In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and, consequently, for the study of bivalve larvae genetics. Culling, by selective sieving of the smallest larvae is an advantageous practice at a phenotypic scale as it reduced variance in larval size, variance of developmental rate and time to settlement. Culling of 50% of the larval population only led to 15% less spat, showing a positive phenotypic correlation between larval growth and settlement success. However, culling represents a substantial risk for diversity loss, because it increases the variance of reproductive success among parental oysters. The effective population sizes of early settling cohorts of settlement were lower than those of later ones. Our results show that the settlement of slow growing larvae significantly contributes to minimizing the variability of reproductive success and therefore to maximizing genetic diversity. These results corroborate the low estimations of variability of broodstocks sampled in several French commercial hatcheries, relative to natural populations. The genetic composition of the larval population and the resulting spat was significantly different between the two tested temperatures, revealing genotype x environment interaction for survival. Similarly, genotype x environment interaction was also observed for larval growth as a higher temperature exerted a positive influence on the expression of genetic variability for this trait. Consequently, we can conclude that a temperature of 26°C coupled with culling, to common practice in oyster hatcheries, is likely to amplify the selection pressure for fast growing larvae. To test for this hypothesis, we compared larval developmental traits in the progeny of a hatchery broodstock closed for 7 generations, with the progeny of wild oysters and the two possible hybrids. Our results show that selection of fast growing larvae can counteract presumed inbreeding depression, due to higher mean relatedness among hatchery broodstock than in the wild. Genetic effects of intensive rearing conditions at larval stage are significant and should be taken into account in hatchery practices, especially in terms of genetic diversity management. Afin d'étudier les conséquences génétiques des pratiques de production en écloserie d'huître creuse, deux facteurs ont été examinés : l'élimination des petites larves et la température. Nos résultats montrent que l'assignation de parenté par marqueurs microsatellites est un outil performant pour les études génétiques en phase larvaire de familles élevées en mélange. Bien qu'avantageux d'un point de vue phénotypique, le tamisage sélectif représente un risque de perte de diversité. La fixation des larves à croissance lente permet en effet de minimiser la variabilité du succès reproducteur et de fait, de maximiser la variabilité génétique. Ces résultats corroborent les estimations de variabilité sur les stocks d'écloseries françaises où l'on constate une diversité allélique inférieure à celle de populations issues du milieu naturel. La température exerce également une influence sur la précocité de l'expression de la variabilité génétique pour la croissance larvaire. Ainsi une température élevée associée à une procédure de tamisage peut amplifier l'effet sélectif. Enfin, la sélection de larves à croissance rapide semble démontrée, s'opposant à la dépression de consanguinité présumée. Les conditions d'élevage peuvent donc avoir un effet génétique significatif qui devrait être pris en considération dans les pratiques d'écloserie, notamment dans la gestion de la diversité génétique. Changes in biochemical composition and fatty acids (FA) profile in the 1st and 4th spawn order females were used as indicators of nutritional condition. Wild population of F. duorarum presented significantly better reproductive outcomes as compared to domesticated ones. Wild spawners achieved significantly higher number of eggs per spawn, number of egg per spawn per g of spawner's body weight and number of nauplii per spawn (P<0.05). Latency period was significantly shorter (P<0.05) for wild than for domesticated females (11 vs 17 days). Wild spawners also presented less mortality (6 vs 12%), higher number of total spawns (51 vs 33), spawns per ablated and per spawning females as well as higher total eggs and nauplii production. On the other hand, domesticated females presented higher number of females that spawned at least once (89 vs 76%). The higher spawning activity was reflected in lower levels of acylglycerides (AG) and cholesterol content in wild hepatopancreas (HP) and ovary (OV) (P<0.05). FA profile, mainly represented by ARA, EPA and sum of n-3 were higher in hepatopancreas and ovary of wild females, however lower levels were detected in eggs as a result of a high spawning activity. A fluctuating quality of broodstock prevents sustainable production of larvae in quantity and quality. The traditional extensive rearing method of broodstock in