SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. These symbioses are of prime importance for the functioning of the ecosystems. Similar symbioses occur in other bivalve species living in shallow and coastal reduced habitats worldwide. In recent years, several deep-sea species have been investigated from continental margins around Europe, West Africa, East America, the Gulf of Mexico, and from hydrothermal vents on the Mid-Atlantic Ridge. In parallel, numerous more easily accessible shallow marine species were studied. We here provide a summary of the current knowledge available on chemosymbiotic bivalves in the area ranging west-to-east from the Gulf of Mexico to Marmara Sea, and north-to-south from the Arctic to the Gulf of Guinea. Characteristics of symbioses in 51 species from the area are summarized for each of the five bivalve families documented to harbor chemosynthetic symbionts (Mytilidae, Vesicomyidae, Solemyidae, Thyasiridae and Lucinidae), and compared among families with special emphasis on ecology, life cycle, and connectivity. Chemosynthetic symbioses are a major adaptation to ecosystems and habitats exposed to reducing conditions, yet relatively little is known regarding their diversity and functioning apart from a few "model species" on which effort has focused over the last 30 yr. L. M. Éditeur(s) : Inter-research Résumé : Environmental conditions experienced by aquatic organisms are archived in biogenic carbonates such as fish otoliths, bivalve shells and coral skeletons. These calcified structures present an accretionary growth and variations in optical properties - color or opacity - that are used to reconstruct time. Full and reliable exploitation of the information extracted from these structures is, however, often limited as the metabolic processes that control their growth and their optical properties are poorly understood. Here, we propose a new modeling framework that couples both the growth of a biogenic carbonate and its optical properties with the metabolism of the organism. The model relies on well-tested properties of Dynamic Energy Budget (DEB) theory. It is applied to otoliths of the Bay of Biscay anchovy (Engraulis encrasicolus) for which a DEB model has been previously developed. The model reproduces well-known otolith patterns and thus provides us with mechanisms for the metabolic control of otolith size and opacity at the scale of an individual lifespan. Two original contributions using this framework are demonstrated. First, the model can be used to reconstruct the temporal variations in the food assimilated by an individual fish. Reconstructing food conditions of past and present aquatic species in their natural environment is key ecological information to better understand population dynamics. Second, we show that non-seasonal checks can be discriminated from seasonal checks, which is a well-recognized problem when interpreting fish otoliths. We discuss further developments of the model and the experimental settings required to test this new promising framework. Drifting FADs used in tuna fisheries: an ecological trap? Auteur(s) : Marsac, F Fonteneau, Alain Menard, Frederic Éditeur(s) : Pêche thonière et dispositifs de concentration de poissons, Caribbean-Martinique, 15-19 Oct 1999 Résumé : This paper discusses the hypothesis that small tunas and the various species found in association with drifting fads (such as "mahi-mahi", rainbow runner, wahoo, etc.) may be biologically trapped by such a strong association. Massive seeding of drifting artificial fads was observed worldwide during recent years. In this hypothesis, we suggest that fads may alter some biological characteristics of epipelagic populations associated with them: migration, growth, condition factors, predation and natural mortality. As fads are most often used in the equatorial currents, they tend to exhibit zonal drift. Therefore, the associated populations would be artificially transferred from one part of the ocean to another, when they would show different movement patterns in the absence of fads. Natural logs were probably beneficial in terms of ecology and evolution, because they tend to accumulate in convergence areas, most often considered as rich forage areas. Now, fads are seeded in offshore areas, which are not necessarily favourable for tuna feeding. This apparently strong association between fishes and drifting fads may then produce an unexpected biological impact on tuna populations and their associated fauna. The plan is to test this hypothesis in the Atlantic, developing an ad hoc research programme based on tagging, biological and physiological studies, in association to an analysis of high resolution fishery data before and after the development of the fad fishery.Original Abstract: Cet article discute l'hypothese selon laquelle les petits thons et les differentes especes associees a des DCP derivants (comme les coryphenes, elagatis, thazards, etc.) pourraient etre pris dans un piege biologique resultant de cette forte association. Le deploiement massif de DCP artificiels derivants a ete constate a l'echelle mondiale au cours des dernieres annees. Dans cette hypothese, ces DCP, colonises en permanence par de grandes fractions de populations epipelagiques, pourraient modifier les caracteristiques biologiques des especes concernees: leur migration, leur croissance, leurs facteurs de condition, la predation et la mortalite naturelle. Les DCP etant plus frequemment utilises dans les courants equatoriaux, ils tendent a deriver zonalement. Ainsi, les populations associees seraient artificiellement transferees d'un bord a l'autre de l'ocean, alors qu'elles manifesteraient d'autres types de deplacements en l'absence de DCP. Les debris naturels etaient probablement benefiques sur le plan de l'ecologie et de l'evolution car ils s'accumulent dans des zones de convergence le plus souvent considerees comme des zones riches en nourriture. Maintenant, les