Inter-specific and ontogenic differences in delta C-13 and delta N-15 values and Hg and Cd concentrations in cephalopods Auteur(s) : Chouvelon, Tiphaine Spitz, Jerome Cherel, Y. Bustamante, Paco Éditeur(s) : Inter-research Résumé : Chemical tracers (e. g. stable isotopes or trace metals) are increasingly used to study trophic ecology and feeding habits of marine organisms and various factors can affect their values. The aim of the present study was to provide information on ontogenic effects on stable carbon and nitrogen values (delta C-13 and delta N-15) and on cadmium (Cd) and mercury (Hg) concentrations in 5 cephalopod species from the Bay of Biscay (North-East Atlantic Ocean). To this end, individuals belonging to 3 species showing the widest range of sizes were analysed for muscle delta C-13 and delta N-15 values and muscle Hg, and digestive gland Cd concentrations. Results showed that stable isotope ratios allowed discrimination of specific feeding strategies during ontogenesis. Segregation between 5 cephalopod species in terms of trophic ecology was also evidenced (different isotopic niches). In contrast, Hg concentrations varied over the same order of magnitude in these 5 cephalopod species, despite higher levels in the benthic octopus Eledone cirrhosa. Consistently, Hg concentrations followed the same ontogenic pattern and increased with increasing body size/age of cephalopods. Finally, Cd concentrations varied over 3 orders of magnitude among the 5 species. Despite possible effects of physiology in terms of metal bioaccumulation, Cd concentrations were likely to reflect specific feeding preferences or feeding zones and ontogenic variability within a single species. Thus, ontogenic effects have to be taken into account when stable isotope ratios of carbon and nitrogen or trace metals are used as ecological tracers; the best recommendation being to focus on a given class age. Nutrition, digestion et développement des larves de crevettes et poissons marins Auteur(s) : Cahu, Chantal Résumé : This report is presented by Chantal Cahu for obtaining accreditation to supervise research. Since 1981, her research activity has addressed the study of reproducers' nutrition and the young stages of development of marine organisms belonging to two different branch lines, crustaceans and fish. This work, performed in a base-lined research body, IFREMER, is placed within a more general programme of aquaculture development. Indeed, whatever species are at issue, the development of aquaculture in France, as at the global level, is based on the production of juveniles in hatcheries. My work has therefore more precisely the goal of acquiring knowledge about the nutritional needs, mechanisms of digestion and the development of these animals in order to improve the production of prawn post-larvae and fish alevin in the hatchery.
This work took place in three main periods that appear in this report:
- The study of reproducers' nutrition and its influence on the quality of the Penaeid prawn's eggs and larvae. My work has focused mainly on the effect of lipids and vitamins on the development of eggs and larvae.
- The study of the ontogenesis of the digestive functions and the nutritional needs of sea fish larvae, with bass as the model. The goal was to determine the specific nutritional needs of the young stages by studying the implementation and regulation of pancreatic and intestinal digestive enzymes.
- The study of the regulation of certain developmental genes by means of nutrients, so as to understand how these nutrients behave during ontogenesis, particularly during development of the skeleton.
Ce mémoire est présentée par Chantal Cahu pour pour l'obtention d'une habilitation a diriger des recherches. Depuis 1981, son activité de recherche concerne l'étude de la nutrition des reproducteurs et des jeunes stades de développement d'organismes marins appartenant à deux embranchements différents, les crustacés et les poissons. Ce travail réalisé dans un organisme de recherche finalisée, l'Ifremer, se situe dans un programme plus général de développement de l'aquaculture. En effet, quelles que soient les espèces en jeu, le développement de l'aquaculture en France comme au niveau mondial repose sur la production de juvéniles en écloserie. Mon travail a donc plus précisément pour objectif l'acquisition de connaissances sur les besoins nutritionnels, les mécanismes de digestion et le développement des ces animaux afin d'améliorer la production de post-larves de crevettes et d'alevins de poissons en écloserie. Ce travail s'est déroulé en trois grandes périodes qui apparaissent dans ce mémoire : - L'étude de la nutrition des reproducteurs et son influence sur la qualité des oeufs et des larves de crevettes Pénéides. Mon travail a essentiellement porté sur l'effet des lipides et des vitamines sur le développement des oeufs et des larves. - L'étude de l'ontogenèse des fonctions digestives et les besoins nutritionnels des larves de poissons marins, avec le bar comme modèle. Le but était de déterminer les besoins nutritionnels spécifiques des jeunes stades, en