CPO) of garnet and pronounced CPO of omphacite, which is characterized by a strong concentration of [001]-axes sub-parallel to the lineation and of (010)-poles perpendicular to the foliation. The asymmetric CPO of omphacite in sample B270 recorded a top-to-the-south shear event during subduction of the Yangtze plate. The calculated fastest V-p is generally sub-parallel to the lineation, but a different deformation environment during exhumation could form second-order variations in omphacite CPO and affect the V-p distribution in eclogites (e.g., the fastest V-p is at similar to 35 degrees from the foliation in sample 13295). Comparison between measured and calculated seismic properties indicates that the CPO of omphacite controls the seismic anisotropy of eclogites at high pressure, and compositional layering and retrograde minerals will increase the anisotropy. Calculated P-wave velocities agree well with velocities measured at 500 MPa and room temperature for fresh eclogites, but much higher than those of retrograde eclogite. As a case study, the laboratory-derived V-p-P and V-p-T relationships were used to estimate P-wave velocities of eclogites and peridotites beneath the Western Superior Province, Canada. The results indicate that besides the fabric-induced anisotropy, the direction dependence of pressure and temperature derivatives of V-p can significantly increase seismic anisotropy of eclogites with depth, which results in eclogites being an important candidate for the seismic anisotropy in the upper mantle. Due to their very high density and velocity, garnet-rich eclogites within peridotite could be detected in seismic reflections in subduction zones.

Objective To guide nosology and classification of patients with eosinophilic granulomatosis with polyangiitis (EGPA) based on phenotype and presence or absence of ANCA. Methods Organ manifestations and ANCA status were retrospectively analyzed based on the presence or not of predefined definite vasculitis features or surrogates of vasculitis in patients asthma, eosinophilia, and at least one systemic organ manifestation attributable to systemic disease. Results The study population included 157 patients (mean age 49.4 ± 14.1), with a follow-up of 7.4 ± 6.4 years. Patients with ANCA (31%) more frequently had weight loss, myalgias, arthralgias, biopsy-proven vasculitis, glomerulonephritis on biopsy, hematuria, leukocytoclastic capillaritis and/or eosinophilic infiltration of arterial wall on biopsy, and other renal disease. A total of 41% of patients had definite vasculitis manifestations (37%) or strong surrogates of vasculitis (4%), of whom only 53% had ANCA. Mononeuritis multiplex was associated with systemic vasculitis (p = 0.005) and with the presence of ANCA (p < 0.001). Overall, 59% of patients had polyangiitis as defined by definite vasculitis, strong surrogate of vasculitis, mononeuritis multiplex, and/or ANCA with at least one systemic manifestation other than ENT or respiratory. Patients with polyangiitis had more systemic manifestations including arthralgias (p = 0.02) and renal disease (p = 0.024), had higher peripheral eosinophilia (p = 0.027), and a trend towards less myocarditis (p = 0.057). Using predefined criteria of vasculitis and surrogates of vasculitis, ANCA alone were found to be insufficient to categorise patients with vasculitis features. Conclusion We suggest a revised nomenclature and definition for EGPA and a new proposed entity referred to as hypereosinophilic asthma with systemic (non vasculitic) manifestations. © 2016 Elsevier B.V.

Résumé : Vibrio splendidus, strain LGP32, is an oyster pathogen associated with the summer mortalities affecting the production of Crassostrea gigas oysters worldwide. Vibrio splendidus LGP32 was shown to resist to up to 10 mu M Cg-Def defensin and Cg-BPI bactericidal permeability increasing protein, two antimicrobial peptides/proteins (AMPs) involved in C. gigas immunity. The resistance to both oyster Cg-Def and Cg-BPI and standard AMPs (polymyxin B, protegrin, human BPI) was dependent on the ompU gene. Indeed, upon ompU inactivation, minimal bactericidal concentrations decreased by up to fourfold. AMP resistance was restored upon ectopic expression of ompU. The susceptibility of bacterial membranes to AMP-induced damages was independent of the ompU-mediated AMP resistance. Besides its role in AMP resistance, ompU proved to be essential for the adherence of V. splendidus LGP32 to fibronectin. Interestingly, in vivo, ompU was identified as a major determinant of V. spiendidus pathogenicity in oyster experimental infections. Indeed, the V. splendidus-induced oyster mortalities dropped from 56% to 11% upon ompU mutation (Kaplan Meier survival curves, P < 0.01). Moreover, in co-infection assays, the ompU mutant was out competed by the wild-type strain with competitive indexes in the range of 0.1-0.2. From this study, ompU is required for virulence of V. splendidus. Contributing to AMP resistance, conferring adhesive properties to V. splendidus, and being essential for in vivo fitness, the OmpU porin appears as an essential effector of the C. gigas/V. splendidus interaction.

