Combined effect of exposure to ammonia and hypoxia on the blue shrimp Litopenaeus stylirostris survival and physiological response in relation to molt stage Auteur(s) : Mugnier, Chantal Zipper, Etienne Goarant, Cyrille Lemonnier, Hugues Éditeur(s) : Elsevier Résumé : The effect of ambient ammonia, hypoxia and combination of both on survival and the physiological and immunological response of the blue shrimp Litopenaeus stylirostris in relation to molt stage was studied. Shrimp were submitted to 44.0-71.5 mg 1(-1) total ammonia-N corresponding to 2.0 mg 1(-1) unionized ammonia NH3-N and/or to 1.5 mg O-2 1(-1) (4.3 kPa) for 24 hours. Survival was recorded and the molt stages of both dead and surviving shrimp determined. Only shrimp in intermolt and premolt stages were sampled for analysis of haemolymph. Haemolymph was assayed for osmoregulatory capacity (OC), magnesium ion (Mg2+), calcium ion (Ca2+), total proteins, oxyhaemocyanin, lactate, glucose and total haemocyte count (THC). Low mortalities were recorded for shrimp submitted independently to ammonia or hypoxia. Seventy five percent of dead shrimp were in early post molt (stage A) in ammonia treatment, while hypoxia affected mainly late premolt animals (stage D-2). A synergic effect of ammonia and hypoxia combination (A+ O-2 treatment) on mortality was observed, affecting nearly exclusively shrimp in late premolt stage D-2. Analysis of molt stage repartition at the end of the experiment suggests that ammonia treatment may have accelerated molting. The common physiological response of shrimp to the different treatments was characterized by a reduced OC and an increase in Ca2+. Increase in Mg2+ could not be validated by the statistical analysis, as well as glycaemia variations. Plasmatic lactate level increased and THC decreased in shrimp submitted to hypoxia and the combination of hypoxia and ammonia. Total proteins concentration was reduced in ammonia and A+O-2 treatments. The effect was more pronounced in late premolt shrimp than in intermolt shrimp. Combination of ammonia and hypoxia led to a physiological response stronger than this observed for ammonia-alone and/or hypoxia-alone treatments, except for oxyhaemocyanin. The effects of each external factor (ammonia, hypoxia) and the combination of both, and internal one (molt stage) are discussed. (c) 2007 Elsevier B.V All rights reserved. Combined effect of external ammonia and molt stage on the blue shrimp Litopenaeus stylirostris physiological response Auteur(s) : Mugnier, Chantal Justou, Carole Éditeur(s) : Elsevier Résumé : The effect of ambient ammonia and molt stage on the physiological response of the blue shrimp Litopenaeus stylirostris was studied. Shrimps were submitted to 54.6 mg l(-1) ammonia-N (1.76 mg l(-1) NH3-N) for 24 h. Only shrimps in stages C, D-0, D-1 and D-2 were used for the analysis. Haemolymph was assayed for osmoregulatory capacity (OC), magnesium ion (Mg ion), total proteins, oxyhaemocyanin, lactate and glucose. Molt stage had an effect on OC, Mg ion and total proteins in control shrimps, and on OC, Mg ion and lactate in treated animals. Ammonia treatment decreased OC and lactate, and increase Mg ion concentration in haemolymph, for all molt stages. It decreased significantly total proteins and oxyhaemocyanin for stages D-1 and D-2, and increased glucose concentration for stages C and Do. There was a combined effect of treatment and molt stage only on total proteins concentration. The effects of an external factor (ammonia), an internal one (molt stage) and the combination of both, and the usefulness of using physiological parameters measured in this study as tools to detect stress, are discussed. New Caledonia is a developing industry, exclusively based on the species Litopenaeus stylirostris. However, it is subject to mortalities during grow out as observed all over the world. Since 1993, shrimp reared during the cold period are affected by a disease named "syndrome 93". As a consequence, the whole industry has become seasonal and produces essentially during the warm season. The climatic conditions during the periods of transition between the two main seasons could be responsible for an environmental stress and the origin reason for mortality outbreaks. Stocking density is the main risk factor. It increases the probability of the appearance and development of the epizooty in the pond. The "Summer Syndrome", a septicemic vibriosis caused by Vibrio nigripulchritudo, was identified for the first time in December 1997 in an intensive shrimp farm (DF). It has become enzootic ever since and is expressed after 50d of rearing. Since 2003, two others farms located near DF have developed the disease. As a consequence, the summer syndrome is considered to be a potential threat for all the industry. Between 1991 and 2005, the increase of feed input and nitrogen levels in feed has led to a significant increase in the growth rate over the years in the farms affected by the disease. In consequence, an early eutrophication of the water is observed, which is concomitant with the beginning of the mortality outbreaks. An early eutrophication and the virulence level of the pathogen are considered as the main risk factors of the disease. Factors may act synergistically to explain the summer syndrome. As part of a multidisciplinary approach (pathology physiology environment), two high-frequency surveys were carried out to analyse the pond ecosystem. Since the feed quantity