Effect of an herbivorous diet on energy balance of Litopenaeus vannamei at selected ontogenetic stages Auteur(s) : Maldonado, Carlos Cuzon, Gerard Guzman, Emilio Brito, Roberto Soto, Luis Arena, Leticia Gaxiola, Gabriela Éditeur(s) : Elsevier Résumé : Herbivorous (20% vegetable protein, 40% carbohydrate) and carnivorous diets (40% marine animal protein, 20% carbohydrate) were experimentally tested to assess their effect on the energy balance and energetic substrates utilized by postlarvae (PL's /15 days) and juvenile shrimp (3-6 g) of Litopenaeus vannamei. Postlarval stage 60 (PL60, early juveniles) shrimps fed HeD and CaD diets, then late juveniles (3-6 g) acclimated to the same diets were tested for their respective energy partitioning potential. No significant differences (p>0.05) on growth were obtained in early juveniles (mean final wet weight of 0.19 g). However in late juvenile stages a significant difference (p<0.05) in growth rate was observed. In terms of energy partitioning, both early and late juveniles seem to spend more energy in respiratory metabolism than in the elimination of excretion products. A change in feed composition based on quality protein sources induced some modifications on shrimp's activity measured by heat increment. Shrimps fed with an herbivorous diet showed a higher heat increment. Interestingly, the early stages of L vannamei display a remarkable capacity to assimilate a plant protein-based and a high carbohydrate level diet. Such capacity with a stimulation of genes would lead to a good adaptation of juveniles receiving all-plant diets to sustain growth performances up to a marketable size. Observation sur la maturation et la reproduction en captivité des crevettes peneides en milieu tropical Auteur(s) : Michel, A Éditeur(s) : Publ. CNEXO (France) (Actes Colloq.)(no. 7) Résumé : The present development of Peneid shrimp culture is limited by the problem set by the routine obtainment of reproduction in captivity. In Polynesia where no species of commercial interest is found naturally, this obtainment is a necessary prerequisite. The observations at the Centre Océanologique du Pacifique of CNEXO were made on 6 species of the genus Penaeus (P. merguiensis, P. aztecus, P. japonicus, P. monodon, P. vannamei and P. stylirostris) raised and maintained in 12 m² outdoortanks. At temperatures, between 25 and 32°, a salinity of 35 and a pH of 8,2, maturations and spawnings are seen throughout the year and several generations were obtained : P. merguiensis, F7 ; P. aztecus et P. japonicus, F3 ;P. monodon, F2 ; P. stylirostris and P. vannamei F1. For P. aztecus et P. mondon maturation is induced by unilateral eyestalk ablation. The animals behavior copulation and external signs of ovarian, development are described. The factors which seem essential to the proper progress of maturation are the temperature, the light intensity, the food and the state of health of the animals. Le développement des élevages de crevettes Pénéidés est actuellement limité par les problèmes que pose l'obtention en routine de la reproduction en captivité. En Polynésie où aucune espèce d'intérêt commercial ne se trouve à l'état naturel, cette obtention était un préalable nécessaire. Les observations effectuées au Centre Océanologique du Pacifique du CNEXO ont porté sUr 6 espèces du genre Penaeus (P. merguiensis, p. aztecus, P. japonicus, P. monodon, P. vannamei et P. stylirostris) élevées et maintenues dans des bacs extérieurs de 12 m² en circulation ouverte. Avec des températures variant de 25 à 32°, une salinité de 35 et un pH voisin de 8,2, maturations et pontes s'obserVent toute l'année et plusieurs générations ont été obtenues : P. merguiensis, F7 ; P. aztecus et P. japonicus, F3 ;P. monodon, F2 ; P. stylirostris et P. vannamei, F1. Pour P. aztecus et P. mondon, la maturation est induite par épédonculation unilatérale. Le comportement des animaux, la copulation et les signes extérieurs du développement des ovaires sont décrits. Survival rate in FLOC and clear water were recorded and a probiotic was tested under both conditions. Daily growth rate (DGR) was higher in FLOC (p < 0.05) than in CW. Survival in each system increased significantly when a probiotic was included in the diet (p < 0.05). The Vibrionaceae community from the hepatopancreas (HP) and the culture medium did not differ between the two culture media. Nevertheless, a novel group of Vibrio strains was found to be unique to