Description : Banana and plantain (Musa spp) are important foreign exchange earners in Guyana and the Windward Islands of the Caribbean. Black Sigatoka Disease (BSD) also referred to as black leaf streak disease is one of the most destructive diseases of banana and plantain and is present within the region. In Guyana within two years following its introduction in 2008, the industry was totally decimated, recording a 100% decline, turning the country to a net banana importer. Also, from 2009 ? 2012 the disease spread in the Windward Islands causing as much as 100% decrease in the export of banana. The disease is caused by the ascomycete fungus, Mycosphaerella fijiensis (anamorph Pseudocercospora fijiensis) and is aggressive, challenging to control and results in enormous crop damage if left uncontrolled. In an effort to develop an integrated management approach to the disease the Caribbean Agricultural Research and Development Institute (CARDI) with financial assistance from the Caribbean Development Bank (CDB) is evaluating five FHIA banana and plantain accessions and eight IITA plantain accessions for tolerance to BSD. The evaluation is presently conducted in four Caribbean countries, Dominica, Guyana, St Lucia and St Vincent and the Grenadines. In each country, four distinct agro- ecological zones were selected for the trial evaluation blocks and using a randomized block design, four blocks were established in each country. Five FHIA accessions: FHIA-01, FHIA-03, FHIA-18, FHIA-21 and FHIA-23 are being evaluated in Dominica, St Lucia and St Vincent and the Grenadines whereas, plantain accessions: PITA-17, PITA-21, PITA-22, PITA-23, PITA-24 and PITA-26 are to be evaluated in Guyana for tolerance to the disease. Cavendish are used as regional controls and as borders rows to increase disease pressure. The disease parameters evaluated are youngest leaf spotted (YLS), disease development time (DDT), leaf emission rate (LER), disease severity at 6 months, bunch emergence and harvest, and index of standing leaves. Agronomic data; days from planting to flowering, bunch weight, number of hands in bunch, average weight of hands and average number of fingers per hand and bunch, as well as organoleptic characteristics of both cooked and ripe fruits are also evaluated. The early data collection, of the FHIA only accessions, show trends in the disease tolerance and agronomic performance as well as their adaptability in the different agro-ecological zone. The trends indicate better disease tolerance of FHIA varieties with higher leaf counts before, at bunching and at harvest. The organoleptic test shows a preference to FHIA 03 and FHIA 21. La banane et le plantain (espèces de Musa) sont les acquéreurs de devises étrangères importants en Guyane et en Îles du Vent des Caraïbe. La maladie noire de Sigatoka (schéma) également désignée sous le nom de la maladie noire de filet de feuille est l'une des maladies les plus destructives de la banane et du plantain et est présente dans la région. En Guyane dans un délai de deux ans après son introduction en 2008, l'industrie a été totalement décimée, enregistrant une baisse de 100%, tournant le pays à un importateur net de banane. L'évaluation est actuellement effectuée quatre pays des Caraïbes, Dominique, Guyane, St Lucia et Saint-Vincent-et-les Grenadines. Dans chaque pays, quatre agro zones écologiques distinctes ont été sélectionnées pour les blocs d'essai d'évaluation et employant une conception de bloc randomisé, quatre blocs ont été établis dans chaque pays. Cinq adhésions de FHIA : FHIA-01, FHIA-03, FHIA-18, FHIA-21 et FHIA-23 sont évalués en Dominique, au St Lucia et au Saint-Vincent-et-les Grenadines tandis que, des adhésions de plantain : PITA-17, PITA-21, PITA-22, PITA-23, PITA-24 et PITA-26 doivent être évalués en Guyane pour la tolérance à la maladie. Cavendish sont employés en tant que contrôles régionaux et comme rangées de frontières pour augmenter la pression de la maladie. Les paramètres de la maladie évalués sont la plus jeune feuille repérée (YLS), le temps d'élaboration de la maladie (DDT), le taux d'émission de feuille (LER), la sévérité de la maladie à 6 mois, émergences et récoltes de groupe, et index des feuilles debout. Données agronomiques ; des jours de la plantation à la floraison, du poids de groupe, nombre de mains dans le poids de groupe et moyen des mains et du nombre moyen de doigts par main et groupe, aussi bien que les caractéristiques organoleptiques des fruits cuits et mûrs sont également évalués. La collecte de données tôt, des adhésions de FHIA seulement, exposition tend dans la tolérance de maladie et la représentation agronomique aussi bien que leur adaptabilité dans la zone agro-écologique différente. Les tendances indiquent qu'une meilleure tolérance de maladie des variétés de FHIA avec une plus haute feuille compte avant, à se rassembler et à la récolte. Résumé : Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in the aquaculture production of this oyster species. Therefore a selective breeding program for resistance to bonamiosis has been undertaken since 1985 bu Ifremer, leading to the production of several selected oyster families. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters (injected with the parasite) to two tested families of oysters originating from the selective breeding program. Mortalities were checked daily, and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection to B. ostreae. The first infections occurred after 4 months of challenge in the tested oysters (Family 1 and Family 2). The cumulative mortalities after 5 monts were 58% for the wil oysters, 9% for Family 1 (20-month old at the beginning of the experiment) and 20% for Family 2 (8-month, old). The parasite could be detected in 66.8% of the dying wild oysters, 67.5% of the dying oysters of Family 1, 89% of the dying oysters of Family 2 and only 11% of the surviving oysters of Family 2. The mortality was significantly higher in Family 2 thant in Family 1 (x2= 20.87, p<0.001, d.f.) as well as the level of infection by the parasite found in heart smear (x2=24.34, p<0.001, 4 d.f.). This result demonstrates that prespawning oysters as yong as 1 year-old can become infected with the parasite and die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. The most probable cause of the dscrepancy in the development of bonamiosis between the 2 tested families is a difference in genetic background.