DCP sont mouilles au large, dans des zones qui peuvent ou non etre favorables a l'alimentation des thons. Cette association apparemment forte entre poissons et DCP derivants pourrait alors causer un impact biologique inattendu sur les populations pelagiques (thons et faune accompagnante). L'objectif est de tester cette hypothese dans l'Atlantique au moyen d'un programme de recherche s'appuyant sur du marquage et des etudes biologiques et physiologiques, parallelement a une analyse des donnees de peche (tailles et CPUE, avant et apres le deploiement des DCP derivants). In rugged landscapes characterized by environmental mosaics, living populations and communities can experience steep ecological gradients over very short geographical distances. In lowland tropical forests, interspecific divergence in edaphic specialization (for seasonally flooded bottomlands and seasonally dry terra firme soils) has been proven by ecological studies on adaptive traits. Some species are nevertheless capable of covering the entire span of the gradient; intraspecific variation for adaptation to contrasting conditions may explain the distribution of such ecological generalists. We investigated whether local divergence happens at small spatial scales in two stands of Eperua falcata (Fabaceae), a widespread tree species of the Guiana Shield. We investigated Single Nucleotide Polymorphisms (SNP) and sequence divergence as well as spatial genetic structure (SGS) at four genes putatively involved in stress response and three genes with unknown function. Significant genetic differentiation was observed among sub-populations within stands, and eight SNP loci showed patterns compatible with disruptive selection. SGS analysis showed genetic turnover along the gradients at three loci, and at least one haplotype was found to be in repulsion with one habitat. Taken together, these results suggest genetic differentiation at small spatial scale in spite of gene flow. We hypothesize that heterogeneous environments may cause molecular divergence, possibly associated to local adaptation in E. falcata. Assessment of tropical shrimp aquaculture impact on the environment in tropical countries, using hydrobiology, ecology and remote sensing as helping tools for diagnosis Auteur(s) : Fuchs, Jacques Martin, Jean-louis M. Populus, Jacques Résumé : After a period of rapid development of the sites (an increase in both reared surface area and production), tropical shrimp aquaculture is currently being faced with critical problems due to economical and ecological constraints. In many countries with favourable conditions for shrimp aquaculture such as indonesia or Vietnam, sites are often badly selected and/or over-exploited. This conducts to sorne extent to decreases or collapses in the production due to the difficulty in predicting the maximum production capacity of the sites. Furthermore, it appears that production sustainability depends on many factors among which socio-economy and ecolo gy are of prime importance.
The STD3 project titled « Assessment of tropical shrimp aquaculture impact on the environment in tropical countries using hydrobiology, ecology and remote sensing as helping tools for diagnosis )) was initiated in 1994, with the aim to study the sustainability of marine shrimp aquaculture in tropical areas, main! y Indonesia and Vietnam, through the following tasks: (i) improve site selection and study the impact of aquaculture on marine environments presenting a variable sensitivity to organic sewage, (ii) analyse the socio-economical aspects and profitability of the aquaculture industry and of common resources (iii) use remote sensing and geographical data bases for diagnosis and monitoring of site degradation (iv) reinforce the capacity of Asian
scientists through training.
Severa! aquaculture sites have been investigated in the Lampung region (South Sumatra, Indonesia), the Mekong delta (Vietnam) and on the West coast of New-Caledonia, providing a large range of typical ecosystems encountered in Asia and in the Pacifie, from coralline sites to coastal plains with mangrove and deltaic areas. The study concerned the spatial structure and seasonal influence of these ecosystems, the temporal variation during the course of the project (3 years) and the relation between the ecological structure and farm productions.
Remote sensing provides a synoptic vision over large land expanses. Severa! scenes were processed for landuse mapping using conventional classification techniques. Concerning water quality assessment, a general relation for the Java sea was found between image and field data in terms of total suspended matter. Applying this relation to a new site in Sumatra has provided an initial approach to water type and, together with land use mapping, a preliminary assessment of the suitability of the area to shrimp aquaculture development.
Socio-economic research has been focused on the identification ofneeds for collective action, including public policy, in the perspective of shrimp farming sustainability. The main concern is in the regulation of shrimp farming intensification and extensification at the scale of coastal ecosystemic entities in a common property resource management perspective. The common considered here is coastal water quality. Comparing the local development profiles and the institutional grounds for the design and implementation of collective management rules shows that variables such as land tenure system, farm owner socio-economic profile, social homogeneity or heterogeneity of the farmers are key factors to analyse the potential for sustainability. The defmition of
water quality and the possible means to ensure its collective management are discussed. The main conclusion is that the economie incentives to farming development are strong but there is no significant difference among traditional, semi-intensive and intensive systems in terms of economie efficiency or wealth distribution.
