We compared PD of tree communities growing (1) on substrates of varying geological age; and (2) in environments with varying ecophysiological barriers to growth and survival.Results PDss is strongly positively correlated with species richness (SR), whereas MNTD has a negative correlation. Communities on geologically young- and intermediate-aged substrates (western and central Amazonia respectively) have the highest SR, and therefore the highest PDss and the lowest MNTD. We find that the youngest and oldest substrates (the latter on the Brazilian and Guiana Shields) have the highest ses.PDss and ses.MNTD. MPD and ses.MPD are strongly correlated with how evenly taxa are distributed among the three principal angiosperm clades and are both highest in western Amazonia. Meanwhile, seasonally dry tropical forest (SDTF) and forests on white sands have low PD, as evaluated by any metric.Main conclusions High ses.PDss and ses.MNTD reflect greater lineage diversity in communities. We suggest that high ses.PDss and ses.MNTD in western Amazonia results from its favourable, easy-to-colonize environment, whereas high values in the Brazilian and Guianan Shields may be due to accumulation of lineages over a longer period of time. White-sand forests and SDTF are dominated by close relatives from fewer lineages, perhaps reflecting ecophysiological barriers that are difficult to surmount evolutionarily. Because MPD and ses.MPD do not reflect lineage diversity per se, we suggest that PDss, ses.PDss and ses.MNTD may be the most useful diversity metrics for setting large-scale conservation priorities. Étude géologique et géophysique des marges continentales passives : exemple du Zaïre et de l'Angola Auteur(s) : Moulin, Maryline Éditeur(s) : Université de Bretagne Occidentale Résumé : The objective of the present work is to study the formation of the passive continental margins of the Central Segment of the South Atlantic, most particularly the Congo and Angola margins. We propose a combined approach, which integrates structural constraints based on geological cross-sections (based on seismic data) and global constraints based on plate kinematic reconstructions. The structural study is based on : i) MCS and refraction data collected during the ZaiAngo programme (a joint project conducted by Ifremer and Total) ; ii) proprietary, industrial seismic data (courtesy of Total) from the Angola margin and iii) on all available seismic lines from the Africa and Brazil conjugated margins, between Walvis Ridge and the Equatorial Fracture Zones. Based on theses data, three structural domains (continental, transitional and oceanic) have been defined, the major characteristics of which are : Crustal thinning occurs abruptly, mostly below the continental slope, over a lateral distance of less than 50 km. The top of the crust deepens as the Moho shallows. Only a few extensional structures are observed ; tilted blocks are very few (one or two, depending on the profile), found only on the upper part of the slope and sealed by a discordance prior to salt deposition. The transitional domain is characterized by the existence of a pre-salt basin lying over a thin crustal layer. No tilted blocks are observed in this domain and reflectors within the pre-salt sediment series are parallel to the base of the Aptian salt, over distances greater than 100 km, precluding the possibility of any significant deformation that would imply large horizontal motions. Two types of crust are observed in the transitional domain. "Type I" crust is found below the undeformed pre-salt sediment series located below the eastern part of the basin ; it is characterized by an upper layer of thickness greater than 5 km and a abnormal velocity layer (7.2 - 7.6 km/s), up to 6 km thick. "Type II" crust is less than 5 km thick and found below the salt compressive front that affects the western part of the basin. The salt cover is continuous (no erosion surface is observed), from the continental shelf to the western termination of the basin. Salt was not deposited in a confined environment (like in the Mediterranean), but in a shallow water, lagunal environment. This imposes the zero-level and constrains the paleo-bathymetry at the time of salt deposition, which dates the latest stage of margin formation. Understanding the formation of a margin cannot be approached without studying the homolog margin. Therefore, it is of major importance to reconstruct the closure of the ocean bordered by these homolog margins and take into account the constraints imposed by the kinematic reconstructions on the lateral motions of the lithospheric plates. In order to assess the relative position of the plates at the ocean closure (prior to crustal thinning), a global study was thus performed, integrating all geophysical and geological constraints, in the ocean and on land. The role of african intra-plate deformation and its limits and their consequences have been thoroughly studied. To juxtapose the margins of the central segment of the Southern Atlantic, it is all the margins bordering the Equatorial Atlantic that need to be adjuste precisely. The kinematic study of this last region shows that the reconstruction obtained are reliable, unambiguous with a quantifiable precision The best fitting poles (obtained using the PLACA software), show that it is impossible to close the margins beyond the superposition of the salt fronts, from the Angola and Brazil margins. The geological cross-sections based on seismic data from the homolog margins indicate that a 330 km wide basin with thin (< 12 km) crust was present at the time of the fit. This basin cannot result from horizontal movement related to pure stretching or simple shear, or any model implying conservative volume. This conclusion is consistent with the existence of presalt reflectors parallel to the salt layer wich extends to the platform: the formation of the pre-salt basin must be related to vertical motions. The scenario that we propose for the evolution of the Congo-Angola margin consist in four stages: the first phase corresponds to extensional deformation limited to the few tilted blocks observed on the upper part of the slope. During the second phase, the main crustal thinning occurs, vertical motions prevailes, resulting in the formation of the continental slope and in the subsidence of the basin. The third phase corresponds to the first stress striction: deformation is concentrated in a limited section of the basin, which corresponds to the salt compression front. A proto-oceanic crust is formed, probably composed of thinned continental crust intruded by mantle material. The second stress striction corresponds to the finale phase, resulting in oceanisation senso stricto. The evolution described shows that we can not apply conservative models for margin formation (such as McKenzie and Wernicke or any of their avatars). In order to explain this thinning, one should investigate non-conservative models (implying geochemical transformation, small scale convection, intrusion...) such as those proposed in marginal or continental basins with no horizontal movments. Ce travail de thèse aborde la formation des marges continentales passives dans le segment central de l'océan Atlantique Sud (plus particulièrement au Congo et en Angola), en intégrant une étude en coupe (étude structurale à partir des coupes sismiques) et une étude en plan (étude cinématique). L'étude structurale de la marge a été réalisée à partir des données de sismique réflexion et réfraction de la campagne Zaïango et d'une compilation de données sismiques réflexion existantes sur toutes les marges africaine et brésilienne entre les zones de fracture équatoriales et la ride de Walvis. L'interprétation de ces données a permis d'individualiser la structure de la marge en trois domaines : continental, transitionnel et océanique et de déterminer quelques points majeurs sur la structuration de la marge. L'amincissement est abrupt, localisé dans la zone de pente continentale et restreint