Description : Drosophila often offers many advantages as an experimental organism. The dead ringer (dri) gene of drosophila melanogaster is a member of the recently discovered ARID-box family of eukaryotic genes that encodes proteins with a conserved DNA-binding domain. dri itself is highly well conserved with specific orthologs from human to yeast genomes. This family include ARID3A, 3B and 3C with have role in embryonic pattening, cell lineage gene regulation, cell cycle control, transcriptional regulation and chromatin structure modification. We first showed that the gene encoding ARID3A also known as Bright (B cell regulator of immunoglobulin heavy chain transcription) play critical roles in B-cell development and SATB1 in T-cell transcription regulation. In this international workshops we will expose a comprehensive understanding of these molecules and better knowledge of their specific targets and function-interactions in controlling cell fate decisions and development. In addition, dri have homology with the Aedes aegypti mosquitoes which had ability to carry and spread disease to human. More than half of the world?s population live in areas where this mosquito species is present, including Martinique. Sustained mosquito control efforts are important to prevent outbreaks from Dengue, Chikungunya, malaria, zika diseases. Discovering the functions of AT-rich genes in preventing illness from mosquito is the central goal for raising working progress hypothesis.

En plus des regroupements de communes, le parc amazonien créé en 2007, poursuit l'organisation spatiale avec ses missions de protection de la nature et de sauvegarde des modes de vie locaux. Il est donc un prescripteur politique majeur et participe ainsi au renforcement du contrôle de l'état dans cette région. Malgré le souhait d'une patrimonialisation de la biodiversité et des modes de vie, les confrontations socio-culturelles sont violentes et peuvent entrainer une aliénation administrative, financière, foncière...; ainsi que fractionner les communautés et créer des déséquilibres de ressources.

STUDY QUESTION: Do freezing and in vitro culture procedures enhance the expression of proteins involved in apoptotic or autophagic pathways in murine pre-pubertal testicular tissue? SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. BackgroundPopulation attributable risks (PARs) are useful tool to estimate the burden of risk factors in cancer incidence. Few studies estimated the PARs of oral cavity cancer to tobacco smoking alone, alcohol drinking alone and their joint consumption but none performed analysis stratified by subsite, gender or age. Among the suspected risk factors of oral cavity cancer, only PAR to a family history of head and neck cancer was reported in two studies. The purpose of this study was to estimate in France the PARs of oral cavity cancer to several recognized and suspected risk factors, overall and by subsite, gender and age.MethodsWe analysed data from 689 oral cavity cancer cases and 3481 controls included in a population-based case–control study, the ICARE study. Unconditional logistic regression models were used to estimate odds ratios (ORs), PARs and 95 % confidence intervals (95 % CI).ResultsThe PARs were 0.3 % (95 % CI −3.9 %; +3.9 %) for alcohol alone, 12.7 % (6.9 %–18.0 %) for tobacco alone and 69.9 % (64.4 %–74.7 %) for their joint consumption. PAR to combined alcohol and tobacco consumption was 74 % (66.5 %–79.9 %) in men and 45.4 % (32.7 %–55.6 %) in women. Among suspected risk factors, body mass index 2 years before the interview <25 kg.m−2, never tea drinking and family history of head and neck cancer explained 35.3 % (25.7 %–43.6 %), 30.3 % (14.4 %–43.3 %) and 5.8 % (0.6 %–10.8 %) of cancer burden, respectively. About 93 % (88.3 %–95.6 %) of oral cavity cancers were explained by all risk factors, 94.3 % (88.4 %–97.2 %) in men and only 74.1 % (47.0 %–87.3 %) in women.ConclusionOur study emphasizes the role of combined tobacco and alcohol consumption in the oral cavity cancer burden in France and gives an indication of the proportion of cases attributable to other risk factors. Most of oral cavity cancers are attributable to concurrent smoking and drinking and would be potentially preventable through smoking or drinking cessation. If the majority of cases are explained by recognized or suspected risk factors in men, a substantial number of cancers in women are probably due to still unexplored factors that remain to be clarified by future studies.