outdoor earthen ponds used in New Caledonia raises several issues: poor water quality control and biosecurity. Therefore it is fundamental to modify the broodstock culture strategy in order to achieve an easier water quality management and maximal biosecurity. Biofloc technology (BFT) offers easier water quality management, higher natural productivity, higher level of biosecurity, and could be a good alternative. Since 2011 a R&D program started under the supervision of IFREMER in New Caledonia and French Polynesia in order to study and develop biofloc technology for rearing shrimp L. stylirostris broodstock. In a first step we showed that biofloc (BFT) improved significantly the reproductive performances of shrimps and the quality of their larvae compared to Clear Water (CW) or Earthen Pond (EP), with: • A better final survival rate of broodstock from BFT compare to CW. • Females from BFT produced more eggs per spawn and spawned more frequently compared to females from CW or EP. • The larvae from BFT females had a significantly higher survival rate compared to larvae from CW females, respectively 70% (n=4) and 45% (n=4). In a second step we studied the underlying biological mechanisms explaining the better reproductive performance and health status of the broodstock from BFT rearing system, and the better quality of their larvae. We showed that the juvenile shrimp fed actively on biofloc, which represented 60% of their whole food (40% coming from pelleted diet). Biofloc represents a significant source of essential nutrients (vitamins, antioxidants, highly unsaturated fatty acids, …) or dietary bacteria which could act as probiotic. These dietary complements could explain the improved health status (immunity and oxidative status status), the better resistance to oxidative stress and a systematically better survival rate of the shrimp reared in BFT compared to those reared in CW. Larval Dispersal Modeling of Pearl Oyster Pinctada margaritifera following Realistic Environmental and Biological Forcing in Ahe Atoll Lagoon Auteur(s) : Thomas, Yoann Dumas, Franck Andrefouet, Serge Éditeur(s) : Public Library Science Résumé : Studying the larval dispersal of bottom-dwelling species is necessary to understand their population dynamics and optimize their management. The black-lip pearl oyster (Pinctada margaritifera) is cultured extensively to produce black pearls, especially in French Polynesia's atoll lagoons. This aquaculture relies on spat collection, a process that can be optimized by understanding which factors influence larval dispersal. Here, we investigate the sensitivity of P. margaritifera larval dispersal kernel to both physical and biological factors in the lagoon of Ahe atoll. Specifically, using a validated 3D larval dispersal model, the variability of lagoon-scale connectivity is investigated against wind forcing, depth and location of larval release, destination location, vertical swimming behavior and pelagic larval duration (PLD) factors. The potential connectivity was spatially weighted according to both the natural and cultivated broodstock densities to provide a realistic view of connectivity. We found that the mean pattern of potential connectivity was driven by the southwest and northeast main barotropic circulation structures, with high retention levels in both. Destination locations, spawning sites and PLD were the main drivers of potential connectivity, explaining respectively 26%, 59% and 5% of the variance. Differences between potential and realistic connectivity showed the significant contribution of the pearl oyster broodstock location to its own dynamics. Realistic connectivity showed larger larval supply in the western destination locations, which are preferentially used by farmers for spat collection. In addition, larval supply in the same sectors was enhanced during summer wind conditions. These results provide new cues to understanding the dynamics of bottom-dwelling populations in atoll lagoons, and show how to take advantage of numerical models for pearl oyster management. Trade has involved, in particular, the following species: - Penaeus japonicus from Japan to Europe, the Pacific Islands and South America - P. monodon from South-East Asia to almost all tropical countries - P. vannamei and P. stylirostris from countries along the Pacific coast of South and Central America to the United States of America, the islands of the South Pacific, countries along the Atlantic coast of the Americas and certain countries in Africa. In the 1980s, research conducted by American and French teams enabled the breeding of stocks of spawners in captivity. This resulted in the establishment of lines of captive broodstock outside natural areas of distribution, and the development of new breeding farms in previously unproductive regions. Trade related to movements of these species is facilitated by the absence of legislation in most countries; even when such legislation does