They are produced in third world countries of the sub-equatorial belt. Among the causes limiting their production, one is the presence of the WSSV (White Spot Syndrome Virus), a pathogenic agent that produces massive mortality. Our aim was to investigate the first stages of the viral infection, in order to be used as target of prophylactic actions. A fish cell line (SSN-1) was used as model to tentatively develop in vitro studies. Only defective particles were produced confirming the high specificity to crustacea of the infection with crustacean virus. Electron microscopy showed structural similarities between the WSSV and B, B2 and Baculo-B viruses of crabs. This suggests B2 may belong also to the family Nimaviridae, genus Whispovirus. This comparison with B2 virus gives the possibility to understand the role played by the tail-like extension of these viruses in the infectious process by attachment to the plasmic membrane at the beginning of the infection in its specific host. Les crevettes de grande taille de la famille des Penaeidae constituent le premier produit aquacole en valeur commerciale à l'échelle planétaire. Elles sont produites à 99% dans les pays en voie de développement de la ceinture sub-équatoriale. Parmi les causes limitant leur production, le White Spot Syndrome Virus est l'agent pathogène ayant provoqué le plus des pertes dans le monde. Notre étude a été orientée vers la reconnaissance des premiers stades de l'infection, susceptibles d'être la cible d'une action prophylactique. Nous avons testé l'utilisation d'une lignée cellulaire de poisson SSN-1 afin d'étudier les possibilités d'un développement in vitro. Seules des particules défectives ont été produites confirmant la haute spécificité des infections à virus de crustacés. Les recherches en microscopie électronique ont montré une similarité structurale du WSSV avec les virus (B, B2 et Baculo-B) des crabes. Ceci suggère que ces agents (B2 et WSSV) seraient tout deux de la famille des Nimaviridae et du genre Whispovirus. La comparaison avec le virus B2 permet de comprendre le rôle clef joué par la partie caudale de ces virus dans l'infection par son attachement à la membrane plasmique lors de l'infection chez son hôte spécifique. IFREMER.]. 1988 Résumé : Since nearly twenty years, world exploitation of crustaceans increases from 1,263,000 tons in 1968 to 3,419,000 tons in 1985, overcoming the predictions of 1970 by more than one million tons. Penaeid prawns fisheries experienced a very fast development. The krill resource proved to be practically commercially unworkable. By now, the major crustaceans stocks appear fully exploited. During the same period, penaeid aquaculture experienced a fast growth, which should continue in the near future. Amélioration génétique expérimentale de la crevette d'élevage de Nouvelle-Calédonie : Sélection d'une population de L. stylirostris résistante à la bactérie pathogène Vibrio penaeicida. The major difficulty to which the industry has been faced for 10 years is the occurrence of the "syndrome 93", which corresponds to mortality phases when the temperature falls down in April-May-June. This mortality is associated to the pathogenic bacteria Vibrio penaecida and is expressed at different levels which are variable from year ta year and from pond to pond. No resistance to this pathology has been developed spontaneously. This is likely due to the protocole used to rear spawners, which does not allow to implement an efficient selective pressure at each generation
An experimental selection on the criteria of survival after picks of syndrome 93 has been conducted at the Laboratoire Aquacole de Calédonie. The 3rd selected generation demonstrates survival rates improved by 20% during experimental infections with V. penaeicida in comparison with a non selected control population of same genetic origin. The comparison of the correlated responses on the level of expression of 5 genes which are potentially implicated in immunity phenomena (Peneidins, lysozyme, transglutaminase. profiline, annexine) shows that the selected population has a level of expression in lyzozyme twice higher than the control population. This result suggests that the lysozyme could be a genetic marker which could be used in a selective breeding program to he developed in relation with the private hatcheries. La filière crevette de Nouvelle-Calédonie repose sur la maîtrise de la reproduction contrôlée de la crevette Litopenaeus stylirostris, espèce introduite dans les années 80. La difficulté majeure que rencontre la filière depuis une dizaine d'années est la récurrence du « syndrome 93 », qui correspond à des épisodes de mortalités lors des baisses de température en avril-mai-juin. Ces mortalités sont associées à la bactérie pathogène Vibrio penaeicida et s'expriment à des niveaux d'intensité variable d'une année à l'autre et d'un bassin à l'autre. Aucune résistance vis-à-vis de cette pathologie ne s'est développée spontanément. Ceci est vraisemblablement lié au protocole employé pour l'élevage des géniteurs qui ne permet pas d'exercer une pression de sélection efficace à chaque génération. Une expérience de sélection sur un critère de survie à des épisodes de syndrome 93 a été menée au Laboratoire Aquacole de Calédonie. La 3ème génération sélectionnée montre des survies améliorées de l'ordre de 20% lors d'infections expérimentales à V. penaeicida par rapport à une population témoin non sélectionnée de même origine génétique. La comparaison des réponses corrélées sur le niveau d'expression de 