Résumé : The Franceville Basin (2.1 Ga) in southeastern Gabon, hosts a black shale series well preserved (FB Formation) which represents an exceptional example of unmetamorphised Paleoproterozoic sediment strata. This basin includes the protore of one of the largest Mn-oxide laterite ore worldwide, mined by Eramet-Comilog. The aim of this this work is to determine formation conditions of this protore and to characterize the global sedimentary evolution of the FB Formation which include the protore. This 75 m thick protore is constituted of Mn carbonate-rich black shales which represent a potential ore deposit for the future. It was drilled, on the Bangombé plateau, during a recent campaign of 24 boreholes with an average depth of 125 m.Through multidisciplinary study on these cores and key outcrops of the basin, this work focuses on : i) the depositional environment of the Mn-protore and FB Formation ; ii) the processes of formation of the Mn-carbonates at 2.1 Ga and iii) the architecture and distribution of Mn-rich levels of the protore on the Bangombé Plateau.A detailed sedimentological and petrological study allowed us to redefine the division of the FB into nine units, named U1 to U9 from the base to the top. These units show a fine upward cycle up to U5 and a reverse coarse loop until U9. This division leads to a new interpretation to propose an evolution from a shoreface depositional environment controlled by deltaic currents (U1) to an offshore depositional environment with anoxic conditions (U2). In this basin, a system of submarine fan is developed, feeding a network of sand injections (injectites) covering the Bangombé plateau (70 km²) and with a thickness of 150 m affecting U4 to U7, including the Mn-protore. The protore depositional environment formed below the wave base limit (U4, U5 and U6) in a starved basin, controlled by biochemical sedimentation allowed the formation of Mn-carbonates. The end of starved basin, would allow the re-opening of the detrital input into the offshore basin (U7). This basin is then filled by storm bar deposits intersected by deltaic sub-aquatic channels (U8). The U9 unit corresponds to a quiet lagoon environment, allowing the multicellular organism development for the first time on the planet.A geochemical study allowed the characterization of the depositional environment of Mn-carbonates as an anoxic to sub-oxic and alkaline environment. For the first time, we showed that the manganese carbonates, present as bacteriomorph forms, are precipitated from the seawater by the mediated photosynthetic cyanobacterial activity, which allows CO2 / HCO3 depression of the environment and a local increase of pH. The cyanobacterial activity is controlled by the absence of bottom currents, which increase the detrital input. This would stop the activity of the cyanobacteria and thus would lead to the decrease of the Mn-concentration along the protore.Finally, well-log and a sequential correlation analyses allowed us to detail spatial and timing repartition of FB Formation deposit, controlled by a tectono-sedimentary model. We propose two- tectono-sedimentary phases. A first syn-tectonic phase (U1 to U3) controls the depocentres and sedimentation gaps. Turbidite coarse deposits are located in the most subsident part, clay deposits rather on the slope and carbonates on shoals. A second post-tectonic phase (U4 to U7) allows isopach deposits. The Mn-protore formed during the post-tectonic phase. However, it shows a variable thickness due to subsidence to the north of the Bangombé plateau. So, on the Bangombé plateau, Mn-contents increase towards the south, whereas the protore thickness increases towards the NNE. Moreover sand injectites decrease Mn-content in the eastern part of the Bangombé plateau and impact on the economic evaluation of the Mn-carbonates. Currently, the protore is structured by post-sedimentary faults, which lead to a non-continuity of the high Mn levels on the Bangombé plateau.
