In the southern part of the Atlantic Ocean, Crassostrea rhizophorae has been observed along the southern coast of America and Crassostrea gasar along the western coast of Africa. Fifteen populations of these oysters were sampled from these two geographic areas. Their polymorphism was studied on the 16S mitochondrial fragment both by par sequencing and RFLP analysis. Some samples' caryotype were also analysed. Two haplotypes were identified: C. gasar was found in Africa, but also for the first time in South America, whereas C. rhizophorae was only found on the coasts of South America. Les huîtres de mangrove englobent plusieurs espèces qui sont présentes sur différentes côtes. Dans la partie sud de l'Océan Atlantique, Crassostrea rhizophorae a été décrite le long des côtes sud américaines et Crassostrea gasar le long des côtes ouest africaines. Quinze populations de ces huîtres ont été échantillonnées dans ces deux zones géographiques. Leur polymorphisme a été étudié sur le fragment mitochondrial 16S par séquençage et analyse en RFLP, le caryotype de certains échantillons a aussi été examiné. These oysters from the French Guyanese coasts host some gregarine Nematopsis sp. spores, characterized by Comps, Grizel and Tige (1972). Through this work, we conducted some experimental infestation trials between Crassostrea rhizophorae spats parasited by these spores and the most common crab on metropolitan farms Carcinus maenas. Poursuivant les objectifs de développement de l'ostréiculture en Guyane et étudiant les possibilités d'accroissement d'un stock constitué de nouvelles espèces d'huîtres, nous avons été amenés à rechercher d'éventuels parasites chez Crassostrea rhizophorae. In the Pacific oyster Crassostrea gigas, the oyster gonadal (og) TGF-beta gene was recently characterized through genome-wide expression profiling of oyster lines selected to be resistant or susceptible to summer mortality. Og TGF-beta appeared specifically expressed in the gonad to reach a maximum when gonads are fully mature, which singularly contrasts with the pleiotropic roles commonly ascribed to most TGF-beta family members. The function of og TGF-beta protein in oysters is unknown, and defining its role remains challenging. In this study, we develop a rapid bacterial production system to obtain recombinant og TGF-beta protein, and we demonstrate that og TGF-beta is processed by furin to a mature form of the protein. This mature form can be detected in vivo in the gonad. Functional inhibition of mature og TGF-beta in the gonad was conducted by inactivation of the protein using injection of antibodies. We show that inhibition of og TGF-beta function tends to reduce gonadic area. We conclude that mature og TGF-beta probably functions as an activator of germ cells development in oyster. Vibrio splendidus LGP32 was shown to resist to up to 10 mu M Cg-Def defensin and Cg-BPI bactericidal permeability increasing protein, two antimicrobial peptides/proteins (AMPs) involved in C. gigas immunity. The resistance to both oyster Cg-Def and Cg-BPI and standard AMPs (polymyxin B, protegrin, human BPI) was dependent on the ompU gene. Indeed, upon ompU inactivation, minimal bactericidal concentrations decreased by up to fourfold. AMP resistance was restored upon ectopic expression of ompU. The susceptibility of bacterial membranes to AMP-induced damages was independent of the ompU-mediated AMP resistance. Besides its role in AMP resistance, ompU proved to be essential for the adherence of V. splendidus LGP32 to fibronectin. Interestingly, in vivo, ompU was identified as a major determinant of V. spiendidus pathogenicity in oyster experimental infections. Indeed, the V. splendidus-induced oyster mortalities dropped from 56% to 11% upon ompU mutation (Kaplan Meier survival curves, P < 0.01). Moreover, in co-infection assays, the ompU mutant was out competed by the wild-type strain with competitive indexes in the range of 0.1-0.2. From this study, ompU is required for virulence of V. splendidus. Contributing to AMP resistance, conferring adhesive properties to V. splendidus, and being essential for in vivo fitness, the OmpU porin appears as an essential effector of the C. gigas/V. splendidus interaction. Oyster population genetics : understanding natural populations and tracing introductions Auteur(s) : Lapegue, Sylvie Boudry, Pierre Éditeur(s) : World Aquaculture Society, Aquaculture 2006 Résumé : Oysters are among the most familiar of all marine invertebrate taxa. However our knowledge of oyster phylogeny and systematics is fragmentary. This