increases with the biomass of shrimp, the eutrophication level of the pond ecosystem will also tend to increase with the duration of rearing. During the first part of the rearing, the abundance of each picoplankton type is exceptionally high and picophytoplankton dominated the autotrophic compartment. The nanophytoplankton dominated the second part of the rearing and was more unstable. The shift from pico to nanophytoplankton could be considered as an environmental stress and was observed in relation to the beginning of the mortalities, whatever the disease. Shift intensity could play a role by inducing directly or indirectly a stress for shrimp and/or a growth and/or virulence factors of the pathogen. Results from sediment studies show that mortality was best characterized by TAN concentration in pore water and even more when the TAN concentration was high. TAN and pH are potential stress factors for shrimp reared in ponds affected by these diseases. Physiological response of the blue shrimp Litopenaeus stylirostris to short-term confinement on a pond bottom Auteur(s) : Mugnier, Chantal Lemonnier, Hugues Legrand, Alexandre Éditeur(s) : Elsevier Résumé : The blue shrimp Litopenaeus stylirostris was exposed to pond sediment for 48 h and its physiological response was studied in relation to molt stage. Shrimp were maintained in cages within a semi-intensive pond after 4 months of rearing. Hypoosmoregulatory capacity (hypo-OC), magnesium ions (Mg ions), glucose, total proteins and oxyhaemocyanin were assayed in haemolymph. An increase of 370% to 500% in Mg ions concentration and 200% to 266% in glucose concentration, and a decrease in hypo-OC in shrimp maintained close to the centre of the pond indicated a short-term stress response in all molt stages. Indicators of water and sediment pore water quality were within normal operating limits, however this area was characterized by high water content and relatively high total ammonia nitrogen in pore water of sediment. To a lesser extent, shrimp maintained in two other stations also presented a stress response. Physiological response of shrimp in molt stage D, compared to C/D-0 and D-2 seems to indicate that shrimp were more sensitive to environment at this stage. The use and choice of physiological parameters to detect shrimp stress in a pond are discussed. Salinité et confort physiologique - Application pratique en élevage larvaire Auteur(s) : Pham, Dominique Mailliez, Jean-rene Peignon, Jean-marie Broutoi, Francis Marteau, Anne-laure Wabete, Nelly Résumé : The caledonian blue shrimp Litopenaeus stylirostris is the second largest export commodity in New Caledonia. This industry is based on 19 production farms but also four hatcheries, two feed production units and two packing plants. The maximum production was reached in 2005 with 2400 tons. Supply of juveniles by hatchery is the bottle neck of the production chain because this species is not present in the natural environment. Recent problems of larval survival led some scientists to work on eco-physiology to propose adaptations of larval rearing protocol. Salinity of the rearing environment is taken as an example to study the influence of abiotic parameters on the animal physiology according to its development stage.
The influence of salinity on survival and growth was evaluated at different development stages in Litopenaeus stylirostris. The study showed that ocean salinity (33-35 ppt) should be maintained during the larval stage, but after the post-larval metamorphosis, better growth could be achieved by lowering the salinity gradually to 27 ppt. This measure is now applied routinely in the Saint Vincent's hatchery and is the result of research in eco-physiology showing that the blue shrimp iso-osmotic point is around 27 ppt. Les capacités osmorégulatrices chez la crevette bleue Litopenaeus stylirostris, au cours de l’ontogenèse Auteur(s) : Pham, Dominique Éditeur(s) : Université de la Polynésie Française Résumé : The ontogeny of osmoregulation was studied in the blue shrimp Litopenaeus stylirostris in New Caledonia. The implication of branchial organs in ionic regulation was first demonstrated in juveniles by immunolocalization of three ion transporting proteins ( namely NKA, co-transporter NKCC1 and CFTR ) and ionocyte detection was done by means of electron microscopy. The role of the main ion transporting enzyme, NKA, was subsequently investigated during post-embryonic development using an integrative approach associating physiological, histological, immunological and biomolecular studies. Results obtained allowed us to propose a model of the osmoregulatory pattern in L. stylirostris based on weak regulation in the early stages, then progressively this species acquires the adult pattern of hyper-hypo-regulation after metamorphosis with the concurrent development of epipodites and gills. Strong NKA immunofluorescence and abundance of NKA transcripts in these two tissues compared to others organs suggest their functional implication in osmoregulatory processes. The results of this study allow us to increase our knowledge of the penaeid shrimp osmoregulation and to propose zootechnical measures to improve the animal physiological comfort. These propositions have been already tested during larval rearing trials, leading a two-fold-growth in 19 days-old postlarvae. Droits : 2011 The author, Univ. Auteur(s) : De Decker, Sophie Résumé : The New-Caledonian shrimp industry is based on the