The description of the functioning of the different kinds of ecosystems make it possible to give an advice concerning the positioning of the activity inside the ecosystem, and furthermore to have a reference state in order to determine the impact of the activity on the coastal environnement. This impact can be summarized as an increase in the concentration of total suspended matt€?r, of particulate organic matter and of sulfate reducing bacterias. This organic ma~.er is either issued from rearing activities or from telluric origine, due to mangrove eradication when building the ponds.
An evaluation of the relationship existing between productivity and characteristics of each ecosystem bas been conducted by correlating ecological indicators data (concentration of total suspended matter, particulate organic matter, chlorophyll, percentage of pheopigments in total pigments and cyanobacteria and sulfatereducing bacteria in water and sediments) with the average production of the farms. This comparison lead to the compilation of a scale of observed production integrating the range of these environmental parameters with shrimp production levels, bound to facilitate the positioning of shrimp farming in relation with the confmement leve! of each ecosystem.
Lastly, ali geographie data originated from the various compartments of the study have been geo-referenced and loaded into a geographical information system. This allows to display any query made on spatial variables and their related statistical data, including the ir variations over the last few years and to reveal patterns and phenomena otherwise not obvious. Le programme mis en oeuvre par la DRV (Paris, CREMA-L'Houmeau, DELIAO Brest, COP Tahiti, GIE/RA Nouvelle Calédonie), dans le cadre d'un projet européen STD3 «(Sciences et Technique au Service du Développement», en collaboration avec l'Université de Montpellier II, l'Ecole Nationale d'Agronomie de Rennes, l'ITC (Hollande), l'Institut Océanographique de Nha Trang (Vietnam), le BADC de Jepara et le BPPT de Jakarta (Indonésie), avait pour but la mise au point de méthodes permettant d'améliorer la sélection des sites ainsi que le suivi du développement et de l'impact de l'aquaculture des crevettes Péneides en milieu tropical pour éviter la surexploitation. Evaluation, scénarios et viabilité écologique et économique des pêcheries côtières tropicales : application au cas de la Guyane Française Auteur(s) : Cisse, Abdoul Éditeur(s) : Université des Antilles et de la Guyane Résumé : The need for an integrated approach to fisheries is now widely affirmed, including the FAO, in particular in the context of tropical artisanal fisheries. These small-scale fisheries, often multispecies are very important in terms of production, employment and food security. However these fisheries and the exploited resources are often characterized by a lack of data making difficult their management within an ecosystem approach, and by the ecological and economic system complexities including trophic and technological interactions. This thesis aims to contribute to the development of bioeconomic tools for small tropical fisheries in a sustainable management perspective based on the ecosystem approach. In this perspective, the French Guiana coastal fishery constitutes an interesting case study. The thesis proposes multi-criteria evaluations, complex bioeconomic models and viable management scenarios for this fishery. At first, multivariate statistical analysis suggests a satisfactory overall status of the fishery in terms of sustainability. However, some performance differences are noted within the fishery, particularly at border areas. Also, management improvements are proposed. Then, through bioeconomic modeling, projections of different fishing scenarios show that, in the long-term, the current exploitation level may not be consistent with the future increase of local demand and a loss of biodiversity may occur. A scenario called "co-viability" reconciling ecological, economic and social objectives, with a high probability of achievement is exhibited. Finally, the comparison of optimal behavior in cooperative and non-cooperative conditions, confirms that harvest levels are greater when actors do not cooperate. Furthermore, it is shown that the state of the ecosystem depends on the fishing strategies and the type of interaction between species. Beyond the diagnosis made for the case study, the method is promising in the context of small tropical fisheries, while the co-viability approach allows finding the exploitation conditions under which ecological and socio-economic sustainability are meet, what the conventional fisheries management generally do not allow. La nécessité d’une approche intégrée des pêches est actuellement largement affirmée, notamment par la FAO, en particulier dans le contexte des pêcheries artisanales tropicales. Ces pêcheries à petites échelles, souvent multi-spécifiques sont très importantes en termes d’emploi et de production, y compris pour la sécurité alimentaire. Néanmoins ces pêcheries et la biodiversité exploitées sont souvent marquées, d’une part, par le manque de données rendant difficile leur gestion dans le cadre d’une approche écosystémique, d’autre part, par la complexité des systèmes écologiques et économiques sous-jacents incluant interactions trophiques et techniques. Cette thèse contribue à la mise au point d’outils bioéconomiques adaptés aux petites pêcheries tropicales dans la perspective d’une gestion durable des pêches fondée sur l’approche écosystémique. Dans