is principally due to the plastic growth patterns of these animals, which result in a wide range of overlapping, ecophenotypic variants that greatly reduce the value of analysis based on shell morphology. Besides that, many intentional or accidental anthropogenic transfers have emphasised this situation. In several cases of misclassification or misidentification of oysters, DNA molecular data, have provided valuable new insights on the tracing of introductions or more generally on the phylogeographic relationships between oyster species. We will illustrate this issue with recent case studies from our lab on several flat and cupped oysters: (1) the Crassostrea gigas Crassostrea angulata couple of Asian species and their successive introductions in Europe, (2) the new species of cupped oysters, Crassostrea honkongensis, described in Hong-Kong and very divergent from the species Crassostrea gigas it was initially supposed to be, (3) the mangrove oyster Crassostrea gasar that was shown not only to be present along the coasts of Western Africa but also along the Atlantic coasts of South America, and (4) the geographic disjunction between Ostreola stentina, described in the Mediterranean Sea, and the genetically closely related species Ostrea aupouria (from New Zealand) and Ostreola equestris (from Mexico Gulf/Atlantic). At the intra specific level, we will focus on one of the main documented case in Europe, the flat oyster, Ostrea edulis. It is a marine bivalve whose natural geographical distribution ranges along the European Atlantic coast from Norway to Morocco, in addition to the Mediterranean and Black Sea. Studies of allozymes, microsatellites and mitochondrial differentiation over the whole range concluded that a significant divergence existed between Mediterranean and Atlantic populations, together with an isolation-by-distance pattern. However, the average mitochondrial haplotypic diversity displayed a high among populations variance, reflecting smaller effective population size in some locations. Additionally, a ten-fold quantitative difference was observed in the same study in Fst between the mitochondrial and the nuclear genomes, which could be due to sex biased differential reproductive success between males and females. In order to further document this hypothesis, two experiments at the population level were conducted. First, brooding females were sampled in a wild population and the number of males fertilizing a given female estimated. Then, parentage analyses were achieved under experimental conditions: successive mass spawnings were collected from a population of potential genitors kept in hatchery, whose genotypes were known, in order to infer a posteriori the relative contribution of each genitor. Moreover, we aimed to better understand the reproduction dynamics of this species. Several patterns of spawning could be distinguished: unique, successive or extended in time. The different parental contributions and reproductive behaviors observed in these experiments are discussed in the context of the hypothesis of a variance in the reproductive success of males and females and consequences in local and temporal reduced effective population sizes. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and, consequently, for the study of bivalve larvae genetics. Culling, by selective sieving of the smallest larvae is an advantageous practice at a phenotypic scale as it reduced variance in larval size, variance of developmental rate and time to settlement. Culling of 50% of the larval population only led to 15% less spat, showing a positive phenotypic correlation between larval growth and settlement success. However, culling represents a substantial risk for diversity loss, because it increases the variance of reproductive success among parental oysters. The effective population sizes of early settling cohorts of settlement were lower than those of later ones. Our results show that the settlement of slow growing larvae significantly contributes to minimizing the variability of reproductive success and therefore to maximizing genetic diversity. These results corroborate the low estimations of variability of broodstocks sampled in several French commercial hatcheries, relative to natural populations. The genetic composition of the larval population and the resulting spat was significantly different between the two tested temperatures, revealing genotype x environment interaction for survival. Similarly, genotype x environment interaction was also observed for larval growth as a higher