INRA, CFCS Description : Banana and plantain (Musa spp) are important foreign exchange earners in Guyana and the Windward Islands of the Caribbean. Black Sigatoka Disease (BSD) also referred to as black leaf streak disease is one of the most destructive diseases of banana and plantain and is present within the region. In Guyana within two years following its introduction in 2008, the industry was totally decimated, recording a 100% decline, turning the country to a net banana importer. Also, from 2009 ? 2012 the disease spread in the Windward Islands causing as much as 100% decrease in the export of banana. The disease is caused by the ascomycete fungus, Mycosphaerella fijiensis (anamorph Pseudocercospora fijiensis) and is aggressive, challenging to control and results in enormous crop damage if left uncontrolled. In an effort to develop an integrated management approach to the disease the Caribbean Agricultural Research and Development Institute (CARDI) with financial assistance from the Caribbean Development Bank (CDB) is evaluating five FHIA banana and plantain accessions and eight IITA plantain accessions for tolerance to BSD. The evaluation is presently conducted in four Caribbean countries, Dominica, Guyana, St Lucia and St Vincent and the Grenadines. In each country, four distinct agro- ecological zones were selected for the trial evaluation blocks and using a randomized block design, four blocks were established in each country. Five FHIA accessions: FHIA-01, FHIA-03, FHIA-18, FHIA-21 and FHIA-23 are being evaluated in Dominica, St Lucia and St Vincent and the Grenadines whereas, plantain accessions: PITA-17, PITA-21, PITA-22, PITA-23, PITA-24 and PITA-26 are to be evaluated in Guyana for tolerance to the disease. Cavendish are used as regional controls and as borders rows to increase disease pressure. The disease parameters evaluated are youngest leaf spotted (YLS), disease development time (DDT), leaf emission rate (LER), disease severity at 6 months, bunch emergence and harvest, and index of standing leaves. Agronomic data; days from planting to flowering, bunch weight, number of hands in bunch, average weight of hands and average number of fingers per hand and bunch, as well as organoleptic characteristics of both cooked and ripe fruits are also evaluated. The early data collection, of the FHIA only accessions, show trends in the disease tolerance and agronomic performance as well as their adaptability in the different agro-ecological zone. The trends indicate better disease tolerance of FHIA varieties with higher leaf counts before, at bunching and at harvest. The organoleptic test shows a preference to FHIA 03 and FHIA 21. La banane et le plantain (espèces de Musa) sont les acquéreurs de devises étrangères importants en Guyane et en Îles du Vent des Caraïbe. La maladie noire de Sigatoka (schéma) également désignée sous le nom de la maladie noire de filet de feuille est l'une des maladies les plus destructives de la banane et du plantain et est présente dans la région. En Guyane dans un délai de deux ans après son introduction en 2008, l'industrie a été totalement décimée, enregistrant une baisse de 100%, tournant le pays à un importateur net de banane. En outre, à partir de 2009-2012 la propagation de maladie en Îles du Vent causant pas moins de la diminution de 100% de l'exportation de la banane. La maladie est provoquée par le champignon d'ascomycète, fijiensis de Mycosphaerella (fijiensis anamorph de Pseudocercospora) et est agressive, contestant pour commander et les résultats dans l'énorme culture endommagent si laissé incontrôlé. Dans un effort de développer une approche intégrée de gestion à la maladie l'institut agricole des Caraïbes de recherche et développement (CARDI) avec l'aide financière de la banque de développement des Caraïbes (BDC) évalue des adhésions de banane et de plantain de cinq FHIA et huit adhésions de plantain d'IITA pour la tolérance au schéma. L'évaluation est actuellement effectuée quatre pays des Caraïbes, Dominique, Guyane, St Lucia et Saint-Vincent-et-les Grenadines. Dans chaque pays, quatre agro zones écologiques distinctes ont été sélectionnées pour les blocs d'essai d'évaluation et employant une conception de bloc randomisé, quatre blocs ont été établis dans chaque pays. Cinq adhésions de FHIA : FHIA-01, FHIA-03, FHIA-18, FHIA-21 et FHIA-23 sont évalués en Dominique, au St Lucia et au Saint-Vincent-et-les Grenadines tandis que, des adhésions de plantain : PITA-17, PITA-21, PITA-22, PITA-23, PITA-24 et PITA-26 doivent être évalués en Guyane pour la tolérance à la maladie. Cavendish sont employés en tant que contrôles régionaux et comme rangées de frontières pour augmenter la pression de la maladie. Les paramètres de la maladie évalués sont la plus jeune feuille repérée (YLS), le temps d'élaboration de la maladie (DDT), le taux d'émission de feuille (LER), la sévérité de la maladie à 6 mois, émergences et récoltes de groupe, et index des feuilles debout. Données agronomiques ; des jours de la plantation à la floraison, du poids de groupe, nombre de mains dans le poids de groupe et moyen des mains et du nombre moyen de doigts par main et groupe, aussi bien que les caractéristiques organoleptiques des fruits