This paper was part of the panel Panama Silver and the Making of Modern Literature, which introduced a collaborative, digital humanities course entitled “Panama Silver, Asian Gold: Migration, Money, and the Making of Modern Caribbean Literature,” and illuminated the significance of West Indians in Panama for the formation of Caribbean literature. The panelists designed and taught the course at Amherst College, the University of Florida, and the University of Miami. In the first paper in this panel, Rhonda Cobham-Sander articulated the overarching goal of the course; namely, that these two, closely related migrations were critical to the development of the Caribbean middle class, political nationalism, and national literatures. In the second paper, Leah Rosenberg outlined primary historical and literary sources on West Indians in Panama now available in the Digital Library of the Caribbean (www.dloc.com) and discussed how this supports collaboration in teaching and research. In the third paper, Margarita Vargas-Betancourt discussed the challenges of finding archival materials concerning West Indians in Panama in, U.S.-oriented archives (which we are using in the course). In the final paper, Donette Francis demonstrated the implications of positioning Panama Silver, the capital accumulated by West Indians, as critical to shaping modern Caribbean literature by reexamining Salkey’s understudied 1971 novel, The Late Emancipation of Jerry Stover.

Description : Under tropical and sub-tropical climates, a sustainable development of agriculture is challenging because of the pest pressure with favorable weather conditions. Implementation of appropriate cropping systems including growing cover crops and crop rotation plays a critical role, which can break up the food chain and life cycles of field pests. Some field trials have demonstrated that among all tested summer cover crops, sunn hemp (Crotalaria juncea L. cv. Tropic sun) can grow vigorously during the rainy summer, cover the land quickly and densely, and it can produce a large quantity of biomass in two to three months. More importantly, sunn hemp can suppress soil root-knot nematodes via releasing some nematicides through its roots and through the decomposition of plant residues. Therefore, growing cover crop â? ? sunn hemp during the crop off season and rotating with valuable vegetable crops in the growing season has become a promising cropping system under the subtropical climate. Such a result may have a potential to be implemented in Caribbean countries to improve the sustainable development of agriculture.

Résumé : From the PNCR, GAP MPs free to vote conscience on gay rights bill By Johann Earle Stabroek News June 11, 2003 The PNCR will allow its members to vote their conscience on the controversial Constitution (Amendment) Act of 2001 which seeks to prevent discrimination against persons on the basis of their sexual orientation and other grounds. A number of religious organisations are opposed to the passage of the bill because it includes sexual orientation as one of the grounds. They are concerned that it could lead to a legalisation of homosexual relations and demands for recognition of gay marriages among other things. PNCR Member of Parliament (MP) Vincent Alexander told a forum at the National Library on Saturday, that the Bill did not seek to legalise homosexuality, but to ensure that persons would not be discriminated against based on their sexual preferences. He was one of two parliamentarians who showed up - the other being PNCR member, Myrna Peterkin. The forum was organised by Students Against Sexual Orientation Discrimination (SASOD), a group comprising fifteen students from the University of Guyana which was formed about two weeks ago. Alexander expects the vote on the bill to come up before the National Assembly in a matter of months. Paul Hardy, Leader of the Guyana Action Party (GAP), told Stabroek News that his party would not be using the so-called parliamentary whip. He added that GAP took a decision that every member should vote according to his or her own conscience. “We have no right to deny the rights of others based on sexual orientation. [The Bill] will guarantee rights to the homosexual.” GAP is in Parliament as part of an alliance with the Working People’s Alliance. GAP/WPA has two MPs. The bill was met with rejection from some members of the religious community in 2001 and as a result of this, the President did not assent to it. In a statement on Monday, the Central Islamic Organisa-tion of Guyana (CIOG) said that it stood in firm opposition to the Bill. The CIOG says the general purpose of the bill may be commendable and that the organisation’s objection is not based on a willingness to promote discrimination. Rather, the CIOG said, it was based on the fact that specific legal protection on the basis of sexual orientation without definition or qualification gives tacit legitimacy to practices which are considered criminal in Islam. “It is foreseeable that such a legal nod of approval (subtle as it may be) of these practices may pave the way for greater social (or even legal) acceptability in the future which, from the perspective of all Muslims including those in Guyana, is an undesirable and sinful outcome,” the CIOG statement said. At Saturday’s event, Muslim teacher Moulana Mohamed Ali Zenjibari spoke of instances of abuse, discrimination and harsh penalties meted out to gay and lesbian persons in countries such as Iran and Saudi Arabia and noted that the Quran did not sanction such punishments for