INRA, CFCS Description : The importance of longevity as an economically trait gives a picture of the flock efficiency and adaptation in a particular environment. A study was conducted in the experimental herd of Creole goats at INRA in Guadeloupe in order to test environmental (year and season at first kidding, age at first kidding and weight at first mating as well as genetic (index of resistance, sire) factors that affect longevity of does. Lifetime data set of 387 Creole does, reared at pasture all year long, was recorded over a period of 11 years (2001-2012). Does were bred for reproduction at 11months of age. Three mating periods were organized per year, corresponding to 3 climatic seasons, using buck effect. Data were analyzed using survival models (Survival Kit 6.1). The average age for culling was 5.03 years. The culling rate was higher for goats between 2 and 3 years (17 and 24%) and then gradually decreased. Year and season at first kidding did not have a long term influence on does? longevity; neither does age at first kidding. In contrast, weight at first mating had a significant effect and it can be recommended to farmers to mate primiparous goats heavier than 17 kg. Heritability was estimated to 0.16 allowing some genetic progress. No significant correlation was shown with the genetic breeding value of resistance to gastrointestinal parasitism. This study gave indications to breeders to improve their female flock management and increase does? longevity. L'importance de la longévité comme trait économique donne une image de l'efficacité et de l'adaptation de troupeau dans un environnement particulier. Une étude a été entreprise dans le troupeau expérimental de chèvres créoles à l'AICN en Guadeloupe afin d'examiner ambiant (l'année et la saison d'abord badiner, âge d'abord badiner et poids d'abord joindre aussi bien que (index de résistance, de père) les facteurs génétiques affectez dont la longévité fait. L'ensemble de données de vie du Créole 387 fait, élevé au pâturage tout au long de l'année, a été enregistré pendant 11 ans (2001-2012). Fait ont été multipliés pour la reproduction à 11months d'âge. Trois périodes d?accouplement ont été organisées par an, correspondant à 3 saisons climatiques, utilisant l'effet de mâle. Des données ont été analysées utilisant des modèles de survie (trousse de survie 6,1). L'âge moyen pour cueillir était de 5,03 ans. Le taux de cueillage était plus haut pour des chèvres entre 2 et 3 ans (17 et 24%) et alors graduellement diminué. L'année et la saison à premier badiner n'ont pas eu une influence à long terme sur la longévité des does ; ni l'un ni l'autre ne vieillit d'abord badiner. En revanche, le poids au premier accouplement a eu un effet significatif et il peut recommander aux agriculteurs de joindre les chèvres primipares des que 17 kilogrammes plus lourds. L'héritabilité a été estimée à 0,16 permettant du progrès génétique. Aucune corrélation significative n'a été montrée avec la valeur d'élevage génétique de la résistance au parasitisme gastro-intestinal. Cette étude a donné des indications aux éleveurs afin d'améliorer la gestion féminine de troupeau et d'augmenter la longévité de ces dernières. INRA, CFCS Description : Carbon dioxide (CO2) accounts for the largest proportion (74%) of total greenhouse gas (GHG) emissions globally, with agricultural activities contributing approximately 25% of this gas. Opportunities to reduce the emissions in the sector can therefore contribute significantly to mitigating global climate change. Ag-lime is used extensively to alleviate problems of soil acidity and the potential for this material to contribute to C sequestration in soils is gaining interest among researchers. A laboratory experiment using C-13 isotopic labelling techniques was therefore established to answer the following questions: 1) Is there a potential for sequestering C using ag-lime in Trinidad acid soils? 2) Does enhancing soil microbial respiration by applying labile organic amendments enhance the C sequestration potential of ag-lime? 3) Are lime-derived C (LDC) estimates from non-isotopic difference technique comparable to that from the C- 13 isotopic labelling technique? The results ascertained for the study showed significant (P<0.05) differences between the LDC estimates from the two techniques with the non-isotopic techniques yielding higher estimates. Over the 32- day experiment, between 16.4 to 85.4% of the ag-lime C was released as CO2 with values being lower for the Piarco soil. The LDC in the CO2 was also significantly (P<0.05) lower for the poultry litter treatments comparison to the other organic amendments for the Piarco soil but not the Nariva soil. This effect may not have been as a result of an enhancement of the carbon sequestration potential of the ag-lime due to an elevation in soil microbial respiration since the LDC in the glucose-amended soils did not differ significantly from the no-amendment control soils. INRA, CFCS Description : The Creole pig has always been part of the rural and suburban landscape of Martinique. Currently, this breed is not integrated into a research and conservation program. The Natural Park of Martinique Region (PNRM) has the objective to maintain and valorize the genetic heritage of Martinique?s Creole pig and develop a niche business. Based on PNRM knowledge, some Creole pigs live freely in the mountains in the North, in the South coast and in a few disparate traditional breeders located in the countryside. It is essential to carry out an inventory of the local pig population to propose a scheme for conservation and economic development. In order to favour the appropriation of the Creole pig niche, the PNRM, as a decision maker, acts