cuits et mûrs sont également évalués. La collecte de données tôt, des adhésions de FHIA seulement, exposition tend dans la tolérance de maladie et la représentation agronomique aussi bien que leur adaptabilité dans la zone agro-écologique différente. Les tendances indiquent qu'une meilleure tolérance de maladie des variétés de FHIA avec une plus haute feuille compte avant, à se rassembler et à la récolte. The dead ringer (dri) gene of drosophila melanogaster is a member of the recently discovered ARID-box family of eukaryotic genes that encodes proteins with a conserved DNA-binding domain. dri itself is highly well conserved with specific orthologs from human to yeast genomes. This family include ARID3A, 3B and 3C with have role in embryonic pattening, cell lineage gene regulation, cell cycle control, transcriptional regulation and chromatin structure modification. We first showed that the gene encoding ARID3A also known as Bright (B cell regulator of immunoglobulin heavy chain transcription) play critical roles in B-cell development and SATB1 in T-cell transcription regulation. In this international workshops we will expose a comprehensive understanding of these molecules and better knowledge of their specific targets and function-interactions in controlling cell fate decisions and development. In addition, dri have homology with the Aedes aegypti mosquitoes which had ability to carry and spread disease to human. More than half of the world?s population live in areas where this mosquito species is present, including Martinique. Sustained mosquito control efforts are important to prevent outbreaks from Dengue, Chikungunya, malaria, zika diseases. Discovering the functions of AT-rich genes in preventing illness from mosquito is the central goal for raising working progress hypothesis. INRA, CFCS Description : Breeding new interspecific banana hybrid varieties with pests and disease resistance characters is instrumental for the development of pesticide-free agroecological banana farming systems. Such breeding relies on the combined use of Musa acuminata and M. balbisiana parents. Unfortunately, infectious alleles of endogenous Banana streak virus (eBSV) sequences are present in the genome of M. balbisiana genitors. Upon activation by biotic and abiotic stresses, these infectious eBSVs lead to spontaneous infections by several species of Banana streak virus (BSV) in interspecific hybrids harbouring both M. acuminata and M. balbisiana genomes. In this work, we show that M. balbisiana diploid genitors available for breeding host at least one infectious eBSV. We also show how we segregated infectious and non-infectious eBSV alleles in seedy M. balbisiana diploids through self- pollination or chromosome doubling of haploid lines. We report on the successful breeding of M. balbisiana diploid genitors devoid of all infectious eBSV alleles following self-pollination and on the potential of breeding additional M. balbisiana diploid genitors free of infectious eBSVs by crossing parents displaying complementary eBSV patterns. This work paves the way to the safe use of M. balbisiana genitors for breeding banana interspecific hybrid varieties with no risk of activation of infectious eBSVs. This core offers instrumentation and expertise in fractionation, purification, and various chemical analyses to support synthetic and natural products drug discovery research groups. Our main equipment are dedicated to chromatographic separation and purification (MPLC, HPLC) and mass-spectrometry for analytic detection, quantification and identification (LC-MS SQ, QqQ and QToF). The CPAS services also include Circular Dichroism spectrometry, general spectroscopic characterization (UV, FTIR), lyophilization (bench top and high capacity freeze-dryers), synthesis (microwave reactor), ADME screening (permeability and drug solubility testing), and high-throughput screening development capability (automated liquid handler and multimode plate reader). Therefore a selective breeding program for resistance to bonamiosis has been undertaken since 1985 bu Ifremer, leading to the production of several selected oyster families. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters (injected with the parasite) to two tested families of oysters originating from the selective breeding program. Mortalities were checked daily, and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection to B. ostreae. The first infections occurred after 4 months of challenge in the tested oysters (Family 1 and Family 2). The cumulative mortalities after 5 monts were 58% for the wil oysters, 9% for Family 1 (20-month old at the beginning of the experiment) and 20% for Family 2 (8-month, old). The parasite could be detected in 66.8% of the dying wild oysters, 67.5% of the dying oysters of Family 1, 89% of the dying oysters of Family 2 and only 11% of the surviving oysters of Family 2. The mortality was significantly higher in Family 2 thant in Family 1 (x2= 20.87, p<0.001, d.f.) as well as the level of infection by the parasite found in heart smear (x2=24.34, p<0.001, 4 d.f.). This result demonstrates that prespawning oysters as yong as 1 year-old can become infected with the parasite and