exist, it is not always applied These practices have led to the rapid development of world shrimp production, but have also contributed to the dissemination of pathogens. In recent years, the occurrence of serious epizootics - occasioning heavy losses world-wide - has led to a more cautious approach involving trade of post-larvae obtained from captive broodstock in which thorough control measures have been implemented for known diseases. Trade in wild shrimp will probably be abandoned progressively, to avoid the risk of introducing new pathogens. The main challenges for research in the near future are the development of diagnostic tools, to enable continuous control of captive broodstock, and the selection of strains which are resistant to the principal pathogens affecting these species. The five first animals have been imported from Fiji Islands and in May 1979, four generations-have been obtained. Under the rearing conditions followed at the Centre, reproduction is achieved all throughout the year. Maturation is induced by unilateral eyestalk ablation on pond reared animals maintained in tanks on adequate food. Results concern mating behaviour, ovarian development, number of spawnings per female and egg viability. The rearing and the maintenance conditions of the captive broodstocks are particularly important to obtain reliable results necessary to sustain commercial hatcheries. Contribution à l'amélioration des survies et performances de reproduction de L. stylirostris en saison fraîche en Nouvelle-Calédonie Auteur(s) : Pham, Dominique Wabete, Nelly Lemaire, Pierrette Mailliez, Jean-rene Broutoi, Francis Chim, Liet Résumé : Since 1993, the New Caledonian shrimp industry has been faced with cold season mortality outbreaks of multifactorial origin. This pathology called “Syndrome 93” affects as well animals in grow-out ponds as broodstock after their transfer in maturation room, with up to 35% of mortality in 48 hours. From literature data and physiological studies on Litopenaeus stylirostris, several combinations of three abiotic parameters (temperature, salinity and feeding) have been tested during the breeders stocking period. This research allowed us to lower stress factors impact and to propose a transfer protocol which leads to an optimal physiological comfort of the animal. Results showed that survival rate is improved and spawning rates are twice higher. These findings have come up with a routine protocol of carrying out any transfer in a tank with water at temperature of 26°C, salinity of 26 ppt and no food supply during 48 hours. The use of this protocol in a commercial hatchery led to a significant broodstock survival improvement. Depuis 1993, la filière crevetticole calédonienne est confrontée aux mortalités de saison froide dont les causes sont multifactorielles. Cette pathologie appelé « Syndrome 93 » affecte non seulement les animaux dans les bassins de grossissement, mais elle touche également les géniteurs après leur transfert en salle de maturation, les pertes pouvant atteindre 35% en 48 heures. A partir de la littérature et d’études physiologiques de la Litopenaeus stylirostris, différentes combinaisons de trois paramètres abiotiques (température, salinité et alimentation) ont été testés lors de la stabulation des reproducteurs en salle de maturation. Ces recherches ont permis de diminuer l’impact des facteurs de stress et d’établir un protocole de transfert des animaux dans les conditions de confort physiologique optimale. Les résultats montrent que la survie est améliorée mais que le pourcentage de pontes est également doublé. Il est maintenant recommandé pour tout transfert, de prévoir des bacs de réception dans lesquels l’eau est à la température de 26°C, la salinité à 26 ppt et de ne fournir aucune alimentation pendant 48 heures. L’application de ce protocole dans une écloserie privée a démontré un gain substantiel sur la survie des géniteurs. The aim was to minimize problems of osmoregulatory imbalance and associated mortalities. The protocol we developed, called the LSD OT protocol (Low Salinity and Diet, Optimal Temperature), was first evaluated on sub-adult shrimp (20-25 g) and then applied to broodstock. Survival after transfer and following eyestalk ablation, as well as reproductive achievement (spawning rate, nauplii number) was considerably improved when shrimps were transferred under "physiological comfort" i.e. warmed isosmotic water (26 degrees C and 26 parts per thousand) and unfed for 3 d. This new handling protocol, based on a better control of salinity, temperature and feeding conditions, has been transferred successfully to private hatcheries and already contributes to an increased profitability of New-Caledonian shrimp industry. Bases biologiques de la production de deux Mollusques d'intérêt économique: reproduction chez Megathura crenulata, reproduction et développement chez Crassostrea gigas Auteur(s) : Cannuel, Rozenn Éditeur(s) : Université de