in a systemic and holistic way by considering the whole Martinican territory and the pig sub-sector: producers involved in the COOPMAR pig farmers? cooperative, researchers of INRA (FWI), the food chain and at least (in a second phase) the consumers and the Martinican society. First of all, the pig farmers are involved (private family farms and specialised pig producers). Researchers and technicians from PNRM and INRA- URZ (Animal production research unit) and INRA-PTEA (Tropical platform in animal experimentation) are performing experimental studies both in controlled conditions and in farms, in order to: i) determine phenotypic and genetic characteristics of Martinique?s Creole pigs in comparison with other pig breeds from the Caribbean area; ii) help at designing genetic management to maintain the population and avoiding inbreeding; iii) help at defining feeding management by a) establishing, at the whole territorial food chain, an inventory of co or by-products available for pig feeding; b) implementing experimental studies in technology for conservation; c) implementing feeding and growing experiments and finally iv) help at defining eco-friendly production systems a) aiming at generate an adequate revenue and b) focusing on ecosystem services such as meat quality, socio-cultural services and circular economy. Le porc créole a toujours fait partie du paysage rural et suburbain de la Martinique. Actuellement, cette race n'est pas intégrée dans un programme de recherches et de conservation. Le parc naturel de la région de la Martinique (PNRM) a l'objectif pour maintenir et valoriser l'héritage génétique du porc créole de la Martinique et pour développer des créneaux. Basé sur la connaissance de PNRM, quelques porcs créoles vivent librement dans les montagnes dans le nord, dans la côte sud et chez quelques éleveurs traditionnels disparates situés dans la campagne. Il est essentiel d'effectuer un inventaire de la population locale de porc pour proposer un plan pour la conservation et le développement économique. Afin de favoriser l'appropriation du créneau créole de porc, le PNRM, comme décideur, agit d'une manière systémique et holistique en considérant tout le territoire de la Martinique et sous-secteur de porc : producteurs impliqués dans la coopérative d'agriculteurs de porc de COOPMAR, les chercheurs d'AICN (FWI), la chaîne alimentaire et au moins (dans une deuxième phase) les consommateurs et la société Martiniquaise. Tout d'abord, les agriculteurs de porc sont impliqués (les fermes privées de famille et les producteurs de porc spécialisés). Therefore, for improvement in crop productivity in Suriname and most other countries, excessive application of pesticides and synthetic fertilizers is used. These result in gradual depletion of soil fertility and microbial diversity. Conventionally managed soils are found to exhibit a poorer micro flora and a lower biological activity then organically managed soils. Recently much attention is paid to organic cultivation and the use of bio fertilizers as a supplement for chemical fertilizers. VAM fungi have the potential to improve the sustainability of tomato production by improving yield and quality. Therefore, the aim of the study was to evaluate the effect of two different concentrations of commercially available vesicular - arbuscular mycorrhiza on the plant growth and fruit production of two tomato varieties (Lycopersicon esculentum Mill.) in the greenhouse.The experiment was set up as a randomized factorial block design with two independent factors and consisted of six treatments and three replications. The factors were two tomato varieties: hybrid 61 and hybrid 501 and two concentrations of VAM namely 2.5 and 10 g. VAM/plant. Biomass was determined two times, at bloom initiation and at the end of the experiment. The growth parameters were measured once per week. The production was recorded only for the first two harvest times. Data was statistically analyzed using three-way ANOVA. The results of this experiment showed that the VAM fungi had a positive influence (p < 0.05) on the production for both varieties of tomatoes. And it also resulted in earlier reproduction. There was no significant difference in production (p > 0.05) between the two concentrations of VAM inoculum used. Therefore a selective breeding program for resistance to bonamiosis has been undertaken since 1985 bu Ifremer, leading to the production of several selected oyster families. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters (injected with the parasite) to two tested families of oysters originating from the selective breeding program. Mortalities were checked daily, and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection to B. ostreae. The first infections occurred after 4 months of challenge in the tested oysters (Family 1 and Family 2). The cumulative mortalities after 5 monts were 58% for the wil oysters, 9% for Family 1 (20-month old at the beginning of the experiment) and 20% for Family 2 (8-month, old). The parasite could be detected in 66.8% of the dying wild oysters, 67.5% of the dying oysters of Family 1, 89% of the dying oysters of Family 2 and only 11% of the surviving oysters of Family 2. The mortality was significantly higher in Family 2 thant in Family 1 (x2= 20.87, p<0.001, d.f.) as well as the level of infection by the parasite found in heart smear (x2=24.34, p<0.001, 4 d.f.). This result demonstrates that prespawning oysters as yong as 1 year-old can become infected with the parasite and die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. The most probable cause of the dscrepancy in the development of bonamiosis between the 2 tested families is a difference in genetic background. SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. The size of the farming earthen ponds (510 ha) and the subsequent production constraints make it difficult to use them as experimental units for random experiments. Moreover, since every pond has its own characteristics, ponds' effects cannot be ruled out, thus making it hard to define true replicates. In order to design future experiments under conditions resembling those used in production, we evaluated the possible use of floating cages as experimental units with the aim of assessing treatment effects with a reasonable statistical power. To this end, two sets of floating cages were placed respectively in two different ponds in a commercial farm. In both cases the zootechnical conditions in the cages were similar in post-larvae origin, management, and diet and feeding regime. The aim of the study was to evaluate on a technical basis the possibility of rearing shrimps in floating cages set up in earthen ponds and to assess this method from a statistical standpoint. Shrimps reared in and outside the cages showed comparable growth and survival rates. The variability in the zootechnical parameters between cages, expressed as the estimated coefficient of variation (CV) for survival rate, total final biomass (g), final body weight (g), daily increment in body weight (g day− 1) and FCR were 11.0, 13.7, 4.4, 5.4, and 7.0%, respectively. On the basis of these figures, we calculated for a given statistical power (80%) the number of replicates (cages) that would be required to reveal significant differences between two treatments, at a 5% level of significance. We found that for expected differences of 20% from the control mean, 3 and 6 floating cages per treatment would be reasonable to determine statistical differences for growth parameters and survival rate, respectively. Moreover, we showed a significant pond effect in regard to survival and growth between the two sets of cages. These results illustrated the within-farm variability among the ponds, and confirmed that the specific characteristics of each pond from the same farm make it difficult to use the ponds themselves as experimental units. The study demonstrates that rearing in floating cages is an economical, powerful and sensitive experimental tool for shrimp culture studies specifically carried out under conditions close to semi-intensive production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the level of infection by B. ostreae. Mortality started after 4 months of cohabitation in the tested oysters. The cumulative mortalities after 6 months were 58% for the wild oysters, 9% for Family 1 (20-month old) and 20% for Family 2 (8-month old). In the dying oysters, the parasite could be detected in 67% of the wild oysters, 68% of Family 1 and 89% of Family 2. It was detected in only 11% of the surviving oysters of Family 2. The mortality and the level of infection by the parasite were significantly higher in Family 2 than in Family 1. Our results demonstrate that prespawning oysters as young as 1 year-old can become infected with the parasite and, most importantly, can die from bonamiosis. This result is inconsistent with the commonly accepted critical age of 2 years-old for the disease development. Additionally, no clear relationship between shell length and level of infection was observed. We also review the different methods for infection of the European flat oyster O. edulis with B. ostreae under experimental conditions and their main results. La bonamiose est une maladie parasitaire (agent causal : Bonamia ostreae) affectant l'huître plate européenne Ostrea edulis, responsable d'un déclin drastique de sa production aquacole. Des programmes de sélection pour la résistance à la bonamiose ont été entrepris pour contrer cette maladie. Dans cette étude, une expérience de 6 mois d'infection par cohabitation a été réalisée de manière à transmettre la maladie à partir d'huîtres sauvages injectées avec le parasite vers deux familles testées d'huîtres (âgées de 20 et 8 mois en début d'expérience, avec des origines génétiques différentes) issues du programme de sélection développé par IFREMER en France. Les mortalités ont été vérifiées quotidiennement et des frottis de coeur ventriculaire réalisés sur les huîtres mortes ou moribondes pour détecter le niveau d'infection par B. ostreae. La mortalité a commencé chez les huîtres testées après 4 mois de cohabitation. Les mortalités cumulées après 6 mois étaient de 58 % chez les huîtres sauvages, 9 % chez la Famille 1 (âgées de 20 mois) et 20 % chez la Famille 2 (âgées de 8 mois). Chez les huîtres mourantes, le parasite a pu être détecté chez 67 % des huîtres sauvages, 68 % de la Famille 1 et 89 % de la Famille 2. Il n'a pu être détecté que chez 11 % des huîtres survivantes de la Famille 2. La mortalité et le niveau d'infection par le parasite étaient significativement plus élevés chez la Famille 2 que chez la Famille 1. Nos résultats démontrent que des huîtres âgées de un an peuvent devenir infectées par le parasite et surtout, peuvent mourir de bonamiose. Ce résultat contraste avec l'âge critique de développement de la maladie communément accepté de 2 ans. De plus, aucune relation claire entre la longueur de la coquille et le niveau d'infection n'a été observée. Probiotic P-acidilactici application in shrimp Litopenaeus stylirostris culture subject to vibriosis in New Caledonia Auteur(s) : Castex, Mathieu Chim, Liet Pham, Dominique Lemaire, Pierrette Wabete, Nelly Nicolas, Jean-louis Schmidely, P Mariojouls, C Éditeur(s) : Elsevier Résumé : We studied the effects of a lactic acid bacterium, Pediococcus acidilactici (strain MA 18/5M, CNCM), as a dietary probiotic on growth performance and