cette perspective, la pêcherie côtière en Guyane Française constitue un cas d’étude particulièrement fécond. La thèse propose ainsi des évaluations multi-critères, des modèles bioéconomiques complexes et des scénarios de gestion viable pour cette pêcherie. Dans un premier temps l’analyse statistique multivariée suggère un statut global satisfaisant de la pêcherie en termes de durabilité. Dengue fever spatio-temporal patterns result from complex interactions between the virus, the host, and the vector. These interactions can be influenced by environmental conditions. Our objectives were to analyse dengue fever spatial distribution over New Caledonia during epidemic years, to identify some of the main underlying factors, and to predict the spatial evolution of dengue fever under changing climatic conditions, at the 2100 horizon. We used principal component analysis and support vector machines to analyse and model the influence of climate and socio-economic variables on the mean spatial distribution of 24,272 dengue cases reported from 1995 to 2012 in thirty-three communes of New Caledonia. We then modelled and estimated the future evolution of dengue incidence rates using a regional downscaling of future climate projections. The spatial distribution of dengue fever cases is highly heterogeneous. The variables most associated with this observed heterogeneity are the mean temperature, the mean number of people per premise, and the mean percentage of unemployed people, a variable highly correlated with people's way of life. Rainfall does not seem to play an important role in the spatial distribution of dengue cases during epidemics. By the end of the 21st century, if temperature increases by approximately 3°C, mean incidence rates during epidemics could double. In New Caledonia, a subtropical insular environment, both temperature and socio-economic conditions are influencing the spatial spread of dengue fever. Extension of this study to other countries worldwide should improve the knowledge about climate influence on dengue burden and about the complex interplay between different factors. This study presents a methodology that can be used as a step by step guide to model dengue spatial heterogeneity in other countries. La diversidad y riqueza de este marco insular ya fue elogiada por grandes científicos como Alexander von Humboldt o Charles Darwin. Sin embargo, actualmente la presión ejercida sobre el medio por parte del desarrollo urbanístico y la industria turística, motor económico del archipiélago, pone en peligro estos frágiles ecosistemas; lo que ha generado un profundo debate entre crecimiento económico y turístico, y desarrollo sostenible y conservación. El presente artículo trata, desde una óptica divulgativa, de acercar al lector al medio que caracteriza las Islas Canarias e introducirlo en el marco donde se desarrolla parte importante de la actividad turística en España, uno de los países que más turistas recibe año a año. Les îles Canaries, situées dans la région de la ‘Macaronésica’ (l’Est de l’Océan Atlantique), abritent des écosystèmes marins et terrestres très précieux, tant du point de vue écologique que paysager. Ils sont représentatifs de les zones biogéographiques de sa région et comptent trois Réserves da Biosphère reconnues par l’UNESCO. De grands scientifiques comme Alexander Von Humboldt et Charles Darwin ont fait l’éloge de la richesse de la biodiversité de cet archipel. Cependant, actuellement, la pression anthropique, notamment en liaison avec le développement touristique et tous les enjeux économiques qui l’accompagnent, mettent en danger les écosystèmes des Canaries ; ce constat a ouvert un large débat sur les enjeux autour des développements économique et touristiques et leurs dérives. Notre article propose de mettre l’accent sur les caractéristiques des îles Canaries et de replacer les processus de développements touristiques dans leur contexte pour ouvrir la réflexion sur les conditions nécessaires pour d’un développement durable. The Canary Islands, located in the region of 'Macaronésica' (Este of Atlantic Ocean), are home to marine and terrestrial ecosystems very valuable, both ecological and landscape. Dynamique de la diversité fonctionnelle des communautés de poissons (lagune de Terminos, Mexique) Auteur(s) : Villeger, Sébastien Éditeur(s) : Montpellier SupAgro Résumé : One of the main challenges in ecology is to understand how global changes affect biodiversity and what are the consequences on ecosystem functioning. In this perspective, the functional diversity of communities is a cornerstone since it allows linking environment, community structure and ecosystem properties. The aim of this thesis is thus to improve the understanding of functional diversity dynamic (i) in relation to natural variability of environmental conditions and (ii) under anthropogenic disturbances. As a first step we have developed a new methodological framework allowing to (1) describe fish functional niches based on functional traits, and (2) to quantify functional diversity within (α) and among (β) sites thanks to new indices. In a second step we have studied the spatiotemporal dynamic of the functional structure of fish and nektonic communities from the Terminos lagoon (Mexico). This tropical estuarine ecosystem is an appropriate ecological model for our problematic as it is characterized by a strong environmental variability, a high biological diversity and is under a strong human pressure. We have put in light the stability of the functional and trophic structures