étudiant la mise en place et la régulation des enzymes digestives pancréatiques et intestinales. - L'étude de la régulation de certains gènes du développement par les nutriments, de façon à comprendre comment ces nutriments agissent lors de l'ontogenèse, notamment lors du développement du squelette. Je suis dorénavant au laboratoire GRIMAAG en Guadeloupe. We focused on the communication problem within multi-agents systems, inspirited by nature and more precisely by the ants that are a perfect example of living integrating learning (ontogenesis) to genetic inheritance (phylogenesis): the Baldwin Effect. We worked with agents represented by classifiers systems using genetic algorithm. We introduced a new fixed knowledge based classifiers system, the simplified Pittsburgh classifier system, which we studied in a spontaneous coordination multi-agents problem. Then, we successfully allowed “homogeneous” agents to communicate their “knowledge” using distributed elitism. Finally, “heterogeneous” agents efficiently exchanged information thanks to a minimal communication model we defined, bound to reality and extended. Nous nous sommes intéressés à la problématique de la communication au sein des systèmes multi-agents en nous inspirant de la nature et plus particulièrement des fourmis qui sont un parfait exemple de l'intégration de l'apprentissage du vivant (ontogenèse) au capital génétique de celui-ci (phylogénie) : l'effet Baldwin. Nous avons choisi de travailler avec des systèmes de classeurs à algorithme génétique pour représenter les agents. Nous avons introduit un nouveau type de système de classeurs à base de connaissance de taille fixe, les systèmes de classeurs de type Pittsburgh simplifié, dont nous avons étudié les propriétés dans un contexte multi-agents de coordination spontanée. Nous avons ensuite proposé avec succès aux agents "homogènes" de communiquer pour échanger leur "connaissance" à l'aide de l'élitisme distribué. Scanning electronic observations show that morphogenetic processes result in a gastrula with two depressions 4 hours after fertilization (T0 + 4h). Two hours later, one depression, located at the animal pole, develops into an open cave, the floor of which becomes the shell field located below the lower face of the prototrochal pad. The invagination located at the vegetal pole features the blastopore. At T0 + 6h, the late gastrula has differentiated into a typical motile trochophore with a shell field synthetizing the organic part of the shell. At T0 + 8h, the shell field, located between the prototroch and the telotroch, appears as a saddle-shaped region with a wrinkled surface extending on both sides of the embryo, establishing bilateral symmetry. At T0 + 12h, the prototroch slides toward the anterior region by outgrowth of the shell material. At T0 + 18h, the prodissoconch I formation is completed and the D-stage larvae possess a calcified shell. At this stage of development, the functional velum is composed of four bands of cilia. Our observations seem to confirm the interpretation of shell differentiation proposed recently by Mouëza et al. (2006) for bivalves even if Transmission Electronic observations will be necessary to validate definitively such a model in the Meretricinae sub-family. Bases biologiques de la production de deux Mollusques d'intérêt économique: reproduction chez Megathura crenulata, reproduction et développement chez Crassostrea gigas Auteur(s) : Cannuel, Rozenn Éditeur(s) : Université de Nantes Résumé : The reproductive cycle of Megathura crenulata was documented and quantification of oocyte lipid reserves with lipo-specific stains was explored in both this species and in Crassostrea gigas. The necessity of providing food to female broodstock C. gigas in spring was re-evaluated, because subsequent larval and post-larval rearing performance was not affected by broodstock starvation during conditioning. Developmental study of the organs responsible of particle processing in C. gigas allowed the identification of critical stages related to the ontogenesis of each organ, as well as a better understanding of the evolution of these organs, highly-modified in the Ostreidae. Ciliary type identification and mucocyte mapping from post-larval to adult stage showed that the heterorhabdic specialization precedes the anatomic formation of gill plicae specific to this condition. Le cycle reproducteur de Megathura crenulata a été documenté et la quantification des réserves ovocytaires par des colorants lipospécifiques a été tentée, sans résultats concluants pour cette espèce, ni pour Crassostrea gigas. L'intérêt d'une alimentation des géniteurs femelles C. gigas au printemps est re-évalué, car la performance ultérieure des élevages larvaires et post-larvaires n'a pas été affectée par le jeûne imposé lors du conditionnement. L'étude du développement des organes impliqués dans le traitement des particules chez C. gigas a permis d'identifier des stades critiques associés à l'ontogenèse de chaque organe, ainsi qu'une meilleure compréhension de l'évolution de ces organes si particuliers chez les