Résumé : The advantages of FLOC over clear water (CW) in rearing juvenile L. vannamei and its effects on Vibrio communities were evaluated. Survival rate in FLOC and clear water were recorded and a probiotic was tested under both conditions. Daily growth rate (DGR) was higher in FLOC (p < 0.05) than in CW. Survival in each system increased significantly when a probiotic was included in the diet (p < 0.05). The Vibrionaceae community from the hepatopancreas (HP) and the culture medium did not differ between the two culture media. Nevertheless, a novel group of Vibrio strains was found to be unique to FLOC. No high level of lesions was observed in shrimp tissues from the FLOC + probiotic treatment; it suggests that the probiotic contributed to homeostasis and prevented outbreak of opportunistic pathogenic species.

Résumé : To improve the knowledge of the survival of enteric viruses in a marine environment, the influence of physico-chemical parameters (temperature, UV, salinity) on the survival of infectious poliovirus 1 and hepatitis A Virus (HAV) in seawater was first studied, the influence of suspended solids (SS) on poliovirus adsorption and survival in seawater was then evaluated and the detection of rotavirus genome in environmental samples (shellfish, river water, treated wastewater) was finally investigated. The results show that temperature has a major impact on virus survival in seawater as the time necessary to inactivate 90 % of the virus (T-90) is 671 days at 4 degrees C and only 25 days at 25 degrees C. Ultraviolet light (42 mW s cm(-2)) rapidly inactivates viruses but HAV is more resistant (T-90 = 2.6 min) than the poliovirus 1 (T-90 = 1.3 min). By contrast, seawater salinity has no effect on virus survival. In presence of SS, 90 % to 99.9 % of the viruses were adsorbed. This adsorption does not provide any protection for viruses with low SS concentrations (3 and 15 mg L-1) but a slight increase in virus survival was observed with a high SS concentration (500 mg L-1). Finally environmental sample analysis indicated that 20 % shellfish, about 40 % river water and 40 % treated wastewater tested positive for the rotavirus genome.
Ce travail a pour but de mieux comprendre le devenir des virus en milieu marin. Dans ce cadre, l'influence de certains paramètres physicochimiques (température, UV et salinité) sur le pouvoir infectieux du poliovirus 1 et du virus de l'hépatite A (VHA) en eau de mer a été évalué, puis l'adsorption du poliovirus 1 sur les matières en suspension (MES) a été étudié, ainsi que les conséquences sur le pouvoir infectieux du virus et enfin le génome des rotavirus a été recherché en milieu naturel (eaux et coquillages). Les résultats montrent que la température est un paramètre important pour la survie des virus puisque le temps nécessaire à inactiver 90 % des virus (T90) en eau de mer est de 671 j à 4 °C alors qu'il est seulement de 25 j à 25 °C. Le rayonnement ultraviolet (42 mW s cm−2) testé en pilote expérimental, permet d'obtenir une inactivation rapide du poliovirus 1 (T90 = 1,3 min). Le VHA est plus résistant avec un T90 = 2,6 min. En revanche la salinité de l'eau de mer n'a pas d'influence sur la survie des virus. En ce qui concerne l'influence des MES, les résultats montrent qu'en présence de 3, 15 et 500 mg L−1 de Na-montmorillonite la majorité des virus est adsorbée (90 à 99,9 %). Cet état adsorbé ne procure aucune protection aux virus pour les faibles concentrations de MES (3 et 15 mg L−1) alors qu'une augmentation significative de survie est observée pour les fortes concentrations (500 mg L−1). Enfin, l'analyse d'échantillon de l'environnement a montré que 20 % des fruits de mer, environ 40 % des eaux de rivière et environ 40 % des eaux usées traitées testées contenaient du génome de rotavirus.