homosexual behaviour. President of the CIOG, Fazeel Ferouz told Stabroek News that a meeting to discuss a strategy with regards to the bill was planned for tomorrow with various religious groups. Stabroek News tried to get a comment on the issue from the PPP/C but to no avail. ROAR leader, Ravi Dev said that his party was now having discussions on the issue. ROAR feels that it is an important question which has to do with morality and should be discussed across the country. He added legislators had to be in tune with their constituents on the issue. SASOD is lobbying for the legislation through the sensitisation of MPs. Keimo Benjamin, a law student at UG, gave a presentation based on the jurisprudential aspects of the discussion on sexual orientation. He argued that morality should not be the only guiding principle on which to base the laws. Sexual activities between two consenting male adults in private could not be equated with a violation of a person’s rights, he said, making the point that the thrust of his presentation was not whether homosexuality was wrong, but whether it violated the rights of others. He said that the attitudes of some towards this subject were based on preconceived notions and prejudices. He cited studies to show that the suppression of certain perceived deviant sexual impulses in persons might do more harm than good. One Harvard University study of teens who said they were gay indicated that those teens were three times more likely to commit suicide. Vidyaratha Kissoon, of Help and Shelter, in his contribution on Saturday, expressed his displeasure at the low turnout at the forum and urged the parliamentarians who showed up to take the message to their colleagues. He noted that because of homophobia, the numerical minority was terrified of speaking out against instances of discrimination. Gays and lesbians in Guyana were subjected to ridicule and abuse, and walk the streets at night not looking for sex necessarily, but for the companionship of persons who empathise with them. During his presentation, Joel Simpson, another member of SASOD, outlined a number of changes made within national jurisdictions that had international implications. One such crucial change was South Africa’s 1996 adoption of a new constitution, making that country the first in the world to expressly include sexual orientation as a prohibited ground of discrimination. It was the first time a developing country had taken the lead with respect to the rights of sexual minorities. He also said that according to Douglas Sanders, a Canadian jurist, the rights of homosexual, bisexual or transsexual men and women had never been officially recognised by the United Nations, despite the fact that international laws on the issue began to emerge at the close of the Second World War. Simpson said that under Article 170 (5), as amended by Section 8 of the Constitution (Amendment) (No. 4) Act 2000, the President is required to assent to any bill which is returned by the National Assembly unaltered after a two-thirds majority within 90 days of its presentation to him. Our party will not say we have to vote for the provision (in the Sexual Orientation Bill). We will allow our members to vote as a matter of conscience,” he told participants gathered in the Conference Room of the National Library. “We feel this is a matter of conscience. You may end up with a collective position but you have to deal with us individually,” Alexander said at a poorly attended public consultation aimed at gaining support for sexual orientation to be considered a fundamental right in Guyana. Alexander, one of the main persons who sat on the Constitutional Reform Commission that addressed this controversial clause, said the legislation was not meant to legalise homosexual activities in Guyana. “It was intended to ensure that persons who have an orientation - a way of thinking - which may or may not lead to a certain activity, to not be discriminated against, in terms of their rights,” he explained. Alexander noted that very often, discussions on the sexual orientation provision in the Bill have led to seepages into other areas where debates centre on the “right to be homosexual.” “I am saying the Bill does not comment on that… However, law is peculiar, especially in a Common Law system. Once you venture out and change the law, very often you open other windows which we cannot definitively say exist or does not exist in advance,” he said. “The fear of some people is that the legislators might say one thing and the Courts will eventually say something else. While some people can’t argue against the law, per say, they will say this has opened a window of opportunity not meant to be opened. So, it is better to stay without a window than open it and then have a possibility of something you didn’t intend to happen - happening sometime in the future.” Alexander added: “I want to say I’ve found this activity to have been rich from the perspective of the amount of research which was done by student (speakers). Without any comment as to whether I agree with the arguments, I would wish that much more research on other issues be done by students, and that at the University (of Guyana) students would find it convenient to have forums on other issues, with the same depth of research for their own intellectual development.” Alexander and other Opposition M.P, Lurlene Nestor were the only three Parliamentarians in attendance at the session organised by the recently formed `Students Against Sexual Orientation Discrimination’ (SASOD), a group that