of communities along environmental gradients, despite a very strong species turnover. This stability is determined by the dominance of a couple of functional groups inside which species are replacing each others according to their environmental preferences. However, at a long-term scale, we have demonstrated a functional diversity loss in a part of the lagoon despite an increase of species richness. This paradox has to be related to the decrease of species associated to seagrass and the increase of more estuarine species. Un des enjeux majeurs de l'écologie est de comprendre comment les changements globaux affectent la biodiversité et quelles en sont les conséquences sur le fonctionnement des écosystèmes. Dans cette perspective, la diversité fonctionnelle des communautés est un outil clé permettant de lier l'environnement, la structure des communautés et les propriétés écosystémiques. L'objectif de cette thèse est donc d'améliorer la compréhension de la dynamique de la diversité fonctionnelle (i) face à des conditions environnementales naturellement variables et (ii) face à des perturbations d'origine anthropique. Dans un premier temps nous avons mis en place un socle méthodologie nous permettant de (1) définir la niche fonctionnelle des poissons à partir de traits fonctionnels et (2) de quantifier les diversités fonctionnelles intra (α) et inter (β) échantillons avec de nouveaux indices. Dans un second temps nous avons étudié la dynamique spatio-temporelle de la structure fonctionnelle des communautés ichtyologiques et nectoniques peuplant la lagune de Terminos (Mexique). Cet écosystème estuarien tropical est un modèle d'étude adapté à notre problématique car il présente une forte variabilité environnementale, une forte diversité biologique et est sous forte pression anthropique. Nous avons mis en évidence une stabilité de la structure fonctionnelle et trophique des communautés face aux gradients environnementaux très marqués, et donc malgré un fort taux de remplacement des espèces entre les communautés. Cette stabilité est due à la dominance de quelques groupes fonctionnels à l'intérieur desquels les espèces se remplacent suivant leurs preferendums environnementaux. Néanmoins, à plus long terme, nous avons démontré qu'une portion de la lagune avait subi une perte de diversité fonctionnelle et ce malgré une augmentation de la richesse spécifique. Ce paradoxe est à relier au remplacement des espèces inféodées aux herbiers de phanérogames par des espèces plus estuariennes. Problems of Development of Tourism and Yachting on the Coast of Gdansk Pomerania (Poland) Auteur(s) : A. Gdansk Bay and the Vistula Lagoon is characterized by very good weather conditions. Numerous advantages of anthropogenic and tourist resorts strengthen the attractiveness of the coast. The marina recently ended in Gdansk, Sopot and Gdynia create very good conditions for the development of yachting. The combination of the Gulf of Gdansk by the river network and channels Żuławy of the Great Masurian Lakes promotes the development of yachting in both the Gulf of Gdansk and Masurian Lake region. However, new infrastructure investments, realized with funding from the EU, like ‘Loop Żuławska’ and the new marina, leading to an increase in tourist traffic and threats to existing reserves and landscape parks. Another problem is the lack of regulation of the right on the crossing of the Strait of Baltiysk, leading from the Vistula Lagoon on the Baltic Sea, which is located in the territorial waters of Russia. The concentration of investments in the region of Żuławy contrasts with too little spending on development of tourist infrastructure in the Hel Peninsula. There are large disparities in tourist traffic. Tourism in Hel Peninsula significantly exceeded the absorption of tourism in the region and leads to the degradation of the local environment. In addition, the seasonality of tourism on the coast favours the trend of maximizing profit tourism enterprises. New, designed ditch the Vistula Spit, which connects directly to the Vistula Lagoon from the Baltic Sea, bypassing the Russian territorial waters, can lead to serious ecological changes. La région de Gdansk en Poméranie est une excellente zone géographique pour le développement du yachting. La baie de Gdansk et la lagune de la Vistule se caractérisent par de très bonnes conditions climatiques. Divers avantages anthropiques et stations touristiques renforcent l’attractivité de la côte.Les marinas nouvellement terminées à Gdansk, Sopot et Gdynia, créent de très bonnes conditions pour le développement du yachting. La combinaison du golfe de Gdansk, du réseau fluvial et des canaux Żuławy qui relient les Grands Lacs de Mazuries, favorise le développement du yachting dans le golfe de Gdansk et la région des lacs. Les nouveaux investissements dans les infrastructures, réalisés grâce à des financements de l’Union européenne, tels "Loop Żuławska" et la nouvelle marina, conduisent à une augmentation de l’activité touristique qui menace les réserves existantes et les parcs naturels. L’autre problème est l’absence de réglementation relative au droit de passage dans le détroit de Baltiysk, menant de la lagune de la Vistule, qui est située dans les eaux territoriales de la Russie. Entre la concentration des investissements dans la région de Żuławy et les faibles dépenses pour le développement de l’infrastructure touristique dans la péninsule d’Hel, il existe de grandes disparités dans le trafic touristique. Les infrastructures touristiques de la péninsule d’Hel ne sont