Bonamia ostreae-induced mortalities in one-year old European flat oysters Ostrea edulis: experimental infection by cohabitation challenge Auteur(s) : Lallias, Delphine Arzul, Isabelle Heurtebise, Serge Ferrand, Sylvie Chollet, Bruno Robert, Maeva Beaumon, Andy Boudry, Pierre Éditeur(s) : EDP Sciences Résumé : Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection by B. ostreae. Mortality started after 4 months of cohabitation in the tested oysters. The cumulative mortalities after 6 months were 58% for the wild oysters, 9% for Family 1 (20-month old) and 20% for Family 2 (8-month old). In the dying oysters, the parasite could be detected in 67% of the wild oysters, 68% of Family 1 and 89% of Family 2. It was detected in only 11% of the surviving oysters of Family 2. The mortality and the level of infection by the parasite were significantly higher in Family 2 than in Family 1. Our results demonstrate that prespawning oysters as young as 1 year-old can become infected with the parasite and, most importantly, can die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. Additionally, no clear relationship between shell length and level of infection was observed. We also review the different methods for infection of the European flat oyster O. edulis with B. ostreae under experimental conditions and their main results. One line of research consists in investigating karst aquifers that represent significant water supplies for the areas around the Mediterranean. This work aims at developing a methodology of predictive evaluation of karst distribution and karst thickness within the carbonate massifs. It is based on the study of the Tethysian carbonate platform from the area of Montpellier, South of France. This area underwent a polyphased, complex, tectonic evolution, involving successive karst forming events. The approach is based on the relationships between eustacy and geodynamics, on one hand, and base-level variation recorded within palaeokarsts, on the other hand. Sedimentary fill of palaeokarsts allows to identify distinct karstification phases and sometimes to date them. It is a marker of base-level variations, which we attempt to quantify, that is responsible for karst formation and karst filling and sealing. For three palaeokarsts of regional extent, we analyze : karst sedimentary-fill, the distribution along upstream-downstream profile, the vertical extension of palaeokarsts, indicative of the base-level lowering. - The " Bauxite palaeokarst " results from the two-stage evolution of the " Isthme Durancien " : i) Bauxitic weathering of the Neocomian cover over the southern Massif Central , related to an inversion during Barremian ; ii) ≥1600 m uplift and exhumation during Aptian-Albian, inducing erosion of alterites, upstream, and deposition of detrital bauxites, downstream. - The " Early Paleocene palaeokarsts with marine sedimentary-fill " indicate base level variations of more than 350m in the upstream area. It is suggested here that such base-level variations, exceeding eustacy and recorded in tabular undeformed series, result from desiccation-flooding events in an endoreic silled basin. - The " Miocene palaeokarst with Cevennes-sourced sediment-fill " records a Serravalian-Tortonian base-level drop, depicted by canyon incision in the hinterland of the Gulf of Lion Margin. Uplift values of ≥ 400m upstream, about 250m in an intermediate area and 10's of m downstream, are measured. The present 3D geometry and distribution of carbonate massifs (exposed and covered) are modeled in the study area, from the Cevennes to the Mediterranean. The 3D geological model integrates seismic reflection, borehole and gravimetric data. Tectonic restoration of a N-S section, extracted from the 3D model, allows to reconstruct paleo-base-levels with respect with the carbonate massifs, at different stages of deformation and successive phases of karstification. The 3D paleo-flows through carbonates across the study area, are thus constrained, especially during the Messinian desiccation stage. Application of the results to hydrogeology leads to the following : - Uplift of upstream hinterland (Cretaceous, Miocene) produces large amplitude karsts, but they do not make significant reservoirs. - Karstification responsible for the development of important and deep karst reservoirs is related to desiccation of endoreic basins (early Paleocene and Messinian). - Theses two events have produced two main deep karst reservoirs, with a gravimetric signature : (1) Upstream of Montpellier Thrust, between Pic St Loup, Les Matelles Fault and Hérault Basin, a deep karsts developed over 900m thickness in the Malm limestone, during Paleocene. (2) Downstream (south) of Montpellier Thrust, the coastal karsts extend offshore over more than 10km, they are sealed by a thick Miocene to Quaternary cover, and they result from the Messinian event.