has a membership of 15 mostly University of Guyana (UG) students from various faculties. SASOD was established two weeks ago. With the exception of members of the press, the consultation only managed to attract 11 persons, a handful of who were SASOD members. While congratulating the students for an “insightful” presentation, Nestor pointed out that whether or not she chooses to agree with their position is “another issue” and she is entitled to her “own view”. “The issue of sexual orientation is very `toucheous’ …while we agree with the human rights provisions and all that as a society, we must also revert to our own society. Some of the very critical questions that I would want to ask is whether or not at the society (level) we are ready for that kind of thing,” she remarked. “We cannot, at (any) time, ignore the religious groups in our society. If we look at statistics going back to 1992 from a survey done by the Bureau of Statistics, we would see that a small section of the Guyanese population might be considered as people who do not subscribe to a religious view. While the laws are not necessarily based on moral values, we must acknowledge the fact that we might want to revert to many of the cases that (concern) laws that protect public morality.” Nestor told the speakers at the consultation these were some of the issues that they needed to deal with. She highlighted, too, that what must be examined is the effects of same-sex marriages on society and how this issue must be tackled. “These are some of the things we should consider and I don’t think you dealt with that in the presentations,” Nestor stated, adding that the issues must be addressed “frontally.” “Do we think that with the coming to being of this Bill that there might be quite a number of challenges to the Constitution in relation to the same issue of a man marrying a man? What do we do at the society (level)? Do we recognise that?” were the questions directed at the five speakers at the session. Nestor continued: “There is some argument that says, `Oh the Bill does not promote homosexuality or does not encourage a man to marry (another) man, but if you look at Section (15) that talks about `non-discrimination’ then how can we not, with the passage of this Bill, allow a man not to marry (another) man.” According to Nestor, matters of this nature constitute some of the “inconsistencies” of the Bill. On the argument raised by presenters at the session on who determines what is morality, Nestor said in the concept of democracy it is the people who are the determinants based on a “line of thinking.” She also rebutted on grounds that put the spotlight on teachers who may have been caught “interfering” with their young students and who may subsequently be dismissed from their jobs. “Could you imagine such a person interfering with a boy below age 10 - and by virtue of the fact that the Bill is there, the judges (in the case) will have to use their discretion in terms of what happens. We will have more Constitutional changes and problems (arising with the passage of the Bill).” SASOD member and law student, Joel Simpson, in reply, said he doesn’t think any homosexual in Guyana wants to “run into a church and ask that people marry them or anything of that sort.” At one point likening the church to a “club”, Simpson claimed the church has the right to exclude whomever it wants. He further stated that in accordance with the Constitution, people of the same sex do not currently have the right to marry, and will also not be able to do such an act with the passage of the Bill. However, Simpson said it is possible that the law, with the passage of the Bill, would have to recognise same-sex domestic partnerships in relation to employment benefits, sharing of properties, etc. Simpson said he believes there should be a realm of “public” and “private” morality between consenting adults, and implied that the Bill did not fully give “rights” to homosexuals. But, Nestor interjected: “I am informing you further…that the Sexual Orientation provision has, in fact, in some way recognised the rights of homosexuals and we must accept that.” Meanwhile, there was no vocal Christian representative(s) at the consultation and apart from the two Opposition representatives none other participant gave comments or directed questions at the presenters. Other speakers in support of the sexual orientation clause were: Moulana Mohammed Ali Zenjiban, Assistant Director of the International Islamic College; Denuka Radzik from Red Thread, Keimo Benjamin, UG law student and Vidyartha Kissoon from Help & Shelter. The Sunday Chronicle has been reliably informed that the Georgetown Ministers Fellowship, representing groups of Christian leaders staunchly against sexual orientation as a right in Guyana, have recently prepared a detailed 16-page document outlining issues arising from research to further boost their argument against the inclusion of the clause. While the global response to HIV and AIDS has realized several successes in achieving universal access to prevention, care and treatment to date, challenges persist in ensuring equal access to these services for most at risk populations. The epidemiology of HIV reflects a stark disparity in access to prevention and treatment services for neglected most-at risk populations, such as men who have sex with men (MSM). World-wide, MSM often experience higher rates of HIV relative to the total population. The reasons for this dynamic are multi-faceted and include high risk behaviors, and cultural as well as structural