Le Bassin de Franceville situé au SE du Gabon présente une série de black shales (Formation FB) d'âge Paléoprotérozoïque (2,1 Ga) surmontant les grès de la Formation FA. Ce bassin peu déformé et non métamorphique représente une archive exceptionnelle des processus de dépôt de cette période de l’histoire de la Terre où la vie commence à se développer. En particulier, la Formation FB contient le protore d'un gisement mondial d'oxydes de manganèse exploité par la société Eramet-Comilog. L’objectif de ce travail est de préciser les conditions de formation de ce protore et de le replacer dans l’histoire du remplissage du bassin. Ce protore d'une épaisseur moyenne de 75 m est constitué de carbonates de manganèse ; il a fait l'objet d'une récente campagne de 24 forages carottés d'une profondeur moyenne de 125 m sur le plateau de Bangombé.A travers une approche pluridisciplinaire comprenant l'étude de ces forages et des affleurements clefs du bassin, ce travail porte sur la caractérisation i) de l'environnement de dépôt du protore et des séries qui l'encadrent (du toit du FA au toit du FB) ; ii) du mode de formation des carbonates de Mn à 2,1 Ga et iii) de l'architecture et de la répartition des hautes teneurs en Mn du protore qui représente un gisement potentiel du futur.L’étude pétro-sédimentaire détaillée a permis de distinguer neuf unités (U1 à U9) au sein de la Formation FB organisées suivant un cycle rétrogradant jusqu’à U5 puis progradant jusqu’à U9. Ce découpage montre une évolution du milieu de dépôt depuis un domaine de shoreface contrôlé par des courants deltaïques (U1) évoluant vers des dépôts d’offshore restreint (U2). Ce bassin enregistre ensuite un système de chenaux-levées turbiditiques (U3) qui alimentent un réseau d'injectites reconnu sur l’ensemble du Pl. de Bangombé et qui traverse 150 m de série (U4 à U7) en affectant le protore et ainsi les teneurs en Mn. L'environnement de dépôt du protore (U5 et U6) marque l’isolement du bassin qui devient affamé et contrôlé par une sédimentation biochimique à l’origine du dépôt des carbonates de Mn. L'Unité 7 correspond à la réouverture des apports détritiques en domaine d'offshore supérieur puis en domaine de shoreface dominé par des dépôts de tempêtes et de chenaux sous-aquatiques deltaïques (U8), et enfin lagunaire (U9) dans lequel apparaissent les premiers métazoaires de l’histoire de la planète, suivant une séquence régressive.Une étude géochimique a permis de préciser les conditions de dépôt du protore qui se forme en milieu alcalin anoxique à sub-oxique. Dans ce milieu, nous montrons pour la première fois une précipitation directe des carbonates de Mn sous forme bactériomorphe induite par l'activité photosynthétique des cyanobactéries qui consomment le CO2/HCO3 et conduisent à une augmentation du pH favorable à la formation des carbonates de Mn. L'activité cyanobactérienne est conditionnée par l’absence de courants de fond qui augmentent la turbidité du milieu et stoppent cette dernière.Les corrélations diagraphiques et séquentielles permettent de préciser la répartition spatiale et temporelle du FB permettant ainsi de replacer le protore dans l’histoire tectono-sédimentaire du bassin. Une première phase syn-tectonique (U1 à U3) contrôle l’épaisseur et la mise en place de dépôts turbiditiques dans les parties subsidentes, de dépôts argileux sur les pentes et de dépôt de carbonates sur les hauts-fonds. La seconde phase post tectonique (U4 à U7) permet la mise en place de dépôts relativement isopaques, bien que le protore enregistre une subsidence plus importante au nord du Pl. de Bangombé. On observe ainsi, sur le Plateau de Bangombé, des teneurs en Mn qui augmentent vers le sud alors que l’épaisseur du protore augmente vers le NNE. Postérieurement au dépôt du FB, le protore est structuré par des failles qui le compartimentent en touches de piano limitant ainsi sa continuité sur le Plateau de Bangombé.

OBJECTIVE: We hypothesized that, in Human Immunodeficiency Virus and Hepatitis C Virus (HIV/HCV) co-infected patients who did not respond to peg-interferon and ribavirin, a maintenance therapy with peg-interferon could induce fibrosis regression. METHODS: This was a randomized study with two parallel groups. HIV/HCV co-infected patients received peg-interferon α-2a at 180 μg/week or remained on observation for 96 weeks. The primary endpoint was the percentage of patients who experienced a decrease of at least one point in their Metavir fibrosis score between initial and final liver biopsies. Secondary endpoints included plasma fibrosis markers at week 96, occurrence of HCV-related complications, and survival. RESULTS: A total of 52 patients were randomized (peg-interferon: 25; control: 27) including 18 with cirrhosis. The median (interquartile range) age was 44 (40-46) years, and 69% were male. A total of 64% had ALT levels >1.5 normal values, and the CD4 cell count was 391 (296-537) cells/mm(3); 67% of patients had HIV RNA <200 copies/mL at entry. The main endpoint was assessed in 41 patients. Response rates were 3/20 (15%) and 4/21 (19%) in the peg-interferon and control groups, respectively (p = 0.99). There was no significant difference between peg-interferon and control groups on plasma fibrosis markers at the final visit. Severe liver-related complications were observed in 2 and 5 patients in peg-interferon