Nantes Résumé : The reproductive cycle of Megathura crenulata was documented and quantification of oocyte lipid reserves with lipo-specific stains was explored in both this species and in Crassostrea gigas. The necessity of providing food to female broodstock C. gigas in spring was re-evaluated, because subsequent larval and post-larval rearing performance was not affected by broodstock starvation during conditioning. Developmental study of the organs responsible of particle processing in C. gigas allowed the identification of critical stages related to the ontogenesis of each organ, as well as a better understanding of the evolution of these organs, highly-modified in the Ostreidae. Ciliary type identification and mucocyte mapping from post-larval to adult stage showed that the heterorhabdic specialization precedes the anatomic formation of gill plicae specific to this condition. Le cycle reproducteur de Megathura crenulata a été documenté et la quantification des réserves ovocytaires par des colorants lipospécifiques a été tentée, sans résultats concluants pour cette espèce, ni pour Crassostrea gigas. L'intérêt d'une alimentation des géniteurs femelles C. gigas au printemps est re-évalué, car la performance ultérieure des élevages larvaires et post-larvaires n'a pas été affectée par le jeûne imposé lors du conditionnement. L'étude du développement des organes impliqués dans le traitement des particules chez C. gigas a permis d'identifier des stades critiques associés à l'ontogenèse de chaque organe, ainsi qu'une meilleure compréhension de l'évolution de ces organes si particuliers chez les Ostreidae. L'examen des types ciliaires et la cartographie des mucocytes de la post-larve à l'adulte ont montré que la spécialisation hétérorhabdique précède même la formation anatomique des plis branchiaux caractéristiques de cette condition. Génotypage des géniteurs de Lates calcarifer de Tahiti : aide à la domestication raisonnée du Loup Tropical pour la filière tahitienne Auteur(s) : Vonau, Vincent Rouxel, Catherine Saulnier, Denis Cochennec-laureau, Nathalie Nedelec, Georges Goyard, Emmanuel Résumé : The tropical seabass Lates calcarifer was introduced in Tahiti in 1984 and then was domesticated without other introduction. Three successive generations have been obtained in captivity. Tissue samples of the 38 animaIs which represent the total Tahitian broodstock were preserved in alcohol to be genotyped with four microsatellite markers (Yue et al., 2001).
Three of four described markers were successfully transferred to the laboratory of genetics of Tahiti (marker LcaM01, LcaM02, LcaM03), as the revelation technology available locally did not allow the use of LcaM04 in routine.
The results show that the genetic diversity of the tahitian broodstock is not equivalent of the one of the species, which could be explained by the small size of the founder population, which is consistant with the history of the Tahitian population. The structuration of the 4th generation into several batches obtained from different parents appears to be the minimum management procedure to limit inbreeding. Le loup tropical Lates calcarifer a été introduit à Tahiti en 1984 puis domestiqué sans apport d'animaux extérieurs. Trois générations successives ont été obtenues en captivité. Des prélèvements de tissus sur les 38 géniteurs qui constituent l'intégralité du stock de reproducteurs tahitiens sont conservés dans l'alcool pour être génotypés à l'aide de quatre marqueurs microsatellites (Yue et al., 2001). Trois des quatre marqueurs décrits ont pu être transférés avec succès au laboratoire de génétique de Tahiti (marqueurs LcaM01, LcaM02, LcaM03), la technique de révélation disponible localement ne permettant pas d'utiliser LcaM04 en routine. Les résultats montrent qu'on ne dispose pas à Tahiti de toute la variabilité génétique de l'espèce, ce qui pourrait s'expliquer, compte tenu de l'histoire de cette population, par un faible nombre de géniteurs fondateurs. La structuration de la 4ème génération en plusieurs lots issus de parents différents apparaît comme la mesure de gestion minimale pour limiter la consanguinité au sein de cette population. Molecular identification of Vibrio tapetis, the causative agent of the brown ring disease of Ruditapes philippinarum Auteur(s) : Paillard, Christine Gausson, Sabrina Nicolas, Jean-louis Le Pennec, Jean Paul Haras, Dominique Éditeur(s) : Elsevier Résumé : Vibrio tapetis is the marine bacterium responsible for the brown ring disease (BRD) affecting die manila clam, Ruditapes philippinarum. Identification of V tapelis has been previously performed using biochemical criteria and serological procedures. All of these methods are time consuming and ill-adapted to individual screening. This study describes an oligonucleotidic probe (Vt446) and two PCR primers, deduced from the 16S rDNA sequence, allowing a fast and specific V tapetis identification using dot blot hybridisation and species-specific