pas en mesure d’absorber les flux touristiques de la région et la situation conduit inéluctablement à la dégradation de l’environnement. La situation de la lagune de la Vistule, dont les eaux sont partagées entre la Russie et la Pologne, peut entraîner de graves changements écologiques. In addition to fundamental knowledge, the study of populations’ evolution has wide applications such as conservation/restoration genetics or epidemiology. Indeed, genetic diversity is the ‘fuel’ for populations’ evolution and adaptation to new conditions, and estimating the extent and the geographic structure of genetic diversity, as well as identifying the evolutionary forces behind (demography, gene flow, and adaptation) are thus major challenges in the current context of global changes. [br/]A particular attention is currently devoted to the process of genetic divergence in wild populations, with emphases on gene flow, local adaptation, and their interactions. Indeed, adaptation in wild populations is supported in many species by many kinds of experiments based on both quantitative phenotypic traits and molecular data. However, the processes governing populations’ evolution are poorly understood in the tropical rainforest of Amazonia, and the relative influence of neutral and adaptive processes are continuously discussed. [br/]I am a young scientist interested in the process of genomic divergence in wild (tree) populations. I aimed at understanding how evolution structures the genetic diversity and governs molecular divergence in wild populations, with particular emphases on gene flow and local adaptation. During five years as a young scientist, I worked on answering the following questions, focusing on two tree species occupying vulnerable areas: mainly [i]Eperua falcata[/i] in Amazonia and, to a lesser extent, [i]Abies alba[/i] in the Mediterranean area.[br/][u]Scientific questions:[/u][br/]- Which evolutionary drivers are responsible of the structure of genetic diversity in wild populations occupying wide areas? This question was addressed at different geographical scales: from large (regional) to very local (microgeographic) scales. [br/]- What is the relative influence of neutral (gene flow, inbreeding) and adaptive drivers in the process of genetic divergence?[br/]- Is microgeographic adaptation possible in spite of extensive gene flow between populations occupying the extreme boundaries of an ecological gradient? Tourism and Biodiversity in Natura 2000 Sites. Therefore, in this context a new challenge is emerging: the need to conserve and protect bio and geo diversity, as well as the economic valorisation of these areas based mainly on tourism and leisure. At EU level, Natura 2000 protected areas have developed, implemented and cultivated the need to practice sport and recreational activities in these areas both for spending leisure time and also as stress antidote. The recreational and sporting practices that can be undertaken within the ecological network Natura 2000 sites adapted to the specificity of each protected area, in close connection with the environmental conditions. For the preservation of the natural environment should develop and maintain a close collaboration with tourism and sports associations that organize such activities in those areas. La réflexion est centrée sur l’importance de la nature et des enjeux du développement de tourisme dans les aires protégées. Un nouveau défi émerge dans le contexte actuel où il s'agit de concilier le besoin de conserver et de protéger la géodiversité et la biodiversité avec la valorisation économique de ces espaces naturels investis par les pratiques touristiques et récréatives. Au niveau de l'Union européenne, les espaces protégés Natura 2000 ont mis en œuvre des programmes permettant de répondre à l’accueil des activités sportives et de loisirs dans ces zones, tant pour passer du temps libre que comme antidote au stress. Ces pratiques entreprises dans le réseau écologique Natura 2000 sites sont adaptés à la spécificité de chaque secteur protégé, en relation avec les conditions environnementales. First, we choose a model species which permitted to develop an appropriate abundance estimation method which take into account the imperfect detection and temporary immigration of mobile species, and to test environmental conditions effects at different spatial scales of analysis. Second, given that bias due to animal detection can be important in the abundance estimation, we determinated the relative role of environmental conditions and species biological traits on detection probability. From this methodological framework, we studied then processes of community organization, by identifying the main determinants involved in community organization, using composition and alpha and beta diversities, and, in a complementary manner, three metrics (taxonomic, functional, and phylogenetic). We sought from these general framework to determinate the relative role of environmental conditions and historical contingency, responsible for the current community organization, and to test the forest refugia hypothesis. Finally, we downscaled the analysis to determinate, and tried to disentangle the effects of environmental conditions and interspecific interactions on species co-occurrence. We used then functional traits to test the relationship between species co-occurrence and similarities.This thesis highlights the important role of historical contingency and facilitation in the structure of regional diversity and the local assembly processes of