For groundwater issues (potential leakages in waste repository, aquifer management ...), the development of modeling techniques is far ahead of the actual knowledge of aquifers. This raises two fundamental issues: 1) which and how much data are necessary to make predictions accurate enough for aquifer management issues; 2) which models remain relevant to describe the heterogeneity and complexity of geological systems. The French observatory H+ was created in 2002 with the twofold motivation of acquiring a large database for validating models of heterogeneous aquifers, and of surveying groundwater quality evolution in the context of environmental changes. H+ is a network of 4 sites (Ploemeur, Brittany, France; HES Poitiers, France; Cadarache, France; Campos, Mallorca, Spain) with different geological, climatic, and economic contexts. All of them are characterized by a highly heterogeneous structure (fractured crystalline basement for Ploemeur, karstified and fractured limestone for Poitiers, Cadarache and Mallorca), which is far to be taken into account by basic models. Ploemeur is exploited as a tap-water plant for a medium-size coastal city (15,000 inhabitants) for 20 years. Each site is developed for long term investigation and monitoring. They involves a dense network of boreholes, detailed geological and geophysical surveys, periodic campaigns and/or permanent measurements of groundwater flow, water chemistry, geophysical signals (including ground motions), climatic parameter, etc. Several large-scale flow experiments are scheduled per year to investigate the aquifer structure with combined geophysical, hydrogeological, and geochemical instruments. All this information is recorded in a database that has been developed to improve the sustainability and quality of data, and to be used as a collaborative tool for both site researchers and modelers. This project lasts now for 5 years. It is a short time to collect the amount of information necessary to apprehend the complexity of aquifers; but it is already enough to obtain a few important scientific results about the very nature of the flow heterogeneity, the origin and residence time of water elements, the kinetic of geochemical processes, etc. We have also developed new methods to investigate aquifers (in-situ flow measurements, flow experiment designs, groundwater dating, versatile in-situ probes, etc.). This experience aiming at building up long term knowledge appears extremely useful to address critical issues related to groundwater aquifers: the structure and occurrence of productive aquifer in crystalline basement, the assessment of aquifer protection area in the context of highly heterogeneous flow, the biochemical reactivity processes, the long term evolution of both water quantity and quality in the context of significant environmental changes, for instance. Schéma directeur pluriannuel 1982-1985 et éléments d'information pour la préparation de la loi de programmation sur la recherche et le développement technologique Résumé : Like numerous industrialized countries, France became aware, throughout the 1960s, of the growing importance of the ocean, of its known and potential resources and of the international issue it represents for nations. In the field of marine research and development, after an initial prompting period from the General Commission for Technical and Scientific Research, the French government decided to extend this action through the creation of a coordinating body, the National Centre for the Exploitation of Oceans. Fish, fishers, seals and tourists: Economic consequences of creating a marine reserve in a multi-species, multi-activity context Auteur(s) : Boncoeur, Jean Alban, Frederique Guyader, Olivier Thebaud, Olivier Éditeur(s) : Wiley Résumé : This paper investigates some economic consequences of creating a marine reserve on both fishing and ecotourism, when the range of controllability of fishing effort is limited and the impact of the reserve on ecosystem is considered. The issue is illustrated by the example of creating a no-take zone in part of a region where fishing is managed through a limited entry license system, and which is inhabited by two interacting stocks: a stock of prey (fish) and a stock of predators (seals). While the former is targeted by commercial fishing, the latter is not subject to harvest but is a potential basis for a commercial non-extractive activity (seal watching). Analysis is conducted with the help of a bioeconomic model combining the features of marine reserve modeling and of multispecies modeling. Following a description of the model, results of several simulation runs are presented. These show that creating a marine reserve has more complex economic implications than predicted in studies focused exclusively on one stock and/or commercial fisheries. More specifically, the model shows that the dynamics of the two interacting stocks reduces the benefits of the no-take zone for the fishing industry, while it makes the creation of this zone provide an opportunity for the development of ecotourism. Due to this dynamics, the model suggests that the optimal size of the reserve is