Aspects moléculaires et biochimiques des stylicines, peptides multifonctionnels identifiés chez la crevette bleue du Pacifique Litopenaeus stylirostris (Crustacea, Decapoda) Auteur(s) : Rolland, Jean-luc Éditeur(s) : Université Montpellier 2 - Sciences et Techniques du Languedoc Résumé : The work reported here was motivated by the economical importance of the pacific blue shrimp Litopenaeus stylirostris farming where high mortality rates are due to bacterial and viral diseases. It consists in the characterisation of two original peptides, the first members of a new multifunctional family of peptides from peneide shrimps, the stylicines. Those two peptides, named stylicines 1 and 2, are negatively charged (pI < 6.0), and characterised by a proline-rich N-terminal region and a C-terminal region containing 13 cysteine residues. Stylicines are synthesized by heamocytes where they are stored within small cytoplasmic granules. To understand the role of these peptides in the immune response of shrimps to a vibrio infection, their recombinant forms were produced in E. coli BL21 (DE3) plysS, purified and characterised. The two rstylicines display biological anti-proliferative and blood clotting activities. Only rstylicine 1 displays antimicrobial activities: antifungal against Fusarium oxysporum (MIC<2.5μM) and bacteriostatic against Gram (−) bacteria, Vibrio sp. (MIC<80μM). Moreover this peptide displays an LPS-binding activity (dissociation constant (Kd) of 9.6×10−8 M) and agglutinate Vibrio. penaeicida "in vitro". Finally, the presence of sequences coding for modified forms of stylicine 1 in some shrimp’s genome may be in relation with their lower ability to survive infections. Les travaux présentés dans ce mémoire ont été motivés par l’importance économique de l’élevage de la crevette bleue du pacifique Litopenaeus stylirostris dont les fortes mortalités sont principalement dues au développement de maladies bactériennes et virales. Ils ont consisté en la caractérisation des deux premiers membres d’une famille originale de peptides multifonctionnels présents chez les crevettes pénéides, les stylicines. Ces peptides, nommés stylicines 1 et 2, sont des peptides anioniques (pI < 6.0), formés d’une région amino-terminale riche en résidus de type proline et d’une région carboxyterminale riche de treize résidus cystéines. Ces molécules sont synthétisées et stockées dans de petits granules présents dans le cytoplasme des hémocytes. Pour mieux appréhender leurs rôles dans la réponse immunitaire des crevettes à une infection par des Vibrio, leurs formes recombinantes ont été produites dans E. coli BL21 (DE3) plysS, purifiées et caractérisées. Les deux rstylicines présentent des activités antiproliférative et anticoagulante. Seule la rstylicine1 présente des activités antimicrobiennes : antifongique sur Fusarium oxysporum (CMI<2.5 μM), et antibactérienne (bactériostatique) sur Vibrio sp (CMI<80 μM). Crustacés profonds capturés aux casiers aux Antilles Françaises Auteur(s) : Paulmier, Gerard Paulmier, Gerard Résumé : For several years the fishermen of the West Indies have directed their activities towards new fishing grounds, notably the bathyales zones adjacent to island plateaux up to approximately 1000 metres in depth. Explorations earned out since 1989 have shown that diverse crustacean populations could be exploited. However, populations and exploitable stocks arc not well known. The use of crustaceans traps as a fishing technique permits the capture of shellfish, shrimps and prawns of which at least six species could have commercial interest. The main objective of this work is do draw up an inventory of the principal commercial species as well as the accompanying species belonging to the subordres Decapoda macrura, anomura, brachyura, and to the orders Isopoda and Amphipoda. It is intended for fishermen and other socio-professionals of the fishing industry. It should enable them to identify and name these previously unexploited varieties of crustacea to facilitate their sale in local markets.

L'emploi du casier comme technique de pêche, permet la capture de crustacés: crevettes et langoustines, dont au moins 6 espèces peuvent avoir un intérêt commercial. Ce travail nomenclatural a pour premier objectif de dresser l'inventaire ces principales espèces commerciales ainsi que des espèces accompagnatrices appartenant aux infra-ordres des Décapodes macroures, anomoures, brachyoures et aux ordres des Isopodes et des Amphipodes. Il est destine aux pécheurs et aux autres personnes des catégories socioprofessionnelles de la filière pêche. Patrones de abundancia y composicion del zooplancton costero a varias escalas temporales en un ciclo de surgencia estacional en la Isla de Margarita, Venezuela Auteur(s) : Hernandez, Ivan Gaspar, Alfredo Éditeur(s) : Interciencia Résumé : Zooplankton abundance, biomass and composition were evaluated in various temporal scales in a seasonal coastal upwelling system at Margarita Island, Venezuela. Scales of temporal variation include weeks, months and seasons (relaxation, upwelling and transition). Seasons were delimitated by upwelling