Aspects moléculaires et biochimiques des stylicines, peptides multifonctionnels identifiés chez la crevette bleue du Pacifique Litopenaeus stylirostris (Crustacea, Decapoda) Auteur(s) : Rolland, Jean-luc Éditeur(s) : Université Montpellier 2 - Sciences et Techniques du Languedoc Résumé : The work reported here was motivated by the economical importance of the pacific blue shrimp Litopenaeus stylirostris farming where high mortality rates are due to bacterial and viral diseases. It consists in the characterisation of two original peptides, the first members of a new multifunctional family of peptides from peneide shrimps, the stylicines. Those two peptides, named stylicines 1 and 2, are negatively charged (pI < 6.0), and characterised by a proline-rich N-terminal region and a C-terminal region containing 13 cysteine residues. Stylicines are synthesized by heamocytes where they are stored within small cytoplasmic granules. To understand the role of these peptides in the immune response of shrimps to a vibrio infection, their recombinant forms were produced in E. coli BL21 (DE3) plysS, purified and characterised. The two rstylicines display biological anti-proliferative and blood clotting activities. Only rstylicine 1 displays antimicrobial activities: antifungal against Fusarium oxysporum (MIC<2.5μM) and bacteriostatic against Gram (−) bacteria, Vibrio sp. (MIC<80μM). Moreover this peptide displays an LPS-binding activity (dissociation constant (Kd) of 9.6×10−8 M) and agglutinate Vibrio. penaeicida "in vitro". Finally, the presence of sequences coding for modified forms of stylicine 1 in some shrimp’s genome may be in relation with their lower ability to survive infections. Les travaux présentés dans ce mémoire ont été motivés par l’importance économique de l’élevage de la crevette bleue du pacifique Litopenaeus stylirostris dont les fortes mortalités sont principalement dues au développement de maladies bactériennes et virales. Ils ont consisté en la caractérisation des deux premiers membres d’une famille originale de peptides multifonctionnels présents chez les crevettes pénéides, les stylicines. Ces peptides, nommés stylicines 1 et 2, sont des peptides anioniques (pI < 6.0), formés d’une région amino-terminale riche en résidus de type proline et d’une région carboxyterminale riche de treize résidus cystéines. Ces molécules sont synthétisées et stockées dans de petits granules présents dans le cytoplasme des hémocytes. Pour mieux appréhender leurs rôles dans la réponse immunitaire des crevettes à une infection par des Vibrio, leurs formes recombinantes ont été produites dans E. coli BL21 (DE3) plysS, purifiées et caractérisées. Les deux rstylicines présentent des activités antiproliférative et anticoagulante. Seule la rstylicine1 présente des activités antimicrobiennes : antifongique sur Fusarium oxysporum (CMI<2.5 μM), et antibactérienne (bactériostatique) sur Vibrio sp (CMI<80 μM). Ce peptide est également capable de se lier aux LPS des bactéries à Gram (-) (Kd= 9.6x10-8 M) et d’agglutiner V. penaeicida "in vitro". Enfin, l’existence de gènes codant des formes modifiées de la stylicine1, chez certaines crevettes, pourrait être en relation avec une diminution de la résistante des individus aux infections. DNA based dietary analysis can supplement predator removal experiments and provide high resolution for prey identification. Here we use a metabarcoding approach to provide initial insights into the diet and functional role of coral-dwelling predatory fish feeding on small invertebrates. Fish were collected in Moorea (French Polynesia) where the BIOCODE project has generated DNA barcodes for numerous coral associated invertebrate species. Pyrosequencing data revealed a total of 292 Operational Taxonomic Units (OTU) in the gut contents of the arc-eye hawkfish (Paracirrhites arcatus), the flame hawkfish (Neocirrhites armatus) and the coral croucher (Caracanthus maculatus). One hundred forty-nine (51%) of them had species-level matches in reference libraries (>98% similarity) while 76 additional OTUs (26%) could be identified to higher taxonomic levels. Decapods that have a mutualistic relationship with Pocillopora and are typically dominant among coral branches, represent a minor contribution of the predators' diets. Instead, predators mainly consumed transient species including pelagic taxa such as copepods, chaetognaths and siphonophores suggesting non random feeding behavior. We also identified prey species known to have direct negative interactions with stony corals, such as Hapalocarcinus sp, a gall crab considered a coral parasite, as well as species of vermetid snails known for their deleterious effects on coral growth. Pocillopora DNA accounted for 20.8% and 20.1% of total number of sequences in the guts of the flame hawkfish and coral croucher but it was not detected in the guts of the arc-eye hawkfish. Comparison of diets among the three fishes demonstrates remarkable partitioning with nearly 80% of prey items consumed by only one predator. Overall, the taxonomic resolution provided by the metabarcoding approach highlights a highly complex interaction web and demonstrates that levels of trophic partitioning among coral reef fishes have likely been underestimated. Therefore, we strongly encourage further empirical approaches to dietary studies prior to making assumptions of trophic equivalency in food web reconstruction. How to cite this article Leray et al. Explorations earned out since 1989 have shown that diverse crustacean populations could be exploited. However, populations and exploitable stocks arc not well known. The use of crustaceans traps as a fishing technique permits the capture of shellfish, shrimps and prawns of which at least six species could have commercial interest. The main objective of this work is do draw up an inventory of the principal commercial species as well as the accompanying species belonging to the subordres Decapoda macrura, anomura, brachyura, and to the orders Isopoda and Amphipoda. It is intended for fishermen and other socio-professionals of the fishing industry. It should enable them to identify and name these previously unexploited varieties of crustacea to facilitate their sale in local markets.