larger when ecotourism is taken into account along with fishing activities. The development of large pelagic fishing in the French Antilles is an integral part of a difficult economic situation of the fishing sector in Martinique where this development can seem like a solution to increasingly acute problems. Indeed, for the regulatory authorities as well as for many participants in the sector, it is imperative to find solutions to the lack of profitability of a part of the fishing ships and to confront the regular increase of the share of imports in supplying Martinique with seafood. This question about the possibilities of developing large pelagic fishing is not new since it is already mentioned in the Notice from the Economic and Social Committee of the Martinique Region in 1982. We will not show in this document a detailed analysis of small-scale fishing in Martinique. Since the latter has been abundantly studied by the team of the Caribbean Oceanological and Halieutic Research Centre (PROHC) between 1985 and 1987, we will take up these principal conclusions (with which we are in full agreement) and bring them up to date based on data collected during our mission from 11 to 24 April 1988. The first section devoted to the supply structure (local fishing and imports) will attempt to define under what conditions large pelagic fishing can be developed. The second section will address marketing and demand. Indeed, at the same time as the problems of developing large pelagic fishing, we are faced with the question of the organisation of seafood markets. This organisation is a fundamental issue for the small-scale fishing sector in the French Antilles since its long-term future depends upon it by setting different prices and amounts exchanged according to the structure it is given. We will come to a conclusion on the economic conditions of developing a large pelagic fishery and organising a market, this based on research and study work already done. We will indicate which investigations must be conducted to complete the set of factors for decision support in this area. To perform this assessment, we reconstructed from the long series 1970-1986 (sometimes with significant inadequacies): these are presented as an appendix. Understanding the disability like a « social object », the thesis analyzes the societal consideration of disabled people in non-urbanized areas with a high issue of touristic, recreational and patrimonial development. In the first place, it's the recent construction of disability as a social object that is discussed by reminding the history of the societal consideration of the disabled person in our occidental world. The French model and his fundamental rule of global accessibility for ail the domains of the social life are explained and the « Nature and Disability » thematic is more precisely analyzed as a development of an active living ideal for people who are in a disability situation. Secondly, the thesis develops this thematic on the particular field of the Reunion Island and tries to answer to a question which is both simple and complicated: « Today, how we live with a disability on the Reunion Island? ». ln a last time and on the basis of this study of the disability inside a Reunion Island society with accelerated transformations, the thesis specifies the current tendencies with the emergence of new public policies for the welcome of disabled people on natural and touristic areas of the island. To conclude and referring to an original survey, the thesis suggests some tools of evaluation and decision-making aids to these areas.
Diffusion, deformation and mineral properties of the Earth's interior. Introduction Auteur(s) : MAINPRICE, David Auteurs secondaires : Géosciences Montpellier ; Université des Antilles et de la Guyane (UAG) - Institut national des sciences de l'Univers (INSU - CNRS) - Université de Montpellier (UM) - Centre National de la Recherche Scientifique (CNRS) Éditeur(s) : HAL CCSD Elsevier Résumé : International audience This special issue in honour of the late Olivier Jaoul brings together contributions from experimental and theoretical mineralogy, the area of most of Olivier's own work, focused for the most part on the Earth's mantle. The volume is divided into three sections; diffusion, deformation and seismic properties. New technological developments have often had a strong impact on research in this area, as again shown by Olivier's constant search to improve the resolution of the short diffusion profiles typical of major cations in mantle silicates. In the section on diffusion the impact of improved computer performance for modelling diffusion and point defect configurations on one hand, and high pressure experimental techniques on the other hand, highlight the continued development in this area. The section on deformation deals with wide range of subjects, including the deformation of olivine and quartz and the relation between deformation and diffusion, both subjects close to Olivier's personal interests. This illustrates the new possibilities to undertake deformation experiments at mantle pressures and temperatures, as well as improvement in high-fidelity