controlled reproduction of the shrimp Litopenaeus stylirostris, a species which was introduced in the 80s. The major difficulty to which the industry has been faced for 10 years is the occurrence of the “Syndrome 93”, which corresponds to mortality phases when the temperature falls down in April-May-June. This mortality is associated to the pathogenic bacteria Vibrio penaecida. An experiment of genetic selection based on the criterion ofsurvival to Syndrome 93 has been conducted at the Laboratory of Aquaculture of New Caledonia. The 3rd selected generation had demonstrated very encouraging results (survival rates improved by around 20% in experimental infections with V. penaeicida). These results have not been confirmed at the 4th generation and no difference in terms of physiology and immunology appears between the selected population and the non-selected control population. The potential causes of these results are examined and proposals for protocol improvements are given. Biological, physiological, immunological and nutritional assessment of farm-reared Litopenaeus stylirostris shrimp affected or unaffected by vibriosis Auteur(s) : Mugnier, Chantal Justou, Carole Lemonnier, Hugues Patrois, Jacques Ansquer, Dominique Goarant, Cyrille Le Coz, Jean-rene Éditeur(s) : Elsevier Science Bv Résumé : Shrimp aquaculture in New Caledonia is subject to seasonal mortalities during grow-out due to highly virulent Vibrio nigripulchritudo (Vn). To understand the mechanisms affecting shrimp resistance and leading to significant mortality, a shrimp ecophysiology and immunology survey was conducted on two farms, the first considered as a "control" farm (HC), the second affected by the disease (DF). Mortality observed during the survey at DF started 50 days after stocking and was typical of this disease. The main observations regarding shrimp were: (a) growth was not affected by the disease and was faster in the DF than in the HC pond; (b) disease did not affect one sex more than the other, or a specific part of the population in terms of weight; (c) the physical condition of shrimp did not specifically allow us to foresee disease outbreak; (d) shrimp at late premolt stage D-2 and early postmolt stage A appeared to be at some points of the mortalities - but not continuously - the most sensitive to disease; (e) physiological, immunological and nutritional parameters of uninfected shrimp in the DF pond were altered, suggesting that environmental stress occurred just before the first mortalities; (f) data suggest that Vn-infected shrimp are more stressed than the presumed healthy shrimp. Combined with pathological and environmental knowledge gained in parallel during this survey, a conceptual model is proposed. Results suggest that an unstable environment induced conditions (i) stressful for the shrimp, increasing their susceptibility to bacterial infections and (ii) favoring the proliferation of the pathogen in the pond. The combination of these two processes could lead to significant mortality. Aquaculture (0044-8486) (Elsevier Science Bv), 2013-04 , Vol. 388 , P. 105-114 Droits : 2013 Elsevier B.V. Influence of sediment characteristics on shrimp physiology: pH as principal effect Auteur(s) : Lemonnier, Hugues Bernard, E Boglio, E Goarant, Cyrille Cochard, Jean-claude Éditeur(s) : Elsevier Résumé : Penaeid shrimp reared in earthen ponds are exposed to sediment, which can, in some instances, induce a stress. In seawater, the osmoregulatory capacity (hyporegulation) is a useful tool to compare the physiological condition of shrimp exposed to various stressors. By keeping some shrimp in cages at different locations of a single pond heterogeneous in terms of sediment quality, it was possible, using osmotic pressure (OP), to identify some locations where the stress was maximum and some others where it was minimum. Simultaneously, sediment samples were taken and analysed in order to evaluate some physico-chemical parameters that could be related to the stress observed in the shrimps kept in the cages. This approach allowed to show a significant positive correlation between the pH of the sediment surface and the shrimp osmotic pressure. This result was confirmed in a study carried out in experimental 70-1 tanks, where osmotic pressure decreased significantly as water pH decreased from 7.0 to 6.5. The methodology developed in this study may be useful to evaluate the stress caused by sediment in shrimp farms. (C) 2004 Elsevier B.V. All rights reserved. Contribution à l'amélioration des survies et performances de reproduction de L. stylirostris en saison fraîche en Nouvelle-Calédonie Auteur(s) : Pham, Dominique Wabete, Nelly Lemaire, Pierrette Mailliez, Jean-rene Broutoi, Francis Chim, Liet Résumé : Since 1993, the New Caledonian shrimp industry has been faced with cold season mortality outbreaks of multifactorial origin. This pathology called “Syndrome 93” affects as well animals in grow-out ponds as broodstock after their transfer in maturation room, with up to 35% of mortality in 48 hours. From literature data and physiological studies on Litopenaeus stylirostris, several combinations of three abiotic parameters (temperature, salinity and feeding) have been tested during the breeders stocking period. This research allowed us to lower stress factors impact and to propose a transfer protocol which leads to an optimal physiological comfort of the animal. Results showed that survival rate is improved and spawning rates