some nutritional and microbiological aspects of the shrimp Litopenaeus stylirostris. Experimental shrimp culture was carried out over 10 weeks, using floating cages of 14 m(2) each set in earthen ponds, in a New Caledonia farm affected by "Summer syndrome", a septicaemic vibriosis caused by Vibrio nigripulchritudo. The experiment design testing probiotic-coated pellets against control was replicated in two earthen ponds. High mortalities, characteristic of Summer syndrome, were observed during the first 2 weeks of the trial. The probiotic improved production in the treated cages from both ponds with increases in the survival rate (7% and 15% respectively) and final biomass (8% and 12% respectively). No differences were recorded among treatments on growth performances, but significant lower food conversion ratios were obtained with probiotic treatment. After 5 weeks of rearing, the Hepatosomatic Index and the adjusted dry weight of the digestive gland were significantly increased by 10% and by 9% respectively in shrimps treated with probiotic. In the meantime, the specific activities of alpha amylase and trypsin in the digestive gland showed a significant effect of the probiotic treatment with respective increases by 35% and 55%. The rise in total trypsin activity following morning feeding was also enhanced by the probiotic treatment (P < 0.001). The highest concentration of probiotic (between 10(4)-10(5) CFU g(-1) of fresh gut) in the shrimp gut was obtained 2 h after feeding. The concentration remained high for 4 h after feeding before decreasing until the next meal. Total bacterial counts on Marine agar and TCBS in the gut were significantly lowered by the probiotic treatment over the 10 weeks of the experiment. Additionally, during the first 2 weeks, prevalence and load of V nigripulchritudo strains in haemolymph was lower in animals fed with the probiotic diet. This study demonstrated, under pond conditions, that feeding shrimp with live terrestrial lactic acid bacteria can be an effective treatment for improving shrimp culture affected by vibriosis. Two oxygen fugacity ranges were investigated: close to the Ni-NiO (NNO) buffer and 2.6 log unit above NNO buffer (a dagger NNO + 2.6). At 1,100A degrees C, olivine is the liquidus phase and co-crystallized with phlogopite and augite at 1,000A degrees C for the whole range of investigated pressure and water content in the melt. At 900A degrees C, the mineral assemblage consists of augite and phlogopite, whereas olivine is not stable. The stability field of both alkali feldspar and plagioclase is restricted to low pressure (100 MPa) at nearly water-saturated conditions (< 3-4 wt.% H2O) and T < 900A degrees C. A comparison between experimental products and natural minerals indicates that mafic durbachites have a near-liquidus assemblage of olivine, augite, Ti-rich phlogopite, apatite and zircon, followed by alkali feldspar and plagioclase, similar to the mineral assemblage of minette magma. Natural amphibole, diopside and orthopyroxene were not reproduced experimentally and probably result from sub-solidus reactions, whereas biotite re-equilibrated at low temperature. The crystallization sequence olivine followed by phlogopite and augite reproduces the sequence inferred in many mica-lamprophyre rocks. The similar fractionation trends observed for durbachites and minettes indicate that mafic durbachites are probably the plutonic equivalents of minettes and that K- and Mg-rich magmas in the Bohemian Massif may have been generated from partial melting of a phlogopite-clinopyroxene-bearing metasomatized peridotite. Experimental melt compositions also suggest that felsic durbachites can be generated by simple fractionation of a more mafic parent and mixing with mantle-derived components at mid crustal pressures. Our models are set up with a viscous plate of silicone (lithosphere) subducting under negative buoyancy in a viscous layer of glucose syrup (mantle). For our study, the lithosphere/upper mantle viscosity contrast has been systematically varied, from similar to 10 to similar to 10(5) in order to explore the parameter space between weak and strong slab dynamics. We found that subduction is characterized by a retreating mode for viscosity ratios > 10(4), by the coexistence of a retreating mode and an advancing mode for viscosity ratios between similar to 10(4) and similar to 10(2), and quasi-stationary, Rayleigh-Taylor like behaviour for ratios < 10(2). By combining our experimental results and kinematic data from current subduction zones in four reference frames which differ in the amount of net rotation, we infer that a lithosphere/upper mantle viscosity contrast of 150-500 is necessary to obtain realistic trench/subducting plate velocity ratios as well as the variability of subduction styles observed in nature. To examine this transition in the context of Icelandic crust, we conducted deformation experiments on a glassy basalt (GB) and a glass-free basalt (GFB) under oceanic crust conditions. Mechanical and micro-structural observations at a constant strain rate of 10(-5) s(-1) and at confining pressure of 100-300 MPa indicate that the rocks are brittle and dilatant up to 700-800 degrees C. At higher temperatures and effective pressures the deformation mode becomes macroscopically ductile, i.e., deformation is distributed throughout the sample and no localized shear rupture plane develops. The presence of glass is a key component reducing the sample strength and lowering the pressure of the BDT. In the brittle field, strength is consistent with a Mohr-Coulomb failure criterion with an internal coefficient of friction of 0.42 for both samples. In the ductile field, strength is strain rate-and