viscoelastic measurements at high temperature. In the last section on seismic properties we discover theoretical developments on predicting properties near the Earth's surface and a new experimental application, which allows the determination of sound velocity at very high pressures with implications for the Earth's inner core. Conséquences génétiques de la production de larves d'huîtres en écloserie : étude des processus de dérive et de sélection Auteur(s) : Taris, Nicolas Sauvage, Christopher Batista, Frederico Baron, Sophie Ernande, Bruno Haffray, Pierrick Boudry, Pierre Éditeur(s) : Actes du 6e colloque national BRG, La Rochelle, 2-3-4 octobre 2006 Résumé : Previous studies have shown heritable variation in larval developmental traits in the Pacific oyster Crassostrea gigas. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and, consequently, for the study of bivalve larvae genetics. Culling, by selective sieving of the smallest larvae is an advantageous practice at a phenotypic scale as it reduced variance in larval size, variance of developmental rate and time to settlement. Culling of 50% of the larval population only led to 15% less spat, showing a positive phenotypic correlation between larval growth and settlement success. However, culling represents a substantial risk for diversity loss, because it increases the variance of reproductive success among parental oysters. The effective population sizes of early settling cohorts of settlement were lower than those of later ones. Our results show that the settlement of slow growing larvae significantly contributes to minimizing the variability of reproductive success and therefore to maximizing genetic diversity. These results corroborate the low estimations of variability of broodstocks sampled in several French commercial hatcheries, relative to natural populations. The genetic composition of the larval population and the resulting spat was significantly different between the two tested temperatures, revealing genotype x environment interaction for survival. Similarly, genotype x environment interaction was also observed for larval growth as a higher temperature exerted a positive influence on the expression of genetic variability for this trait. Consequently, we can conclude that a temperature of 26°C coupled with culling, to common practice in oyster hatcheries, is likely to amplify the selection pressure for fast growing larvae. To test for this hypothesis, we compared larval developmental traits in the progeny of a hatchery broodstock closed for 7 generations, with the progeny of wild oysters and the two possible hybrids. Our results show that selection of fast growing larvae can counteract presumed inbreeding depression, due to higher mean relatedness among hatchery broodstock than in the wild. Genetic effects of intensive rearing conditions at larval stage are significant and should be taken into account in hatchery practices, especially in terms of genetic diversity management. Consider tourism as a remedy requires first to question the nature of evil. It is therefore necessary to analyze at first, poverty without reducing to its economic approach. Poverty is a multidimensional concept, which should be studied in a systematic approach. Tourism attracted international institutions and many politicians (at local and national levels) by its development opportunities. Tourism is distinguished by its adaptability and its ability to transform host environments. These are consumers who move from producers (home territories). This activity therefore provides direct revenue for the host societies.Hence the tourism capabilities to create specific jobs and stimulate investment. However, if the economic potential is undeniable, this activity should be placed in a real development project that benefits the poorest territories. By contrast tothe complete mantle-crustal sections of Semail-type ophiolite sheets, Tethyan ophiolites are characterized by asmaller amount of mafic rocks (gabbros and basalts), by the absence of any sheeted dyke complex and by thefrequent occurrence of oceanic sediments stratigraphically overlying mantle-derived peridotites and associatedgabbroic intrusions. Therefore, they are considered as typical remnants of oceanic lithosphere formed in slowspreadingenvironment or in ocean–continent transition at distal passive margins. In the very first models offormation of the Tethyan ophiolites, in the years 1980, the geodynamical processes leading to mantle unroofingwere poorly understood due to the paucity of data and concepts available at that time from the present-dayoceans. In particular, at that time, little work had focused on the distribution, origin and significance ofmafic rocks with respect to the dominant surrounding ultramafics. Here, we reconsider the geology of sometypical metaophiolites from the Western Alps and Corsica, and we show how results from the past decadeobtained in the current oceans ask for reassessing the significance of the Tethyan ophiolites in general. Revisitedexamples include a set of representative metaophiolites from the blueschists units of the Western Alps (Queyrasregion) and from