Links between architectural stages of development (ASD), photosynthetic capacities and leaf characteristics in Tachigali melinonii (Caesalpiniaceae) were investigated in the tropical forest of French Guiana. Three ASD were sampled under three light conditions to elucidate the impact of ASD- and light-related changes on photosynthetic functional traits. Photosynthetic parameters (maximum rate in RubisCO carboxylation (Vcmax) and maximum rate of electron transport (Jmax) were estimated by adjusting a biochemical model (Farquhar et al., 1980) to response curves of CO2 assimilation rate to intercellular CO2 concentration, under saturated light. In addition, N and C foliar concentrations were analysed and N allocations to the carboxylation and bioenergetic processus of photosynthesis were calculated using the model of Niinemets and Tenhunen (1997). The variations in ecophysiological parameters observed were due to leaf anatomical and structural changes instead of changes in photosynthetic components, suggesting adjustments to light conditions. Tachigali melinonii thus appears to display morphological and anatomical plasticity at different ASDs and under different light conditions.
Une espèce forestière de Guyane Française, Tachigali melinonii (Caesalipiniaceae), a été choisie afin de regarder la variation des traits fonctionnels relatifs à la photosynthèse en fonction de la disponibilité en lumière et en fonction de son stade de développement architectural. Du fait du tempérament de l'espèce vis-à-vis de la lumière de type «struggling gambler» (ombrage nécessaire aux jeunes stades puis développement en pleine lumière), trois stades clefs de développement parmi la séquence de différenciation de l'espèce ainsi que trois environnements lumineux ont été choisis. Les capacités photosynthétiques foliaires ont été estimées à partir de l'ajustement de paramètres du modèle biochimique de photosynthèse foliaire (Farquhar et al. 1980) sur des courbes de réponses de l'assimilation de CO² (A) à des concentrations intracellulaires en CO² (Ci) croissantes, en condition de lumière saturante. La vitesse maximale de carboxylation Vcmax et le flux maximal d'électrons photosynthétiques Jmax ont ainsi été estimés et ensuite reliés aux caractéristiques structurales foliaires, notamment la concentration en azote, en carbone ainsi qu'avec la densité et l’épaisseur. Les fractions d'azote allouées aux processus photosynthétiques de carboxylation et de bioénergétique ont été calculées. Des variations, dues à un effet du stade de développement sur les paramètres étudiés ainsi qu’à un effet de la quantité de lumière disponible, ont été constatées. Ces variations sont surtout liées à des ajustements anatomiques et structuraux foliaires et non à des modifications de l’appareil photosynthétique. Tachigali melinonii semble donc modifier ses caractéristiques structurales foliaires en fonction de son stade de développement, ce qui suppose un contrôle ontogénique partiel, et en fonction de la disponibilité en lumière.

Amélioration des productions phytoplanctoniques en écloserie de mollusques : caractérisation des microalgues fourrage

Résumé : in mollusc hatcheries. Consequently, we assessed themost commonly used species to establish their growth and size profiles, their cytomorphological, biochemical, biomolecular identification as well as their cytometer signs, and also their ability to grow under extreme conditions of temperature and salinity. This work aimed at matching the conformity of the strains used in Argenton (dispatched to most of the French commercial hatcheries) to those of a referenced culture collection. A previous screening preceded this study, because of the 45 species and/or clones held originally, Argenton kept no more than 15 species. As most of the latter are also represented in referenced collection, our strains are therefore correctly identified. On the other hand, the combination of several approaches (characteristic of growth and size, cytomorphological description, establishment of biochemical and biomolecular profiles) to discriminate the different species was broadly successful. Nevertheless, flow cytometer measurements were extremely variable from sample to sample, dueto their high sensitivity to the microalga's physiological state. Accordingly, we have developed a method of identification which could be transferred at a national or international level, to the Institutions in charge of microalgae collections. Furthermore, we achieved relevant data to establish an identity card for each of the microalgae suitable for mollusc hatcheries.
La caractérisation des microalgues fourrage constituait l'un des points perfectibles au regard de ce qui est appliquée en microbiologie. Nous avons alors cherché à définir, pour les espèces les plus utilisées en écloserie, leurs profils de croissance et de taille, leur caractéristiques cytomorphologique, cytofluorimétrique, biochimique et biomoléculaire, et enfin, précisé leur aptitude au développement sous des conditions extrêmes de température et de salinité (écotolérance). Ce travail avait pour but de vérifier, d'une part, la conformité des souches utilisées à l'écloserie d'Argenton (auprès de laquelle se fournissent la plupart des écloseries commerciales françaises) en comparant les espèces utilisées à celles issues d'une collection référencée. Un premier balayage avait été réalisé avant la mise en place de cette étude puisque sur les 45 espèces et clones détenus originellement, Argenton ne possédait plus que 15 espèces. La plupart de ces dernières ayant leur équivalent en collection référencée notre souchier est désormais parfaitement identifiée. D'autre part, la confrontation de plusieurs approches, caractéristique de la croissance et de taille, description cytomorphologique, établissement des profils biochimique et biomoléculaire, écotolérance, signature cytofluorimétrique, pour discriminer les différentes espèces de microalgues fourrage a été globalement couronnée de succès. Seule cette dernière technique n'a pas permis de caractériser les espèces entre elles, les paramètres mesurés en cytométrie de flux étant particulièrement sensibles à l'état physiologique des microalgues, très variable d'un échantillon à un autre. Nous disposons dorénavant d'une méthode d'identification qu'il serait souhaitable de transférer, au niveau national voire international, auprès des organismes en charge des collections. De plus, nous disposons dorénavant de données pertinentes pour établir pour chacune des microalgues d'intérêt aquacole une véritable carte d'identité.