temperature exerted a positive influence on the expression of genetic variability for this trait. Consequently, we can conclude that a temperature of 26°C coupled with culling, to common practice in oyster hatcheries, is likely to amplify the selection pressure for fast growing larvae. To test for this hypothesis, we compared larval developmental traits in the progeny of a hatchery broodstock closed for 7 generations, with the progeny of wild oysters and the two possible hybrids. Our results show that selection of fast growing larvae can counteract presumed inbreeding depression, due to higher mean relatedness among hatchery broodstock than in the wild. Genetic effects of intensive rearing conditions at larval stage are significant and should be taken into account in hatchery practices, especially in terms of genetic diversity management. Afin d'étudier les conséquences génétiques des pratiques de production en écloserie d'huître creuse, deux facteurs ont été examinés : l'élimination des petites larves et la température. Nos résultats montrent que l'assignation de parenté par marqueurs microsatellites est un outil performant pour les études génétiques en phase larvaire de familles élevées en mélange. Bien qu'avantageux d'un point de vue phénotypique, le tamisage sélectif représente un risque de perte de diversité. La fixation des larves à croissance lente permet en effet de minimiser la variabilité du succès reproducteur et de fait, de maximiser la variabilité génétique. Ces résultats corroborent les estimations de variabilité sur les stocks d'écloseries françaises où l'on constate une diversité allélique inférieure à celle de populations issues du milieu naturel. La température exerce également une influence sur la précocité de l'expression de la variabilité génétique pour la croissance larvaire. Ainsi une température élevée associée à une procédure de tamisage peut amplifier l'effet sélectif. Enfin, la sélection de larves à croissance rapide semble démontrée, s'opposant à la dépression de consanguinité présumée. Les conditions d'élevage peuvent donc avoir un effet génétique significatif qui devrait être pris en considération dans les pratiques d'écloserie, notamment dans la gestion de la diversité génétique. L'importation de toute espèce est conditionnée par les résultats d'une enquête préliminaire dont l'objet est de rechercher les exigences écologiques de l'espèce et la situation épidémiologique dans son milieu d'origine. Le travail du LGP et de ses partenaires a été partagé en différentes actions: 1. Recherche de contacts dans divers pays étrangers, en vue de l'importation d'huîtres présentant des caractéristiques intéressantes. 2. Importation : L'importation se fait selon les normes du CIEM (Conseil International pour l'Exploration de la Mer) : contrôles pathologiques sur les lots d'animaux importés, confinement en salle de quarantaine des géniteurs avec stérilisation systématique des eaux de rejet, sacrifice des géniteurs après la production de la première génération. 3. Production d'une première descendance: La ponte est également réalisée en salle de quarantaine. Des contrôles pathologiques auront lieu tout au long de l'élevage. 4. Contrôle des performances : ils se font, dans un premier temps, uniquement au laboratoire avec stérilisation des eaux de rejet. Cette caractérisation intègre différentes approches : résistance aux parasites présents dans le milieu français, caractères quantitatifs (poids, taille, rendement, vitesse de croissance), polymorphisme enzymatique ou ADN, tests physiologiques (mesure de la respiration, du rendement d'assimilation ... ). 5. Hybridation: Des hybridations entre espèces ont été tentées, avec contrôle par marqueurs génétiques. 6. Natural hybridization between genetically differentiated populations of Crassostrea gigas and C-angulata highlighted by sequence variation in flanking regions of a microsatellite locus Auteur(s) : Huvet, Arnaud Fabioux, Caroline Mccombie, Helen Lapegue, Sylvie Boudry, Pierre Éditeur(s) : Inter-Research Résumé : The marine environment is of special interest for studying hybridization between closely related taxa because of the high dispersal potential of planktonic larvae, such as those of most bivalve species. The oysters Crassostrea angulata and C. gigas are known to be very close genetically and entirely inter-fertile under controlled conditions. However, hybridization in the wild had not been investigated, mainly due to the lack of nuclear diagnostic markers. In the present paper, we first estimated genetic differentiation between