Alpine Corsica (Golo Valley). Field relationships between the ophiolitic basement andthe metasedimentary/metavolcanic oceanic cover are described, outlining a typical character of the Tethyanophiolite lithological associations. Jurassic marbles and polymictic ophiolite metabreccias are unconformablyoverlying the mantle-gabbo basement, in a way strictly similar to what is observed in the non-metamorphicAppennine ophiolites or Chenaillet massif. This confirms that very early tectonic juxtaposition of ultramafic andmafic rocks occurred in the oceanic domain before subduction. This juxtaposition resulted from tectonic activitythat is now assigned to the development of detachment faults and to the formation of Oceanic Core Complexes(OCCs) at the axis of slow spreading ridges. This fundamental Plate Tectonics process is responsible for the exhumationand for the axial denudation of mantle rocks and gabbros at diverging plate boundaries. In addition, fieldrelationships between the discontinuous basaltic formations and the ultramafic–mafic basement indicate that thistectonic stage is followed or not by a volcanic stage. We discuss this issue in the light of available field constraints. Nutrition, digestion et développement des larves de crevettes et poissons marins Auteur(s) : Cahu, Chantal Résumé : This report is presented by Chantal Cahu for obtaining accreditation to supervise research. Since 1981, her research activity has addressed the study of reproducers' nutrition and the young stages of development of marine organisms belonging to two different branch lines, crustaceans and fish. This work, performed in a base-lined research body, IFREMER, is placed within a more general programme of aquaculture development. Indeed, whatever species are at issue, the development of aquaculture in France, as at the global level, is based on the production of juveniles in hatcheries. My work has therefore more precisely the goal of acquiring knowledge about the nutritional needs, mechanisms of digestion and the development of these animals in order to improve the production of prawn post-larvae and fish alevin in the hatchery.
This work took place in three main periods that appear in this report:
- The study of reproducers' nutrition and its influence on the quality of the Penaeid prawn's eggs and larvae. My work has focused mainly on the effect of lipids and vitamins on the development of eggs and larvae.
- The study of the ontogenesis of the digestive functions and the nutritional needs of sea fish larvae, with bass as the model. The goal was to determine the specific nutritional needs of the young stages by studying the implementation and regulation of pancreatic and intestinal digestive enzymes.
- The study of the regulation of certain developmental genes by means of nutrients, so as to understand how these nutrients behave during ontogenesis, particularly during development of the skeleton.
Assessment of tropical shrimp aquaculture impact on the environment in tropical countries, using hydrobiology, ecology and remote sensing as helping tools for diagnosis Auteur(s) : Fuchs, Jacques Martin, Jean-louis M. Populus, Jacques Résumé : After a period of rapid development of the sites (an increase in both reared surface area and production), tropical shrimp aquaculture is currently being faced with critical problems due to economical and ecological constraints. In many countries with favourable conditions for shrimp aquaculture such as indonesia or Vietnam, sites are often badly selected and/or over-exploited. This conducts to sorne extent to decreases or collapses in the production due to the difficulty in predicting the maximum production capacity of the sites. Furthermore, it appears that production sustainability depends on many factors among which socio-economy and ecolo gy are of prime importance.
The STD3 project titled « Assessment of tropical shrimp aquaculture impact on the environment in tropical countries using hydrobiology, ecology and remote sensing as helping tools for diagnosis )) was initiated in 1994, with the aim to study the sustainability of marine shrimp aquaculture in tropical areas, main! y Indonesia and Vietnam, through the following tasks: (i) improve site selection and study the impact of aquaculture on marine environments presenting a variable sensitivity to organic sewage, (ii) analyse the socio-economical aspects and profitability of the aquaculture industry and of common resources (iii) use remote sensing and geographical data bases for diagnosis and monitoring of site degradation (iv) reinforce the capacity of Asian
scientists through training.
Severa! aquaculture sites have been investigated in the Lampung region (South Sumatra, Indonesia), the Mekong delta (Vietnam) and on the West coast of New-Caledonia, providing a large range of typical ecosystems encountered in Asia and in the Pacifie, from coralline sites to coastal plains with mangrove and deltaic areas. The study concerned the spatial structure and seasonal influence of these ecosystems, the temporal variation during the course of the project (3 years) and the relation between the ecological structure and farm productions.