Analyse des résultats d'une ferme d'élevage de crevettes sur 20 années : SODACAL 1984-2004. "Elasticité" de la capacité de production d'un écosystème bassin crevetticole

Résumé : SODACAL prawn farm which was set up as a pilot farm for the development of a prawn industry in New Caledonia, saw its production exponentially rise from 1984 to 1992 (to 400 t/year, then experienced a sharp drop in 1993 (250 t/year) which carried on until 1997, and finally rose back to a stabilizing level around 400 t/year until 1999. The study analyses the difficulties observed before and during the drop of production. Among them, mortality outbreaks, other biological perturbations (benthic mat clearing and crab and fish mortality) and focus on the noxious state of the bottom (reduced and smelling sulfide hydrogen). It particularly shows that Syndrome 93 outbreaks could be observed on this farm from 1990. The analysis highlights the particular climatic conditions in New Caledonia from 1990 to 1995 (drought) and their potential impact on the production. The development of this very particular process of physical and topographic differential evolution of the pond bottom under wind action (accumulation and digging) and its consequences on pond preparation is explained. This work takes a close look at the grow-out management : age and quality of post larvae, pond stocking with pre-grown animals, feed characteristics and management, water exchange management and pumping capacity, strategy on density and size of harvested animals. The results have been assessed using a distinctive tool, the "survival differential", allowing inter-pond comparisons within a site, inter-farms comparisons over pluri-annual periods of time. >From the commented farm results over a 20 year period, we could distinguish the principal factors responsible for the drop of grow-out capacity of some ponds. We have outlined a conceptual description of the different parameters leading to the decrease of survival, an epidemiological scenario and a synoptic table of the history of events and their relative severity. The study draws the hypothesis of a cumulative effect of positive factors, which could have led back to satisfying production results: a physical and biochemical restoration of pond bottoms, a modified and standardized grow-out management, an appropriate pumping capacity and the come back of adequate climatic conditions. This case study illustrates the concept of « flexibility » of the farming capacity and the sustainability of prawn farming in New Caledonia. The clear objective of this 20 year historic investigation was to get relevant information, in order to improve the understanding on the "summer syndrome" which hits two farms at the moment, and to participate in the hypothesis under study by the DAC (2005). We also tried to make the new farmers aware of the fragility of the pond ecosystem, and give some common-sense advices in the hope that such problems will not happen again in other places.
La ferme de SODACAL, conçue à l'origine comme une ferme de démonstration pour le développement d'une filière d'aquaculture exportatrice de crevettes en Nouvelle-Calédonie a connu une phase de croissance exponentielle de sa production de 1984 à 1992 (jusqu'à 400 t/an), puis une chute brutale en 1993 (250 t/an) qui s'est prolongée jusqu'en 1997 et enfin une reprise à partir de 1998 pour arriver à une stabilisation autour de 420 t/an depuis 1999. L'étude inventorie les dysfonctionnements observés avant et pendant la crise de production. Parmi ceux-ci sont évoqués les épisodes de mortalité de crevettes, d'autres désordres biologiques (décollements de tapis benthique et « hécatombes » de crabes et de poissons), l'état délétère des fonds (sédiments réduits et odeur d'hydrogène sulfuré). Elle permet notamment de montrer que des épisodes de mortalité typiques du Syndrome 93 pouvaient être observés sur cette ferme dès 1990. L'étude met en évidence les conditions climatiques remarquables qui ont sévi en Nouvelle-Calédonie de 1990 à 1995 (sécheresse) et leur impact probable sur les élevages. La genèse du phénomène très particulier d'évolution physique et topographique des fonds des bassins par l'effet du vent (accumulation et creusement) et des conséquences sur la préparation des bassins est explicitée. Ce travail aborde la gestion zootechnique : l'âge et la « qualité » des post larves, la pratique des ensemencements à partir d'animaux pré grossis, la gestion de l'alimentation et la nature de la provende, le contrôle du renouvellement d'eau et la capacité de pompage, le choix des densités et du calibre des animaux récoltés. Les résultats ont pu être évalués à travers un outil original «le différentiel de survie », permettant des comparaisons inter-bassins au sein du site, inter-fermes à des périodes pluri annuelles distinctes. A partir des résultats commentés de la ferme obtenus sur 20 années, on a pu distinguer les principaux facteurs qui ont provoqué cette chute d'aptitude culturale de certains bassins. Un schéma conceptuel des différents paramètres ayant conduit à la baisse des survies, un scénario épidémiologique ainsi qu'un tableau synoptique de la chronologie des évènements et de leur relative sévérité sont esquissés. Le rapport ébauche l'hypothèse d'un cumul de causes favorables qui ont permis le retour à des résultats zootechniques satisfaisants : une réhabilitation physique et biochimique des fonds de bassins, une zootechnie modifiée et standardisée, une capacité de pompage adéquate et une climatologie de nouveau favorable. Cette étude de cas permet ainsi d'illustrer les notions d « élasticité » de l'aptitude culturale et de durabilité de la crevetticulture en Nouvelle-Calédonie. Le but de cette enquête historique sur 20 ans est l'acquisition d'informations pertinentes permettant d'améliorer la compréhension du « Syndrome d'été » qui affecte actuellement deux fermes de production et ainsi aider à compléter le scénario hypothétique avancé par le Dac (2005). Enfin, au travers de cet historique, on a cherché également à sensibiliser les nouveaux opérateurs de la filière sur la fragilité de l'écosystème-bassin et présenter quelques conseils de bon sens pour éviter que de tels phénomènes ne se reproduisent ailleurs.

