'Syndrome 93' is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Litopenaeus stylirostris in grow-out ponds in New Caledonia. This study assessed the toxic activities of extracellular products (ECPs) from V: penaeicida, V. alginolyticus and V, nigripulchritudo using in vivo injections in healthy juvenile L. stylirostris (= Penaeus stylirostris) and in vitro assays on shrimp primary cell cultures and the fish cell line epithelioma papulosum cyprini (EPC). Toxic effects of ECPs were demonstrated for all pathogenic Vibrio strains tested both in vivo and in vitro, but for shrimp only; no effect was observed on the fish cell line. ECP toxicity for New Caledonian V. penaeicida was found only after cultivation at low temperature (20 degrees C) and not at higher temperature (30 degrees C). This points to the fact that 'Syndrome 93' episodes are triggered by temperature drops. The assays used here demonstrate the usefulness of primary shrimp cell cultures to study virulence mechanisms of shrimp pathogenic bacteria. Salinité et confort physiologique - Application pratique en élevage larvaire Auteur(s) : Pham, Dominique Mailliez, Jean-rene Peignon, Jean-marie Broutoi, Francis Marteau, Anne-laure Wabete, Nelly Résumé : The caledonian blue shrimp Litopenaeus stylirostris is the second largest export commodity in New Caledonia. This industry is based on 19 production farms but also four hatcheries, two feed production units and two packing plants. The maximum production was reached in 2005 with 2400 tons. Supply of juveniles by hatchery is the bottle neck of the production chain because this species is not present in the natural environment. Recent problems of larval survival led some scientists to work on eco-physiology to propose adaptations of larval rearing protocol. Salinity of the rearing environment is taken as an example to study the influence of abiotic parameters on the animal physiology according to its development stage.
The influence of salinity on survival and growth was evaluated at different development stages in Litopenaeus stylirostris. The study showed that ocean salinity (33-35 ppt) should be maintained during the larval stage, but after the post-larval metamorphosis, better growth could be achieved by lowering the salinity gradually to 27 ppt. This measure is now applied routinely in the Saint Vincent's hatchery and is the result of research in eco-physiology showing that the blue shrimp iso-osmotic point is around 27 ppt. La crevette bleue calédonienne Litopenaeus stylirostris est au deuxième rang des produits exportés en Nouvelle-Calédonie. L’industrie crevetticole calédonienne est basée sur 19 fermes de production mais également 4 écloseries, 2 provendiers et 2 usines de conditionnement. La production maximale a été atteinte en 2005 avec 2400 tonnes. La fourniture de juvéniles par les écloseries est le point crucial de la chaîne de production car cette espèce n’est pas présente dans le milieu naturel. Les récents problèmes de survie larvaire ont orienté une partie de la recherche vers l’éco-physiologie en vue de proposer des adaptations du protocole d’élevage. La salinité du milieu d’élevage est prise comme exemple pour étudier l’influence d’un paramètre abiotique sur la physiologie de l’animal en fonction de son stade de développement. L’influence de la salinité sur la survie et la croissance a été évaluée aux différents stades de développement chez Litopenaeus stylirostris. L'étude a permis de montrer que la salinité océanique (33-35 ppt) devait être maintenue durant la phase larvaire mais qu'après la métamorphose en post-larve, une meilleure croissance pouvait être obtenue en abaissant la salinité progressivement à 27 ppt. Cette mesure est maintenant appliquée en routine en élevage larvaire à l'écloserie de Saint-Vincent et découle des recherches menées en éco-physiologie qui situe le point iso-osmotique de la crevette bleue autour de 27 ppt. Eutrophication in a tropical pond: Understanding the bacterioplankton and phytoplankton dynamics during a vibriosis outbreak using flow cytometric analyses Auteur(s) : Lucas, Ronan Courties, C. Although difficult to maintain, its stability is required to guarantee the success of the culture. A 110-day period of phytoplankton and bacterioplankton stock and dynamics in an earthen pond (1.2 ha area, 1 m depth) was monitored using flow cytometry to provide baseline information on community characteristics and ecosystem instability. Seven autotrophic cell types were identified over the whole sampling period. Prokaryotic cells included Synechococcus sp., a group named UNK which presented an atypical new flow cytometric signature and picoeukaryotes (PEUK). Nanophytoplankton cells were represented by 4 groups: NAN1, NAN2, NAN3 and Cryptophytes. During the first part of the survey, picophytoplankton dominated the phytoplanktonic assemblage. The mean abundance of total cells (up to 8 x 10(6) cells mL(-1)) was among the highest recorded in marine and brackish waters. Bacterial abundance and production ranged from 0.8 to 5.1 x 10(7) cells mL(-1) and from 30 to 110 mu g C L-1 h(-1). A shift from pico to nanophytoplankton abundance was observed for a few days from d 96. During this period, heterotrophic bacteria production and abundance suddenly dropped, implying a change in the functioning of the microbial loop. This shift was concomitant with a significant shrimp mortality outbreak due to Vibrio penaeicida, the etiological agent of a disease known as Syndrome 93, which affects the shrimp industry in New Caledonia. This survey suggests that flow cytometric analysis could be used for the monitoring of aquaculture systems to improve our understanding of the complex phytoplankton and bacterial dynamics of these systems and its potential influence on disease development. A white spot disease-like syndrome in the Pacific blue shrimp (Litopenaeus stylirostris) as a form of bacterial shell disease Auteur(s) : Goarant, Cyrille Brizard, Raphael Marteau, Anne-laure Éditeur(s) : Elsevier Résumé : In May 1997, some white lesions evoking the white spot syndrome disease were observed in Litopenaeus stylirostris broodstock in New Caledonia. The occurrence of these lesions was neither associated with mortality, nor with histological evidence of white spot syndrome baculovirus (WSBV), The evidence suggests that these lesions result from a form of bacterial disease and are associated with an increased bacterial flora on the outer surface of the cuticle, as well as an increased incidence throughout the molt cycle. A microscopic observation of these lesions allows them to be differentiated from WSBV-associated lesions. Acquisition of susceptibility to Vibrio penaeicida in Penaeus stylirostris postlarvae and juveniles Auteur(s) : Goarant, Cyrille Regnier, France Brizard, Raphael Marteau, Anne-laure Éditeur(s) : Elsevier Résumé : Vibriosis is a major disease problem in shrimp aquaculture, affecting all developmental stages, from larvae in hatchery tanks to juveniles and broodstock in growout ponds. However, bacterial strains responsible for vibriosis in the successive stages are usually considered to be different, and virulence specificity has been reported both at the species and at the stage levels. The so-called « Syndrome 93 » is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Penaeus stylirostris in growout ponds and broodstock tanks in New Caledonia. This pathology does not cause any mortality in hatchery or nursery phases. An experimental infection design using balneation of postlarvae and early juveniles in V. penaeicida suspensions was used to evaluate the developmental stage at which shrimp become sensitive to this vibriosis. We demonstrated that the acquisition of susceptibility to this pathogen is very sudden and correlated with the acquisition of the definitive rostral formula, and from this draw conclusions regarding virulence mechanisms of V. penaeicida in P. stylirostris.