Remote sensing provides a synoptic vision over large land expanses. Severa! scenes were processed for landuse mapping using conventional classification techniques. Concerning water quality assessment, a general relation for the Java sea was found between image and field data in terms of total suspended matter. Applying this relation to a new site in Sumatra has provided an initial approach to water type and, together with land use mapping, a preliminary assessment of the suitability of the area to shrimp aquaculture development.
Socio-economic research has been focused on the identification ofneeds for collective action, including public policy, in the perspective of shrimp farming sustainability. The main concern is in the regulation of shrimp farming intensification and extensification at the scale of coastal ecosystemic entities in a common property resource management perspective. The common considered here is coastal water quality. Comparing the local development profiles and the institutional grounds for the design and implementation of collective management rules shows that variables such as land tenure system, farm owner socio-economic profile, social homogeneity or heterogeneity of the farmers are key factors to analyse the potential for sustainability. The defmition of
water quality and the possible means to ensure its collective management are discussed. The main conclusion is that the economie incentives to farming development are strong but there is no significant difference among traditional, semi-intensive and intensive systems in terms of economie efficiency or wealth distribution.
The description of the functioning of the different kinds of ecosystems make it possible to give an advice concerning the positioning of the activity inside the ecosystem, and furthermore to have a reference state in order to determine the impact of the activity on the coastal environnement. This impact can be summarized as an increase in the concentration of total suspended matt€?r, of particulate organic matter and of sulfate reducing bacterias. This organic ma~.er is either issued from rearing activities or from telluric origine, due to mangrove eradication when building the ponds.
An evaluation of the relationship existing between productivity and characteristics of each ecosystem bas been conducted by correlating ecological indicators data (concentration of total suspended matter, particulate organic matter, chlorophyll, percentage of pheopigments in total pigments and cyanobacteria and sulfatereducing bacteria in water and sediments) with the average production of the farms. This comparison lead to the compilation of a scale of observed production integrating the range of these environmental parameters with shrimp production levels, bound to facilitate the positioning of shrimp farming in relation with the confmement leve! of each ecosystem.
Lastly, ali geographie data originated from the various compartments of the study have been geo-referenced and loaded into a geographical information system. This allows to display any query made on spatial variables and their related statistical data, including the ir variations over the last few years and to reveal patterns and phenomena otherwise not obvious. This issue of the Revue Etudes Caribéennes, devoted to tourism, aims to lay the foundations of a reflection on the functioning tourist places, their development programs and the different challenges presented to them. Exposed/Open or closed location, concentrated or diffused flows, the nature of these tourist places with their traditional or original customs, orientate different types of potential impacts. The choices made in tourism development are essential: in which conditions should be opted for the traditional seaside tourism, or for mass tourism, or for alternative tourism? Are certain properties linked to specific territories? Should we oppose these two approaches or consider them as complementary in their environments and can they share the same place? The objective of this thematic issue is to call for various and renewed approaches which develop analysis in terms of the economy, regional planning, sociology, geography etc. Articles may propose a theoretical reflection or focus on case studies, dealing with the relations of mass tourism, alternative tourism, or to illustrate one of these two aspects. Priorities fields will not exclusively be Caribbean and American but can come from any part of the world and can illustrate post and/or present situations. The prioritized analysis will include: 1. The concepts of mass tourism and alternative tourism (identification, characteristics, spatial logics, and economic, social and environmental issues). 