For groundwater issues (potential leakages in waste repository, aquifer management ...), the development of modeling techniques is far ahead of the actual knowledge of aquifers. This raises two fundamental issues: 1) which and how much data are necessary to make predictions accurate enough for aquifer management issues; 2) which models remain relevant to describe the heterogeneity and complexity of geological systems. The French observatory H+ was created in 2002 with the twofold motivation of acquiring a large database for validating models of heterogeneous aquifers, and of surveying groundwater quality evolution in the context of environmental changes. H+ is a network of 4 sites (Ploemeur, Brittany, France; HES Poitiers, France; Cadarache, France; Campos, Mallorca, Spain) with different geological, climatic, and economic contexts. All of them are characterized by a highly heterogeneous structure (fractured crystalline basement for Ploemeur, karstified and fractured limestone for Poitiers, Cadarache and Mallorca), which is far to be taken into account by basic models. Ploemeur is exploited as a tap-water plant for a medium-size coastal city (15,000 inhabitants) for 20 years. Each site is developed for long term investigation and monitoring. They involves a dense network of boreholes, detailed geological and geophysical surveys, periodic campaigns and/or permanent measurements of groundwater flow, water chemistry, geophysical signals (including ground motions), climatic parameter, etc. Several large-scale flow experiments are scheduled per year to investigate the aquifer structure with combined geophysical, hydrogeological, and geochemical instruments. All this information is recorded in a database that has been developed to improve the sustainability and quality of data, and to be used as a collaborative tool for both site researchers and modelers. This project lasts now for 5 years. It is a short time to collect the amount of information necessary to apprehend the complexity of aquifers; but it is already enough to obtain a few important scientific results about the very nature of the flow heterogeneity, the origin and residence time of water elements, the kinetic of geochemical processes, etc. We have also developed new methods to investigate aquifers (in-situ flow measurements, flow experiment designs, groundwater dating, versatile in-situ probes, etc.). This experience aiming at building up long term knowledge appears extremely useful to address critical issues related to groundwater aquifers: the structure and occurrence of productive aquifer in crystalline basement, the assessment of aquifer protection area in the context of highly heterogeneous flow, the biochemical reactivity processes, the long term evolution of both water quantity and quality in the context of significant environmental changes, for instance.