temperature-dependent and both samples were characterized by the same stress exponent in the range 3 < n < 4.2 but by very different activation energy Q(GB) = 59 +/- 15 KJ/mol and Q(GFB) = 456 +/- 4 KJ/mol. Extrapolation of these results to the Iceland oceanic crust conditions predicts a BDT at similar to 100 degrees C for a glassy basalt, whereas the BDT might occur in non-glassy basalts at deeper conditions, i.e., temperatures higher than 550 +/- 100 degrees C, in agreement with the Icelandic seismogenic zone. Entre préservation et/ou mise en valeur de la ressource, quel avenir pour les Aires Marines Protégées en Province Nord de la Nouvelle-Calédonie ? En effet, les écosystèmes marins sont essentiels d’autant plus, qu’ils présentent en Nouvelle-Calédonie une grande richesse, notamment en termes de patrimoine culturel. Ce sont majoritairement les Kanak qui habitent sur la Côte Est de la Province nord, où la Collectivité et le WWF collaborent à la mise en place de plusieurs aires marines protégées (AMP). Jusqu’à présent, la gestion coutumière Kanak a permis une relative préservation de ces écosystèmes dans un secteur littoral non soumis aux pressions de l’urbanisation, du tourisme ou de la navigation. L’étude s’attache à souligner les enjeux autour de la mise en place des AMP et des perspectives de développement local, sur le milieu marin et les populations locales. Il s’agit de voir comment cette expérimentation est envisagée par les différents acteurs (État, Province Nord, associations, coutumiers, etc.) et de s’interroger sur les mécanismes de gouvernance. This study allows a distinct approach to thinking about the marine resources in a protected area. Indeed, marine ecosystems are essential, especially as they are in New Caledonia, particularly in terms of cultural heritage. It is mainly Kanak who live on the East Coast of the North Province, where the Community and WWF collaborate on the installation of several marine protected areas (MPAs). So far, the Kanak customary management allowed a relative preservation of these ecosystems in a coastal area not subject to the pressures of urbanization, tourism and navigation. The study aims to highlight the issues surrounding the establishment of MPAs and opportunities for local development on the marine environment and local populations. This is to see how this experiment is considered by the various actors (State, Northern Province, associations, etc. customary). And to consider the mechanisms of governance. Effects of oceanic ridge subduction on accretionary wedges: Experimental modeling and marine observations Auteur(s) : E. The first experiment incorporates a massive ridge within a sand wedge. The wedge thickens and shortens when the forward propagation of the basal decollement ceases. The wedge thickening results in taper change, reactivation of preexisting thrusts, and retreat of the frontal part of the sand wedge. Similar mechanisms may have affected some margins that have undergone ridge subduction such as the Tonga margin after the subduction of the oblique Louisville oceanic ridge. The second experiment shows the effects of an active basement thrust slice as it enters a subduction zone. This process may have happened in the eastern Nankai accretionary wedge. Initially, the wedge in this experiment behaved similarly to that of the first experiment. Rapidly the topographic slope changed, the wedge thickened above the basement slice generating a slope break in the topography; a deeper propagating accretionary wedge again characterized by a small taper developed. These results, when compared with observations made in the Eastern Nankai Trench, are in agreement with the past subduction of a basement thrust slice in this area. Using specific searches of the Aquatic Sciences and Fisheries Abstracts ( ASFA) database from 1978 to December 2003 and a classical search of the pre-1978 literature, we collected 407 references on FADs. Publications before 1980 were predominantly peer-reviewed, although non-peer reviewed literature has dominated since 1980, due to the numerous technical reports produced as FADs became more widely used in artisinal and large-scale industrial. sheries in the 1980s. Most studies of the ecology of FAD-associated fish were descriptive, with few mensurative experimental studies and even fewer manipulative experimental studies that tested specific hypotheses, due to inherent difficulties in working in the open ocean on objects that are temporary in space and time. Publications presenting information on moored FADs outnumbered papers on drifting FADs by a ratio of 3.5: 1. That biomass triggers physical and chemical processes which greatly modify the durability and the sustainability of the sites. These processes, mainly from oxidative/reductive reactions, are poorly understood. This is mainly due to the fact that former studies were done at the macroscopic level far away from the micrometric scale where relevant processes induced by bacteria take place. Investigations at microscopic level are needed. Thus, we developed an experimental set-up based on the combined use of optical microscopy (transmission), atomic force microscopy (AFM) and electrical and electro-chemical AFM microscopy (EC_AFM) in order to get simultaneous information on topographic and electro-chemical processes. The first highly sensitive step was to use AFM with biological samples in liquid environment: wepresent here a study about AFM imaging of living, moving or self-immobilized bacteria, in their genuine physiological liquid medium and in true in vivo conditions. No external immobilization protocol, neither chemical nor mechanical was needed. For the first time, the native gliding movements of Gram