à 50 km. La marge montre peu de structures distensives : seuls un ou deux blocs basculés sont observés en haut de pente continentale. Le domaine transitionnel est caractérisé par la géométrie particulière de la sédimentation anté-salifère, l'absence de blocs basculés et la faible épaisseur de croûte. La couche sédimentaire anté-salifère montre des réflecteurs plans jusqu'à la base du sel, continus sur 100 km, éliminant toutes possibilités de déformation du socle pendant et après son dépôt. La croûte du domaine transitionnel peut-être divisée en deux types : une croûte de type I sur laquelle se déposent les sédiments non déformés, et une croûte de type II sur laquelle se superposent les limites du « front compressif salifère » bien exprimé dans les séries postsalifères. Enfin le sel, que l'on observe depuis la plate-forme jusqu'au bassin profond, ne se dépose pas dans un bassin confiné (comme en Méditerranée) mais à un niveau proche de 0 m (ressemblant probablement à un dépôt de type lagunaire) et donne la paléo-bathymétrie au moment de son dépôt qui marque la fin de la période de formation de la marge. La compréhension de la genèse d'une marge ne peut être approchée sans son homologue. Cette simple constatation, cette évidence, montre toute l'importance que l'on doit apporter à la reconstruction cinématique initiale de l'océan qui borde ces marges homologues et aux contraintes imposées par les reconstructions cinématiques sur les mouvements horizontaux des plaques lithosphériques. Afin d'étudier la position des marges au moment de cette fermeture, c'est-à-dire avant amincissement, une étude globale intégrant l'ensemble des données disponibles, géophysiques et géologiques, océaniques et continentales, a été réalisée. Le rôle de la déformation intraplaque africaine, ses limites et leurs conséquences a, en particulier, été l'objet d'une attention poussée. Pour juxtaposer les marges du segment central, ce sont toutes les marges de l'océan Atlantique Equatorial qui doivent être ajustées précisément. L'étude cinématique réalisée de la région équatoriale montre que l'on obtient une reconstruction fiable et sans ambiguïté, avec une précision que l'on peut quantifier. Les pôles issus de cette étude (et calculés avec le Logiciel PLACA) indiquent qu'il est impossible d'obtenir une fermeture plus serrée que celle qui conduit à la superposition des fronts salifères brésilien et angolais : les coupes issues de la sismique réflexion des deux marges indiquent qu'il subsiste un bassin aminci, large de plus de 330 km et dont la croûte n'excède jamais 13 kilomètres d'épaisseur. La formation de ce bassin ne peut résulter de mouvements horizontaux, ce qui exclut un amincissement par étirement (pure stretching) ou par l'existence d'une faille de détachement (simple shear) ou par quelque modèle conservatif que ce soit. Cette constatation corrobore l'observation de la présence d'horizons anté-salifère parallèles, entre eux et au sel, couche salifère que l'on retrouve sur la plate-forme : la création de ce bassin anté-salifère ne peut être que liée à un mouvement vertical. Le schéma d'évolution que nous proposons à partir des données structurales et des contraintes cinématiques présente quatre étapes : le premier stade correspond à une phase de déformation distensive limitée aux quelques rares blocs basculés observés en haut de pente continentale. C'est durant la deuxième étape que se déroule la phase d'amincissement principal, les mouvements verticaux prévalent, aboutissant à la formation de la pente continentale et à la subsidence du bassin. La troisième phase correspond à une première striction des contraintes : la déformation se concentre sur une partie réduite du bassin, coïncidant avec le front salifère compressif. Une proto-croûte océanique se forme, probablement composée de croûte continentale amincie et intrudée de matériel mantellique. La seconde striction correspond à la phase finale de formation de la marge et aboutit à l'océanisation sensu stricto. L'étude cinématique et la description de l'évolution de la marge à partir des données sismiques montre donc que l'on ne peut envisager l'application d'un modèle de genèse des marges avec conservation de volume (type McKenzie ou Wernicke et leurs avatars) : pour expliquer l'amincissement du bassin, il faudrait probablement nous intéresser aux modèles non-conservatifs (impliquant transformation, convection à petite échelle, ...) qui sont déjà invoqués pour la formation des bassins marginaux ou continentaux, sans mouvements horizontaux. Ce texte aborde deux types de réserves de conservation environnementale: une « réserve extractiviste » conquise par des populations rurales riveraines au confluent des fleuves Xingu et Amazone; et une « réserve marine» de populations rurales côtières – les deux à l’État du Pará. Au Brésil comme en Europe, ces initiatives ont visé un meilleur mode de vie et le développement des populations locales et, éventuellement, par surplus, régionales ou nationales. Au Brésil, ces objectifs ont été liés au droit à l’usage collectif des forêts, des bois, des cours d’eau et des mangroves. À des degrés différents, ces territoires sont issus de luttes pour la défense des ressources qui étaient la source de la subsistance de leurs habitants, de leur mode de vie et culture, en un mot, leur patrimoine – contre la pénétration d’« étrangers » venus d’ailleurs en dépit de la loi et de la légitimité: entreprises du bois, de grands élevages et de pêche. Ce texte cherche à confronter, au niveau de l’analyse sociologique, des intérêts de groupes, des acteurs, des stratégies des uns et des autres et de leurs perspectives pour les actuelles et futures générations. This article refers to what some writers call, in Brazil, "new territories". Since the debates during the Federal Constitution elaboration, in 1988, they have been implemented, wholly or partly, within the jurisdiction of a state, a municipality, or a district. This paper addresses two types of environmental conservation reserves: an "extractive reserve" conquered by rural people at the confluence of the rivers Xingu and Amazon; and a "marine reserve" of rural populations on the northern coast of Brazil - both in the federal state of Pará. In Brazil as in Europe, these initiatives aim at better living standards and collective development of local populations. In Brazil, these have been linked to the respect of their collective rights to use the forests, wood, water streams and mangroves. To varying degrees, these territories result from struggles in defense of natural resources that were source of livelihood, lifestyle and culture of the inhabitants, in short, their heritage – against the invasion of "foreigners", timber companies, large farms and fishers. This text contrasts, through sociological analysis, the interest groups, the stakeholders, the challenges and their development prospects vis-à-vis the present and future generations. Comparison between three implementations of automatic identification algorithms for the quantification and characterization of mesoscale eddies in the South Atlantic Ocean Auteur(s) : Souza, Joao Marcos Azevedo Correia De De Boyer Montegut, Clement Le Traon, Pierre-yves Éditeur(s) : Copernicus Gesellschaft Mbh Résumé : Three methods for automatic detection of mesoscale coherent structures are applied to Sea Level Anomaly (SLA) fields in the South Atlantic. The first method is based on the wavelet packet decomposition of the SLA data, the second on the estimation of the Okubo-Weiss parameter and the third on a geometric criterion using the winding-angle approach. The results provide a comprehensive picture of the mesoscale eddies over the South Atlantic Ocean, emphasizing their main characteristics: amplitude, diameter, duration and propagation velocity. Five areas of particular eddy dynamics were selected: the Brazil Current, the Agulhas eddies propagation corridor, the Agulhas Current retroflexion, the Brazil-Malvinas confluence zone and the northern branch of the