these 2 closely related taxa using 8 microsatellite markers. Interestingly, 5 markers displayed significant differences of allele size between taxa. The subsequent sequencing of alleles of one of these microsatellites showed several mutational events, which suggested null alleles and homoplasy. The presence of 1 insertion/deletion event in its 5' flanking sequence enabled us to design a new bi-allelic ('C' and 'NC') nuclear PCR-restriction fragment length polymorphism (-RFLP) marker (CG44R). This, together with a mitochondrial DNA marker, was used to analyze populations of C. angulata and C. gigas. The CG44R allele frequencies were very different between C. angulata (f[C] = 0.91) and C. gigas (f[NC] = 0.92) populations. This analysis also provided evidence for hybridization between C. angulata and C. gigas in a wild Portuguese population where the 2 taxa are in contact due to recent transportation of C. gigas stocks for aquacultural production. Our results represent the first indication of hybridization between these 2 taxa in the natural environment, and contribute to knowledge of the evolutionary history of the Crassostrea genus. Effects of short-term environmental disturbances on living benthic foraminifera during the Pacific oyster summer mortality in the Marennes-Oléron Bay (France) Auteur(s) : Bouchet, Vincent Debenay, Vincent Sauriau, Pierre-guy Radford Knoery, Joel Soletchnik, Patrick Éditeur(s) : Elsevier Résumé : Sediment cores were collected from April to August 2004 on tidal mudflats of the macrotidal Marennes-Oléron Bay (SW France), famous for the cultivation of Pacific oysters (Crassostrea gigas). The response of living (stained) benthic foraminifera to short-term biogeochemical disturbances in the sediment and overlying water, which may be involved in oyster summer mortality, was monitored. Short-term hypoxia occurred in early June, in conjunction with a sudden rise in temperature. In mid-June, the ammonia content of sediment porewater increased, leading to potentially maximal flux towards overlying waters. Foraminiferal assemblages, particularly in the topmost layer, were altered. Ammonia tepida was the most tolerant to temperature increase and hypoxic conditions whereas Brizalina variabilis and Haynesina germanica were sensitive to organic degradation and hypoxia. Cribroelphidium gunteri was the most opportunistic during recolonisation. Benthic foraminifera showed that short-term biochemical changes in the sediment are toxic and may be involved in the summer mortality of Pacific oysters. Phylogénie moléculaire des huîtres du genre Crassostrea. Approche par les techniques de restriction enzymatique et de séquençage Auteur(s) : Delandes, Hélène Résumé : En France, ainsi que dans de nombreux autres pays, l'histoire de l'ostréiculture est émaillée d'une succession de phases de développement, de surexploitation et d'apparition de maladies qui perturbent lourdement la profession. Depuis la disparition massive de l'huître portugaise, Crassostrea angulata, victime entre 1969 et 1971 d'une épidémie virale et l'effondrement de l'élevage de l'huître plate Ostrea edulis dû à deux parasitoses, la marteiliose et la bonamiose, l'ostréiculture française connaît actuellement une situation de monoculture de l'huître creuse japonaise Crassostrea gigas, suite à l'introduction de l'espèce entre 1971 et 1975 (Grizel & al. 1991). Pour tenter de remédier à cette situation de monoculture, qui reste une des inquiétudes majeures des professionnels, la région Poitou-Charentes a apporté une aide financière pour la création d'un "conservatoire de souches", implanté dans les locaux du Laboratoire Génétique, Aquaculture et Pathologie de La Tremblade. Les principaux objectifs sont d'étudier les possibilités d'acclimatation des huîtres étrangères dans les eaux françaises afin de diversifier la production ou de préparer une éventuelle substitution à C. gigas en cas d'épizootie majeure, de développer des hybrides intra et inter-spécifiques au sein du genre Crassostrea (performants en ce qui concerne la résistance aux maladies parasitaires, la croissance et la fertilité) et enfin d'effectuer une étude de la différenciation génétique (intra et inter spécifique). Phylogéographie des huîtres creuses des mangroves de l'Atlantique Sud par l'apport des marqueurs moléculaires Auteur(s) : Verdon, Beatrice Résumé : La taxonomie des huîtres est longtemps restée basée sur des critères morphologiques tel que la coquille et les organes internes ou sur la répartition géographique. Ces critères se révèlent aujourd'hui peut fiable