index and sea temperature. The sampling period comprise Sept/2002 - July/2003. Zooplankton samplings were made by horizontal trawls (1-10m depth), using 200,mu m nets, 2-6km from the coastal margin. Total abundance show important variation at week scales, with different patterns between locations, while seasonal differences of abundance are marginal. Biomass only shows seasonal variation due significant reduction in the transition season. Zooplankton assemblages show changes at scale of weeks, with different local patterns, and seasonal changes. Changes in assemblage between the relaxation and upwelling seasons include reduction (during upwelling) of abundance in copepods, cladocerans, salpidae, chaetognates, fish and crustacea larvae. Except by cirripedia larvae, those were important in the upwelling assemblages. Transition shows high variation, with reduction of biomass and assemblage composition similar to upwelling, except by an isolated event. Small scale variation in the zooplankton could be related to physical processes at small scales, eg. turbulence, local circulation, small eddies and filaments, and biological response. Additional sources of nutrient input, eg. riverine input and internal waves, could support local plankton productivity and abundance during relaxation. Detection of high variation in zooplankton abundance and composition at small scales, with local interaction, bring a new light over zooplankton estimation in time series in this system. La abundancia, la biomasa y la composición del zooplancton costero fueron analizadas a varias escalas temporales en un área afectada por surgencia costera estacional (Isla de Margarita, Venezuela); considerando para ello escalas de semanas, meses y periodos, así como la variación local. La estimación del zooplancton fue realizada a partir de arrastres horizontales (1-10m de profundidad) utilizando redes de 200μ entre 2 y 6 km de la línea de costa, entre set/2002 y jul/2003. La abundancia total presentó alta variabilidad a escala de semanas, con patrones diferentes entre localidades, mientras que la diferencia entre periodos es marginal. La biomasa únicamente mostró variación a escala de periodos por una disminución durante el periodo de transición. La composición presentó fluctuaciones a escalas temporales pequeñas, con patrones de variación diferentes localmente, así como a escala de periodos. Los cambios en composición entre el periodo de relajación y el de surgencia incluyen disminución en la abundancia de copépodos, cladóceros, salpas, quetognatos, larvas de peces y crustáceos; excepto por las larvas de cirrípedos que forman un componente importante durante la surgencia. Se postula que procesos físicos a pequeña escala, como la turbulencia, la circulación local o la formación de pequeños giros o filamentos, y los procesos biológicos, condicionan esta elevada variabilidad a escalas pequeñas. La existencia de fuentes de enriquecimiento adicionales como aportes de ríos y ondas internas pudiera condicionar la presencia de elevada abundancia y biomasa, en especial durante periodos de relajación. The penaeidins, initially characterized from the shrimp Litopenaeus vannamei, are a family of antimicrobial peptides that appear to be expressed in all penaeid shrimps. As of recent, a large number of penaeid nucleotide sequences have been identified from a variety of penaeid shrimp species and these sequences currently reside in several databases under unique identifiers with no nomenclatural continuity. To facilitate research in this field and avoid potential confusion due to a diverse number of nomenclatural designations, we have made a systematic effort to collect, analyse, and classify all the penaeidin sequences available in every database. We have identified a common penaeidin signature and subsequently established a classification based on amino acid sequences. In order to clarify the naming process, we have introduced a 'penaeidin nomenclature' that can be applied to all extant and future penaeidins. A specialized database, PenBase, which is freely available at http://www.penbase.immunaqua.com, has been developed for the penaeidin family of antimicrobial peptides, to provide comprehensive information about their properties, diversity and nomenclature. Eel mainly feeds on benthic organisms, primarily small Amphipoda, Crustacea (mainly Gammarus gr. locusta), Polychaeta, insect larvae (Chironomidae) and small fish. Based on variations in the eels feeding regime, seasonal influence is illustrated by decreased feeding activity in summer, which progressively increases during the following seasons. The trophic quality available to eels in the Mauguio lagoon depends more on allochtonous inputs from the catchment area than on its own hydrodynamics on which water of marine origin has little influence due to its long distance from the sea. Variations observed during the four seasons illustrate the opportunist type feeding behaviour of eels. They consume the most available benthic preys at a given moment without using a particular feeding strategy like specialisation or generalisation. Eel adapts its