Chlordecone is a persistent organochlorine pesticide that was used in the French West Indies until the early 1990s for banana weevil borer control. Human exposure to this chemical in this area still occurs nowadays due to consumption of contaminated food. Although adverse effects on neurodevelopment, including tremors and memory deficits, have been documented in experimental studies conducted with rodents exposed during the gestational and neonatal periods, no study has been conducted yet to determine if chlordecone alters child development. This study examines the relation of gestational and postnatal exposure to chlordecone to infant development at 18 months of age in a birth-cohort of Guadeloupean children. In a prospective longitudinal study conducted in Guadeloupe (Timoun mother-child cohort study), exposure to chlordecone was measured at birth from an umbilical cord blood sample (n=141) and from a breast milk sample collected at 3 months postpartum (n=75). Toddlers were assessed using an adapted version of the Ages and Stages Questionnaire. Higher chlordecone concentrations in cord blood were associated with poorer fine motor scores. When analyses were conducted separately for boys and girls, this effect was only observed among boys. These results suggest that prenatal exposure to chlordecone is associated with specific impairments in fine motor function in boys, and add to the growing evidence that exposure to organochlorine pesticides early in life impairs child development.

Résumé : During the present study, we aimed at providing a first look at the elemental composition of the early stages of cephalopods as an approach to their elemental requirements in culture. Essential and non-essential elemental profiles of the European cuttlefish Sepia officinalis, the European squid Loligo vulgaris and the common octopus Octopus vulgaris laboratory hatchlings and wild juveniles were analysed. In addition, for O. vulgaris we determined elemental profiles of mature ovary, eggs in different stages of development and followed possible effects of four dietary treatments during paralarval rearing, also analyzing elemental content of the live preys Artemia nauplii and Maja brachydactyla hatchling zoeae. Content was determined for essential (As, Ca, Cr, Co, Cu, Fe, K, Mg, Mn, Na, Ni, P, Rb, S, Sr, Zn) and non-essemial (Ag, Al, Ba, Cd, Hg, Pb) elements. The content in non-essential elements found in hatchlings and juveniles of the three species analyzed here seems to be far lower in comparison with subadult and adult stages of coastal cephalopods. In the octopus eggs, the non-essential element concentrations remained globally low compared to hatchlings and juveniles indicating the absorption of these elements along the ontogenefic development. The elemental composition of the octopus ovary and of the eggs, hatchlings and juveniles of the three cephalopod species analyzed here showed a high content in S. As expected, the calcified internal shell of the cuttlefish, rich in Ca and Sr, originates the main difference between species. It is remarkable the richness in Cu of hatchling octopus, that may indicate a particular nutritional requirement for this element during the planktonic life. The reared octopus paralarvae feed on Artemia nauplii, a prey with relatively low Cu content, showed nearly half Cu content that the "natural" profile of octopus hatchlings or wild juveniles. This suggests a dietary effect and/or an indication of the poor physiological stage of the Artemia-fed paralarvae. At the present, the percentage of essential element absorption by food or seawater is unknown for cephalopods and should be determined in the future to understand their feeding requirements in culture. Sperm and embryos cryopreservation is a commonly applied technique for several years. Recently authorized in France, vitrification tends to replace gradually the conventional technique of slow freezing, so upsetting the practices in the management of patients. It allows from now on the cryopreservation of oocytes and opens new perspectives in egg donation either still in fertility preservation. This review thus attempted to examine the contribution of vitrification in the freezing of oocytes and human embryos at various stages of development. If obviously vitrification appears as the current method of choice for the cryopreservation of oocytes as well as blastocysts, the results are less cut as regards embryos to early stages. No increase in adverse obstetric and perinatal outcomes in children conceived from vitrified oocytes or embryos is noted in the literature

Objectives To define the different stages of spontaneous labour. To determine the indications, modalities of use and the effects of administering synthetic oxytocin. And to describe undesirable maternal and perinatal outcomes associated with the use of synthetic oxytocin. Method A systematic review was carried out by searching Medline database and websites of obstetrics learned societies until March 2016. Results The 1st stage of labor is divided in a latence phase and an active phase, which switch at 5 cm of cervical dilatation. Rate of cervical dilatation is considered as abnormal below 1 cm per 4 hour during the first part of the active phase, and blow 1 cm per 2 hours above 7 cm of dilatation. During the latent phase of the first stage of labor, i.e. before 5 cm of cervical dilatation, it is recommended that an amniotomy not be performed routinely and not to use oxytocin systematically. It is not recommended to expect the active phase of labor to start the epidural analgesia if patient requires it. If early epidural analgesia was performed, the administration of oxytocin must not be systematic. If dystocia during the active phase, an amniotomy is recommended in first-line treatment. In the absence of an improvement within an hour, oxytocin should be administrating. However, in the case of an extension of the second stage beyond 2 hours, it is recommended to administer oxytocin to correct a lack of progress of the presentation. If dynamic dystocia, it is recommended to start initial doses of oxytocin at 2 mUI/min, to respect at least 30 min intervals between increases in oxytocin doses delivered, and to increase oxytocin doses by 2 mUI/min intervals without surpassing a maximum IV flow rate of 20 mUI/min. The reported maternal adverse effects concern uterine hyper-stimulation, uterine rupture and postpartum haemorrhage, and those of neonatal adverse effects concern foetal heart rate anomalies associated with uterine hyper-stimulation, neonatal morbidity and mortality, neonatal jaundice, weak suck/poor breastfeeding latch and autism. Conclusion The widespread use of oxytocin during spontaneous labour must not be considered as simply another inoffensive prescription without any possible deleterious consequences for mother or foetus. Conditions for administering the oxytocin must therefore respect medical protocols. Indications and patient consent have to be report in the medical file. © 2016 Elsevier Masson SAS