barriers. For MSM in developing countries, for example, basic services for prevention and treatment of HIV infection have yet to reach the large majority of men. Homophobia and discrimination limit access of MSM to prevention services and markedly increase vulnerability, as do criminalization of same-sex behavior. Decriminalization of same-sex behavior is a structural intervention for prevention of HIV infection and has recently been embraced by a nonbinding statement from the United Nations (Beyrer C,Clin Infect Dis. 2010 May 15;50 Suppl 3:S108-13) In the Caribbean, all of the above mentioned barriers exist. In fact, recent evidence demonstrates that there is a correlation between the decriminalization of homosexuality and lower rates of HIV (UNAIDS). This correlation is attributed to improved access to services. Therefore, The United Nations Development Program (UNDP) has recently launched an initiative to address the current situation regarding men who have sex with men in Guyana where the HIV prevalence in this population is 19.4% in contrast to 1.8% in the total population. This rapid assessment has been commissioned to supplement two previous studies that were conducted in Guyana in informing the design of upcoming activities to support local capacity building to enhance access to services and respect for the human rights protections for MSM. Unfortunately, in Guyana, like much of the Caribbean, MSM tend to be a hard to reach population, perhaps due to elevated levels of stigma and discrimination and difficulty in accessing men who might participate in high risk same sex behaviors yet do not self-identify as an MSM. As a result, there is a relative paucity of data concerning the knowledge, attitudes and behaviors of this total population in Guyana. (i.e. those who self –identify and those who do not) The baseline report is organized as follows: the methodology is explained followed by the study limitations, the review (literature and desk). The report then proceeds with describing the reality of work and working with sexual and gender minorities in Guyana, the status quo and entry points. The main findings of the capacity assessment and focus groups are then discussed. Issues relating to the uniformed forces and health services providers are then presented followed by discussions and conclusions.

STUDY QUESTION: Do freezing and in vitro culture procedures enhance the expression of proteins involved in apoptotic or autophagic pathways in murine pre-pubertal testicular tissue? SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. Résumé : Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection by B. ostreae. Mortality started after 4 months of cohabitation in the tested oysters. The cumulative mortalities after 6 months were 58% for the wild oysters, 9% for Family 1 (20-month old) and 20% for Family 2 (8-month old). In the dying oysters, the parasite could be detected in 67% of the wild oysters, 68% of Family 1 and 89% of Family 2. It was detected in only 11% of the surviving oysters of Family 2. The mortality and the level of infection by the parasite were significantly higher in Family 2 than in Family 1. Our results demonstrate that prespawning oysters as young as 1 year-old can become infected with the parasite and, most importantly, can die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. Additionally, no clear relationship between shell length and level of infection was observed. We also review the different methods for infection of the European flat oyster O. edulis with B. ostreae under experimental conditions and their main results.
La bonamiose est une maladie parasitaire (agent causal : Bonamia ostreae) affectant l'huître plate européenne Ostrea edulis, responsable d'un déclin drastique de sa production aquacole. Des programmes de sélection pour la résistance à la bonamiose ont été entrepris pour contrer cette maladie. Dans cette étude, une expérience de 6 mois d'infection par cohabitation a été réalisée de manière à transmettre la maladie à partir d'huîtres sauvages injectées avec le parasite vers deux familles testées d'huîtres (âgées de 20 et 8 mois en début d'expérience, avec des origines génétiques différentes) issues du programme de sélection développé par IFREMER en France. Les mortalités ont été vérifiées quotidiennement et des frottis de coeur ventriculaire réalisés sur les huîtres mortes ou moribondes pour détecter le niveau d'infection par B. ostreae. La mortalité a commencé chez les huîtres testées après 4 mois de cohabitation. Les mortalités cumulées après 6 mois étaient de 58 % chez les huîtres sauvages, 9 % chez la Famille 1 (âgées de 20 mois) et 20 % chez la Famille 2 (âgées de 8 mois). Chez les huîtres mourantes, le parasite a pu être détecté chez 67 % des huîtres sauvages, 68 % de la Famille 1 et 89 % de la Famille 2. Il n'a pu être détecté que chez 11 % des huîtres survivantes de la Famille 2. La mortalité et le niveau d'infection par le parasite étaient significativement plus élevés chez la Famille 2 que chez la Famille 1. Nos résultats démontrent que des huîtres âgées de un an peuvent devenir infectées par le parasite et surtout, peuvent mourir de bonamiose. Ce résultat contraste avec l'âge critique de développement de la maladie communément accepté de 2 ans. De plus, aucune relation claire entre la longueur de la coquille et le niveau d'infection n'a été observée. Nous faisons également la revue des différentes méthodes d'infection de l'huître plate européenne O. edulis avec B. ostreae en conditions expérimentales et leurs principaux résultats.