diet according to the available resources offered by the ecosystem. Because of this eel, as an amphihaline migrant species, can be characterised as an obvious indirect bioindicator of the trophic capacity and very marked level of confinement in the brackish lagoon ecosystem of Mauguio. Peches experimentales des crustaces profonds dans les eaux de la martinique ( pandalidae, nephropidae ). Prospections, rendements et biologie des especes. Auteur(s) : Paulmier, Gerard Gervain, Paul Résumé : The principal objective of the work carried out in the deep waters adjoining the island shelf around Martinique between July 1991 and april 1992 was to establish an initial inventory of potentially exploitable deep benthic resources.The cylindrical lobster pot used during fishing experiments allowed the catch of a varied carcinological marine organisms of which six species at least have economic interest : 5 species of shrimp all belonging to the sub-order of Caridea of the Pandalidae family : Plesionika edwardsi, P. (macropoda) polyacanthomerus, P.laevis, Heterocarpus ensifer and H.laevigatus, and one species of prawn of the Nephropidae family : Eunephrops cadenasi. The yields, compared to those of commercial fishermen, were medium or good for Plesionika edwardsi and P. (macropoda) polyacanthomerus between depths of 200 - 300 metres ; for Plesionika laevis and Eunephrops cadenasi in the vicinity 500 metres ; for Heterocarpus laevigatus below 700 metres. The species Heterocarpus ensifer , of which the vertical distribution is very spread out from 170 to 600 metres, seems divided into two populations which provide good yields about 300 metres and below of 450 mètres. Data on the environment and the biology of the species where also established, especially on growth and breeding. For example growth of the pandalids is relatively rapid and their life cycle rather short, between three and four or five years according to the species. At least 2 principal egg-laying seasons occur in the annual cycle. Information is given on the isopoda of the family of Cirolanidae, Bathynomus giganteus, which could eventually, under certain conditions, become of interest to the fishermen. Les pêches expérimentales effectuées dans les eaux profondes circum-martiniquaises,attenantes au plateau insulaire, entre juillet 1991 et avril 1992, avaient pour principal objectif d'établir un premier inventaire des ressources benthiques profondes potentiellement exploitables. Le casier cylindrique , a permis la capture d'une faune carcinologique variée dont 6 espèces au-moins ont présenté un intérêt économique : 5 espèces de crevettes appartenant toutes à l'infra-ordre des Caridea, famille des Pandalidae, Plesionika edwardsi, P. (macropoda) polyacanthomerus, P.laevis, Heterocarpus ensifer et H.laevigatus, et une espèce de langoustine, famille des Nephropidae: Eunephrops cadenasi. Les rendements, comparés à ceux des professionnels, sont moyens ou bons pour Plesionika edwardsi et P.(macropoda) polyacanthomerus entre 200 et 300 mètres de profondeur; pour Plesionika laevis et Eunephrops cadenasi autour de 500 mètres ; pour Heterocarpus laevigatus en-dessous de 700 mètres. L'espèce Heterocarpus ensifer dont la distribution verticale est très étendue, de 170 à 600 mètres, semble divisée en 2 populations pour lesquelles de bons rendements peuvent être obtenus vers 300 mètres et en-dessous de 450 mètres. Des données sur l'environnement et la biologie des espèces ont également été acquises, notamment sur la croissance et la reproduction. Ainsi la croissance des pandalides est relativement rapide et leur longévité plutôt courte, entre 3 et 4 ou 5 années selon les espèces. Au moins deux pontes principales interviennent sur un cycle annuel. This analysis based on exploitation of stomachal contents shows a majority of Fishes and Brachyura Crustaceans consumed. The alimentary diet with a Fish dominance in winter area (off Belle-Ille island) becomes a Crustacea dominance when adult cuttlefishes come in Morbihan Bay, in March, for reproduction. As for newly-born, they eat exclusively Crustacean Amphipoda and Macrura. L'analyse spécifique des proies permet de déterminer la composition du régime alimentaire des seiches en milieu naturel. Cette analyse basée sur l'exploitation des contenus stomacaux montre une consommation majoritaire de Poissons et de Crustacés Brachyoures. Le régime alimentaire à dominance Poisson en zone d'hivernage (large de Belle-Ille) devient à dominance Crustacé lorsque les seiches adultes entrent dans le Golfe du Morbihan, en mars, pour s'y reproduire. Transmission per os du white spot syndrome virus : facteur limitant l'extension de la maladie chez les crustacés marins Auteur(s) : Hernandez Herrera, Rosa Idalia Éditeur(s) : Université de Montpellier II Résumé : Penaeid family shrimp constitute the first aquaculture product in the world in terms of commercial value. They are produced in third world countries of the sub-equatorial belt. Among the causes limiting their production, one is the presence of the WSSV (White Spot Syndrome Virus), a pathogenic agent that produces massive mortality. Our