negative Nostoc cyanobacteria upon the surface were studied by AFM. AFM height and mechanical stiffness data were simultaneously acquired. From these, mechanical parameters, inner turgor pressure and Young modulus, were derived for different bacterial species (Anabaenopsis circularis, Rhodococcus wratislaviensis). Our study revealed that AFM imaging is thus possible on moving living species. These results open a large window on new studies of both dynamical phenomena of practical and fundamental interests such as the formation of biofilms and dynamic properties of bacteria in real physiological conditions. The second delicate step was to combine AFM and optical measurements with electrical ones. We mounted a new experimental set-up coupling real-time (i) monitoring of optical properties as the optical density (OD) evolution related to bulk bacterial growth in liquid or as the counting of number of bacteria adhering on the surface of the sample as well and (ii) electrical and electrochemical measurements. Furthermore, these results will shortly be applied to the optimized monitoring of the in-situ activity of bacteria consuming oil pollutants, following this way, in real-time, the bioremediation of an oilcontaminated soil (ANR ECOTECH_BIOPHY program). La présence de matière biologique (biofilms) dans les sites de stockage géologique profond, d'éléments toxiques ou encore de l'eau potable des aquifères est maintenant clairement démontrée. Cette biomasse est à l'origine de processus physiques et chimiques qui modifient considérablement la durabilité et la pérennité des sites concernés. Ces processus, principalement de type oxydo-réductif, sont encore mal compris. Ceci est principalement dû aux méthodes d'investigation, principalement macroscopiques, loin de l'échelle micrométrique caractéristique des bactéries. Seules des études, basées sur des méthodes d'investigation locale, peuvent apporter les informations requises. Ainsi, nous avons développé un dispositif expérimental basé sur l'utilisation combinée de la microscopie optique (en transmission), la microscopie à force atomique (AFM) et la microscopie AFM en mode électrique et électrochimique (EC_AFM) afin d'obtenir des informations simultanées sur la topographie de l'échantillon et sur les processus électrochimiques à l'échelle des bactéries. La première étape sensible consistait à utiliser l'AFM sur des échantillons biologiques en milieu liquide: nous présentons ici les résultats de l'imagerie AFM en milieu liquide de plusieurs types de bactéries dans leurs conditions physiologiques naturelles (conditions in vivo). Aucun protocole d'immobilisation, ni chimique ni mécanique, n'a été nécessaire; et pour la première fois, les mouvements de reptation de cyanobactéries Nostoc ont été étudiés par l'AFM. Les études AFM ont permis d'acquérir des données topographiques mais aussi mécaniques : nous avons pu ainsi mesurer le module d'Young, la pression de turgescence de différentes souches bactériennes (Anabaenopsis circularis, Rhodococcus wratislaviensis). Cette étude complète, a révélé que l'imagerie AFM est donc possible sur des espèces vivantes en mouvement. Ces résultats ouvrent une grande fenêtre sur de nouvelles études d'intérêts tels que la formation de biofilms et les propriétés dynamiques de bactéries dans des conditions physiologiques réelles. La deuxième étape délicate était de combiner l'AFM aux mesures optiques et électriques. Nous avons développé un nouveau dispositif expérimental permettant (i) le suivi de l'évolution de la croissance bactérienne par la mesure des propriétés optiques comme la densité optique DO (pour le développement bactérien en volume - milieu planctonique) , ou l'analyse de l'image du substrat par comptage du nombre de bactéries sur la surface de l'échantillon (biofilm), et (ii) les mesures électriques et électrochimiques. Understanding the navigation capacities of the Eastern Caribbean pre-Columbian population is crucial for the community working in the Antilles. We have chosen to focus our work on an experimental approach of these techniques. Our ambition was to set up an experimental program including all the aspects associated with the building and the utilization of the kanawa. Thus, we have conducted an ethno-archaeological study of the kanawa building process followed by the Kali'ña from French Guyana. Then, the « Akayouman » kanawa was made into a 3D model to support a hydrostatic analyse. Finally, the heart of the project was composed of gathering and studying the data collected during our expeditions in the kanawa. Hemos optado por centrar nuestro trabajo en una practica experimental de estas técnicas. We tested the effects of depth and ultraviolet radiation on turf algae at different levels of successional stages using two experiments. Depth-related differences were found for all turf algal communities, characterized by a higher amount of the cyanobacteria taxon Dichothrix and the red filamentous genera Poly-/Herposiphonia in the shallow and the appearance of oscillating cyanobacteria in deeper waters. In the first experiment, cross-depth transplantation of 153 days old communities influenced percentage cover, biomass and taxa composition. Downward transplantation lowered overall biomass and abundance of the foraminifera Sorites, whereas the crustose green alga Pringsheimiella and filamentous cyanobacteria colonized the communities. A nearly reverse pattern was observed in upward transplanted communities. Overall we distinguished between sensitive taxa, like Oscillatoria, and taxa able to acclimate to alterations in their environment, like Pringsheimiella, Poly/Herposiphonia and Dichothrix. In the second experiment, algae grown for 285 days at 5 m were