FLOC. No high level of lesions was observed in shrimp tissues from the FLOC + probiotic treatment; it suggests that the probiotic contributed to homeostasis and prevented outbreak of opportunistic pathogenic species. Pathogenicity of Vibrio penaeicida or white shrimp Litopenaeus vannamei: a cysteine protease-like exotoxin as a virulence factor Auteur(s) : Aguirre Guzman, G Ascencio, F Saulnier, Denis Éditeur(s) : Inter-Research Résumé : The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. Toxicity and median lethal doses (LD50) of Fast Protein Liquid Chromatography (FPLC) products were evaluated on L. vannamei. The first FPLC fraction sample (A) from PFs40 of the AM101 strain displayed LD50 values of 1.68 and 5.61 mu g protein ind.(-1), respectively. The second FPLC process from Fraction A showed a peak (A1) also with toxic effects to shrimp. PFs40, Fraction A, and Peak At showed a 38.5 kDa molecular band (SDS-PAGE), with activity on a gelatin protease zymogram. The lethal effect of PFs40 and Fraction A was inhibited by Proteinase K, CuCl2, E-64, and heat (60 and 100 degrees C) treatments, but was not inhibited by EDTA-Na-2, aprotinin, and soy trypsin treatments. These results and the zymogram inhibition test suggest the presence of a cysteine protease-like proteinaceous exotoxin as a dominant protease, secreted by V. penaeicida Strain AM101. Its osmoregulation pattern and variations in its tolerance to salinity are partially known from several studies under different experimental conditions (developmental stages, salinities and acclimation procedures). Although L. vannamei is recognized as one of the most euryhaline penaeid species, with adults and juveniles exhibiting an hyper-hypo-osmoregulatory pattern and being able to tolerate a wide salinity range, little is known on larval and early postlarval strategies to cope with salinity fluctuations. In order to establish their euryhalinity range and to fully understand the ontogenetic changes in L. vannamei osmoregulatory pattern, we evaluated the effect of six salinities (5, 10, 20, 32, 45 and 60 psu) on 17 developmental stages by directly exposing them to experimental salinities and conducting observations during the next 48 hours. At 5 hours post osmotic shock (hps), all developmental stages survived (> 20%) in 20, 32 and 45 psu. The euryhalinity and osmoregulation pattern changed at some developmental stages of L. vannamei. The hyper-hypo osmoregulatory pattern exhibited by Juvenile and Adult appears to be established early in the first post-larval stage PL1 (ontogenetic osmoregulation pattern type 3), with higher tolerance to salinity variations observed in PL2, PL4 and PL22 suggesting that L. vannamei shows a progressive increase in the efficiency of osmoregulatory mechanism following last metamorphosis. Analyse économique d'une nouvelle technique d'élevage de crevettes en cages flottantes au Brésil Auteur(s) : Paquotte, Philippe Éditeur(s) : Actes de la 8ème Conférence de l'IIFET (Marrakech, juillet 1996) Résumé : In the framework of French-Brazil co-operation, the French Institute of research for exploitation of the sea (IFREMER), the State of Bahia and the company Littoral Sul Maricultura have developed a new technique for shrimp farming (Penaeus vannamei) using floating cages. After trials carried out during three years, this technique turns out very interesting from several viewpoints. The growth speed and the survival rate are very good, the negative effects on the environmental quality are weak and local fishermen have proved eager to adopt this technique as a part-time activity. In the same time, a technico-economic analysis, using a computerised simulation tool, as well as a market analysis in the State of Bahia have been carried out in order to assess the feasibility of such a project and to help research and development decisions. Despite low first investment and good biological results, the ex-farm production cost of shrimps weighing 15 g is as high as 4,5 US$ per kilo. This price is due to the low productivity of the labour and to the cost of nets which have a short life-time. It is possible to consider this activity as profitable on the market of Salvador de Bahia. Nevertheless, the question is how to face the international competition, which is particularly strong in the case of tropical shrimps. The conditions allowing enterprises to develop a strategy based on product quality and environmentally friendliness are