die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. The most probable cause of the dscrepancy in the development of bonamiosis between the 2 tested families is a difference in genetic background. Effet des conditions environnementales sur le développement des pathologies à Vibrio dans les élevages de crevettes en Nouvelle-Calédonie Auteur(s) : Lemonnier, Hugues Éditeur(s) : Université de La Rochelle Résumé : Shrimp aquaculture in New Caledonia is a developing industry, exclusively based on the species Litopenaeus stylirostris. However, it is subject to mortalities during grow out as observed all over the world. Since 1993, shrimp reared during the cold period are affected by a disease named "syndrome 93". As a consequence, the whole industry has become seasonal and produces essentially during the warm season. The climatic conditions during the periods of transition between the two main seasons could be responsible for an environmental stress and the origin reason for mortality outbreaks. Stocking density is the main risk factor. It increases the probability of the appearance and development of the epizooty in the pond. The "Summer Syndrome", a septicemic vibriosis caused by Vibrio nigripulchritudo, was identified for the first time in December 1997 in an intensive shrimp farm (DF). It has become enzootic ever since and is expressed after 50d of rearing. Since 2003, two others farms located near DF have developed the disease. As a consequence, the summer syndrome is considered to be a potential threat for all the industry. Between 1991 and 2005, the increase of feed input and nitrogen levels in feed has led to a significant increase in the growth rate over the years in the farms affected by the disease. In consequence, an early eutrophication of the water is observed, which is concomitant with the beginning of the mortality outbreaks. An early eutrophication and the virulence level of the pathogen are considered as the main risk factors of the disease. Factors may act synergistically to explain the summer syndrome. As part of a multidisciplinary approach (pathology physiology environment), two high-frequency surveys were carried out to analyse the pond ecosystem. Since the feed quantity increases with the biomass of shrimp, the eutrophication level of the pond ecosystem will also tend to increase with the duration of rearing. During the first part of the rearing, the abundance of each picoplankton type is exceptionally high and picophytoplankton dominated the autotrophic compartment. The nanophytoplankton dominated the second part of the rearing and was more unstable. The shift from pico to nanophytoplankton could be considered as an environmental stress and was observed in relation to the beginning of the mortalities, whatever the disease. Shift intensity could play a role by inducing directly or indirectly a stress for shrimp and/or a growth and/or virulence factors of the pathogen. Results from sediment studies show that mortality was best characterized by TAN concentration in pore water and even more when the TAN concentration was high. TAN and pH are potential stress factors for shrimp reared in ponds affected by these diseases. L'aquaculture mondiale de crevettes est un secteur à très forte croissance. Toutefois, les problèmes liés aux maladies dans les élevages ne cessent d'augmenter depuis 1980. Au cours du développement de la filière de Nouvelle-Calédonie, deux épisodes saisonniers de mortalités associées à la présence de Vibrio pathogènes sont apparus et ont fortement perturbé les techniques et les schémas de production. Les premières mortalités hivernales, décrites sous le nom de « syndrome 93 » ont été observées en mai 1993. Elles ont affecté depuis tous les élevages de saison froide de toutes les fermes de production. Les conditions climatiques pendant les périodes de transitions entre les deux principales saisons caractérisant le climat de la Nouvelle-Calédonie, seraient responsables d'un stress environnemental à l'origine du déclenchement du syndrome 93. La densité initiale d'ensemencement, apparaît comme le principal facteur de risque, augmentant la probabilité d'apparition de la maladie et l'ampleur de ses effets. Le « syndrome d'été » a été identifié pour la première fois en 1997 dans une ferme pratiquant une production intensive. Il s'est déclaré depuis 2003 dans deux autres fermes géographiquement proches. Les mortalités apparaissent 58 jours en moyenne après l'ensemencement des post-larves dans les bassins. La recherche d'une croissance maximale des animaux a induit, avec les années, une intensification progressive du système caractérisée par une augmentation de la quantité d'aliment distribuée. L'examen des données historiques fait apparaître une eutrophisation du milieu de plus en plus précoce concomitante au déclenchement des mortalités. En l'état actuel de nos connaissances, cette évolution précoce de l'écosystème en présence de façon récurrente de la souche hautement pathogène du Vibrio incriminé et à une période où la fréquence des mues des crevettes est élevée, constitue le principal facteur de risque de la maladie. Avec l'apport croissant en aliment, la colonne d'eau évolue vers une autotrophie croissante alors que le sédiment devient de plus en plus