Résumé : Herbivorous (20% vegetable protein, 40% carbohydrate) and carnivorous diets (40% marine animal protein, 20% carbohydrate) were experimentally tested to assess their effect on the energy balance and energetic substrates utilized by postlarvae (PL's /15 days) and juvenile shrimp (3-6 g) of Litopenaeus vannamei. Postlarval stage 60 (PL60, early juveniles) shrimps fed HeD and CaD diets, then late juveniles (3-6 g) acclimated to the same diets were tested for their respective energy partitioning potential. No significant differences (p>0.05) on growth were obtained in early juveniles (mean final wet weight of 0.19 g). However in late juvenile stages a significant difference (p<0.05) in growth rate was observed. In terms of energy partitioning, both early and late juveniles seem to spend more energy in respiratory metabolism than in the elimination of excretion products. A change in feed composition based on quality protein sources induced some modifications on shrimp's activity measured by heat increment. Shrimps fed with an herbivorous diet showed a higher heat increment. Interestingly, the early stages of L vannamei display a remarkable capacity to assimilate a plant protein-based and a high carbohydrate level diet. Such capacity with a stimulation of genes would lead to a good adaptation of juveniles receiving all-plant diets to sustain growth performances up to a marketable size. The implications of these findings for the shrimp farming feeding costs are briefly discussed. (C) 2009 Elsevier B.V. All rights reserved.

Résumé : The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. Toxicity and median lethal doses (LD50) of Fast Protein Liquid Chromatography (FPLC) products were evaluated on L. vannamei. The first FPLC fraction sample (A) from PFs40 of the AM101 strain displayed LD50 values of 1.68 and 5.61 mu g protein ind.(-1), respectively. The second FPLC process from Fraction A showed a peak (A1) also with toxic effects to shrimp. PFs40, Fraction A, and Peak At showed a 38.5 kDa molecular band (SDS-PAGE), with activity on a gelatin protease zymogram. The lethal effect of PFs40 and Fraction A was inhibited by Proteinase K, CuCl2, E-64, and heat (60 and 100 degrees C) treatments, but was not inhibited by EDTA-Na-2, aprotinin, and soy trypsin treatments. These results and the zymogram inhibition test suggest the presence of a cysteine protease-like proteinaceous exotoxin as a dominant protease, secreted by V. penaeicida Strain AM101.

Résumé : The advantages of FLOC over clear water (CW) in rearing juvenile L. vannamei and its effects on Vibrio communities were evaluated. Survival rate in FLOC and clear water were recorded and a probiotic was tested under both conditions. Daily growth rate (DGR) was higher in FLOC (p < 0.05) than in CW. Survival in each system increased significantly when a probiotic was included in the diet (p < 0.05). The Vibrionaceae community from the hepatopancreas (HP) and the culture medium did not differ between the two culture media. Nevertheless, a novel group of Vibrio strains was found to be unique to FLOC. No high level of lesions was observed in shrimp tissues from the FLOC + probiotic treatment; it suggests that the probiotic contributed to homeostasis and prevented outbreak of opportunistic pathogenic species.

Résumé : A study was conducted to evaluate variations of digestive enzyme activities in Litopenaeus vannamei (Boone) reared in commercial ponds under semi-intensive conditions. Shrimp were collected at each body weight increase of 2 g. As the shrimp grew (2-12 g), significant increases in the activities of lipase and chymotrypsin were observed. The total protease activity decreased from 6 g onwards. Trypsin activity showed a peak at 6 g and amylase activity increased two-fold after 2 g. Additionally, the stomach contents were analysed microscopically for shrimp between 2 and 10 g. Plant matter contributed above 30% of the total stomach content in 6-, 8- and 10-g shrimp. Detritus represented 58% and 62% of the total stomach content in 2- and 4-g shrimp, respectively, decreasing to 33-43% at greater shrimp weights. Artificial feed showed a maximum contribution of 20% in 6-g shrimp. The present results show changes in the enzyme activity after the shrimp reach 6 g in body weight, evidenced by a decrease in total protease and an increase in lipase and amylase activities. The amylase/protease ratio was 2.6 in 2-g shrimp and increased steadily to 9.6 in 12-g shrimp. These findings suggest an adaptation of the enzymatic activity to diets with lower protein content as body weight increases, and may be related to the variation of the different items found in the stomach.