aim was to investigate the first stages of the viral infection, in order to be used as target of prophylactic actions. A fish cell line (SSN-1) was used as model to tentatively develop in vitro studies. Only defective particles were produced confirming the high specificity to crustacea of the infection with crustacean virus. Electron microscopy showed structural similarities between the WSSV and B, B2 and Baculo-B viruses of crabs. This suggests B2 may belong also to the family Nimaviridae, genus Whispovirus. This comparison with B2 virus gives the possibility to understand the role played by the tail-like extension of these viruses in the infectious process by attachment to the plasmic membrane at the beginning of the infection in its specific host. Les crevettes de grande taille de la famille des Penaeidae constituent le premier produit aquacole en valeur commerciale à l'échelle planétaire. Elles sont produites à 99% dans les pays en voie de développement de la ceinture sub-équatoriale. Notre étude a été orientée vers la reconnaissance des premiers stades de l'infection, susceptibles d'être la cible d'une action prophylactique. Nous avons testé l'utilisation d'une lignée cellulaire de poisson SSN-1 afin d'étudier les possibilités d'un développement in vitro. Seules des particules défectives ont été produites confirmant la haute spécificité des infections à virus de crustacés. Les recherches en microscopie électronique ont montré une similarité structurale du WSSV avec les virus (B, B2 et Baculo-B) des crabes. Ceci suggère que ces agents (B2 et WSSV) seraient tout deux de la famille des Nimaviridae et du genre Whispovirus. La comparaison avec le virus B2 permet de comprendre le rôle clef joué par la partie caudale de ces virus dans l'infection par son attachement à la membrane plasmique lors de l'infection chez son hôte spécifique. Composition chimique et ornementation de Cyprideis torosa (Crustacea, Ostracoda) dans le domaine paralique Auteur(s) : Bodergat, Am Rio, M Andreani, Am Éditeur(s) : Gauthier-Villars Résumé : Cyprideis torosa (Crustacea, Ostracoda), which is well known for its ability to live in a wide range of salinity and for the variability of its ornamentation, was sampled in the paralic environment. in the Camargue (France) and in the area of Santa-Pola (south-east Spain), in ambient waters whose salinity varies from 1 to 120 g/l. The collected valves. which are smooth, punctuated, reticulated or have nodes on their surface, were analyzed by means of spark-source mass spectrometry. We detected twenty-six chemical elements, and found a clear relationship between the chemical composition, the ornamentation of the valves and the environmental parameters. In the oligohaline environment, the carapaces are thick, reticulated or node-bearing ; they are enriched in Si, Al, Fe, Mn and Ba which are terrigenous elements introduced by fresh water. In an open hypersaline environment, the carapaces are reticulated or punctuated ; they are chemically characterized by P, Sr and Li, typical for concentrated seawater. Finally, in a closed hypersaline environment. the collected individuals are smooth, and their carapaces are very thin ; their content S is related to the abundant organic matter provided by the development of algal and cyanobacterial mats. Salinity and confinement, which are the main parameters characteristic of the studied ecosystems, are also the two factors that control the ornamentation of the valves. The chemical analysis and the field observations reveal that the granulometric characteristics of the microhabitats must be considered because they control the exchanges at the water-sediment interface. The anthropic influences on the chemical composition of the carapace must be emphasized: it is possible to detect industrial and domestic wastes. But the ornamentation of the carapace is not related to these pollutants - chiefly heavy metals (Pb, Ni Sn) - and depends mainly on the open or closed character of the ambient environment. The present method highlights, on the basis of the chemical composition, a relationship between ornamentation and environmental parameters. It is particularly efficient in the interpretation of the morphology of the valves in relation to the palaeoenvironment. Species of the genus Munidopsis (Crustacea, Decapoda, Galatheidae) from the deep Atlantic Ocean, including cold-seep and hydrothermal vent areas Auteur(s) : Macpherson, E Segonzac, Michel Éditeur(s) : Magnolia Press Résumé : Twenty-two species of the genus Munidopsis from the Atlantic Ocean are studied. Five new species are described (M. anemia, M. exuta, M. hirtella, M. laurentae and M. truculenta), and diagnoses and illustrations of some relatively rare species (M. acuminata, M. bairdii, M. livida, M. parfaiti and M. thieli) are provided. None of the species discussed here is restricted to a particular habitat, including species collected from hydrothermal vent (M. acutispina, M. exuta) and cold seep areas (M. acutispina, M. geyeri, M. hirtella, M. livida, M. marionis). The new records of some species greatly extend the previously known distribution