Antarctic Circumpolar Current (ACC). For these areas, mean propagation velocities and amplitudes were calculated. Two regions with long duration eddies were observed, corresponding to the propagation of Agulhas and ACC eddies. Through the comparison between the identification methods, their main advantages and shortcomings were detailed. The geometric criterion presents the best performance, mainly in terms of number of detections, duration of the eddies and propagation velocities. The results are particularly good for the Agulhas Rings, which have the longest lifetimes of all South Atlantic eddies. Conflicting structural and geochronological data from the Ibituruna quartz-syenite (SE Brazil): Effect of protracted "hot" orogeny and slow cooling rate? We have measured the Anisotropy of Magnetic Susceptibility (AMS) in samples from the Ibituruna sill to unravel its magnetic fabric that is regarded as a proxy for its magmatic fabric. A large magnetic anisotropy, dominantly due to magnetite, and a consistent magnetic fabric have been determined over the entire Ibituruna massif. The magmatic foliation and lineation are strikingly parallel to the solid-state mylonitic foliation and lineation measured in the country-rock. Altogether, these observations suggest that the Ibituruna sill was emplaced during the high temperature (similar to 750 degrees C) regional deformation and was deformed before full solidification coherently with its country-rock. Unexpectedly, geochronological data suggest a rather different conclusion. LA-ICP-MS and SHRIMP ages of zircons from the Ibituruna quartz-syenite are in the range 530-535 Ma and LA-ICP-MS ages of zircons and monazites from synkinematic leucocratic veins in the country-rocks suggest a crystallization at similar to 570-580 Ma, i.e., an HT deformation >35My older than the emplacement of the Ibituruna quartz-syenite. Conclusions from the structural and the geochronological studies are therefore conflicting. A possible explanation arises from Ar-40-Ar-39 thermochronology. We have dated amphiboles from the quartz-syenite, and amphiboles and biotites from the country-rock. Together with the ages of monazites and zircons in the country-rock, Ar-40-Ar-39 mineral ages suggest a very low cooling rate: <3 degrees C/My between 570 and similar to 500 Ma and similar to 5 degrees C/My between 500 and 460 Ma. Assuming a protracted regional deformation consistent over tens of My, under such stable thermal conditions the fabric and microstructure of deformed rocks may remain almost unchanged even if they underwent and recorded strain pulses separated by long periods of time. This may be a characteristic of slow cooling "hot orogens" that rocks deformed at significantly different periods during the orogeny, but under roughly unchanged temperature conditions, may display almost indiscernible microstructure and fabric. Public use and Ecotourism Policies in Brazilian and Canadian Protected Areas Auteur(s) : Matheus, Fabricio Scarpeta Raimundo, Sidnei Éditeur(s) : Université des Antilles Études caribéennes Résumé : Brazil and Canada have two of the biggest protected area systems in the world, both of them based in the protected areas model established by the USA in the XIX century. However, they have distinct policies when it comes to public use and ecotourism inside those areas. Canadian guidelines have a greater focus on leisure and recreation activities, in a more permissive use of its resources with activities geared to large scale tourism. On the other hand, Brazil has a focus more limited on low-impact activities with a strong component of environmental education and restrictions of activities within its protected areas. From that approach, this article analyzes the Brazilian and the Canadian public policies for public use development inside protected areas and how they address the three main aspects of ecotourism: environmental conservation, environmental awareness of visitors and involvement of local community. Brésil et le Canada ont deux des plus grands réseaux d´aires protégées du monde, tous les deux basés dans le modèle d’aires protégées établi par les États-Unis au XIXe siècle. Cependant, ils ont des politiques distinctes quand il s´agit de l'usage du public et de l'écotourisme à l'intérieur de ces zones. Les lignes directrices canadiennes ont une plus grande focalisation sur les activités de loisirs et de détente, dans une utilisation plus permissive de ses ressources à des activités visant à grande échelle le tourisme. D'autre part le Brésil a un objectif plus limité sur les activités à faible impact avec une forte composante de l'éducation et les restrictions d'activités au sein de ses aires protégées de l'environnement. A partir de cette approche, cet article analyse les politiques publiques brésiliennes et canadiennes pour le développement du ecotourisme à l'intérieur d’aires protégées et de la façon dont ils abordent les trois principaux aspects de l'écotourisme: conservation de l'environnement, education environmental et la participation de la communauté locale. Réalité socio-spatiale en marges à toutes les échelles malgré le différentiel socio-économique fort qu’elle marque, la frontière franco-brésilienne est devenue, depuis les années 1990, une interface politique, support de la relance des relations internationales franco-brésiliennes et des relations entre Guyane et Amapá.La thèse pose la question de cet écart en se concentrant sur la partie estuarienne de la frontière fluviale de l’Oyapock, autour des bourgs riverains de Saint-Georges (Guyane française) et d’Oiapoque (Brésil), entre lesquels un pont binational d’envergure est en construction, malgré leur rattachement routier à la marge aux polarités régionales de Cayenne et de Macapá. Brazil and France share a common border of over 700 km via French Guiana, a French Overseas monodepartmental region, and Amapá, State of the Federal Republic of Brazil. Socio-spatial margin at all scales despite the strong socio-economic differential it marks, the Franco-Brazilian border has become since the 1990s a political interface, support for the revival of the Franco-Brazilian international relations and of the relations between French Guiana and Amapá. The thesis raises the question of this gap, focusing on the estuarine portion of the border river of the Oyapock, around the towns of Saint-Georges (French Guiana) and Oiapoque (Brazil) ; a significant bi-national bridge is under construction between these two towns, despite their road connection as margins to the regional polarities of Cayenne and Macapá. 1914-2014, un siècle d’anthropophagie féminine dans l’art brésilien : pertinence et actualité ? ; 1914-2014, a century of female anthropophagy in brazilian art : relevance and topicality ? Cette modernité se démarque de celle d’Europe par la prise de conscience des distances géographiques, culturelles et politiques vis à vis de l’Europe et surtout de l’ancienne puissance coloniale, le Portugal, par la recherche de son identité à travers la multiplicité des métissages du Nouveau Monde, ses mythes et son histoire réelle décolonisée. L’avant-garde brésilienne émerge autour de 1928, à travers les Manifestes anthropophages d’Oswald de Andrade (1890-1954) qui est donc le fondateur des anthropophagies, que l’on peut définir comme un indianisme à rebours. Le ″mauvais sauvage″ exerce sa critique contre les impostures du monde. « La ″Descente anthropophagique″ n’est pas une révolution littéraire, ni sociale, ni politique, ni religieuse. Elle est tout cela à la fois ! Sa loi est simple : Ne m’intéresse que ce qui n’est pas à moi ! Loi de l’homme, loi de l’anthropophagie ! » . Elle prescrit donc la dévoration des modèles importés et leur digestion dans l’hybridation au nom de l’identité brésilienne, par déplacement de concepts freudiens : « L’anthropophagie c’est la transformation permanente du Tabou (manger de l’homme) en Totem (de l’identité brésilienne) ! » . Ici l’influence notable de la psychanalyse et de l’anthropologie est à resituer : le déplacement du tabou anthropophage demeure une transgression symbolique, une métaphore, mais la référence anthropophagique ne concerne pas que la période précolombienne, car elle se réactualise. Nous nous proposons d’étudier ce phénomène à travers quatre questions : 1°- Quelles relations existent entre l’anthropophagie, l’histoire, l’esthétique et l’idéologie ? 