primed PCR (SSP-PCR). The probe and primers have been tested on 60 strains, including referenced Vibrio sp., Gram negative and positive bacteria, marine bacteria samples and isolated clam bacteria. For all the 19 V tapetis strains, the results of PCR assays consistently corroborated those of the agglutination tests. The detection limit was estimated to be 10(2) CFU ml(-1). The SSP-PCR method has resulted in V tapetis detection in larvae, m diseased clams, and in asymptomatic broodstock clams that later developed BRD. In conclusion, the two SSP-PCR primers were useful for direct and fast identification of V tapetis strains isolated in clams, and are well suited for the screening of individual R. philippilnarum broodstock clams and larvae from the hatchery. Acquisition of susceptibility to Vibrio penaeicida in Penaeus stylirostris postlarvae and juveniles Auteur(s) : Goarant, Cyrille Regnier, France Brizard, Raphael Marteau, Anne-laure Éditeur(s) : Elsevier Résumé : Vibriosis is a major disease problem in shrimp aquaculture, affecting all developmental stages, from larvae in hatchery tanks to juveniles and broodstock in growout ponds. However, bacterial strains responsible for vibriosis in the successive stages are usually considered to be different, and virulence specificity has been reported both at the species and at the stage levels. The so-called « Syndrome 93 » is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Penaeus stylirostris in growout ponds and broodstock tanks in New Caledonia. This pathology does not cause any mortality in hatchery or nursery phases. An experimental infection design using balneation of postlarvae and early juveniles in V. penaeicida suspensions was used to evaluate the developmental stage at which shrimp become sensitive to this vibriosis. We demonstrated that the acquisition of susceptibility to this pathogen is very sudden and correlated with the acquisition of the definitive rostral formula, and from this draw conclusions regarding virulence mechanisms of V. penaeicida in P. stylirostris. Etude des processus de dérive et de sélection liés aux pratiques d'élevage en écloserie d'huître creuse Auteur(s) : Boudry, Pierre Résumé : Genetic consequences of production of Pacific oyster larval in hatchery: drift and selective pressures related to rearing practices. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors were examined: the effects of discarding the smallest larvae (i.e. culling) and temperature effects. A mixed-family approach was used in order to infer the genetic composition of the larval population. The results show that high polymorphic microsatellite-based family assignment is a powerful tool for the study of bivalve larvae genetics. Culling by selective sieving is an advantageous practice at a phenotypic scale, but also represents a substantial risk for diversity loss if parentage assignment is not introduced as a breeding practice. Settlement of slow growing larvae contributes to minimizing the variability of reproductive success and therefore to maximizing genetic diversity. These results corroborate the lower estimations of variability made on broodstocks from French commercial hatcheries relative to natural populations. Temperature exerts an influence on the expression of genetic variability for larval growth. A temperature of 26°C, coupled with culling could amplify the selective effect. Furthermore, selection of fast growing larvae has proven to counteract inbreeding depression at this stage. Genetic effects of intensive rearing conditions are significant and should be taken into account in hatchery practices, especially in terms of genetic diversity management. Afin d'étudier les conséquences génétiques des pratiques de production de larves en écloserie d'huître creuse, deux facteurs ont été examinés : l'effet de l'élimination des plus petites larves et l'effet de la température. Une approche de familles élevées en mélange a été utilisée afin d'avoir accès à l'information génétique au stade larvaire. Les résultats obtenus montrent que l'assignation de parenté basée sur des marqueurs microsatellites hautement discriminants est un outil performant pour les études génétiques en phase larvaire. Bien qu'avantageuse d'un point de vue phénotypique, la pratique de tamisage sélectif représente un risque substantiel de perte de diversité si cette pratique n'est pas associée à une assignation de parentée par empreintes génétiques. La fixation des larves à croissance lente permet de minimiser la variabilité du succès reproducteur et de fait, de maximiser la variabilité génétique. Ces résultats corroborent les estimations de variabilité sur les stocks d'écloseries commerciales françaises où l'on constate une diversité allélique inférieure à celle de populations issues du milieu naturel. La température exerce également une influence sur la précocité de l'expression de la variabilité génétique pour la croissance larvaire. Ainsi une température