Résumé : High P-wave velocities (7.1-7.8 km/s) lower crustal bodies (LCBs) imaged along volcanic margins are commonly interpreted as plume and break-up-related thick mafic underplating. This interpretation is partly challenged in this paper based on new seismic observations and modelling of the outer Voring Basin (Norway). An exceptional strong amplitude reflection, the T Reflection, is particularly well defined below the North Gjallar Ridge (NGR) between 7 and 8 s TWT. The T Reflection is located near the volcanic lava flows emplaced during the NE Atlantic breakup (similar to 55-54 Ma ago) and coincides with the top of the LCB, forming a mid-crustal dome. Based on structural and temporal relationships, we show that the dome clearly influences the structural development of the NGR and predates the continental breakup at least by 10-15 Ma. Using a thermo-kinematical model, we tried also to investigate and quantify the relationships between the extension, LCB and the magmatic production. Modelling suggests that significant Paleocene-Early Eocene magmatism can be produced without any temperature anomaly in the mantle if differential stretching occurs during the break-up initiation. The conclusion of 2D thermo-kinematical parametric analysis is that the magmatic model predicts, either little extension (beta < 2) with no melting or high extension (beta > 5) with significant melting along the outer Voring Basin. We suggest that the continental part of the LCB could not necessarily be breakup-related and so magmatic, as has often been stated previously. It is concluded here that the continental part of the LCB observed beneath the outer Voring Basin may be partly (or fully) attributed to inherited, high-pressure granulite/eclogite lower crustal rocks. The real amount of mafic material emplaced along the outer Voring Basin could be 20-40% less than thought.

Résumé : The transforming growth factor (TGF)-beta superfamily is a group of important growth factors involved in multiple processes such as differentiation, cell proliferation, apoptosis and cellular growth. In the Pacific oyster Crassostrea gigas, the oyster gonadal (og) TGF-beta gene was recently characterized through genome-wide expression profiling of oyster lines selected to be resistant or susceptible to summer mortality. Og TGF-beta appeared specifically expressed in the gonad to reach a maximum when gonads are fully mature, which singularly contrasts with the pleiotropic roles commonly ascribed to most TGF-beta family members. The function of og TGF-beta protein in oysters is unknown, and defining its role remains challenging. In this study, we develop a rapid bacterial production system to obtain recombinant og TGF-beta protein, and we demonstrate that og TGF-beta is processed by furin to a mature form of the protein. This mature form can be detected in vivo in the gonad. Functional inhibition of mature og TGF-beta in the gonad was conducted by inactivation of the protein using injection of antibodies. We show that inhibition of og TGF-beta function tends to reduce gonadic area. We conclude that mature og TGF-beta probably functions as an activator of germ cells development in oyster.

Résumé : Understanding the genetic mechanisms of speciation and basis of species differences is among the most important challenges in evolutionary biology. Two questions of particular interest are what roles divergent selection and chromosomal differentiation play in these processes. A number of recently proposed theories argue that chromosomal rearrangements can facilitate the development and maintenance of reproductive isolation and species differences by suppressing recombination within rearranged regions. Reduced recombination permits the accumulation of alleles contributing to isolation and adaptive differentiation and protects existing differences from the homogenizing effects of introgression between incipient species. Here, we examine patterns of genetic diversity and divergence in rearranged versus collinear regions in two widespread, extensively hybridizing sunflower species, Helianthus annuus and Helianthus petiolaris, using sequence data from 77 loci distributed throughout the genomes of the two species. We find weak evidence for increased genetic divergence near chromosomal break points but not within rearranged regions overall. We find no evidence for increased rates of adaptive divergence on rearranged chromosomes; in fact, collinear chromosomes show a far greater excess of fixed amino acid differences between the two species. A comparison with a third sunflower species indicates that much of the nonsynonymous divergence between H. annuus and H. petiolaris probably occurred during or soon after their formation. Our results suggest a limited role for chromosomal rearrangements in genetic divergence, but they do document substantial adaptive divergence and provide further evidence of how species integrity and genetic identity can be maintained at many loci in the face of extensive hybridization and gene flow.

Résumé : Ecosystem-based management (EBM) requires more extensive information than single-species management. Active underwater acoustic methods provide a means of collecting a wealth of ecosystem information with high space-time resolution. Worldwide fisheries institutes and agencies are carrying out regular acoustic surveys covering many marine shelf ecosystems, but these data are underutilized. In addition, more and more acoustic data collected by vessels of opportunity are becoming available. To encourage their use for EBM, we provide a brief introduction to acoustic and complementary data collection methods in the water column, and review current and potential contributions to monitoring population abundance and biomass, spatial distributions, and predator-prey relationships. Further development of acoustics-derived indicators is needed. We review and propose indicators for assessing and monitoring zooplankton, population dynamics of fish and other nekton, and changes in diversity and food-web functioning. Acoustic methods have the potential to make a strong contribution to EBM. Evaluation of new indicators and suitable reference points in different ecosystems are the current challenges.