Ostreidae. L'examen des types ciliaires et la cartographie des mucocytes de la post-larve à l'adulte ont montré que la spécialisation hétérorhabdique précède même la formation anatomique des plis branchiaux caractéristiques de cette condition. Transmission (TEM) and scanning (SEM) electron microscopy show that morphogenetic processes result in a gastrula with two depressions, 4 h after fertilization (T014 h). Two hours later, one depression, located at the animal pole, develops into an open cave, the floor of which becomes the shell field, characterized by scarce microvilli, located below the lower face of the prototrochal pad. The invagination located at the vegetal pole is characterized by regular microvilli and constitutes the digestive tract. At T018 h, the late gastrula differentiates into a typical motile trochophore, possessing a well-developed prototroch delimiting anterior and posterior embryonic regions. At T019h, the shell field, located between the prototroch and the telotroch, appears as a saddle-shaped region with a wrinkled surface extending on both sides of the embryo, establishing bilateral symmetry. From TEM observations, the apical surface of the underlying cells possesses a modified glycocalyx featuring a dense pellicle overlapping the cells of the shell field. At T0112 h, the prototroch is displaced toward the anterior region by outgrowth of the shell material. TEM views show that the wrinkled surface observed with SEM corresponds to the periostracum, attached dorsally to the hinge and ventrally to the cells of the future mantle edge. At T0115 h, an alveolar matrix of proteinaceous material, which could contain calcite crystals, appears between the periostracum and the mantle. At T0124 h, prodissoconch I formation is completed and the D-stage larvae possess a calcified shell. At this stage of development, only one mantle fold is present, representing the outer fold. The periostracumsecreting cells are in contact with the marginal cells of the velum. This functional velum is composed of four bands of cilia. Our TEMobservations confirm previous descriptions of the velum ciliary bands obtained only from SEM analyses, such as described in other bivalves. The apical sense organ and the digestive tract are poorly developed. From observations at the ultrastructural level of shell differentiation in C. cancellata, we propose a new interpretation of shell differentiation, including hinge and ligament for bivalves as opposed to that described for gastropods. Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale Auteur(s) : Breuil, Gilles Éditeur(s) : Université de Montpellier II Résumé : The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under the chorionic membrane. Secondly, we have studied the development of the immunity system on larvae, preparing IgM assays with ELISA method, the study on the organogenesis of the main lymphoid organs, and using flow cytometry for the ontogenesis of IgM producing cell. Based on this experience, we can propose a date for the immunocompetence onset of the bar, i.e. from the 54th day of the hatchery phase. Finally, we made the most of these different results to study a viral pathology (nodavirus) in spawners and larvae. We defined an experimental contamination model for larvae and eggs, and we highlighted that eggs born from vaccinated hen fishes were protected against the nodavirus. Le bar, Dicentrarchus labrax, fait l'objet d'une aquaculture intensive sur le littoral Méditerranéen depuis une dizaine d'années. Les écloseries marines représentent un des maillons clefs de cette filière aquacole, et sont confrontées à différentes pathologies, dont une d'origine virale (nodavirus), pouvant remettre en cause leur faisabilité. Aussi, nous avons tenté d'apporter quelques données concernant la protection des larves et des géniteurs reproducteurs. Dans un premier temps, nous avons étudié les immunoglobulines, éléments de l'immunité spécifique humorale ainsi que leur rôle anticorps. Chez les bars adultes ces immunoglobulines sont des molécules tétramériques de haut poids moléculaire (850 kD), composées de chaînes lourdes (H) de 80 kD et légères (L) de 26 kD. Dans les oeufs, des formes plus simples, monomériques ou dimériques, sont mises en évidence et sont localisées sous la membrane chorionique. Dans un second temps, nous avons étudié la mise en place du système immunitaire spécifique chez les larves, en effectuant le dosage des IgM par ELISA, l'étude de l'organogenèse des principaux organes lymphoïdes, puis celle de l'ontogenèse des cellules productrices d'IgM par cytométrie de flux. L'ensemble de cette étude nous permet de proposer une date d'acquisition de l'immunocompétence à partir du 54ème jour d'élevage chez le bar. Enfin, ces différents résultats ont été mis à profit dans le cadre d'une pathologie virale (nodavirus) touchant les reproducteurs et les larves. Après avoir défini un modèle de contamination expérimental des larves et des oeufs, nous avons mis en évidence une protection des oeufs lorsque ceux-ci proviennent d'une femelle vaccinée (vaccin expérimental) contre le nodavirus.