range of the species (i.e., M. abyssicola, M. aries, M. bairdii, M. geyeri, M. livida, M. parfaiti, M. thieli). Finally, some biological and biogeographical data for the different species are discussed. Al Blasco, F Éditeur(s) : Taylor & Francis Résumé : This study is an attempt to produce an assessment of the impact of shrimp aquaculture in the Mekong Delta (Viet Nam) on mangrove ecosystems. For this exercise we selected two sub-areas (Ca Mau and Tra Vinh provinces) encompassing a variety of land uses and ecological conditions. Twenty stations in Tra Vinh and 15 stations in Ca Mau have been surveyed several times from September 2000 to March 2002. Field investigations included mangrove soils studies, measurements of pH and salinity of the water, analysis of mangrove flora, and density and structure of the vegetation. Four Systeme Probatoire de l'Observation de la Terre (SPOT) scenes were used for the discrimination of mangrove types and for the delineation of landscape units. For the first time, five ecologically distinct landscape classes were identified and delineated. Their possible links with the farming and yields of high valued species of shrimps, especially the giant tiger shrimp (Penaeus monodon) destined for export markets, need further studies. Since 1965, about 30% of mangrove ecosystems have been lost in Ca Mau Province and more than 30% of present mangroves are replanted monospecific stands. To the best of our knowledge, this is the first study which demonstrates that, in spite of deep and ancient man interactions in the Mekong Delta, five ecologically distinct classes of land use can be defined. Satellite surveys confirm a clear distribution of landscape units with possible links with shrimp aquaculture potentialities. Etude des pecheries, reflexions sur l'environment et la gestion multispecifique. These phenomenons are exposed in the first part. About consequences of human interventions, both of these aspects must be taken into account: the introduction of new species and fishing. Second part deals with the use of models in stock management. Une réflexion sur la gestion des stocks halieutiques devait nécessairement aboutir à la reconnaissance des relations liant la ressource à son environnement naturel : action des facteurs abiotiques sur l'état des stocks et la distribution des espèces, et inter-relation entre les composantes biologiques ; à l'appréciation des conséquences de l'intervention humaine sur l'équilibre des écosystèmes. Ce sont ces phénomènes qui ont été présentés dans une première partie. En ce qui concerne l'influence des facteurs abiotiques, il est rappelé que la mise en place de réseaux de surveillance systématique a permis de progresser dans le domaine des recherches hydroclimatiques et dans la compréhension des connexions pouvant exister entre facteurs abiotiques et phénomènes biologiques. d'ordre biologique sont principalement envisagées au niveau des relations de prédation. Celles-ci ne peuvent être correctement appréhendées que par le biais du comportement trophique des diverses espèces et des problèmes généraux de nutrition, Les conséquences de l'intervention humaine sont envisagées sous deux aspects : l'introduction d'espèces nouvelles et la pêche. L'action de pêche entraîne des interactions d'ordre technologique dans les pêcheries plurispécifiques, dont les effets ne peuvent être ignorés et doivent être pris en compte par les modèles de gestion. La deuxième partie traite de l'utilisation des modèles dans la gestion commune de plusieurs stocks. Les modèles dits « multispécifiques », tenant compte des interactions d'ordre biologique (prédation), supposent un support de connaissance scientifique qui fait actuellement défaut. Leur utilisation comme outil de gestion devrait être précédée d'une amélioration des connaissances générales halieutiques. Les interactions d'ordre technologique ont été prises en compte dans l'approche plurispécifique de LE GUEN et FoNTANA (1971) qui permet de rechercher le meilleur compromis dans l'exploitation simultanée de plusieurs espèces, sur des bases pécuniaires plutôt que pondérales. GOHIN et LAUREC (1981) ont amélioré l'approche en tenant compte en particulier de l'ensemble de la courbe de sélectivité et non seulement du point L50. Quoi qu'il en soit, les modèles classiques mono-spécifiques, bien qu'imparfaits, restent encore très utiles ; ils peuvent donner des réponses satisfaisantes à des questions simples mais essentielles. Un développement de la recherche en hydrobioclimatologie est indispensable pour prévoir la variabilité dans le recrutement, la disponibilité, la capturabilité... Les relations de prédation entre espèces méritent d'être mieux cernées. Enfin, chaque fois que cela sera possible, les composantes socio-économiques et politiques devront être prises en compte, le bon sens remplaçant au besoin les modèles. Cet ouvrage n'a pas la prétention de traiter dans le fond tous les sujets abordés. Aussi, le lecteur qui désirera avoir une réflexion plus approfondie sur certains sujets trouvera à la fin du document une bibliographie, sinon exhaustive du moins très fournie.