Previous studies have reported respiratory, cardiac and muscle changes at rest in triathletes 24 h after completion of the event. To examine the effects of these changes on metabolic and cardioventilatory variables during exercise, eight male triathletes of mean age 21.1 (SD 2.5) years (range 17-26 years) performed an incremental cycle exercise test (IET) before (pre) and the day after (post) an official classic triathlon (1.5-km swimming, 40-km cycling and 10-km running). The IET was performed using an electromagnetic cycle ergometer. Ventilatory data were collected every minute using a breath-by-breath automated system and included minute ventilation (V(E)), oxygen uptake (VO2), carbon dioxide production (VCO2), respiratory exchange ratio, ventilatory equivalent for oxygen (V(E)/VO2) and for carbon dioxide (V(E)/VCO2), breathing frequency and tidal volume. Heart rate (HR) was monitored using an electrocardiogram. The oxygen pulse was calculated as VO2/HR. Arterialized blood was collected every 2 min throughout IET and the recovery period, and lactate concentration was measured using an enzymatic method. Maximal oxygen uptake (VO2max) was determined using conventional criteria. Ventilatory threshold (VT) was determined using the V-slope method formulated earlier. Cardioventilatory variables were studied during the test, at the point when the subject felt exhausted and during recovery. Results indicated no significant differences (P > 0.05) in VO2max [62.6 (SD 5.9) vs 64.6 (SD 4.8) ml x kg(-1) x min(-1)], VT [2368 (SD 258) vs 2477 (SD 352) ml x min(-1)] and time courses of VO2 between the pre- versus post-triathlon sessions. In contrast, the time courses of HR and blood lactate concentration reached significantly higher values (P < 0.05) in the pre-triathlon session. We concluded that these triathletes when tested 24 h after a classic triathlon displayed their pre-event aerobic exercise capacity, bud did not recover pretriathlon time courses in HR or blood lactate concentration.

This study investigated the possibility of there being differences in respiratory muscle strength and endurance in elite and competition triathletes who have similar maximal oxygen uptakes (V_O2max) and ventilatory thresholds (Thvent). Five internationally-ranked elite, [mean (SD) age 23.8 (1.4) years] and six nationallyand regionally-ranked competition [age 21.1 (1.1) years] male triathletes performed two successive trials: first an incremental cycle test to assess V_O2max and Thvent and second 20 min of cycling followed by 20 min of running (C-R) at intensities higher than 85% V_O2max. Cardioventilatory data were collected every minute during the two trials, using an automated breath-by-breath system. Maximal expiratory and inspiratory (PImax) strength were assessed before and 10 min after C-R from the functional residual capacity. Respiratory muscle endurance was assessed 1 day before and 30 min after C-R by measuring the time limit (tlim). The results showed firstly that during C-R, the competition triathletes had significantly (P<0.05) higher minute ventilation [mean (SEM) 107.4 (3.1) compared to 99.8 (3.7) lÆmin-1], breathing frequency [44.4 (2.0) compared to 40.2 (3.4) Æmin-1] and heart rate [166 (3) compared to 159 (4) beatsÆmin-1] and secondly that after C-R, they had significantly lower PImax [127.1 (4.2) compared to 130.7 (3.0) cmH2O] and tlim [2:35 (0:29) compared to 4:12 (0:20) min] than the elite triathletes. We conclude that, despite similar V_O2max and Thvent, the competition triathletes showed less extensive adaptive mechanisms, including those in the respiratory muscles, than did the elite triathletes. This led to higher ventilation, which appeared to be the cause of the faster development of fatigue in the inspiratory muscles in this group.

B-type olivine fabrics are pervasive within highly depleted dunites of the small-sized Imono peridotite body located within the subduction-type Sanbagawa metamorphic belt of the southwest Japan arc. The dunites contain various microstructures, ranging from porphyroclastic to fine-grained intensely sheared textures. The Mg/(Mg+Fe) atomic ratios (Fo number) of olivine within these dunites are consistently around 0.9, as are the Cr/(Cr+Al) atomic ratios (Cr number) of chromian spinel, suggesting their evolution from a highly depleted magma (boninite). These data provide strong thermal constraints on the formation of the highly depleted dunites,as their formation requires hot, hydrous, shallow mantle (> 1250 degrees C at < 30 km depth) in the mantle wedge. Because the Sanbagawa metamorphic belt finally entrained these peridotites during progressive retrogression, B-type olivine fabrics probably developed in the fore-arc side of the subduction zone, above or along the subducting slab, possibly in association with dehydration fluids derived from the slab. The previously documented small magnitude of S-wave splitting can be explained by the seismic properties of B-type peridotites within an anisotropic layer of approximately several kilometers in thickness, oriented by flow parallel to the subducting slab, under maximum temperatures of 880-1030 degrees C depending on the flow stress. These findings indicate that such a B-type layer could constitute a dominant source of seismic S- and P-wave anisotropy in mantle wedge regions. (c) 2008 Elsevier B.V. All rights reserved.