2°- Quelle est la place des femmes artistes brésiliennes dans l’émergence de ce mouvement, compte tenu de leur présence décisive dès l’origine ?3°- Compte tenu de la résurgence rhizomique de l’anthropophagie dans la 2e moitié du XXe siècle, y compris jusqu’à aujourd’hui, quelle est la place des femmes artistes dans ce phénomène ? Y a-t-il continuité avec l’époque fondatrice ?4°- Etant donné ce qui précède, peut-on déduire qu’il existe un courant spécifiquement féminin dans l’anthropophagie d’hier et d’aujourd’hui ? Quelle est son importance réelle ? Quelles sont ses relations et postures par rapport à la postmodernité et à la mondialisation de l’art contemporain ? At the beginning of the twentieth century, in Brazil, a radical artistic rupture took place, which marked the beginning of a new era. The resulting modernity differed from its European counterpart by the awareness of geographical, cultural and political distances that alienated Brazil from Europe and more specifically from its former colonial owner, Portugal. Brazilian modernity sought to define its identity through important basic elements that constitute the stuff that the New World is made of: cross-breeding, mythology and post-colonialism. The Brazilian avant-garde emerged around 1928 with the publication of The Anthropophagy Manifesto by Oswald de Andrade (1890-1954), who is thus the founder of the Anthropophagy, that we can define as a backward step into a reinvented form of ″Amerindianness″. The ″bad savage″ voices his criticism against impostures of the world. « Anthropophagy art is not a literary revolution, nor is it a social plea, nor a political pamphlet, nor a religious tract. It is all these things at the same time. Its law is simple: everything that is not me is of interest to me. The law of men is the law of Anthropophagy ». It thus prescribes eating up imported models and digesting them through the process of hybridization in the name of Brazilian identity. By displacing Freudian concepts, «Anthropophagy is the permanent transformation of the Taboo (man-eating) into a Totem (Brazilian identity) ». The permeating influences of psychoanalysis and anthropology need to be put in perspective: the displacement of the anthropophagous taboo remains a symbolic act of transgression, a metaphor, but the anthropophagic reference does not concern the pre-Columbian period, because it is updated. We propose to study this phenomenon through four questions: 1°- What are the relations between Anthropophagy, history, esthetics and ideology? 2°- What is the place of women Brazilian artists in the emergence of this movement, taking into account their decisive presence right from the start? 3°- Taking into account the resurgence of Anthropophagy from the second half of the 20th century onwards, what is the place of women artists in this phenomenon? Are they pursuing the same interests as the founders of the movement?4°- Can we deduce that there exists a specifically female genre within the anthropophagic movement of yesterday and of today? If so, what is its relevance? Analyse économique d'une nouvelle technique d'élevage de crevettes en cages flottantes au Brésil Auteur(s) : Paquotte, Philippe Éditeur(s) : Actes de la 8ème Conférence de l'IIFET (Marrakech, juillet 1996) Résumé : In the framework of French-Brazil co-operation, the French Institute of research for exploitation of the sea (IFREMER), the State of Bahia and the company Littoral Sul Maricultura have developed a new technique for shrimp farming (Penaeus vannamei) using floating cages. After trials carried out during three years, this technique turns out very interesting from several viewpoints. The growth speed and the survival rate are very good, the negative effects on the environmental quality are weak and local fishermen have proved eager to adopt this technique as a part-time activity. In the same time, a technico-economic analysis, using a computerised simulation tool, as well as a market analysis in the State of Bahia have been carried out in order to assess the feasibility of such a project and to help research and development decisions. Despite low first investment and good biological results, the ex-farm production cost of shrimps weighing 15 g is as high as 4,5 US$ per kilo. This price is due to the low productivity of the labour and to the cost of nets which have a short life-time. It is possible to consider this activity as profitable on the market of Salvador de Bahia. Nevertheless, the question is how to face the international competition, which is particularly strong in the case of tropical shrimps. The conditions allowing enterprises to develop a strategy based on product quality and environmentally friendliness are reviewed. [NOT CONTROLLED OCR] Dans le cadre de la coopération Franco-Brésilienne, l'IFREMER, l'Etat de Bahia et la société Littoral Sul Maricultura ont développé une nouvelle technique d'élevage de crevettes pénéides (Penaeus vannamei) en utilisant des cages flottantes. Après trois années de mise au point, cette technique s'est avérée intéressante sur plusieurs aspects. La vitesse de croissance et le taux de survie sont très satisfaisants, les effets négatifs sur la qualité du milieu naturel sont faibles et les pêcheurs locaux ont accueilli favorablement cette nouvelle pratique en complément de leur activité traditionnelle. Parallèlement, une analyse technico-économique utilisant un outil de simulation informatique ainsi qu'une analyse du marché de la crevette dans l'état de Bahia ont été conduites afin d'évaluer la faisabilité d'un tel projet et d'orienter les actions de recherche et développement. En dépit d'investissements initiaux plus faibles que dans les élevages en bassins et de bonnes performances zootechniques, le coût de production de crevettes de 15 g atteint 4,7 US$ par kilo. Dans les conditions de marché de l'Etat de Bahia, la rentabilité d'un projet d'élevage de crevettes en cage serait assurée, mais la faible productivité du travail et le coût de remplacement des filets ne permettent pas d'avoir des coûts de production aussi bas que dans les élevages en bassins. C'est pourquoi la question se pose de la possibilité pour ce type de production innovante d'affronter une concurrence internationale très forte comme dans le cas des crevettes tropicales. To constrain strain distribution in a pervasively molten crust, we analysed the deformation in an eroded analogue of these orogens. The Ribeira-Araçuaí orogen (SE Brazil) comprises a stack of allochthons containing large volumes of anatectic and magmatic rocks. The upper allochton (∼300 km long, 50–100 km wide and >10 km thick) involves peraluminous diatexites and leucogranites resulting from partial melting of the middle crust. It overlies another allochthon containing huge early- to syn-collisional plutons intruding metasediments. Both anatexites and magmatic intrusions display a pervasive strain-induced magmatic fabric. Homogeneous strain distribution suggests inefficient localization. U–Pb ages of ∼575 Ma imply that anatexite melting was synchronous to the early- to syn-collisional magmatism. Similarity in ages magmatic and solid-state fabrics indicates that intrusions and anatexites deformed coherently with solid-state rocks while still molten, in response to a combination of gravity-driven and collision-driven deformation. Shear wave splitting in SE Brazil: an effect of active or fossil upper mantle flow, or both? Measurements were performed on seismic data recorded in the Ribeira and Brasilia Neoproterozoic belts, which wrap around the southern termination of the São Francisco craton and disappear westward under the Paraná