du fait de la grande plasticité des caractères morphologiques des huîtres en fonction des conditions environnementales, et du transport d'espèce par l'ostréiculture ou accidentellement. La biologie moléculaire et en particulier les marqueurs génétiques sont des outils très utiles pour différencier les espèces. La phylogéographie des huîtres creuses de l'Atlantique Sud a ainsi été étudié grâce à différentes méthodes d'observation du polymorphisme de l'ADN. Ces techniques ont dans un premier temps été appliquées à des fragments mitochondriaux (16S) pui à des fragments nucléaires (ITS). La PCR6RFLP a été utilisée d'une part sur les fragments 16S pour déterminer les haplotypes de 5 nouvelles populations du genre Crassostrea provenant du Sénégal, du Nigeria et du Brésil. Il a été ainsi possible d'identifier une nouvelle population de Crassostrea gasar en Amérique du Sud. Nous nous sommes ensuite intéressés aux polymorphisme pouvant exister entre les différentes populations de C. gasar de part et d'autre de l'Atlantique. Pour cela des fragments nucléaires ITS, à priori plus polymorphes que les fragments mitochondriaux 16S ont été étudiés. Différentes mises au point ont été réalisées afin de révéler un éventuel polymorphisme entre les populations. Toutes les informations recueuillies tendent vers cette hypothèse sans toutefois pouvoir la confirmer. La présence d'une zone de sympatrie entre les espèces C. gasar en Amérique du Sud poste la question de son introduction, soit par l'homme soit accidentellement. Des études complèmenaires sont indispensables, par un échantillonnage plus large, pour évaluer l'étendue de la présence des espèces de part et d'autre de l'océan. Il est également nécessaire de continuer la mise au point sur les marqueurs nucléaires afin de confirmer le polymorphisme suspecté entre les populations C. gasar d'Afrique et d'Amérique. La publication récente d'un article séparant les espèces C. brasiliana et C. rhizophorae pose également la question de la position de C. brasiliana par rapport aux deux espèces que nous avons étudié. Experimental assessment of oyster transfers as a vector for macroalgal introductions Auteur(s) : Mineur, Frederic Belsher, Thomas Johnson, Mark P. Maggs, Christine A. In shellfish farming, transfer of livestock, especially oysters, is a common practice and potentially constitutes a pathway for non-indigenous introductions. Many species of seaweeds are believed to have been accidentally introduced in association with these transfers, but there is little direct evidence. We experimentally simulated the transfer of oysters from the Thau Lagoon (France). These transfers involved increasing periods of aerial emersion and additional brine and hot water treatments. The brine and hot water treatments were evaluated as a means of reducing the probability of algal introductions with oyster transfers. Shells were cultured for 40 days in experimental tanks to identify the macroalgae likely to be introduced with any oyster transfer. A total of 57 macroalgal taxa, including 16 taxa not indigenous to the Thau Lagoon, were recorded across all treatments and experiments. The abundance of some species increased in several cases following aerial emersion. Elimination treatments (immersion in brine or hot water) significantly reduced algal diversity, with hot water treatments resulting in no species or only tubular Ulva spp. present. The results support the hypothesis that oyster transfers are effective as primary and secondary vectors of macroalgal introductions. Relatively simple changes to the transfer practice (particularly hot water treatments) are suggested as a means of reducing the risk of non-indigenous algal introductions. Bases biologiques de la production de deux Mollusques d'intérêt économique: reproduction chez Megathura crenulata, reproduction et développement chez Crassostrea gigas Auteur(s) : Cannuel, Rozenn Éditeur(s) : Université de Nantes Résumé : The reproductive cycle of Megathura crenulata was documented and quantification of oocyte lipid reserves with lipo-specific stains was explored in both this species and in Crassostrea gigas. The necessity of providing food to female broodstock C. gigas in spring was re-evaluated, because subsequent larval and post-larval rearing performance was not affected by broodstock starvation during conditioning. Developmental study of the organs responsible of particle processing in C. gigas allowed the identification of critical stages related to the ontogenesis of each organ, as well as a better understanding of the evolution