exposed together with a set of sterile settlement tiles to three UVR regimes at 2 m for 22 days. UVR had no effect on turf algal communities regardless of successional stage. This study highlights the presence of high light and UV tolerant species. The high UV tolerance of turf communities may confer a competitive advantage over other more sensitive coral reef biota, such as corals. This study demonstrates that turf algae are dynamic communities exhibiting species-specific resistance to environmental changes. Etude bactériologique des sédiments abyssaux. Etude de la dynamique des communautés microbiennes et de leurs activités hétérotrophes dans les parcs conchylicoles Auteur(s) : Bianchi, A. Résumé : Bacteriological study of abyssal sediments: During the Demeraby mission of the N/O Jean Charcot in the western Atlantic Ocean, a study of the microflora of the digestive tracts of echinoderms (Holothuroidea of various genera, Asteroidea, Echinoidea) was compared with that of the surrounding sediment. The bacterial concentration observed in the digestive tracts of all the animals studied is higher than that in the surrounding sediments. The largest numbers were always found in the most anterior part of the digestive tracts of the holothurians. In some cases, the increase is considerable (7.10 versus 5.2. 10 cells per millilitre): It indicates a proliferation of bacterial cells in the anterior part of the digestive tract. As the sediment moves through the digestive tract, the bacterial level progressively decreases. In the most posterior part, the concentration is, however, always clearly greater than that in the surrounding sediment. In the case of the holothurian of the genus Deima, analysis according to the Adamsonian principle of comparison shows that the microflora isolated from the various parts of the digestive tract is composed of taxonomic groups showing little resemblance to those isolated from the sediment. In the case of the holothurian of the genus Pseudostichopus, no specialisation of the microflora from digestive tract contents appears with respect to that of the ingested sediment. Examination of metabolic activities suggests a juxtaposition of two types of communities: Free sediment - sediment in transit that complement one other in their enzymatic and nutritional behaviour Indeed, the strains of free sediment seem to be rather suitable for hydrolysing and using carbohydrates, while those of enteric communities prefer the hydrolysis of lipids and proteins and the assimilation of fatty acids, dicarboxylic acids, hydroxylic acids and certain amino acids. Moreover, the enteric microflorae possess a greater nutritional versatility that, linked with its total numbers, has a sizeable catabolic potential. .../... -2- Measurements of heterotrophic activity done by means of 14 C glucose tracers make it possible to show that the metabolic activities of the bacterial communities of superficial abyssal sediments vary according to zones. In the Amazon Cone, a clear glucose mineralization gradient over a distance of a few kilometres can be observed. In the abyssal plain of Demerara heterotrophic activities are slowed in relation to those of the superficial sediments of the Cone of the Amazon, even though the bacterial numbers and the depths are of the same order of magnitude in the two zones. The abundant microflorae associated with the various divisions of digestive tract contents of the Deima abyssal holothurian manifest little metabolic activity as regards the glucose. However, the bacterial communities of Pseudostichopus and Psychropotes manifest very strong heterotrophic activities, increasing from the oesophagus towards the cloaca. Current results are too fragmentary to allow us to observe possible physiological specialisation of enteric bacterial communities enabling the facilitation of digestion processes in the host. Conclusion: In the abyssal environment, characterised by poorly developed bacterial communities, microflorae associated with the digestive tract contents of echinoderms have considerably greater cellular concentrations. By means of their enzymatic equipment, these enteric communities, which complement those of the free microflorae of superficial sediments, can participate in the transformation of organic matter in the abyssal area.
Study of the dynamics of microbial communities and their heterotrophic activities in shellfish farms: From experiments done on the water in the Arcachon basin, trials to quantify and describe the reactivity potential of bacterial communities were performed. Bacterial communities were defined by direct counts, counts after setting them up for cultivation, structure and diversity of these communities, dosage of AMP, ADP, ATP. Two opposite ecological conditions were considered: Autumn conditions (high temperature) and a winter conditions (low temperature). Tubs of sea water of 6 and 60 litres were enriched either with ammonium chloride or a mixture of amino acids, at about 500 uAtg N.L. The experiments lasted 24 hours, with sampling every 3 hours. The autumn community reacted rapidly (6 hours), the response to the enrichment in organic nitrogen being the highest. The structure of the community has unstable immature stages. On the contrary, during the winter experiment, no response could be perceived. This may mean that the latency phase is greater than 24 hours. The structure of this community has mature stages. In conclusion, bacterial communities react in the same way as macroscopic communities. Stable communities are diversified and correspond to mature stages of the ecosystem; the immature stages are unstable and less diversified. This type of experiment has made it possible to bring to light the rapidity of response of bacterial populations, and can be used to define the stability of a bacterial community in a precise biotope.