élevée (26°C) associée à une procédure de tamisage peut amplifier l'effet sélectif. Enfin, la sélection de larves à croissance rapide semble démontrée, s'opposant à la dépression de consanguinité présumée en phase larvaire. Les conditions d'élevage peuvent donc avoir un effet génétique significatif qui devrait être pris en considération dans les pratiques d'écloserie, notamment dans la gestion de la diversité génétique. A white spot disease-like syndrome in the Pacific blue shrimp (Litopenaeus stylirostris) as a form of bacterial shell disease Auteur(s) : Goarant, Cyrille Brizard, Raphael Marteau, Anne-laure Éditeur(s) : Elsevier Résumé : In May 1997, some white lesions evoking the white spot syndrome disease were observed in Litopenaeus stylirostris broodstock in New Caledonia. The occurrence of these lesions was neither associated with mortality, nor with histological evidence of white spot syndrome baculovirus (WSBV), The evidence suggests that these lesions result from a form of bacterial disease and are associated with an increased bacterial flora on the outer surface of the cuticle, as well as an increased incidence throughout the molt cycle. A microscopic observation of these lesions allows them to be differentiated from WSBV-associated lesions. March 2 - 6, 2009 Tahiti, French Polynesia Résumé : New Caledonian shrimp farming feasibility studies started in the mid-seventies. Today ca. 2000 tons of Litopenaeus stylirostris, introduced in 1978, are semi-intensively produced each year. Since fifteen years, production has been impacted by two seasonal vibriosis, the “Summer” syndrome caused by Vibrio nigripulchritudo in relation to pond bottom and water column quality, and the “Winter Syndrome” caused by Vibrio penaeicida in relation to drastic temperature drops . With the ambition of elucidating processes possibly leading to mortality outbreaks in ponds, Ifremer has been conducting a research program since 2003 under the DESANS project (2003-2006) and the present DEDUCTION project, equally funded by the South and North Provinces and by the Government of New Caledonia. This program has been designed in a multidisciplinary approach integrating the suspected multifactorial origin of shrimp mortalities. Environment quality in ponds (either sediment or water column) plays a major role in the shrimp ecophysiological status, influencing sensitivity to pathogens . DEDUCTION focuses on a better knowledge of phytoplankton composition and biogeochemical fluxes at the sediment-water interface in ponds. Impact of farm discharges is also investigated. In addition, a more fundamental work is conducted to understand the pathogenicity mechanisms of these two Vibrio species but also to prevent the occurrence of viral infections through a RNA interference approach. Markers of the ecophysiological status of broodstock, larvae, post-larvae and reared animals, such as enzymes testifying from an oxidative stress, are developed. Gain of performance (resistance to pathogens and growth) were obtained on hybrids of the New Caledonian and introduced Hawaiian strains. Finally the database Stylog is exploited in view of discriminating “good” from “bad” rearings on the basis of relevant indicators. Les études de faisabilité de l'aquaculture de crevettes ont débuté en Nouvelle-Calédonie dans les années soixante dix. Aujourd'hui environ 2000 tonnes de Litopenaeus stylirostris, introduite en 1978, sont produites chaque année de façon semi-intensive. Depuis quinze ans la production a été impactée par deux vibrioses : le syndrome d'été provoqué par Vibrio nigripulchritudo en relation avec la qualité de la tranche d'eau et des fonds de bassins et le syndrome d'hiver provoqué par Vibrio penaeicida en relation avec des chutes rapides de température. Dans le but d'élucider les processus entrainant des mortalités dans les élevages, Ifremer a conduit un programme de recherche depuis 2003 dans le cadre du projet DESANS (2003-2006) puis du présent projet DEDUCTION co-financé par les provinces Nord et Sud et le Gouvernement de la Nouvelle-Calédonie. Ce programme se construit dans une démarche multidisciplinaire intégrant les origines supposées multifactorielles des mortalités de crevettes. DEDUCTION se focalise sur une meilleure compréhension de la composition phytoplanctonique et des flux bio-géochimiques à l'interface eau/sédiment dans les bassins.L'impact des rejets des fermes est également étudié. En outre, un travail plus fondamental est conduit pour connaître les mécanismes de pathogénicité de ces deux espèces de Vibrio mais aussi pour prévenir les occurences d'infections virales par des approches ARN Interférence. Des marqueurs de l'état physiologique des oeufs, larves, post-larves et animaux en élevage, tels que les enzymes du stress oxydant sont utilisés.Les gains de performance (résistance aux pathogènes et croissance) sont obtenus sur des hybrides issus du croisement de la souche locale avec une souche hawaienne. Finalement