medium- and large-sized vertebrates, respectively, in French Guiana. Cette thèse a pour but d’étudier l'influence de l'hétérogénéité des forêts de terre ferme en Guyane sur l'organisation des communautés de moyens et grands vertébrés. Nous avons, d'abord, choisi une espèce modèle afin de déterminer une méthode adéquate d'estimation d'abondance prenant en compte la détection imparfaite et l'immigration temporaire des espèces, et de tester l'effet des conditions environnementales à différentes échelles d'analyse. Puis, partant du constat que les biais dus à la détection dans l'estimation d'abondance pouvaient être importants, nous avons déterminé l'importance relative des conditions environnementales et des traits biologiques des espèces sur la probabilité de détection. A partir de ce cadre d'étude, nous avons étudié la composition et les diversités alpha et beta en utilisant de façon complémentaire les métriques taxonomique, fonctionnelle, et phylogénétique pour déterminer le rôle relatif de l'environnement et de la contingence historique dans l'organisation des communautés, et de tester l’hypothèse des refuges forestiers. Enfin, en descendant dans l'échelle d'analyse, nous avons tenté de démêler l'effet de l'environnement et des interactions interspécifiques sur la cooccurrence des espèces, et utilisé les traits fonctionnels des espèces pour tester le lien entre cooccurrence et similitude fonctionnelle. Effects of Oceanic Salinity on Body Condition in Sea Snakes Auteur(s) : Brischoux, Francois Rolland, Virginie Bonnet, Xavier Caillaud, Matthieu Shine, Richard Éditeur(s) : Oxford Univ Press Inc Résumé : Since the transition from terrestrial to marine environments poses strong osmoregulatory and energetic challenges, temporal and spatial fluctuations in oceanic salinity might influence salt and water balance (and hence, body condition) in marine tetrapods. We assessed the effects of salinity on three species of sea snakes studied by mark-recapture in coral-reef habitats in the Neo-Caledonian Lagoon. These three species include one fully aquatic hydrophiine (Emydocephalus annulatus), one primarily aquatic laticaudine (Laticauda laticaudata), and one frequently terrestrial laticaudine (Laticauda saintgironsi). We explored how oceanic salinity affected the snakes' body condition across various temporal and spatial scales relevant to each species' ecology, using linear mixed models and multimodel inference. Mean annual salinity exerted a consistent and negative effect on the body condition of all three snake species. The most terrestrial taxon (L. saintgironsi) was sensitive to salinity over a short temporal scale, corresponding to the duration of a typical marine foraging trip for this species. In contrast, links between oceanic salinity and body condition in the fully aquatic E. annulatus and the highly aquatic L. laticaudata were strongest at a long-term (annual) scale. The sophisticated salt-excreting systems of sea snakes allow them to exploit marine environments, but do not completely overcome the osmoregulatory challenges posed by oceanic conditions. Future studies could usefully explore such effects in other secondarily marine taxa such as seabirds, turtles, and marine mammals. Etude des processus hydrodynamiques et écophysiologiques de la dynamique d'une algue invasive : Valonia aegagropila (C. Agardh) dans la lagune de Salses-Leucate Auteur(s) : Cesmat, Ludovic Éditeur(s) : Universite Montpellier II Résumé : Since 1980, the expansion of a green macro-alga: Valonia aegagropila (C Agardh) is observed in Salses-Leucate lagoon (France). It is currently distributed in the north of the lagoon with important biomasses (40 gDW/m²) eroding the cover of Zostera noltii. The free form of the alga can be transported with the surface or bottom currents depending on seasons. It multiplies mainly by vegetative reproduction. There are few studies on the ecophysiological capacities of V. aegagropila. This work tries to answer why (physiological adaptation) and how (mechanical process of transport) the alga invaded the lagoon, with an approach including observations, experiments in controlled environment and biological and hydrodynamic models. The growth capacities depend on temperature and light intensities, the nutrients absorption rates and internal nitrogen and phosphorus internal quotas were measured and modelled using a Droop model based on internal quota. Vertically transport was studied with laboratory experiments and measurements of biomass transport were made into the lagoon. The biological model (growth, vertical transport, mortality) was coupled to the MARS 3D hydrodynamic model with a meshgrid of 160m. The results show an adaptation of the alga to low availability of phosphorus and an annual growth of biomass controlled by the availability of nutrient during winter. The alga seems to occupy the whole zones where growth is optimal and the risk of invasion of the southern part of the lagoon is limited. This work opens on research prospects on an ecological model (confronting physiological capacities and competition of V. aegagropila, Zostera noltii, green algae and primary production), new physiological experiments, ecological observations and potential valorisation of V. aegagropila. Depuis 1980, est observée dans l'étang de Salses-Leucate l'expansion d'une macro-algue verte : Valonia aegagropila (C. Agardh). En 2004, elle était présente sur la presque totalité de la moitié nord de la lagune avec des biomasses importantes (jusqu'à 600 gPS/m²). L'algue envahit et érode également