Résultats des importations, acclimatation et hybridation. a) Rappel des conditions d'introduction de nouvelles souches. L'importation de toute espèce est conditionnée par les résultats d'une enquête préliminaire dont l'objet est de rechercher les exigences écologiques de l'espèce et la situation épidémiologique dans son milieu d'origine. Le travail du LGP et de ses partenaires a été partagé en différentes actions: 1. Recherche de contacts dans divers pays étrangers, en vue de l'importation d'huîtres présentant des caractéristiques intéressantes. 2. Importation : L'importation se fait selon les normes du CIEM (Conseil International pour l'Exploration de la Mer) : contrôles pathologiques sur les lots d'animaux importés, confinement en salle de quarantaine des géniteurs avec stérilisation systématique des eaux de rejet, sacrifice des géniteurs après la production de la première génération. 3. Production d'une première descendance: La ponte est également réalisée en salle de quarantaine. Des contrôles pathologiques auront lieu tout au long de l'élevage. 4. Contrôle des performances : ils se font, dans un premier temps, uniquement au laboratoire avec stérilisation des eaux de rejet. Cette caractérisation intègre différentes approches : résistance aux parasites présents dans le milieu français, caractères quantitatifs (poids, taille, rendement, vitesse de croissance), polymorphisme enzymatique ou ADN, tests physiologiques (mesure de la respiration, du rendement d'assimilation ... ). 5. Hybridation: Des hybridations entre espèces ont été tentées, avec contrôle par marqueurs génétiques. 6. Conservatoire de souches : Les générations F1 des différentes espèces ont systématiquement été conservées. Une salle a été spécialement équipée au LGP dans cet objectif. Determination of gender in the pearl oyster pinctada margaritifera Auteur(s) : Chavez-villalba, Jorge Soyez, Claude Huvet, Arnaud Gueguen, Yannick Lo, Cedrik Le Moullac, Gilles Éditeur(s) : Natl Shellfisheries Assoc Résumé : The pearl industry in French Polynesia is based on exploitation of natural stocks of the black-lip pearl oyster (Pinctada margaritifera); it generates an annual turnover of 90 million euros. Improvements in pearl quality need genetic studies to improve the populations. This pearl oyster is a protandric species, where the sex-ratio normally is biased towards males. There is an increasing interest in gender control to find the mechanisms to augment female proportions for management purposes. This review summarizes information on exogenous and endogenous factors regulating gender in this and other bivalves and concludes that P. margaritifera is a protandric hermaphrodite, developing as a male during the first two years and without evidence of an effect from abiotic and biotic factors on gender during this phase. Later, pearl oysters progressively change to females, reaching a sex ratio close to 1:1 in specimens >8 years; at this stage, gender is apparently influenced by environmental parameters, but particularly by stress. Future research should seek to accurately determine the effect of temperature and food on sex ratios. Studies should be performed to characterize genes responsible for expression of gender. The use of hormones is a path that might be explored to influence the gender of pearl oysters. In the Pacific oyster Crassostrea gigas, the oyster gonadal (og) TGF-beta gene was recently characterized through genome-wide expression profiling of oyster lines selected to be resistant or susceptible to summer mortality. Og TGF-beta appeared specifically expressed in the gonad to reach a maximum when gonads are fully mature, which singularly contrasts with the pleiotropic roles commonly ascribed to most TGF-beta family members. The function of og TGF-beta protein in oysters is unknown, and defining its role remains challenging. In this study, we develop a rapid bacterial production system to obtain recombinant og TGF-beta protein, and we demonstrate that og TGF-beta is processed by furin to a mature form of the protein. This mature form can be detected in vivo in the gonad. Functional inhibition of mature og TGF-beta in the gonad was conducted by inactivation of the protein using injection of antibodies. We show that inhibition of og TGF-beta function tends to reduce gonadic area. We conclude that mature og TGF-beta probably functions as an activator of germ cells development in oyster. Natural hybridization between genetically differentiated populations of Crassostrea gigas and C-angulata highlighted by sequence variation in flanking regions of a microsatellite locus Auteur(s) : Huvet, Arnaud Fabioux, Caroline Mccombie, Helen Lapegue, Sylvie Boudry, Pierre Éditeur(s) : Inter-Research Résumé : The marine environment is of special interest for studying hybridization between closely related taxa because of the high dispersal potential of planktonic larvae, such as those of most bivalve species. The oysters Crassostrea angulata and C. gigas are known to be very close genetically and entirely inter-fertile under controlled conditions. However, hybridization in the wild had not been investigated, mainly due to the lack