la base de données STYLOG est exploitée dans le but de distinguer les bonnes des mauvaises pratiques sur la base d'indicateurs fiables. Enzymatic polymorphism study as help for constitution of initial broodstock for a new cultivated finfish species. To constitute an initial broodstock of L. friderici from local wild populations, the genetic structure of these species was studied. Fifteen enzyme systems, representing twenty loci, were screened. La biosécurité à l'Ifremer LEAD,station de Saint-Vincent Auteur(s) : Herlin, Jose Résumé : Penaeus stylirostris shrimp imported from Hawaii for a genetic program and kept at the Saint-Vincent research facility had progressively developed a high sensitivity to IHHNV, the etiological agent of the Infectious, Hypodermic, and Hematopoietic Necrosis. Fears were that the virus could not only be transferred to their offspring, but also that the New Caledonian strain of Penaeus stylirostris, so far considered as resistant, could become sensitive by an off balance of its hosts/pathogen equilibrium. In October 2008, a joint decision between Uprac (shimp owner), Davar (New Caledonian Animal Health Authority) and Ifremer (scientific partner) was taken to terminate all pure breed and hybrid Hawaiian populations remaining in the ponds, and to implement an intensive fallowing protocol, and controlled restocking. IHHNV was also suspected to trouble experiments carried out in the research facility (mortalities, deformities, slow growth), thus affecting research programs. Difficulties pointed out by IHHNV have triggered since 2008 some awareness on biosecurity issues at some level of the New Caledonian shrimp industry. At the St-Vincent research station, it led to a risk assessment and implementation of biosecurity procedures (fallowing, propagation, grow out and use of screened animals) which minimizes the risk of expression of IHHNV.
The use of a brand new hatchery building on site also helped in resetting routine hygiene procedures during production and in controlling incoming and outgoing items (shrimp, water, feed, personnel, equipment, pests and wastes).
IHHNV has not been detected on analysed samples since then nor have any symptoms been found during experimental or broodstock growout. Keeping this control alive is considered as a useful tool to lower the risk of potential health threats, to get a better understanding in case of an outbreak, and to increase the reliability of experimental results. Les crevettes originaires d’Hawaii et conservées au LEAD St-Vincent, dans le cadre du programme d’introduction de variabilité génétique dans le cheptel Calédonien, ont progressivement montré une sensibilité importante au virus IHHNV, agent de la Nécrose Hypodermale et Hématopoïétique Infectieuse (mortalité, croissance ralentie, déformations). Les risques d’une transmission génétique de cette sensibilité à leur descendance à moyen terme, et de son développement par rupture de l’équilibre hôte/pathogène sur la souche dite « Calédonienne » jusqu’alors considérée comme résistante à l’IHHN, ont conduit à partir d’octobre 2008, avec l’accord et l’appui de l’UPRAC et de la DAVAR, à la suppression des animaux hawaiiens ou hybrides encore présents dans les bassins de terre du LEAD St-Vincent, au vide sanitaire de ces derniers, et au repeuplement contrôlé du site. D’autre part, le virus IHHN est soupçonné d’avoir pu affecter les animaux utilisés dans les expérimentations menées au LEAD-St-Vincent (mortalités, déformations, croissance réduite), influençant donc potentiellement leur bon déroulement et celui des programmes de recherches. Les obstacles soulevés par l’expression du virus IHHN ont donc initié depuis 2008 une prise de conscience et la mise en place de démarches de biosécurité dans certains points de la filière. A la station de St-Vincent, cette démarche s’est traduite par une identification/évaluation des risques et l’application de mesures (vide sanitaire, reproduction, élevage et utilisation des animaux contrôlés) chacune dotée de procédures qui regroupées, minimisent le risque d’introduction et de dissémination de l’IHHNV. La prise en main de la nouvelle structure d’écloserie sur le site de la SASV a également été mise à profit pour revoir les procédures d’hygiène de routine dans la conduite des productions et dans le contrôle des flux intrants et sortants (animaux, eau, aliments, personnel, équipement, nuisibles, déchets). Aucune détection d’IHHNV sur les échantillons prélevés n’a depuis été observée, de même qu’aucun symptôme de l’expression de cette maladie sur les animaux en élevage sur le site de Saint-Vincent. La poursuite de ces mesures est envisagée comme un outil d’aide à la minimisation des problèmes potentiels, à leur compréhension en cas d’apparition, et à la fiabilisation des résultats. The research has been focused on the complete control of the whole cycle: constitution of broodstock, maturation and reproduction, larval rearing, and grow-out.