Résumé : The euryhaline white shrimp Litopenaeus vannamei lives in both coastal and oceanic areas through ontogeny. Its osmoregulation pattern and variations in its tolerance to salinity are partially known from several studies under different experimental conditions (developmental stages, salinities and acclimation procedures). Although L. vannamei is recognized as one of the most euryhaline penaeid species, with adults and juveniles exhibiting an hyper-hypo-osmoregulatory pattern and being able to tolerate a wide salinity range, little is known on larval and early postlarval strategies to cope with salinity fluctuations. In order to establish their euryhalinity range and to fully understand the ontogenetic changes in L. vannamei osmoregulatory pattern, we evaluated the effect of six salinities (5, 10, 20, 32, 45 and 60 psu) on 17 developmental stages by directly exposing them to experimental salinities and conducting observations during the next 48 hours. At 5 hours post osmotic shock (hps), all developmental stages survived (> 20%) in 20, 32 and 45 psu. The euryhalinity and osmoregulation pattern changed at some developmental stages of L. vannamei. The hyper-hypo osmoregulatory pattern exhibited by Juvenile and Adult appears to be established early in the first post-larval stage PL1 (ontogenetic osmoregulation pattern type 3), with higher tolerance to salinity variations observed in PL2, PL4 and PL22 suggesting that L. vannamei shows a progressive increase in the efficiency of osmoregulatory mechanism following last metamorphosis.

Résumé : Many questions remain unanswered regarding RNAi-based mechanisms and dsRNA-induced antiviral immune responses in penaeid shrimp. In this study, we report the characterization in the white leg shrimp Litopenaeus vannamei of RNAi pathway associated proteins Lv-Ago 1 and Lv-Ago 2, two members of the Argonaute family of proteins, as well as Lv-sid 1, the first shrimp homologue of Sid-1, a membrane channel-forming protein implicated in the cellular import of dsRNA. To decipher their functional implication in RNAi-related phenomena, we monitored their relative expression following stimulation by specific and non-specific RNA duplexes of diverse length. The findings show that the length of small RNA duplexes plays a critical role in the activation of both RNAi-related and innate antiviral responses. They also suggest that these two mechanisms of antiviral response may activate the same pathway, requiring Lv-Sid 1 and Lv-Ago 2 induction. (C) 2010 Elsevier Ltd. All rights reserved.

Résumé : Antimicrobial peptides play a major role in innate immunity. The penaeidins, initially characterized from the shrimp Litopenaeus vannamei, are a family of antimicrobial peptides that appear to be expressed in all penaeid shrimps. As of recent, a large number of penaeid nucleotide sequences have been identified from a variety of penaeid shrimp species and these sequences currently reside in several databases under unique identifiers with no nomenclatural continuity. To facilitate research in this field and avoid potential confusion due to a diverse number of nomenclatural designations, we have made a systematic effort to collect, analyse, and classify all the penaeidin sequences available in every database. We have identified a common penaeidin signature and subsequently established a classification based on amino acid sequences. In order to clarify the naming process, we have introduced a 'penaeidin nomenclature' that can be applied to all extant and future penaeidins. A specialized database, PenBase, which is freely available at http://www.penbase.immunaqua.com, has been developed for the penaeidin family of antimicrobial peptides, to provide comprehensive information about their properties, diversity and nomenclature. Résumé : injection of non-specific dsRNA initiates a broad-spectrum innate antiviral immune response in the Pacific white shrimp, Litopenaeus vannamei, however, the receptor involved in recognition of this by-product of viral infections remains unknown. In vertebrates, dsRNA sensing is mediated by a class of Toll-like receptors (TLRs) and results in activation of the interferon system. Because a TLR (lToll) was recently characterized in L. vannamei, we investigated its potential role in dsRNA recognition. We showed that injection of non-specific RNA duplexes did not modify lToll gene expression. A reverse genetic approach was therefore implemented to study its role in vivo. Silencing of lToll did not impair the ability of nonspecific dsRNA to trigger protection from white spot syndrome virus and did not increase the shrimp susceptibility to viral infection, when compared to controls. In contrast, gene-specific dsRNA injected to specifically silence lToll expression activated an antiviral response. These data strongly suggest that shrimp lToll plays no role in dsRNA-induced antiviral immunity. (C) 2009 Elsevier Ltd. All rights reserved.