2. Places of mass tourism (resorts, historic cities, specific forms of site layout and reception facilities) 3. Alter-tourism and the renewal of tourism practices (ecotourism, community tourism, agrotourism) 4. The development of tourist spots and of management methods concerning protected environments (conservation, protection, tourism practices and recreational practices) 5. Economic, social, and environmental issues surrounding different types of tourist places. Assays for this purpose, commonly based on P-gp-expressing cell lines and digoxin as a reference P-gp substrate probe, unfortunately exhibit high variability, raising thus the question of developing alternative or complementary tests for measuring inhibition of P-gp activity. In this context, the present study was designed to investigate the use of the fluorescent dye rhodamine 123 as a reference P-gp substrate probe for characterizing P-gp inhibitory potential of 16 structurally-unrelated drugs known to interact with P-gp. 14/16 of these P-gp inhibitors were found to increase rhodamine 123 accumulation in P-gp-overexpressing MCF7R cells, thus allowing the determination of their P-gp inhibitory potential, i.e., their half maximal inhibitor concentration (IC50) value towards P-gp-mediated transport of the dye. These IC50 values were in the range of variability of previously reported IC50 for P-gp and can be used for the prediction of clinical P-gp inhibition according to Food and Drug Administration (FDA) criteria, with notable sensitivity (80%). Therefore, the data demonstrated the feasibility of the use of rhodamine 123 for evaluating the P-gp inhibitory potential of drugs Pharmaceutics hal-01305488 https://hal-univ-rennes1.archives-ouvertes.fr/hal-01305488 DOI : 10.3390/pharmaceutics8020012 PUBMED : 27077878 | Partager |

Éditeur(s) : HAL CCSD Society for the Study of Reproduction Résumé : International audience Matrix-assisted laser desorption/ionization (MALDI) tissue imaging mass spectrometry is particularly promising among the numerous applications of mass spectrometry. It is used for probing and analyzing the spatial arrangement of a wide range of molecules, including proteins, peptides, lipids, drugs, and metabolites, directly in thin slices of tissue. In the field of proteomics, the technology avoids tedious and time-consuming extraction and fractionation steps classically required for sample analysis. MALDI imaging mass spectrometry is increasingly recognized as a powerful method for clinical proteomics, particularly in cancer research. The technology has particular potential for the discovery of new tissue biomarker candidates, classification of tumors, early diagnosis or prognosis, elucidating pathogenesis pathways, and therapy monitoring. Over recent years, MALDI imaging mass spectrometry has been used for molecular profiling and imaging directly in male and female reproductive tissues. This review will consider some of the recent publications in the field, addressing a range of issues covering embryo development, gene expression product profiling during gametogenesis, and seeking and identifying biomarkers of reproductive cancers. The wealth of advances in mass spectrometry imaging will inevitably attract biologists and clinicians as the advantages and power of this technology become more widely known. This review will also discuss bottlenecks and the many technical issues that remain to be resolved before laboratories in the field can adopt the technology. We foresee that MALDI imaging mass spectrometry will have a major impact in reproductive research by opening new avenues to the understanding of various molecular mechanisms and the diagnosis of reproductive pathologies. Fishing strategies, economic performance and management of moored fishing aggregating devices in Guadeloupe Auteur(s) : Guyader, Olivier Bellanger, Manuel Reynal, Lionel Demaneche, Sebastien Berthou, Patrick Éditeur(s) : Edp Sciences S A Résumé : Moored FADs (MFADs) have been a key area for development in Guadeloupe (Lesser Antilles) since the late 1980's. The main objectives underpinning the development of MFADs were to (i) reduce fishing pressure on coastal fishing resources; (ii) boost fishermen's incomes; and (iii) increase local consumption of locally generated fishery products as a proportion of overall consumption. This paper gives an overview of the current structure of Guadeloupe's fishing fleet and fisheries, with a specific focus on MFAD fleets in a relatively unstructured environment with regard to regulation, and so how these fleets operate in a context of unrestricted development of private MFADs. In addressing the following key issues, this paper aims to: assess the economic performance of fleets operating on MFADs, understand why the MFAD fishing activity has not developed more than it has, and suggest how better MFAD management could improve the situation. Socio-economic information on a trip and fleet level basis were collected and stored on a fisheries information system to analyse the relative attractiveness of MFAD fishing and assess the overall economic performance of MFAD fleets as compared to other non-MFAD fleets. Non-monetary variables were also tested to explain the degree of dependence on MFADs. In 2008, economic returns from FADs were relatively low, both in terms of wages for the crews and in terms of profit for vessel owners. Earnings were higher than coastal fishing activities, but seem to have been very sensitive to the indicators used in our analysis. MFADs are expected to become much more attractive as a result of MFAD regulation, especially through the establishment of collective MFADs.