basin. In the northern Ribeira belt, dominated by thrust tectonics, the fast shear wave polarization planes trend on average N080°E, whereas in the central domain, dominated by strike-slip tectonics, fast shear waves are polarized parallel to the structural trend (N065°E). Stations located above the main transcurrent fault display large delay times (> 2.5 s). Such values, among the largest in the world, require either an unusually large intrinsic anisotropy frozen within the lithosphere, or a contribution from both the lithospheric and asthenospheric mantle. Within the southern Brasilia belt, fast split shear waves are polarized parallel to the structural trend of the belt, at a high angle from the APM. Although part of our data set strongly favors an origin of anisotropy related to a fabric frozen in the lithospheric mantle since the Neoproterozoic, a contribution of the asthenospheric flow related to the present day plate motion is also required to explain the observed splitting parameters. V. Jean Charcot (1973) several marine geophysical lines, including bathymetry, magnetism, continuous seismic as well as sonobuoy profiles have been run across a deep marginal plateau extending on the southeastern Brazilian margin and known as the Sao Paulo Plateau. These data indicate the presence of a thick sedimentary cover (more than 4s DTT) on the upper slope and up to 3s DTT thick on the lower slope and rise. The entire slope is highly disturbed by salt diapiric structures mostly apparent between the 2 000-3 000 bathymetriclines and comparable to the ones described in the western Mediterranean sea. The Sao Paulo scarp, bordering the deep marginal plateau to the South-West, is believed to be a discontinuous and "en échelon» structure probably related to numerous and closely spaced early fracture zones. The overal structures Of two areas - the Sao Paulo and the Angolan margin - are briefly cOmpared and Preliminary results of DSDP holes 356 and 364 suggest a similar sedimentary history, during mesozoic times, for both margins. Despite the ethnopharmacological significance of the species, no phytochemical or cytotoxic studies of its latex have been published. In this context, the aim of the present study was to assess the cytotoxic activity of crude extracts, fractions and isolated compound from the latex of C. urucurana and to investigate the effects of crourorb A1 on the viability, apoptosis, cell cycle and cell migration of human hepatocarcinoma cell lines (Huh-7). Also, we have evaluated the viability of Huh-7 cells treated with crourorb A1 in 3D collagen gels. The cytotoxicity was detected using the sulforhodamine B (SRB) assay against six human cancer cells lines and one nontumor NIH/3T3 cell line (murine fibroblast). Studies on the mechanism of cell death of crourorb A1 were conducted in different test systems in vitro. The bioactive ethyl acetate phase obtained from the latex of C. urucurana afforded a novel cyclic peptide, [1−9-NαC]-crourorb A1, that proved active against NCIADR/ RES (ovary, multidrug-resistance phenotype) cells with the same potency as doxorubicin (positive control) and inactive up to the highest concentration tested against nontumor NIH/3T3 cells (GI50 = > 250 μg/mL). Crourorb A1 treatment decreases the cell viability of Huh-7 at dose and time dependent manner associated with induction of apoptosis by caspase 3/7 activation and increasing expression of pro-apoptotic proteins: Bak, Bid, Bax, Puma, Bim, and Bad and clived-caspase3 protein. The JNK/MAP pathway is involved in this induced cell death. Crourorb A1 effect is also associated with G2/M phase cell cycle arrest. In parallel, we observed that crourorb A1 treatment resulted in an increase of cyclin-dependent kinase (CDK1), cyclin B1 and cyclin D1 expressions. A significant reduction of Huh-7 cells migration by crourorb A1 can be assessed and we also show that cells grown on 3D collagen gels exhibited increased resistance to crourorb A1 when compared to 2D cell culture. Croton urucurana Baillon (Euphorbiaceae), est une plante très connue et utilisée en médecine traditionnelle au Brésil, la «sangra-d’água» retrouvée dans l'État du Mato Grosso do Sul (région Centre-Ouest du Brésil). Son latex est abondamment utilisé en médecine traditionnelle dans le traitement de plusieurs types de cancer. En dépit de son importance ethnopharmacologique, on ne trouve aucune étude sur sa composition chimique ou sur sa potentielle activité anti-cancéreuse. L’objectif de ce travail a été d'évaluer l'activité cytotoxique des extraits totaux, fractions d’extraits et de molécules purifiées, isolées du latex de C. urucurana. Nous avons étudié plus particulièrement les effets du crourorb A1 sur la viabilité, l'apoptose, le cycle cellulaire et la migration de cellules issues de carcinome hépatocellulaire humaine (Huh-7). Nous avons également évalué l’effet du crourorb A1sur la viabilité des cellules Huh-7 cultivées dans les gels de collagène en 3D. L' activité cytotoxique ont été réalisées par le test à la sulforhodamine B sur six lignées cellulaire cancéreuses et sur une lignée cellulaire non cancéreuse de fibroblastes murins, les cellules NIH/3T3. L'étude phytochimique de la fraction ‘acétate d'éthyle’ du latex de C. urucurana bioguidé a permis d’obtenir un nouveau cyclopeptide que nous avons nommé [1-9-NαC]-crourorb A1. Le crourorb présente une cytotoxicité puissante sur la lignée cellulaire cancéreuse NCI-ADR/RES (ovaire, le phénotype de multirésistance) à un niveau équivalent à celle de la doxorubicine (témoin positif). Ce composé est de plus pratiquement inactif (GI50 = >250 μg/mL) sur la lignée non cancéreuse NIH/3T3. L'évaluation de la cytotoxicité in vitro du crourorb A1 (test wst1) sur les cellules Huh-7 a démontré que la viabilité des cellules est inhibée de manière dose et temps-dépendant. Nous avons mesuré une forte activation des caspases 3 et 7 et une nette augmentation de l’expression des protéines pro-apoptotiques (Bak, Bid, Bax, Puma, Bim et Bad) ainsi que de la caspase 3 clivée par le crourorb A1. Nous avons également déterminé que la voie de signalisation JNK-MAPkinase semble nécessaire à l’induction de la mort cellulaire des cellules Huh-7 traitées au crourorb A1. Le crourob A1 induit un arrêt du cycle cellulaire en phase G2/M. En parallèle, les expressions de la cycline-dépendante kinase CDK1, des cyclines B1 et D1 sont augmentées après traitement au crourorb A1. De plus, le crourorb A1 a un effet inhibiteur significatif de la migration des cellules Huh-7, en présence ou absence de mitomycine C. Nous avons également confirmé, l’effet inhibiteur du crourob A1 sur la viabilité des cellules Huh7 cultivées en 3D dans les gels de collagène 1 et démontré que les cellules cultivées en 3D semblent plus résistantes au crourorb A1 par rapport aux cultures en monocouche 2D. Apesar de sua importância etnofarmacológica, não há relatos de estudos relacionados à sua composição química ou atividades anticâncer na literatura. Nesse contexto, o objetivo do presente estudo foi avaliar a atividade citotóxica de extratos, fases, frações e da substância isolada do látex de C. ururucurana e investigar os mecanismos celulares e moleculares in vitro responsáveis pela citotoxicidade da substância isolada considerando seu potencial apoptótico e influência sobre a progressão do ciclo celular em uma linhagem de hepatocarcinoma celular humano (Huh-7), bem como, os efeitos desta substância sobre a migração e viabilidade de células Huh-7 cultivadas em geis de colágeno em 3D. O ensaio de citotoxicidade in vitro foi realizado com o corante sulforrodamina B (SRB) em seis linhagens de células neoplásicas humanas e uma linhagem de células não neoplásicas NIH/3T3 (fibloblasto murinho). Os estudos de mecanismos de ação com crourorb A1 foram conduzidos em diferentes sistemas-teste in vitro. O estudo fitoquímico biomonitorado da fase bioativa acetato de etila do látex de C. urucurana resultou no isolamento de um novo ciclopeptídeo denominado [19-NαC]-crourorb A1, que