hétérotrophe au fur et à mesure que l'élevage progresse. Au cours du processus d'eutrophisation, le compartiment autotrophe montre une succession de deux assemblages. Le picophytoplancton domine sur la première partie de l'élevage et laisse place à du nanophytoplancton en seconde partie. Cette phase de transition, concomitante à l'apparition des mortalités quel que soit le syndrome, caractérise un stress environnemental dont l'intensité pourrait favoriser ou non le déclenchement des mortalités. Les suivis de différents indicateurs dans les sédiments - pH, potentiel d'oxydoréduction et concentration en ammoniaque dans l'eau interstitielle indiquent dans les bassins déclarant les épizooties des conditions qui ont été définies expérimentalement comme potentiellement plus stressantes pour les animaux. She is an Assistant Professor of Medicine at Harvard Medical School and an Associate Physician in the Division of Global Health Equity at Brigham and Women's Hospital (BWH). She completed her residency in Internal Medicine at Massachusetts General Hospital (MGH) and a fellowship in Infectious Diseases at the combined MGH/BWH program. Dr. Ivers also received a diploma in Tropical Medicine and Hygiene from the London School of Hygiene & Tropical Medicine and a Master of Public Health from the Harvard School of Public Health. Dr. Ivers implements health programs, and is interested in improving the delivery of healthcare in resource poor settings, the provision of care to the rural and urban poor, as well as patient-oriented investigation that offers solutions to barriers to healthcare. She balances her time between management of PIH Haiti, direct clinical service, and operational research. Dr. Ivers has contributed to published articles on HIV/AIDS, food insecurity, and the prevention of mother-to-child transmission of HIV. Current projects include investigating the impact of targeted food assistance to people with HIV infection in rural Haiti, the effectiveness of ARV therapies on viral suppression in community-based programs, and humanitarian assistance in response to the January 2010 earthquake. Dr. Ivers has served as a Technical Advisor to the WHO and also mentors Haitian and American physicians. Bonamia ostreae-induced mortalities in one-year old European flat oysters Ostrea edulis: experimental infection by cohabitation challenge Auteur(s) : Lallias, Delphine Arzul, Isabelle Heurtebise, Serge Ferrand, Sylvie Chollet, Bruno Robert, Maeva Beaumon, Andy Boudry, Pierre Éditeur(s) : EDP Sciences Résumé : Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection by B. ostreae. Mortality started after 4 months of cohabitation in the tested oysters. The cumulative mortalities after 6 months were 58% for the wild oysters, 9% for Family 1 (20-month old) and 20% for Family 2 (8-month old). In the dying oysters, the parasite could be detected in 67% of the wild oysters, 68% of Family 1 and 89% of Family 2. It was detected in only 11% of the surviving oysters of Family 2. The mortality and the level of infection by the parasite were significantly higher in Family 2 than in Family 1. Our results demonstrate that prespawning oysters as young as 1 year-old can become infected with the parasite and, most importantly, can die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. Additionally, no clear relationship between shell length and level of infection was observed. We also review the different methods for infection of the European flat oyster O. edulis with B. ostreae under experimental conditions and their main results. La bonamiose est une maladie parasitaire (agent causal : Bonamia ostreae) affectant l'huître plate européenne Ostrea edulis, responsable d'un déclin drastique de sa production aquacole. Des programmes de sélection pour la résistance à la bonamiose ont été entrepris pour contrer cette maladie. Dans cette étude, une expérience de 6 mois d'infection par cohabitation a été réalisée de manière à transmettre la maladie à partir d'huîtres sauvages injectées avec le parasite vers deux familles testées d'huîtres (âgées de 20 et 8 mois en début d'expérience, avec des origines génétiques différentes) issues du programme de sélection développé par IFREMER en France. Les mortalités ont été vérifiées quotidiennement et des frottis de coeur ventriculaire réalisés sur les huîtres mortes ou moribondes pour détecter le niveau d'infection par B. ostreae. La mortalité a commencé chez les huîtres testées après 4 mois de cohabitation. Les mortalités cumulées après 6 mois étaient de 58 % chez les huîtres sauvages, 9 % chez la Famille 1 (âgées de 20 mois) et 20 % chez la Famille 2 (âgées de 8 mois). Chez les huîtres mourantes, le parasite a pu être détecté chez 67 % des huîtres sauvages, 68 % de la Famille 1 et 89 % de la Famille 2. Il n'a pu être détecté que chez 11 % des huîtres survivantes de la Famille 2. La mortalité et le niveau d'infection par le parasite étaient significativement plus élevés chez la Famille 2 que chez la Famille 1. Nos résultats démontrent que des huîtres âgées de un an peuvent devenir infectées par le parasite et surtout, peuvent mourir de bonamiose. Ce résultat contraste avec l'âge critique de développement de la maladie communément accepté de 2 ans.