Résumé : The effect of ambient ammonia, hypoxia and combination of both on survival and the physiological and immunological response of the blue shrimp Litopenaeus stylirostris in relation to molt stage was studied. Shrimp were submitted to 44.0-71.5 mg 1(-1) total ammonia-N corresponding to 2.0 mg 1(-1) unionized ammonia NH3-N and/or to 1.5 mg O-2 1(-1) (4.3 kPa) for 24 hours. Survival was recorded and the molt stages of both dead and surviving shrimp determined. Only shrimp in intermolt and premolt stages were sampled for analysis of haemolymph. Haemolymph was assayed for osmoregulatory capacity (OC), magnesium ion (Mg2+), calcium ion (Ca2+), total proteins, oxyhaemocyanin, lactate, glucose and total haemocyte count (THC). Low mortalities were recorded for shrimp submitted independently to ammonia or hypoxia. Seventy five percent of dead shrimp were in early post molt (stage A) in ammonia treatment, while hypoxia affected mainly late premolt animals (stage D-2). A synergic effect of ammonia and hypoxia combination (A+ O-2 treatment) on mortality was observed, affecting nearly exclusively shrimp in late premolt stage D-2. Analysis of molt stage repartition at the end of the experiment suggests that ammonia treatment may have accelerated molting. The common physiological response of shrimp to the different treatments was characterized by a reduced OC and an increase in Ca2+. Increase in Mg2+ could not be validated by the statistical analysis, as well as glycaemia variations. Plasmatic lactate level increased and THC decreased in shrimp submitted to hypoxia and the combination of hypoxia and ammonia. Total proteins concentration was reduced in ammonia and A+O-2 treatments. The effect was more pronounced in late premolt shrimp than in intermolt shrimp. Combination of ammonia and hypoxia led to a physiological response stronger than this observed for ammonia-alone and/or hypoxia-alone treatments, except for oxyhaemocyanin. Résumé : On the western coast of the Atlantic, according to the literature, 2 distinct groups of Semibalanus balanoides occur with a distinct interface near the Miramichi Estuary, New Brunswick, in the Gulf of St. Lawrence. On each side of this interface, the groups are characterized by clinal variations for MPI (mannose-6-phosphate isomerase) and GPI (glucose-6-phosphate isomerase). The present study was carried out to determine whether selection occurs at this interface, to establish how early in the sessile life period it occurs and to examine the selecting forces involved. Reciprocal transplant experiments of newly settled individuals to both sides of the interface were carried out. No significant differences specifically linked to source or destination were observed in growth or fecundity for the 2 groups at the sites studied for either control or transplanted individuals. However, differences in survival were observed; individuals transplanted south of the estuary showed lower survival than individuals transplanted north. An allozyme analysis of barnacle survivors for MPI and GPI, 2 alleles whose frequencies are known to vary abruptly in this region, indicated a change of allele frequency in transplanted individuals. The transplants’ allele frequencies came to resemble those of adults from target sites, while no change occurred in transplanted individuals at control sites. Taken together with previous results, our study suggests that selection occurs very early in the newly settled individuals (spat).

TMPRSS2/ERG fusion resulting in ERG overexpression occurs in 30 to 50% of prostate cancer (PCa) in Caucasian patients, but its prognostic relevance remains controversial. In the present study, we investigated ERG expression in all stages of PCa progression, and evaluated the prognostic impact of ERG status in clinically localized PCa (CLC) and in castration resistant disease (CRPC). ERG and AR expressions were evaluated by immunohistochemistry on tissue microarrays containing samples of high grade PIN (n = 57), CLC surgically treated (n = 299, including 185 Caucasians and 114 African-Caribbeans), metastases (n = 17), and CRPC (n = 41). In Caucasians, ERG expression significantly increased from high grade PIN (17.5%) to pT2 (27%) and pT3 CLC (43%), then to metastases (53%). In CLC, stainings for ERG and AR were correlated, and ERG expression was less frequent in African-Caribbeans compared to Caucasians (11.5% vs. 33%). In Caucasians CLC, ERG was associated with longer recurrence free survival, after adjusting for classical prognostic markers. In CRPC, ERG was expressed in 29% of cases, and was associated with a longer overall survival. Our results confirm that ERG expression is less frequent in PCa from patients of African descent. Although ERG expression increases during PCa natural history, positive ERG status is associated with better outcome in both CLC and CRPC. This paradox could be explained in part by the fact that ERG expression is AR dependant, then ERG positive cancers are likely to progress in a rich androgen environment, with a better response to androgen suppression.