apresentou potente atividade citotóxica frente à linhagem de células neoplásicas NCI-ADR/RES (ovário com fenótipo de resistência a múltiplos fármacos), com a mesma potência que a doxorrubicina (controle positivo), e foi inativa até a maior concentração testada (GI50 = >250 μg/mL) frente à linhagem de células não neoplásicas 3T3. A exposição de células Huh-7 ao crourorb A1 resultou em uma diminuição da viabilidade celular de maneira dose e tempo-dependentes. Nós observamos um aumento da atividade de caspases 3 e 7 e um aumento da expressão de proteínas próapoptóticas (Bak, Bid, Bax, Puma, Bim e Bad) e caspase 3 clivada após tratamento com crourorb A1. Além disso, observamos que a via de sinalização celular MAP/JNK foi necessária para induzir a morte celular em células Huh-7 tratadas com crourorb A1. Os efeitos de crourorb A1 foram também associados com o acúmulo de células na fase G2/M do ciclo celular. Em paralelo, as expressões da cinase dependente da ciclina (CDK1), ciclina B1 e ciclina D1 foram aumentadas após tratamento com crourorb A1. Foi demonstrado também o envolvimento de crourorb A1 na redução da migração de células Huh-7 cultivadas na presença e ausência de mitomicina C. Meanwhile, ecosystem-based management of this fishery is constrained by a paucity of information regarding the interaction of FAD-associated tuna aggregations with their local environment. This paper reports the results of a nine-month study around a FAD moored near Martinique Island, aimed at assessing the effects of the local environment on the variability of monthly estimates of proximate tuna biomass. Dual-frequency, active acoustics provided highresolution quantitative data on the pelagic community around the FAD, from fish to micronekton forage. Geostatistics were used to compute biomass estimates of the tuna aggregation comprising most of the FADassociated fish biomass, with a sampling error of 27%. Environmental variability was summarized by a small set of principal components (PCs) derived from profiles of temperature, salinity and dissolved oxygen vs. depth; and maps of chlorophyll a derived from remotely sensed, sea-surface colour. A generalized linear model was used to relate tuna biomass to environmental PCs and revealed a positive correlation between tuna abundance and: i) a micronekton layer sensed at 38 kHz and potentially consisting of preferred prey at about 180 m depth; and ii) low subsurface salinity (6080 m). These favourable environmental conditions may be related to thepresence of North Brazilian Current eddies that migrating tuna follow when not temporally associated with the FADs. A Lagrangian numerical investigation of the origins and fates of the salinity maximum water in the Atlantic - art. no. 3163 Auteur(s) : Blanke, Bruno Arhan, Michel Lazar, A Prevost, Gwenaelle Éditeur(s) : American Geophysical Union Résumé : The origins and fates of the Atlantic salinity maximum water (SMW), formed through excess evaporation in the tropics and subtropics of both hemispheres, are studied using monthly mean outputs of a numerical simulation of the world ocean climatological circulation. After defining formation domains from the surface salinity field and the vertical stratification, a Lagrangian technique is used to estimate the formation rates and main pathways in each hemisphere and the role of this water in the framework of the warm water return flow of the meridional overturning cell. Formation rates around 9 and 11 Sv are found in the Southern and Northern Hemispheres, respectively. While the export of the southern SMW from its formation area is realized by the western boundary currents, that of the northern SMW mainly results from interior subduction. Equatorward of the formation regions, a fraction of each SMW variety is entrained in the subtropical cells that connect the subtropics to the equatorial region. Poleward of them, both varieties are seen to feed the regions of subtropical mode water (STMW) formation around 35degrees of latitude in both hemispheres. The bulk of the transport associated with each variety eventually turns northward: This amounts to similar to6 Sv of southern SMW gathered in the North Brazil Undercurrent (NBUC), and similar to10 Sv of northern SMW found in the Gulf Stream at 35degreesN, of which 8 Sv have gone through the Caribbean Sea. Of the 13.4-Sv northward transport of the meridional overturning cell estimated by the model at 47degreesN, more than 50% (6.9 Sv) is found to have transited through at least one of the SMW regions. This gives an indication of the likely important role of SMW formation in the observed northward salinity increase of the upper Atlantic Ocean. Biologie et écologie du mérou géant (Epinephelus itajara) en Guyane française ; Biology and ecology of Goliath Grouper (Epinephelus itajara) in French Guiana Auteur(s) : Artero, Céline Auteurs secondaires : Antilles-Guyane Bouchon, Claude Résumé : Le mérou géant, Epinephelus itajara, espèce en danger critique d’extinction au niveau mondial, est présent dans les eaux turbides et saumâtres de la Guyane française où l’espèce est exploitée par une pêcherie côtière. Dans la partie est du plateau continental guyanais, les mérous géants se localisent autour de tous les sites rocheux marins où ils se répartissent en fonction de la profondeur et du faciès (éboulis ou tombant). Les analyses des isotopes stables du carbone et de l’azote suggèrent qu’environ 30 % des mérous géants < 115 cm présents sur ces sites rocheux, proviendraient des mangroves alentours. Les analyses de contenus stomacaux révèlent un changement ontogénétique du régime alimentaire. Les petits individus (< 90 cm) s’alimentent principalement de Crustacés puis intègrent progressivement les poissons à leur alimentation jusqu’à devenir essentiellement piscivores au-delà de 140 cm. La population de Guyane est principalement composée d’individus juvéniles : seulement 8,5 % des individus capturés en Guyane a plus de 6 ans. Ces jeunes mérous sont très territoriaux durant les 4-5 premières années de leur vie puis ne sont plus retrouvés sur les sites rocheux. Le devenir de ces juvéniles est incertain, soit ils sont pêchés, soit ils migrent vers d’autres sites ou vers les agrégations de reproduction. Cependant, les mérous géants ne semblent pas se reproduire en Guyane. Il est possible que les conditions environnementales (forte turbidité et température) ne favorisent pas le développement gonadique, ce qui engendrerait une omission totale de reproduction. Le suivi par tags satellite suggère que les mérous géants migrent dans le sens des courants marins vers Trinidad-et-Tobago, potentiellement pour la reproduction. Aucun mouvement vers le Brésil n’a été mis en évidence bien que les stocks de mérous géants du Brésil et de Guyane semblent liés. En effet, ces deux stocks évoluent conjointement avec un déclin des populations au début des années 90. Le maintien de la population de mérou géant en Guyane se ferait grâce à l’apport de larves des sites d’agrégation du Brésil par le courant marin. La mortalité totale des mérous géants (0,65) est aussi élevée au sein de la réserve marine ainsi que sur les sites de pêche. Cela suggère que l’interdiction de pêche à la réserve n’est pas respectée et que les grands individus quittent la réserve pour des sites plus spacieux. En supposant que la population de mérous géants adultes a été réduite à cause de la pêche, une protection régionale des individus > 150 cm doit être instaurée afin de régénérer le stock d’individus matures. Il semble nécessaire de développer une politique de gestion internationale de l’espèce, surtout entre la Guyane et le Brésil, afin de conserver les populations de mérous géants d’Amérique du Sud. The critically endangered Goliath Grouper, Epinephelus itajara, is found in the turbid and brackish waters of French Guiana, where the species is subject to a coastal fishery. Goliath Grouper populations inhabit all rocky habitats and exhibit increasing abundance with depth. Around 30% of individuals less than 115 