Objectives To investigate the association between priapism in men with sickle cell anemia (SCA) and hemorheological and hemolytical parameters. Materials and Methods Fifty-eight men with SCA (median age: 38 years) were included; 28 who had experienced priapism at least once during their life (priapism group) and 30 who never experienced this complication (control group). Twenty-two patients were treated with hydroxycarbamide, 11 in each group. All patients were at steady state at the time of inclusion. Hematological and biochemical parameters were obtained through routine procedures. The Laser-assisted Optical Rotational Cell Analyzer was used to measure red blood cell (RBC) deformability at 30 Pa (ektacytometry) and RBC aggregation properties (laser backscatter versus time). Blood viscosity was measured at a shear rate of 225 s(-1) using a cone/plate viscometer. A principal component analysis was performed on 4 hemolytic markers (i.e., lactate dehydrogenase (LDH), aspartate aminotransferase (ASAT), total bilirubin (BIL) levels and reticulocyte (RET) percentage) to calculate a hemolytic index. Results Compared to the control group, patients with priapism exhibited higher ASAT (p = 0.01), LDH (p = 0.03), RET (p = 0.03) levels and hemolytic indices (p = 0.02). Higher RBC aggregates strength (p = 0.01) and lower RBC deformability (p = 0.005) were observed in patients with priapism compared to controls. After removing the hydroxycarbamide-treated patients, RBC deformability (p = 0.01) and RBC aggregate strength (p = 0.03) were still different between the two groups, and patients with priapism exhibited significantly higher hemolytic indices (p = 0.01) than controls. Conclusion Our results confirm that priapism in SCA is associated with higher hemolytic rates and show for the first time that this complication is also associated with higher RBC aggregate strength and lower RBC deformability.

Introduction: Type 1 diabetes mellitus, a T cell-mediated autoimmune disease, is a complex process that results from the loss of tolerance to insulin and other beta-cell-specific antigens. Diabetes is associated with an increased risk of death from infectious disease. Hyperglycaemia, oxidative stress and inflammation have been identified as the main factor contributing to the development of diseases associated with diabetes mellitus. However, experimental evidence indicates individual susceptibility to develop complications of diabetes. Various genetic and environmental factors have been studied so far, but precise causation has yet to be established. Numerous studies in rodents and human subjects have been performed in order to elucidate the role of B and T cells, which determine the risk of development and progression of diabetes. Moreover, the therapy management of diabetes includes nutritional recommendations and exercise training. The effects of exercise training on glycemia, oxidative stress, inflammation and immune response are well known and favorable to limit the development of diabetes and its complications. More interestingly, ?-galacto-oligosaccharides (?-GOS) are prebiotics that selectively alter the composition of gut microbiota and exhibit strong antioxidant, anti- inflammatory and insulin-sensitive effects in vitro (Efstathiou and Fathi 2010) and in vivo (Boucher J et al. 2003). Thus, ? -GOS is a potential functional food for the treatment of diabetes. But, less is known about the effect of training and ? GOS on immune response in diabetes. In this context, the aim of this work was to study the immune response in a streptozotocin-induced type 1 diabetes with two therapeutic means: training and ?-galacto-oligosaccharides (?-GOS) alone or combined. Methods: Forty-two male Wistar rats were divided into control placebo, streptozotocin-diabetic rat (STZ) groups supplemented or not with ?-GOS (20 mg/kg/day) for 8 weeks and/or following endurance training. The training program was individualized and consisted in a 1h/day, 5days/week running during 8 weeks on a motor-driven treadmill. At the end of the protocol, blood was obtained by venous puncture into heparin-containing tubes for further analyses (glucose, insulin, fructosamine and immune responses) Results: Relative to the control group, STZ rats exhibited hyperglycemia, insulin deficiency and a trend to higher plasma fructosamine levels, a marker of glycated proteins. They also exhibited a decrease in T and B cells and an increase in natural killers (NK). ?-GOS treatment reduced plasma fructosamine levels and increased B cells compared to STZ rats. Training induced a decrease in plasma glucose, T cells and an increase in B cells in STZ rats. The combined effect of ?-GOS and training didnt potentiate the immune response on B and T cells but inhibited the effects of training on NK. Discussion: Our results suggest that training and ?-GOS alter positively plasma glucose and/or fructosamine and the immune response. But the effects of combined treatments only concern limited parameters. ? -GOS effects could be partly attributable to its immunomodulating activity.