are twice higher. These findings have come up with a routine protocol of carrying out any transfer in a tank with water at temperature of 26°C, salinity of 26 ppt and no food supply during 48 hours. The use of this protocol in a commercial hatchery led to a significant broodstock survival improvement. A soft technology to improve survival and reproductive performance of Litopenaeus stylirostris by counterbalancing physiological disturbances associated with handling stress Auteur(s) : Wabete, Nelly Chim, Liet Pham, Dominique Lemaire, Pierrette Massabuau, J Éditeur(s) : Elsevier Résumé : The consequences of handling stress (fishing, transfer, eyestalk ablation) on shrimp broodstock are poorly documented. The weakness of farmed shrimp, Litopenaeus stylirostris, during winter is a major problem in New Caledonia, because of seasonal climate (tropical-sub-temperate). The transfer of broodstock in winter from earthen outdoor ponds to indoor maturation tanks in the hatchery (T=20 degrees C, Salinity=35 parts per thousand, fed shrimp) usually leads, after 48 h, to high mortality (up to 70%). Eyestalk ablation to induce ovarian maturation in females leads to further mortality. Starting from a background analysis of physiological disturbances (initial osmoregulatory imbalance) associated with handling stress (Wabete, N., Chim, L., Lernaire, P., Massabuau, J.-C., 2004. Ed. Ifremer. Actes Colloq. 38, 75-84.), we developed aprotocol using a soft technology, based on modifications of water salinity, temperature and feeding regime. The aim was to minimize problems of osmoregulatory imbalance and associated mortalities. The protocol we developed, called the LSD OT protocol (Low Salinity and Diet, Optimal Temperature), was first evaluated on sub-adult shrimp (20-25 g) and then applied to broodstock. Survival after transfer and following eyestalk ablation, as well as reproductive achievement (spawning rate, nauplii number) was considerably improved when shrimps were transferred under "physiological comfort" i.e. warmed isosmotic water (26 degrees C and 26 parts per thousand) and unfed for 3 d. This new handling protocol, based on a better control of salinity, temperature and feeding conditions, has been transferred successfully to private hatcheries and already contributes to an increased profitability of New-Caledonian shrimp industry. Marine crustacean farming: present status and perspectives Auteur(s) : Laubier, Annie Laubier, Lucien Éditeur(s) : EDP Sciences Résumé : For centuries, several species of prawns and crabs have been raised from wild-caught juveniles in coastal brackish-water fish ponds in various countries of south east Asia. The Indonesian "tambaks" are well known examples of such traditional practices. In western countries, since the turn of the century, advances of marine biology and fast increase of marine fisheries enabled the development of large-scale production and release of larval stages of American and European lobsters in a fruitless attempt to restock natural populations. After the Second World War, the increasing demand for crustaceans in United States and Japan was satisfied by opening new prawn fisheries all over the world. A major breakthrough was achieved with the development of hatchery technologies for the penaeid prawn Penaeus japonicus (Hudinaga, 1942 in Japan) and the caridean prawn Macrobrachium rosenbergii (Ling, 1969 in Malaysia), which occurred during the first decades of the second half of this century. Together with the increasing market demand in developed countries for sea food, this led to a considerable interest of both public agencies and private investors in marine shrimp and prawn culture. In western countries, a large number of pioneering commercial ventures, often based on assumptions not scientifically founded, failed. Nevertheless, the aquaculture production of prawns mainly based on wild-caught juveniles increased in South-East Asia and Central America during the 1980s. This overall positive trend should not hide important failures which occurred at a local scale, such as the Taiwanese crisis of 1988 due principally to environmental degradation, resulting in severe disease problems and a near collapse of the farming activity. Following the early period of hatchery technology development, the major scientific achievements were related to food requirements and formulation of compound diets for larvae, juveniles and adults and to a better knowledge of diseases caused by bacteria and several viruses which have been identified from hatcheries and intensive farming ponds. Additional new technological advances have emerged from recent research in the fields of physiology (endocrinology) and genetics. By far, the major part of the world production of marine crustaceans relies on penaeid prawns and, to a lesser extent, on Macrobrachium species. However, some other species of marine crustaceans have potential for aquaculture. The economic aspects of marine crustacean aquaculture should be considered together with those of the fishing industry: market prices are rather similar, depending on the quality of the product. The balance between market demand and production is an important constraint which, in turn, establishes the success of prawn farming. Since the early 1980s, crustacean aquaculture has increased tremendously in both Asia and America: the world production for 1991 approximated 700,000 tons, with more than 600,000 tons from penaeid prawn culture.