cm may derive from the abundant mangrove habitat along the french Guiana coast. There is a ontogenetic shift of the diet, primarily crustacivorous, individuals >140 cm become piscivorous. The Goliath Grouper population in French Guiana is composed primarily og juvelines. The fate of the juvelines is unknonw, some may be caught in the fishery, others may move to other habitat. No evidence of spawning or gonadal readiness was found in french Guiana. it is possible that environmental conditions (high turbidity and temperature) are not conducive to reproductive activity. Goliath Grouper may migrate down current from French Guiana to Trinidad and Tobago or further. No movement of Goliath Grouper was found against the current toward Brazil. French Guiana juveniles may be derived from spawns occuring off Brazil. Calculated total mortality of Goliath Grouper off French Guiana is high (0.65) in both fished and protected areas suggesting that illegal fishing is occuring within the protected area, or that emigration rates are high. Leur nombre a beaucoup augmenté ces dernières années et les modes d’action qu’ils recouvrent se sont considérablement diversifiés. Pour autant, les écosystèmes continuent à se dégrader, et l’efficacité de ces dispositifs est souvent remise en cause. Cette thèse vise à analyser les facteurs influant sur l’efficacité des espaces protégés pour la conservation d’écosystèmes amazoniens encore peu dégradés. Elle étudie pour cela la mise en œuvre de dispositifs relevant de doctrines de gestion de l’environnement différentes, basées sur (i) la limitation réglementaire maximale des activités humaines impactantes pour les écosystèmes, (ii) la gestion des ressources par des populations locales ou traditionnelles ou (iii) la mise en place d’une gestion forestière durable sur des terres publiques. Nous nous plaçons dans une posture comparative entre la Guyane (France) et l’Amapá (Brésil), territoires partageant une frontière commune, principalement matérialisée par le fleuve Oyapock. La comparaison internationale entre ces deux régions présentant une couverture exceptionnelle en espaces protégés, dans des conditions écologiques et géographiques relativement similaires, nous permet d’observer l’influence du contexte historique et sociopolitique sur les modes de prise en charge de la gestion de l’environnement par les aires protégées. Nous nous sommes basés sur une approche constructiviste, appuyée sur une production de données principalement qualitatives (entretiens semi-directifs, analyse de documents, observation participante…). Nous avons ainsi procédé à une déconstruction critique des dispositifs « aires protégées », permettant de mettre en lisibilité les enjeux environnementaux qu’ils portent, et d’analyser l’efficacité environnementale des stratégies mises en œuvre. Cette analyse s’est articulée autour d’une lecture à la fois diachronique et multiscalaire des processus de gestion.Nous montrons que la mise en place des espaces protégés de Guyane et d’Amapá a été portée par des coalitions d’acteurs structurées autour de doctrines de gestion, c’est à dire de conceptions partagées des conditions de mise en œuvre d’une « bonne gestion environnementale ». Les dispositifs créés ont hérité des ressources stratégiques d’action et de la légitimité de ces coalitions. Les coalitions porteuses de la création des espaces protégés en Amapá sont fortement articulées aux mouvements sociaux et environnementaux plus généraux de l’Amazonie brésilienne, notamment pour les revendications socio-environnementales émergeant à la sortie de la période dictatoriale. En Guyane française, les espaces naturels protégés sont principalement la résultante de compromis entre d’une part une volonté d’exemplarité de l’action de la France en Amazonie, et d’autre part une recherche de minimisation des conflits avec les acteurs politiques locaux. Nowadays, natural protected areas are one of the main public policies implemented at the international level in order to prevent the destruction of ecosystems. During the last decades, they became very numerous, and mobilized more and more diversified modes of action. However, natural ecosystems continue to deteriorate, and the effectiveness of protected areas is often questioned. The aim of this thesis was to analyse the factors influencing the effectiveness of protected areas in a context of Amazonian ecosystems that are still well preserved. We observed the implementation of environmental management arrangement (protected areas) that are based on various doctrines: (i) the strong limitation of all harmful human activities through legislation; (ii) the management of resources by local or traditional populations; and (iii) the implementation of sustainable forest management on public lands. We adopted a comparative approach between Guyana (France) and Amapá (Brazil), territories that share a common border, mainly identifiable by the Oyapock River. These two territories present quite similar ecological and geographical conditions, and both have an exceptional coverage of protected areas. Therefore this international comparison allowed us to observe the influence of the historical and socio-political context on the implementation of protected areas. We used a constructivist approach, mainly based on qualitative data (semi-structured interviews, document analysis, participant observation...). We effected a critical deconstruction of "protected areas" management arrangements in order to make their environmental objectives intelligible, and we analysed the environmental effectiveness of the strategies implemented. This analysis was based on a diachronic and multiscalar lecture of management processes. We show that the establishment of protected areas in Guiana and Amapá was led by coalitions of actors structured around management doctrines, i.e. shared conceptions of "good environmental management". Management arrangements created inherited the strategic resources of action and the legitimacy of these coalitions. In Amapá, coalitions were strongly articulated with wider social and environmental movements of the Brazilian Amazon, especially in the case of socio-environmental claims rising at the end of the dictatorship. In French Guiana, the natural protected areas are mainly the result of compromise between the French willingness to serve as an example in the Amazon region, and the minimisation of conflicts with local politicians. http://www.theses.fr/2014AGUY0744/document | Partager |
Composés antimicrobiens ou cytotoxiques à partir de micro-organismes endophytes foliaires ; Substâncias antimicrobianas ou citotóxicas em micro-organismos endofíticos foliares ; Antimicrobial or cytotoxic compounds from leaf endophytic microorganisms Auteur(s) : Meirelles Casella, Thiago Auteurs secondaires : Antilles-Guyane Universidade de Brasília Stien, Didier Espindola, Laïla Salmen Résumé : En raison de la nature symbiotique des micro-organismes endophytes, cette étude visait à étudier l'activité antibactérienne, antifongique et cytotoxique chez les métabolites secondaires des extraits endophytes foliaires de plantes de l'Amazonie brésilienne et du Cerrado. Dans ce travail de thèse 147 micro-organismes cultivables ont été isolés (130 champignons, 3 bactéries e 14 champignons non-identifiés ou inconnus) à partir de 28 plantes (4 espèces collectées au Brésil et 24 en Guyane Française). Tous les micro-organismes furent identifiés par analyse moléculaire des régions spécifiques de l'ADNr en utilisant des techniques de séquençage génomique. Des champignons endophytes de l’ordre des Xylariales furent ceux de plus importante fréquence d’isolation dans cette étude, représentés par 25 isolats. Des extraits bruts à l’AcOEt furent produits à partir de cultures de chaque micro-organisme isolé. Une proportion relative signifiante (23,1%) des extraits démontra une activité sur Candida albicans ATCC 10213, pendant que 4% furent actifs sur Staphylococcus aureus ATCC 29213. Le potentiel cytotoxique des extraits fut évalué