la couverture d'herbiers garant de la biodiversité, de la productivité et de la stabilité de l'écosystème. V. aegagropila se reproduit essentiellement de manière végétative (fragmentation des thalles) et possède la particularité de retenir de l'oxygène de la photosynthèse dans sa structure sphérique. Cet oxygène piégé lui donne la possibilité de mouvement sur la verticale et d'être transportée dans le domaine par les courants. Les conditions physico-chimiques de l'étang présentent de fortes variations saisonnières en salinité, en température et en turbidité, ainsi que des apports nutritifs faibles : la production primaire est principalement limitée par le phosphore. Il existe malheureusement peu d'informations sur V. aegagropila ainsi que trop peu de connaissances sur son écophysiologie. Elle semble vivre dans des milieux plutôt oligotrophe dans des régions tropicales à sub-tropicales. Ce travail basé sur une approche d'observations de terrain, d'expérimentations en milieu contrôlé (en laboratoire) et de modélisation des processus, a permis d'une part d'explorer les capacités adaptatives physiologiques de V. aegagropila aux conditions environnementales de l'étang et d'autre part de mettre à jour les modes de fonctionnement de la dynamique spatio-temporelle de la population. Les capacités de croissance ont été mesurées sur un large spectre de température et de lumière. Les capacités d'absorption des sels nutritifs dans l'eau et les facultés de l'algue à stocker l'azote et le phosphore dans ses tissus ont également été étudiées. D'autre part, un modèle de dynamique des populations a été conceptualisé puis développé. Les processus pris en compte dans ce modèle sont la croissance de l'algue (modèle à quota interne), la flottabilité de l'algue et la mortalité par surdensité. Ces processus ont été paramétrés en majeure partie par des expérimentations réalisées en laboratoire. Ce modèle 0D a par la suite était couplé au modèle hydrodynamique MARS3D (modèle Ifremer) mis en place sur la lagune de Salses-Leucate, utilisant deux grilles de calcul (160m ou de 80m de résolution). Les simulations réalisées permettent d'appréhender les capacités spatio-temporelles de la croissance de l'algue dans l'étang, le turnover annuel de la biomasse totale et les attracteurs de biomasse du système sous différents évènements météorologiques. Ces résultats sont comparés avec les répartitions cartographiques de la population observées in situ en 1999 et 2004 et des flux de biomasses mesurés au cours d'observations de terrain entre juin 2003 et septembre 2004. L'ensemble des résultats montre d'une part une adaptation de l'algue à évoluer dans un milieu pauvre en phosphore et d'autre part une croissance annuelle de la biomasse totale qui est principalement contrôlée par les apports hivernaux en sels nutritifs dans la lagune. Les résultats des simulations montrent également que V. aegagropila semble occuper à présent la majeure partie des zones favorables à sa croissance. Il est également montré que si sa capacité de mouvement peut favoriser la dispersion d'individus dans la lagune et accélérer la croissance de la population pour de faibles biomasses initiales, cette même capacité de mouvement joue plutôt un rôle de régulation de la population lorsque la biomasse totale devient importante. On peut penser que le risque de colonisation de l'herbier à zostère de la partie sud de la lagune (bassin de Salses) est limité. Ce travail ouvre enfin des perspectives de recherche sur l'amélioration du modèle de dynamique des populations de l'espèce, sur de nouvelles expérimentations à réaliser ou encore sur la surveillance écologique du milieu. Des propositions sont faites pour concevoir un modèle écologique confrontant les capacités physiologiques des macrophytes (algues vertes, Valonia et zostères) afin d'apréhender les phénomènes de compétitions et les modes de fonctionnement du réseau trophique de la lagune de Salses- Leucate. Des pistes sur la valorisation de la matière algale de V. aegagropila sont enfin proposées. There are reports of no effect as well as both decreases and increases in cortisol after chronic stressors. Therefore, the first aim of the present study was to determine how measures of compromised welfare, such as chronic pain and haematological anomalies, related to cortisol levels in domestic horses (Equus caballus). Domestic horses are an informative model to investigate the impact of chronic stress (due to environment, pain, work, housing conditions…) on the HPA axis. The second aim was to determine whether levels of fecal cortisol metabolites (FCM) may be used as an indicator of welfare measures. The present study used fifty-nine horses (44 geldings and 15 mares), from three riding centres in Brittany, France. The primary findings show that horses whose welfare was clearly compromised (as indicated by an unusual ears backward position, presence of vertebral problems or haematological anomalies, e.g. anaemia) also had lower levels of both FCM and plasma cortisol. This work extends our previous findings showing that withdrawn postures, indicators of depressive-like behavior in horses, are associated with lower plasma cortisol levels. We also found that evening plasma cortisol levels positively correlated with FCM levels in horses. Future research aims to determine the extent to which factors of influence on welfare, such as living conditions (e.g. single stalls versus group housing in pasture or paddocks), early life factors, and human interaction, act as mediators of cortisol levels in horses.