ETUDE BACTERIOLOGIQUE DES SEDIMENTS ABYSSAUX RESUME Au cours de la mission DEMERABY du n/o Jean CHARCOT en atlantique occidental , une étude de la microflore des tractus digestifs d'Echinodermes (Holothurides de différents genres, Astérides, Echinides) a été comparée à celle du sédiment environnant. La concentration bactérienne observée dans les tractus digestifs de tous les animaux étudiés est supérieure à celle des sédiments environnants. Les effectifs les plus importants sont toujours observés dans la partie la plus antérieure des tractus digestifs des holothuries. Dans certains cas, l'accroissement est considérable (7.10 contre 5,2. 10 cellules/ml) : il indique une prolifération des cellules bactériennes dans la partie antérieure du tractus digestif. Au.cours de la progression du sédiment dans le tractus digestif, la charge bactérienne décroit progressivement. Dans la partie la plus postérieure, la concentration est cependant toujours nettement supérieure à celle du sédiment environnant-. Dans le cas d'une holothurie du genre Deima, l'analyse selon le principe adansonien de comparaison montre que la microflore isolée des différentes parties du tractus digestif est constituée de groupes taxonomiques présentant peu de ressemblance avec ceux isolés du sédiment. Dans le cas d'une holothurie du genre Pseudostichopus il n'apparait pas de spécialisation de la microflore du contenu du tractus digestif par rapport à celle du sédiment ingéré. ¿ ¿ . L'étude des activités métaboliques suggère une juxtaposition de deux types de communautés : sédiment libre-sédiment en transit qui se complètent dans leur comportement enzymatique et nutritionnel. En effet les souches du sédiment libre semblent être plutôt aptes à hydrolyser et à utiliser les glucides, tandis que celles des communautés entériques portent leur préférence sur l'hydrolyse des lipides et protéines et 1'assimilation des acides gras, acides dicarboxyl iques, hydro'xy-acides et certains acides aminés. D'autre part les microflores entériques possèdent une versatilité nutritionnelle plus importante qui, liée à leurs effectifs, offre une potentialité catabolique non négligeable. .../... -2- Les mesures d'activité hétérotrophe effectuées au moyen de traceurs glucose 14 C permettent de montrer que les activités métaboliques des communautés bactériennes des sédiments abyssaux superficiels varient selon les zones. Dans le Cone de l'Amazone on observe un net gradient de minéralisation du glucose sur une distance de quelques kilomètres. Dans la plaine abyssale de Demerara les activités hétérotrophessont ralenties par rapport à celles des sédiments superficiels du Cone de l'Amazone, bien que les effectifs bactériens et les profondeurs soient du même ordre de grandeur dans ces deux zones. Les abondantes microflores associées aux différentes fractions du contenu du tractus digestif de l'holothurie abyssale Deima ne manifestent que peu d'activités métaboliques vis à vis du glucose. Par contre les communautés . bactériennes de Pseudostichopus et P syahropotes manifestent des activités hétérotrophes très fortes, croissant de 1'oesophage vers le cloaque. Les résultats actuels sont trop fragmentaires pour permettre d'observer une éventuelle spécialisation physiologique des communautés bactériennes entëriques permettant de faciliter les processus de digestion chez l'hôte. Conclusion: dans le milieu abyssal, caractérisé par des communautés bactériennes faiblement développées, les microflores associées aux contenus de tractus digestif des Echinodermes présentent des concentrations cellulaires considérablement plus importantes. Par leurs équipements enzymâtiques ces communautés entëriques complémentaires de ceux des microflores libres des sédiments superficiels, peuvent participer à la transformation des matières organiques du domaine abyssal. ETUDE DE LA DYNAMIQUE DES COMMUNAUTES MICROBIENNES ET DE LEURS ACTIVITES HETEROTROPHES DANS LES PARCS CONCHYLICOLES RESUME A partir d'expérimentations effectuées sur l'eau du bassin d'Arcachon, des essais de quantification et qualification du potentiel de réactivité des communautés bactériennes ont été effectuées. Les communautés bactériennes ont été définies par comptage directs, comptages après mise en culture, structure et diversité de ces communautés, dosage des AMP, ADP, ATP. Deux situations écologiques opposées ont étées considérées . Une situation automnale (température élevée) et une situation hivernale (température faible). Des bacs d'eau de mer de 6 et 60 litres ont été enrichis , soit avec du chlorure d'ammonium, soit avec un mélange d'acides aminés, à environ 500 uAtg N.l" . Les expériences ont duré 24 heures, avec des prélèvements toutes les 3 heures. La communauté automnale réagit rapidement (6 heures), la ' réponse à l'enrichissement en azote organique étant la plus importante. La structure de la communauté présente des stades immatures instables. Au contraire, au cours de l'expérimentation hivernale, aucune réponse n'a pu être perçue. Ceci peut signifier que la phase de latence est supérieure à 24 heures. La structure . de cette communauté présente des stades matures. En Conclusion, les communautés bactériennes réagissent de la même façon que les communautés macroscopiques. Les communautés stables sont diversifiées et correspondent à des stades matures de l'écosystème, les stades immatures sont instables et moins diversifiés. Ce type d'expérience a permi de mettre en évidence la rapidité de réponse des populations bactériennes, et peut être utilisé pour définir la stabilité d'une communauté bactérienne dans un biotope précis.