reviewed. [NOT CONTROLLED OCR] Dans le cadre de la coopération Franco-Brésilienne, l'IFREMER, l'Etat de Bahia et la société Littoral Sul Maricultura ont développé une nouvelle technique d'élevage de crevettes pénéides (Penaeus vannamei) en utilisant des cages flottantes. Après trois années de mise au point, cette technique s'est avérée intéressante sur plusieurs aspects. La vitesse de croissance et le taux de survie sont très satisfaisants, les effets négatifs sur la qualité du milieu naturel sont faibles et les pêcheurs locaux ont accueilli favorablement cette nouvelle pratique en complément de leur activité traditionnelle. Parallèlement, une analyse technico-économique utilisant un outil de simulation informatique ainsi qu'une analyse du marché de la crevette dans l'état de Bahia ont été conduites afin d'évaluer la faisabilité d'un tel projet et d'orienter les actions de recherche et développement. En dépit d'investissements initiaux plus faibles que dans les élevages en bassins et de bonnes performances zootechniques, le coût de production de crevettes de 15 g atteint 4,7 US$ par kilo. Dans les conditions de marché de l'Etat de Bahia, la rentabilité d'un projet d'élevage de crevettes en cage serait assurée, mais la faible productivité du travail et le coût de remplacement des filets ne permettent pas d'avoir des coûts de production aussi bas que dans les élevages en bassins. C'est pourquoi la question se pose de la possibilité pour ce type de production innovante d'affronter une concurrence internationale très forte comme dans le cas des crevettes tropicales. However, selection for bacterial infection resistance is not documented. In New Caledonia, "Syndrome 93" is a vibriosis that affects cultured shrimps during the cool season with survival from 15 to 30%.
Current practises for breeders rearing at low densities do not improve the resistance of the offsprings mainly because selection pressures for "Syndrome 93" are low. Even after the fifth generation, survival improvement is not significant. La sélection génétique de lignées de crevettes résistantes à certains virus a démontré son efficacité en particulier chez L. vannamei et L. stylirostris. En revanche, la sélection de crevettes résistantes à une bactérie du genre Vibrio est peu documentée. En Nouvelle-Calédonie, le « Syndrome 93 », vibriose à V. penaeicida, affecte toutes les fermes de crevettes L. stylirostris lors des élevages de saison fraîche. Les survies en bassins de grossissement, de l’ordre de 40 à 60% dans un élevage sain, tombent à des valeurs de 15 à 30%. L’élevage de géniteurs à faible densité, s’il maximise les performances de reproduction, ne semble pas optimiser la qualité de la descendance du fait des faibles pressions de sélection qui s’exercent vis-à-vis du syndrome 93. Les améliorations significatives du nombre d’animaux survivant aux infections et aux épisodes de mortalité en bassin restent faibles en valeur absolue à la 5ème génération de sélection expérimentale en conditions d’élevage, mais sur le long terme la stratégie testée pourrait se révéler économiquement efficace. Lack of evidence for Litopenaeus vannamei Toll receptor (lToll) involvement in activation of sequence-independent antiviral immunity in shrimp Auteur(s) : Labreuche, Yannick O'Leary, Nuala A. Éditeur(s) : Elsevier Résumé : injection of non-specific dsRNA initiates a broad-spectrum innate antiviral immune response in the Pacific white shrimp, Litopenaeus vannamei, however, the receptor involved in recognition of this by-product of viral infections remains unknown. In vertebrates, dsRNA sensing is mediated by a class of Toll-like receptors (TLRs) and results in activation of the interferon system. Because a TLR (lToll) was recently characterized in L. vannamei, we investigated its potential role in dsRNA recognition. We showed that injection of non-specific RNA duplexes did not modify lToll gene expression. A reverse genetic approach was therefore implemented to study its role in vivo. Silencing of lToll did not impair the ability of nonspecific dsRNA to trigger protection from white spot syndrome virus and did not increase the shrimp susceptibility to viral infection, when compared to controls. In contrast, gene-specific dsRNA injected to specifically silence lToll expression activated an antiviral response. These data strongly suggest that shrimp lToll plays no role in dsRNA-induced antiviral immunity. (C) 2009 Elsevier Ltd. All rights reserved.