pour des lignées cellulaires humaines KB (carcinome cervical utérin), MDA-MB-435 (mélanome), et MRC-5 (fibroblastes de poumon normal), résultant en proportion signifiante avec activité d’inhibition de la prolifération cellulaire (24,4%, 23,1% e 16,3%, respectivement).Dix-huit métabolites secondaires furent isolés et identifiés à partir du fractionnement des extraits bruts de huit endophytes. Dix-sept de ces substances furent déjà décrites précédemment dans la littérature: l’acide piliformique (24) et la griséofulvine (25) isolés à partir de Xylaria cubensis SNB-GCI02; La phomopirone (26), la pyrenocine (27), l’alterpérilenol (28) et la novae-zelandine (29), isolés de Lewia infectoria SNB-GTC2402; 5-métilmelleina (31) et la dihidrosporothrioride (32) isolées à partir de Xylaria sp. SNB-GTC2501; La mycoleptodiscine (34) et la mycoleptodiscine B (35) isolées à partir de Mycoleptodiscus sp. SNB-GTC2304; le mycoepoxidiène (36) et la altiloxina A (37) isolés à partir de Diaporthe pseudomangiferae SNB-GSS10; L’acremonisol (40), le semicochliodinol (41) et le cochliodinol (42) isolés à partir de Chaetomium globosum SNB-GTC2114; la colletofragarone A2 (43) isolée à partir de Colletotrichum sp. SNB-GTC0201; la flavoglaucine (44) isolée à partir de Eurotium rubrum BBS01. Parmi ces substances, la flavoglaucine (44) isolée à partir de E. rubrum BBS01 démontra une activité comparable au contrôle fluconazol en C. albicans (CIM de 4 µg.mL-1). La substance (44) présenta IC50 >10 µM sur cellules normales MRC-5, ce qui en fait un candidat pour des études ultérieures. Dans ce travail, fut identifié pour la première fois l’activité de la colletofragarone A2 (43) isolée à partir de Colletotrichum sp. SNB-GTC0201. La substance inédite nommée pyrrocidine C (30) fut isolée à partir de L. infectoria SNB-GTC2402 et identifiée par des analyses spectroscopiques (Casella et al., 2013). La pyrrocidine C (30) fut active en S. aureus ATCC 10213 (CIM de 2 µg.mL-1) et ne fut pas considérées cytotoxique pour les cellules normales MRC-5 (IC50 >10 µM), démontrant une sélectivité dans l’action antimicrobienne. Ces résultats démontrent la grande diversité des endophytes fongiques chez les plantes de l'Amazonie brésilienne et du Cerrado et la grande chimiodiversité associée aux métabolites secondaires de ces micro-organismes. Des champignons endophytes tropicaux, comme ceux présentés dans cette étude, peuvent apparaître comme une nouvelle source de substances antimicrobiennes et cytotoxiques. Because of the symbiotic nature of endophytes, this survey aims to investigate the probality of discovering antibacterial, antifungal and cytotoxic activities in secondary metabolites of leaf endophytes isolated from plants of Amazon and Cerrado biomes. In this study, 147 cultivable microorganisms were isolated (130 fungi, 3 bacteria and 14 unidentified or unknown microbes) from 28 plant species (4 species collected in Brazil and 24 in French Guyana). All endophytes were identified by molecular analyses of specific rDNA regions, with genomic sequencing techniques. Fungal endophytes belonging to Xylariales order were the most frequently isolated in this study, represented by 25 isolates. Crude AcOEt extracts were produced from cultures of each isolated endophyte. A significant relative proportion (23,1%) of extracts showed activity in Candida albicans ATCC 10213, while 4% were active in Staphylococcus aureus ATCC 29213. The cytotoxic potencial of the extracts was evaluated for human cell lines KB (uterin cervical carcinome), MDA-MB-435 (melanoma) and MRC-5 (normal lung fibroblasts), and a significant proportion of them showed cellular proliferation inhibition (24,4%, 23,1% e 16,3%, respectively).Eighteen secondary metabolites were isolated by the fractionation of eight endophytic extracts. Seventeen of these substances had already been previously described in the literature: piliformic acid (24) and griseofulvin (25) isolated from Xylaria cubensis SNB-GCI02; phomopiron A (26), pyrenocin A (27), alterperilenol (28) and novae-zelandin A (29) isolated from Lewia infectoria SNB-GTC2402; 5-metilmellein (31) and dihidrosporothriorid (32) isolated from Xylaria sp. SNB-GTC2501; mycoleptodiscin A (34) and mycoleptodiscin B (35) isolated from Mycoleptodiscus sp. SNB-GTC2304; mycoepoxidien (36) and altiloxin A (37) isolated from Diaporthe pseudomangiferae SNB-GSS10; acremonisol A (40), semicochliodinol A (41) and cochliodinol (42) isolated from Chaetomium globosum SNB-GTC2114; colletofragaron A2 (43) isolated from Colletotrichum sp. SNB-GTC0201; flavoglaucin (44) isolated from Eurotium rubrum BBS01. Among these substances, flavoglaucin (44), isolated from E. rubrum BBS01, showed comparable antifungal activity with the positive control fluconazol in C. albicans (MIC of 4 µg.mL-1). Flavoglaucin (44) also showed IC50 >10 µM in normal MRC-5 cells, becoming a good candidate for further studies. In this work, the cytotoxic activity of colletofragaron A2 (43), isolated from Colletotrichum sp. SNB-GTC0201 was described for the first time. The unpublished substance named pyrrocidin C (30) isolated from L. infectoria SNB-GTC2402 was identified by spectroscopic analyses (Casella et al., 2013). The pyrrocidin C (30) was active in S. aureus ATCC 10213 (MIC of 2 µg.mL-1), and was not considered cytotoxic for normal MRC-5 cells (IC50 >10 µM), showing selectivity in antimicrobial activity. These results demonstrate the great endophytic fungal diversity in plants of Amazon and Cerrado biomes, along with the chemodiversity associated to the secondary metabolites of these endophytes. Tropical fungal endophytes, like those seen in this work, may emerge as a new source of antimicrobial and cytotoxic substances. Devido à natureza simbiótica dos micro-organismos endofíticos, este estudo teve como objetivo investigar a atividade antibacteriana, antifúngica e citotóxica em metabólitos secundários de extratos de fungos endofíticos foliares de plantas do bioma Amazônia e Cerrado. Neste trabalho de tese foram isolados 147 micro-organismos cultiváveis (130 fungos, 3 bactérias e 14 fungos não-identificados ou desconhecidos) a partir de 28 plantas (4 espécies coletadas no Brasil e 24 na Guiana Francesa). Todos os micro-organismos foram identificados por análise molecular de regiões específicas de DNAr, com uso de técnicas de sequenciamento genômico. Fungos endofíticos da ordem Xylariales foram os de maior frequência de isolamento neste estudo, representados por 25 isolados. Extratos brutos em AcOEt foram produzidos a partir de culturas de cada micro-organismo isolado. Uma proporção relativa significante (23,1%) dos extratos demonstrou atividade em Candida albicans ATCC 10213, enquanto 4% foram ativos em Staphylococcus aureus ATCC 29213. O potencial citotóxico dos extratos foi avaliado para as linhagens celulares humanas KB (carcinoma cervical uterino), MDA-MB-435 (melanoma), e MRC-5 (fibroblastos de pulmão normal), resultando em proporção significante com atividade de inibição da proliferação celular (24,4%, 23,1% e 16,3%, respectivamente). Dezoito metabólitos secundários foram isolados a partir do fracionamento de oito extratos brutos endofíticos. Dezessete destas substâncias já tinham sido descritas anteriormente na literatura: ácido pilifórmico (24) e griseofulvina (25) isoladas de Xylaria cubensis SNB-GCI02; phomopirona A (26), pyrenocina A (27), alterperilenol (28) e novae-zelandina A (29) isoladas de Lewia infectoria SNB-GTC2402; 5-metilmelleina (31) e Dihidrosporothriolida (32) isoladas de Xylaria sp. SNB-GTC2501; mycoleptodiscina A (34) e mycoleptodiscina B (35) isoladas de Mycoleptodiscus sp. A pyrrocidina C (30) foi ativa em S. aureus ATCC 10213 (CIM de 2 µg.mL-1), e não foi considerada citotóxica para as células normais MRC-5 (IC50 >10 µM), demonstrando seletividade na ação antimicrobiana. Estes resultados demonstram a grande diversidade fúngica endofítica em plantas do bioma Amazônia e Cerrado, e a quimiodiversidade associada aos metabólitos secundários destes micro-organismos.