of these organs, highly-modified in the Ostreidae. Ciliary type identification and mucocyte mapping from post-larval to adult stage showed that the heterorhabdic specialization precedes the anatomic formation of gill plicae specific to this condition. Le cycle reproducteur de Megathura crenulata a été documenté et la quantification des réserves ovocytaires par des colorants lipospécifiques a été tentée, sans résultats concluants pour cette espèce, ni pour Crassostrea gigas. L'intérêt d'une alimentation des géniteurs femelles C. gigas au printemps est re-évalué, car la performance ultérieure des élevages larvaires et post-larvaires n'a pas été affectée par le jeûne imposé lors du conditionnement. L'étude du développement des organes impliqués dans le traitement des particules chez C. gigas a permis d'identifier des stades critiques associés à l'ontogenèse de chaque organe, ainsi qu'une meilleure compréhension de l'évolution de ces organes si particuliers chez les Ostreidae. L'examen des types ciliaires et la cartographie des mucocytes de la post-larve à l'adulte ont montré que la spécialisation hétérorhabdique précède même la formation anatomique des plis branchiaux caractéristiques de cette condition. Within the scope of the growth of oyster farming in Guiana, we researched possible parasites of these oysters that are still little studied. This work, conducted based on living and set samples, allowed us to highlight the presence of nematops spores. Convention 95 RPC R 62 - Programme Pathologie - Rapport Scientifique année 1995 Auteur(s) : Renault, Tristan Berthe, Franck Chollet, Bruno Cochennec, Nathalie Haffner, Philippe Le Deuff, Rose-marie Thuillier, Benoit Résumé : Le but de ce programme pathologie est de rechercher et d'étudier les agents pathogènes connus et inconnus chez les mollusques bivalves marins, plus particulièrement chez l'huître creuse, Crassostrea gigas, afin d'identifier d'éventuels agents causant des mortalités, de proposer des mesures prophylactiques et de définir des conditions zootechniques permettant aux professionnels de limiter l'impact des maladies sur leur cheptel. La présence de maladies, chez les mollusques bivalves marins dont l'impact socio économique peut être catastrophique pour les productions conchylicoles, et l'accroissement des échanges de produits d'aquaculture entre les pays de la CEE et avec les pays tiers nécessitent le renforcement des recherches dans les domaines de la pathologie. En particulier, du fait du mono élevage de l'huître japonaise, Crassostrea gigas, en France, il semble aujourd'hui indispensable d'effectuer des contrôles zoosanitaires réguliers chez cette espèce. Ces travaux ont d'autant plus d'importance qu'aujourd'hui, devant les pathologies rencontrées, les professionnels restent extrêmement démunis (traitements inapplicables ou inexistants). Dans ce cadre, la promptitude du diagnostic ainsi que l'acquisition de données épidémiologiques et biologiques concernant les agents pathogènes impliqués sont les meilleurs moyens d'éviter leur dissémination chez les mollusques bivalves marins. Les travaux entrepris dans ce cadre par l'URPIG, semblent d'autant plus indispensables qu'il n'existait pas jusqu'à ces dernières années de pathologies identifiées chez l'huître creuse, Crassostrea gigas, sur le littoral français. Cependant depuis 1991, sont apparus dans certaines écloseries françaises des épisodes de mortalité sur des larves de cette espèce. Il a été possible de détecter un virus apparenté à la famille des Herpesviridae par ses caractères morphologiques (NICOLAS et al., 1992) et de démontrer sa pathogénicité pour les larves (LE DEUFF et al., 1994). Par ailleurs, en 1993, 1994 et 1995, un virus de même type, a été observé en association à de fortes mortalités sur différents lots de naissain de la même espèce, originaires d'écloseries et de captage naturel. Par ailleurs, en 1993, suite à l'observation d'anomalies au niveau des branchies chez des huîtres creuses provenant de Charente Maritime et du bassin d'Arcachon associées à de médiocres performances de croissance, il a été entrepris des analyses de divers échantillons, en histologie classique et en microscopie électronique à transmission. Ces examens ont permis de révéler la présence au niveau des branchies, chez un nombre non négligeable d'animaux, d'un microorganisme de type chlamydien (RENAULT and COCHENNEC, 1995). Il a également été possible, chez cette même espèce, de détecter la présence d'un organisme de type rickettsien au niveau des branchies (RENAULT, COCHENNEC and CHOLLET, 1995).