Résumé : Structural, metamorphic, and geochronologic work shows that the Ampelos/Dilek nappe of the Cycladic blueschist unit in the eastern Aegean constitutes a wedge of high-pressure rocks extruded during early stages of orogeny. The extrusion wedge formed during the incipient collision of the Anatolian microcontinent with Eurasia when subduction and deep underthrusting ceased and the Ampelos/Dilek nappe was thrust southward over the greenschist-facies Menderes nappes along its lower tectonic contact, the Cycladic-Menderes thrust, effectively cutting out a ∼30- to 40-km-thick section of crust. The upper contact of the Ampelos/Dilek extrusion wedge is the top-to-the-NE Selçuk normal shear zone, along which the Ampelos/Dilek nappe was exhumed by ∼30–40 km. Detailed Rb-Sr and 40Ar/39Ar dating of mylonites demonstrates that both shear zones operated between 42 and 32 Ma. There is no evidence for episodic motion during the ∼10 Myr life span of the shear zones, suggesting that both shear zones operated in a steady, nonepisodic fashion. Our data provide supporting evidence that simultaneous thrust-type and normal sense shearing can accomplish the early exhumation of deep-seated rocks

Cytochrome P450 aromatase (P450arom; aromatase) is a microsomal enzyme involved in the production of endogeneous sex steroids by converting testosterone into oestradiol. Aromatase is the product of the cyp19a1 gene and plays a crucial role in the sexual differentiation of the brain and in the regulation of reproductive functions. In the brain of mammals and birds, expression of cyp19a1 has been demonstrated in neuronal populations of the telencephalon and diencephalon. By contrast, a wealth of evidence established that, in teleost fishes, aromatase expression in the brain is restricted to radial glial cells. The present study investigated the precise neuroanatomical distribution of cyp19a1 mRNA during brain development in Xenopus laevis (late embryonic to juvenile stages). For this purpose, we used in situ hybridisation alone or combined with the detection of a proliferative (proliferating cell nuclear antigen), glial (brain lipid binding protein, Vimentin) or neuronal (acetylated tubulin; HuC/D; NeuroβTubulin) markers. We provide evidence that cyp19a1 expression in the brain is initiated from the very early larval stage and remains strongly detected until the juvenile and adult stages. At all stages analysed, we found the highest expression of cyp19a1 in the preoptic area and the hypothalamus compared to the rest of the brain. In these two brain regions, cyp19a1-positive cells were never detected in the ventricular layers. Indeed, no co-labelling could be observed with radial glial (brain lipid binding protein, Vimentin) or dividing progenitors (proliferating cell nuclear antigen) markers. By contrast, cyp19a1-positive cells perfectly matched with the distribution of post-mitotic neurones as shown by the use of specific markers (HuC/D, acetylated tubulin and NeuroβTubulin). These data suggest that, similar to that found in other tetrapods, aromatase in the brain of amphibians is found in post-mitotic neurones and not in radial glia as reported in teleosts.

The mucosal events of HIV transmission have been extensively studied, but the role of infected cells present in the genital and rectal secretions, and in the semen, in particular, remains a matter of debate. As a prerequisite to a thorough in vivo investigation of the early transmission events through infected cells, we characterized in detail by multi-parameter flow cytometry the changes in macaque seminal leukocytes during SIVmac251 infection, focusing on T cells, macrophages and dendritic cells. Using immunocytofluorescence targeting SIV proteins and real-time quantitative PCR targeting SIV DNA, we investigated the nature of the infected cells on sorted semen leukocytes from macaques at different stages of infection. Finally, we cocultured semen CD4(+) T cells and macrophages with a cell line permissive to SIV infection to assess their infectivity in vitro. We found that primary infection induced strong local inflammation, which was associated with an increase in the number of leukocytes in semen, both factors having the potential to favor cell-associated virus transmission. Semen CD4(+) T cells and macrophages were productively infected at all stages of infection and were infectious in vitro. Lymphocytes had a mucosal phenotype and expressed activation (CD69 & HLA-DR) and migration (CCR5, CXCR4, LFA-1) markers. CD69 expression was increased in semen T cells by SIV infection, at all stages of infection. Macrophages predominated at all stages and expressed CD4, CCR5, MAC-1 and LFA-1. Altogether, we demonstrated that semen contains the two major SIV-target cells (CD4+ T cells and macrophages). Both cell types can be productively infected at all stages of SIV infection and are endowed with markers that may facilitate transmission of infection during sexual exposure.