Résumé : Terrestrial and marine biodiversity provides the basis for both ecosystems functioning and numerous commodities or services that underpin human well-being. From several decades, alarming trends have been reported worldwide for both biodiversity and ecosystem services. Therefore the sustainable management of biodiversity requires a double viewpoint balancing ecological conservation with the welfare of human societies. Understanding the underlying trade-offs, synergies and interactions imposes the development of interdisciplinary researches and methods. In that respect, bio-economic or ecological economic modeling is likely to play a major role. The present paper intends to elicit the key features, strengths and challenges of bio-economic approaches especially in mathematical and computational terms. It first recall the main bio-economic methods, models and decisional instruments used in these types of analyses. Then the paper shows to what extent bio-economic sustainability lies between equilibrium, viability and optimality mathematical frameworks. It ends up by identifying new major challenges among which the operationalization of ecosystem based management, the precautionary principle and the implementation of governance are especially important. Coral reef fisheries support the development of local and national economies and are the basis of important cultural practices and worldviews. Transitioning economies, human development, and environmental stress can harm this livelihood. Here we focus on a transitioning social-ecological system as a case study (Moorea, French Polynesia). We review fishing practices and three decades of effort and landing estimates with the broader goal of informing management. Fishery activities in Moorea are quite challenging to quantify because of the diversity of gears used, the lack of centralized access points or markets, the high participation rates of the population in the fishery, and the overlapping cultural and economic motivations to catch fish. Compounding this challenging diversity, we lack a basic understanding of the complex interplay between the cultural, subsistence, and commercial use of Moorea's reefs. In Moorea, we found an order of magnitude gap between estimates of fishery yield produced by catch monitoring methods (2 t km −2 ∼ year −1) and estimates produced using consumption or participatory socioeconomic consumer surveys (∼24 t km −2 year −1). Several lines of evidence suggest reef resources may be overexploited and stakeholders have a diversity of opinions as to whether trends in the stocks are a cause for concern. The reefs, however, remain ecologically resilient. The relative health of the reef is striking given the socioeconomic context. Moorea has a relatively high population density, a modern economic system linked into global flows of trade and travel, and the fishery has little remaining traditional or customary management. Other islands in the Pacific in similar contexts in Polynesia such as Hawaii, that continue to develop economically, may have small-scale fisheries that increasingly resemble Moorea. Therefore, understanding Moorea's reef fisheries may provide insight into their future.

Advances in technology and scientific development in forensic handwriting examination (FHE) require a review in response to the challenges published in the NAS Report. This survey summarizes the advances made in theoretical and methodological approaches to handwriting examination including a review of research on the proficiency of FHEs. Technology has impacted FHE including analysis of e-signatures and use of technology for signature and handwriting authentication. A review of legal cases in the U.S. confirms how research and technological advances have met legal challenges and impacted decisions in the courtroom.

Accumulating evidence suggests that developmental exposure to environmental chemicals may modify the course of brain development, ultimately leading to neuropsychiatric / neurodegenerative disorders later in life. In the present study, we assessed the impact of one of the most frequently used pesticides in both residential and agricultural applications - the synthetic pyrethroid cypermethrin (CYP) - on developmental neurotoxicity (DNT). Female mice were perinatally exposed to low doses of CYP (5 and 20 mg/kg body weight) from gestation to postnatal day 15. Behavioral analyses were performed during the offspring's early life and during adulthood. Postnatal analyses revealed that perinatal exposure to CYP disturbed motor development without modifying sensory and communicative skills. We found that later in life, CYP-exposed offspring expressed maladaptive behaviors in response to highly challenging tasks and abnormal sociability. Transcriptomic analyses performed in the offspring's brain at the end of the exposure, highlighted mitochondrial dysfunction as a relevant pathomechanism underlying CYP-induced DNT. Interestingly, several genes involved in proteostasis maintenance were also shown to be dysregulated suggesting that alterations in biogenesis, folding, trafficking and degradation of proteins may significantly contribute to CYP-related DNT. From a regulatory perspective, this study highlights that behavioral and transcriptomic analyses are complementary tools providing useful direction for better DNT characterization, and as such, should be used together more systematically.