Des campagnes exploratoires menées depuis 1989. on montré que diverses populations carcinologiques pouvaient l'aire l'objet d'une exploitation. Toutefois, populations et stocks exploitables ne sont pas bien connus. L'emploi du casier comme technique de pêche, permet la capture de crustacés: crevettes et langoustines, dont au moins 6 espèces peuvent avoir un intérêt commercial. Ce travail nomenclatural a pour premier objectif de dresser l'inventaire ces principales espèces commerciales ainsi que des espèces accompagnatrices appartenant aux infra-ordres des Décapodes macroures, anomoures, brachyoures et aux ordres des Isopodes et des Amphipodes. Il est destine aux pécheurs et aux autres personnes des catégories socioprofessionnelles de la filière pêche. II devrait leur permettre d'identifier et de nommer ces produits nouveaux pour faciliter leur vente sur les marchés locaux. Species of the genus Munidopsis (Crustacea, Decapoda, Galatheidae) from the deep Atlantic Ocean, including cold-seep and hydrothermal vent areas Auteur(s) : Macpherson, E Segonzac, Michel Éditeur(s) : Magnolia Press Résumé : Twenty-two species of the genus Munidopsis from the Atlantic Ocean are studied. Five new species are described (M. anemia, M. exuta, M. hirtella, M. laurentae and M. truculenta), and diagnoses and illustrations of some relatively rare species (M. acuminata, M. bairdii, M. livida, M. parfaiti and M. thieli) are provided. None of the species discussed here is restricted to a particular habitat, including species collected from hydrothermal vent (M. acutispina, M. exuta) and cold seep areas (M. acutispina, M. geyeri, M. hirtella, M. livida, M. marionis). The new records of some species greatly extend the previously known distribution range of the species (i.e., M. abyssicola, M. aries, M. bairdii, M. geyeri, M. livida, M. parfaiti, M. thieli). Finally, some biological and biogeographical data for the different species are discussed. Al Blasco, F Éditeur(s) : Taylor & Francis Résumé : This study is an attempt to produce an assessment of the impact of shrimp aquaculture in the Mekong Delta (Viet Nam) on mangrove ecosystems. For this exercise we selected two sub-areas (Ca Mau and Tra Vinh provinces) encompassing a variety of land uses and ecological conditions. Twenty stations in Tra Vinh and 15 stations in Ca Mau have been surveyed several times from September 2000 to March 2002. Field investigations included mangrove soils studies, measurements of pH and salinity of the water, analysis of mangrove flora, and density and structure of the vegetation. Four Systeme Probatoire de l'Observation de la Terre (SPOT) scenes were used for the discrimination of mangrove types and for the delineation of landscape units. For the first time, five ecologically distinct landscape classes were identified and delineated. Their possible links with the farming and yields of high valued species of shrimps, especially the giant tiger shrimp (Penaeus monodon) destined for export markets, need further studies. Since 1965, about 30% of mangrove ecosystems have been lost in Ca Mau Province and more than 30% of present mangroves are replanted monospecific stands. To the best of our knowledge, this is the first study which demonstrates that, in spite of deep and ancient man interactions in the Mekong Delta, five ecologically distinct classes of land use can be defined. Satellite surveys confirm a clear distribution of landscape units with possible links with shrimp aquaculture potentialities. In this study, we have specifically analysed the expression in circulating hemocytes of antimicrobial peptides (AMPs) encoding genes, such as PEN2 and PEN3, ALF, crustin, lysozyme and a putative cysteine-rich peptide. We evidenced a relationship between the level of expression of some AMPs and the successful response of the shrimp, Litopenaeus stylirostris, to circumvent a pathogenic Vibrio penaeicida infection. Additionally, significant differences in some AMP transcript amounts are evidenced between control, non-selected shrimp line and the third generation breeding of shrimp selected for their survival to natural V. penaeicida infections. On the basis of these results, it will now be of great interest to determine if these AMPs are directly involved in the resistance of shrimp to infection or if they only reflect other acquired defence mechanisms which can confer a resistance.