of nuclear diagnostic markers. In the present paper, we first estimated genetic differentiation between these 2 closely related taxa using 8 microsatellite markers. Interestingly, 5 markers displayed significant differences of allele size between taxa. The subsequent sequencing of alleles of one of these microsatellites showed several mutational events, which suggested null alleles and homoplasy. The presence of 1 insertion/deletion event in its 5' flanking sequence enabled us to design a new bi-allelic ('C' and 'NC') nuclear PCR-restriction fragment length polymorphism (-RFLP) marker (CG44R). This, together with a mitochondrial DNA marker, was used to analyze populations of C. angulata and C. gigas. The CG44R allele frequencies were very different between C. angulata (f[C] = 0.91) and C. gigas (f[NC] = 0.92) populations. This analysis also provided evidence for hybridization between C. angulata and C. gigas in a wild Portuguese population where the 2 taxa are in contact due to recent transportation of C. gigas stocks for aquacultural production. Our results represent the first indication of hybridization between these 2 taxa in the natural environment, and contribute to knowledge of the evolutionary history of the Crassostrea genus. Ressources génétiques et phylogéographie des huîtres creuses Crassostrea gigas et C. angulata : variabilité, différenciation et adaptation des populations naturelles et introduites Auteur(s) : Huvet, Arnaud Éditeur(s) : Université de Tours Résumé : L 'huître creuse japonaise Crassostrea gigas et portugaise C. angulata ont été respectivement décrites par Thunberg (1793) et Lamarck (1819). Mais les similitudes morphologiques et l 'homogénéité des fréquences allozymiques entre populations ont conduit de nombreux auteurs à proposer l'existence d'une seule et mênle espèce. L'analyse récente d'un gène mitochondrial a pennis de déterminer l'origine asiatique de C. angulata, soulignant l'intérêt de l'étude de la différenciation génétique et phénotypique entre les deux taxons. La notion biologique d'espèce a tout d'abord été examinée. Des croisements de 1ère et 2ème génération ont montré la viabilité et fertilité des hybrides. L'analyse de la compétition spennatique par marqueurs microsatellites ne montre pas d'isolement reproductif, mênle partiel et le séquençage du fragment ITS 1 confirme la forte proximité génétique des deux taxons. Les patrons de différenciation génétique observés par marqueurs microsatellites et mitochondriaux apparaissent sitnilaires. L'origine asiatique de C. angulata est donc confinnée. Le fort niveau de polymorphisme des marqueurs microsatellites explique partiellement les plus faibles valeurs observées de différenciation. Une bimodalité des tailles d'allèles microsatellites entre taxons a pennis le développement d'un nouveau marqueur nucléaire, mettant en évidence des phénomènes d'hybridation naturelle dans la zone du sud de l'Europe où l'activité ostréicole met aujourd'hui les 2 taxons en contact. L'étude de caractères phénotypiques a été réalisée en milieu naturel et contrôlé sur des descendances d'écloserie. Les résultats montrent une meilleure croissance de C. gigas et une période de reproduction plus longue chez C. angulata. Des mesures physiologiques montrent un plus fort taux de respiration du naissain C. gigas, ainsi qu'un temps de filtration supérieur chez les adultes. La différence de croissance entre taxons pourraient donc être expliquée par le temps de filtration et l'effort de reproduction. Vibrio aestuarianus zinc metalloprotease causes lethality in the Pacific oyster Crassostrea gigas and impairs the host cellular immune defenses Auteur(s) : Labreuche, Yannick Le Roux, Frederique Henry, Joel Zatylny, Celine Huvet, Arnaud Lambert, Christophe Soudant, Philippe Mazel, Didier Éditeur(s) : Academic Press Ltd- Elsevier Science Ltd Résumé : Extracellular products (ECPs) of the pathogenic Vibrio aestuarianus 01/32 were previously reported to display lethality in Crassostrea gigas oysters and to cause morphological changes and immunosuppression in oyster hemocytes. To identify the source of this toxicity, biochemical and genetic approaches were developed. ECP protease activity and lethality were shown to be significantly reduced following incubation with metal chelators, suggesting the involvement of a zinc metalloprotease. An open reading frame of 1836 bp encoding a 611-aa metalloprotease (designated yam) was identified. The deduced protein sequence showed high homology to other Vibrio metalloproteases reported to be involved in pathogenicity. To further confirm the role of this enzyme in ECP toxicity, a plasmid carrying the yarn gene under the control of an araC-P-BAD expression cassette was transferred to a Vibrio splendidus related strain, LMC20012(T), previously characterized as non-pathogenic to oysters. Expression of Vam conferred a toxic phenotype to LMG20012(T) ECPs in vivo and cytotoxicity to oyster hemocytes in vitro. Collectively, these data suggest that the Vam metalloprotease is a major contributor to the toxicity induced by V aestuarianus ECPs and is involved in the impairment of oyster hemocyte functions.