Résumé : The tremendous development of penaeid shrimp culture across the world over the past twenty years has led to international trade in eggs, larvae and spawners of the best shrimp species for aquaculture. Trade has involved, in particular, the following species: - Penaeus japonicus from Japan to Europe, the Pacific Islands and South America - P. monodon from South-East Asia to almost all tropical countries - P. vannamei and P. stylirostris from countries along the Pacific coast of South and Central America to the United States of America, the islands of the South Pacific, countries along the Atlantic coast of the Americas and certain countries in Africa. In the 1980s, research conducted by American and French teams enabled the breeding of stocks of spawners in captivity. This resulted in the establishment of lines of captive broodstock outside natural areas of distribution, and the development of new breeding farms in previously unproductive regions. Trade related to movements of these species is facilitated by the absence of legislation in most countries; even when such legislation does exist, it is not always applied These practices have led to the rapid development of world shrimp production, but have also contributed to the dissemination of pathogens. In recent years, the occurrence of serious epizootics - occasioning heavy losses world-wide - has led to a more cautious approach involving trade of post-larvae obtained from captive broodstock in which thorough control measures have been implemented for known diseases. Trade in wild shrimp will probably be abandoned progressively, to avoid the risk of introducing new pathogens. The main challenges for research in the near future are the development of diagnostic tools, to enable continuous control of captive broodstock, and the selection of strains which are resistant to the principal pathogens affecting these species.

Résumé : The object of this article is to offer a synthesis of the knowledge clarifying the originality and the diversity of the submarine geological formations of the New Caledonian EEZ. A reminder is given onto the geologic history of the region, based upon the dislocation of the Gondwana Continent, the opening of the Tasman Sea, the presence of submarine volcanism, the setting up of heavy peridotitic rocks, the building up of the Fairway and Lord Howe Ridges and of sedimentary basins, the formation of volcanic domes and sea-mounts or guyots, the subduction along the Vanuatu Trench, the uplift of the Loyalty Islands, the development of carbonated ptatforms around the islands etc.... Of this complex history ensues an exceptional submarine geo-diversity. After this presentation the article clarifies the methods and the tools used in marine geosciences to map, explore, observe, analyze, sample, that they are embarked or are autonomous, teleoperated or inhabited systems. A chapter describes the programs which were set up since about forty years and the results obtained as well as the scientific teams concerned with. In conclusion, based on the unpublished character of this geo-diversity, the article introduces the second text to come in a next issue of theTai Kona magazine, text which will focus on the scientific, economic (mineral resources), environmental (deep biodiversity) and risks (seismicity) challenges that are worn by one of the most original submarine geologic context in the world.
L’objet de cet article est de proposer une synthèse des connaissances explicitant l’originalité et la diversité des formations géologiques sous-marines de la ZEE calédonienne. Un rappel est donné sur l’histoire géologique de la région basée sur la dislocation du continent Gondwana, l’ouverture de la Mer de Tasman, la présence de volcanisme sous-marin, la mise en place des péridotites, la formation des rides de Fairway et Lord Howe et de bassins sédimentaires, la formation de dômes et monts sous-marins ou guyots, la subduction le long de la fosse de Vanuatu, la surrection des Loyautés, la mise en place de plateformes carbonatées etc... De cette histoire complexe découle une géo-diversité sous-marine exceptionnelle. Après cette présentation l’article explicite les méthodes et les outils utilisés en géosciences marines pour cartographier, explorer, observer, analyser, prélever, qu’ils soient embarqués ou soient des systèmes autonomes, télé-opérés ou habités. Un chapitre décrit les programmes qui ont été mis en place depuis une quarantaine d’années et les résultats obtenus ainsi que les équipes scientifiques en place. Cumulative human impacts have led to the degradation of marine ecosystems and thedecline of biodiversity in the European and contiguous seas. Effective conservationmeasures are urgently needed to reverse these trends. Conservation must entail societalchoices, underpinned by human values and worldviews that differ between the countriesbordering these seas. Social, economic and political heterogeneity adds to the challenge ofbalancing conservation with sustainable use of the seas. Comprehensive macro-regionalcoordination is needed to ensure effective conservation of marine ecosystems andbiodiversity of this region. Under the European Union Horizon 2020 framework programme,the MarCons COST action aims to promote collaborative research to support marinemanagement, conservation planning and policy development. This will be achieved bydeveloping novel methods and tools to close knowledge gaps and advance marineconservation science. This action will provide support for the development of macro-regional and national policies through six key actions: to develop tools to analysecumulative human impacts; to identify critical scientific and technical gaps in conservationefforts; to improve the resilience of the marine environment to global change and biologicalinvasions; to develop frameworks for integrated conservation planning across terrestrial,freshwater, and marine environments; to coordinate marine conservation policy acrossnational boundaries; and to identify effective governance approaches for marine protectedarea management. Achieving the objectives of these actions will facilitate the integration ofmarine conservation policy into macro-regional maritime spatial planning agendas for theEuropean and contiguous seas, thereby offsetting the loss of biodiversity and ecosystemservices in this region.