5 gènes potentiellement impliqués dans les phénomènes de défense immunitaire (penaeidine, lysozyme, transglutaminase, profiline, annexine) montre que la population sélectionnée a un niveau d'expression en lysozyme deux fois plus élevé que la population témoin. Ce résultat suggère que le lysozyme pourrait être un marqueur génétique utilisable dans un programme de sélection à développer en relation avec les écloseries de production. Giffard-mena, Ivone Éditeur(s) : Elsevier Science Bv Résumé : The euryhaline white shrimp Litopenaeus vannamei lives in both coastal and oceanic areas through ontogeny. Its osmoregulation pattern and variations in its tolerance to salinity are partially known from several studies under different experimental conditions (developmental stages, salinities and acclimation procedures). Although L. vannamei is recognized as one of the most euryhaline penaeid species, with adults and juveniles exhibiting an hyper-hypo-osmoregulatory pattern and being able to tolerate a wide salinity range, little is known on larval and early postlarval strategies to cope with salinity fluctuations. In order to establish their euryhalinity range and to fully understand the ontogenetic changes in L. vannamei osmoregulatory pattern, we evaluated the effect of six salinities (5, 10, 20, 32, 45 and 60 psu) on 17 developmental stages by directly exposing them to experimental salinities and conducting observations during the next 48 hours. At 5 hours post osmotic shock (hps), all developmental stages survived (> 20%) in 20, 32 and 45 psu. The euryhalinity and osmoregulation pattern changed at some developmental stages of L. vannamei. The hyper-hypo osmoregulatory pattern exhibited by Juvenile and Adult appears to be established early in the first post-larval stage PL1 (ontogenetic osmoregulation pattern type 3), with higher tolerance to salinity variations observed in PL2, PL4 and PL22 suggesting that L. vannamei shows a progressive increase in the efficiency of osmoregulatory mechanism following last metamorphosis. Nutrition, digestion et développement des larves de crevettes et poissons marins Auteur(s) : Cahu, Chantal Résumé : This report is presented by Chantal Cahu for obtaining accreditation to supervise research. Since 1981, her research activity has addressed the study of reproducers' nutrition and the young stages of development of marine organisms belonging to two different branch lines, crustaceans and fish. This work, performed in a base-lined research body, IFREMER, is placed within a more general programme of aquaculture development. Indeed, whatever species are at issue, the development of aquaculture in France, as at the global level, is based on the production of juveniles in hatcheries. My work has therefore more precisely the goal of acquiring knowledge about the nutritional needs, mechanisms of digestion and the development of these animals in order to improve the production of prawn post-larvae and fish alevin in the hatchery.
This work took place in three main periods that appear in this report:
- The study of reproducers' nutrition and its influence on the quality of the Penaeid prawn's eggs and larvae. My work has focused mainly on the effect of lipids and vitamins on the development of eggs and larvae.
- The study of the ontogenesis of the digestive functions and the nutritional needs of sea fish larvae, with bass as the model. The goal was to determine the specific nutritional needs of the young stages by studying the implementation and regulation of pancreatic and intestinal digestive enzymes.
- The study of the regulation of certain developmental genes by means of nutrients, so as to understand how these nutrients behave during ontogenesis, particularly during development of the skeleton.
Ce mémoire est présentée par Chantal Cahu pour pour l'obtention d'une habilitation a diriger des recherches. Depuis 1981, son activité de recherche concerne l'étude de la nutrition des reproducteurs et des jeunes stades de développement d'organismes marins appartenant à deux embranchements différents, les crustacés et les poissons. Ce travail réalisé dans un organisme de recherche finalisée, l'Ifremer, se situe dans un programme plus général de développement de l'aquaculture. En effet, quelles que soient les espèces en jeu, le développement de l'aquaculture en France comme au niveau mondial repose sur la production de juvéniles en écloserie. Mon travail a donc plus précisément pour objectif l'acquisition de connaissances sur les besoins nutritionnels, les mécanismes de digestion et le développement des ces animaux afin d'améliorer la production de post-larves de crevettes et d'alevins de poissons en écloserie. Ce travail s'est déroulé en trois grandes périodes qui apparaissent dans ce mémoire : - L'étude de la nutrition des reproducteurs et son influence sur la qualité des oeufs et des larves de crevettes Pénéides. Mon travail a essentiellement porté sur l'effet des lipides et des vitamines sur le développement des oeufs et des larves. - L'étude de l'ontogenèse des fonctions digestives et les besoins nutritionnels des larves de poissons marins, avec le bar comme modèle. Le but était de déterminer les besoins nutritionnels spécifiques des jeunes stades, en étudiant la mise en place et la régulation des enzymes digestives pancréatiques et intestinales. - L'étude de la régulation de certains gènes du développement par les nutriments, de façon à comprendre comment ces nutriments agissent lors de l'ontogenèse, notamment lors du développement du squelette.