and control groups, respectively. Three deaths were observed, all in the control group. CONCLUSIONS: A maintenance therapy with peg-interferon α-2a over 96 weeks in HIV/HCV co-infected patients, who were non-responders to HCV treatment, did not change liver fibrosis. Résumé : The green turtle (Chelonia mydas) is an emblematic species of marine life. However, nowadays it is subject to many threats (poaching, by-catch). Even if there is deep growing measures for its protection, the green turtle still is an endangered species and it is listed in Appendix I of Washington Convention (CITES). In order to elaborate efficient conservation and management plans, perfect knowledge of green turtle biology, but also of its population structure and their characteristics, are needed. In this thesis, we have assessed genetic structure of green turtle populations in the South-Western Indian Ocean by using genetic tools. In all, 1551 tissue samples have been collected from our study zone and from our control site French Polynesia (37 samples). All kinds if individuals were sampled (except males in reproductive phase) from 15 sampling sites including nesting, foraging, and immature development site. We used both control region of mitochondrial DNA and 6 microsatellite loci to better infer maternal and paternal lineages. We identified 29 haplotypes in the South-Western Indian Ocean. They are distributed in 3 independent and highly divergent clades, including one composed with haplotypes from Atlantic Ocean. For 7 of these haplotypes, it was the first time they were detected in the study zone. Fifteen haplotypes were previously undescribed, distributed in all the 3 clades. These new haplotypes seem to be specific to the South-Western Indian Ocean, which is then an original zone. Besides, we found a high allelic richness. These results show the South-western Indian Ocean is rich and very diversified. This region plays an important role in the global diversity of the species. The South-Western Indian Ocean is one of the two contact zones presently known between the two metapopulations of green turtles (Atlantic-Mediterranean and Indo-Pacific). This contact induces a genetic cline based on CM8 (Atlantic) and C3 (Indo-Pacific) haplotype frequencies. Analysis of the microsatellite differentiation between individuals provides evidence of genetic exchanges between the two metapopulations in the region. The South-Western Indian Ocean participates to green turtle global genetic mixing. Studying the influence of several intrinsic and extrinsic factors on population structuring provides useful information for management plan elaboration. We found no significant difference between genetic structures of foraging females and males, contrary to immature turtles which seem to be organised in 'regional pools'. This organisation could be due to both immature natal homing and influence of oceanic currents. High mitochondrial differentiation of nesting females and low global microsatellite differentiation of our samples indicate male-mediated gene flow among populations of the study zone. The genetic composition of a sampling site presents no significant variation along the year, even if we could notice some trends. Nevertheless, it can be significantly different from a year to an other one. This may result from alternation of distinct populations on the same site. We noticed different evolution in 10 or 20 years of the genetic composition depending on the sampling site. Geographic distance seems not to have significant influence on population structuring concerning microsatellite markers. Nesting females of Saziley Beach (Mayotte Island, Comoros Archipelago) present genetic divergence from females nesting in the two other sampled beaches of this island. The observed population structure shows no contradiction with the organisation of oceanic currents in the South-Western Indian Ocean. Comparing the results from the two genetic markers used, we identified 8 genetic differentiated clusters of turtles in the study zone and at least 6 distinct populations. These clusters constitute 8 potential management units (MUs) which could serve as basis in the elaboration of conservation and management plans.
La tortue verte (Chelonia mydas) constitue l'un des espèces emblématiques de la vie marine, pourtant de nombreuses menaces pèsent de nos jours encore sur sa survie (braconnage, captures accidentelles). Ainsi, malgré l'essor de mesures de protection menées à travers pour sa sauvegarde, la tortue verte constitue une espèce 'en danger d'extinction' et figure dans l'Annexe I de la Convention de Washington (CITES). Afin d'élaborer des plans de gestion et de conservation qui soient efficaces, il est important d'avoir une parfaite connaissance de la biologie de la tortue verte, mais aussi de la structure de ses populations et de leurs caractéristiques. C'est dans ce cadre que s'inscrit la présente étude. L'objectif de cette étude était d'acquérir des connaissances sur la structure des populations de tortues vertes dans le sud-ouest de l'océan Indien grâce à l'utilisation de l'outil génétique. Au total, 1551 échantillons de tissu ont été collectés dans la zone d'étude et dans notre site témoin la Polynésie française (37 échantillons). Toutes les catégories d'individus ont été échantillonnées (excepté les mâles en phase de reproduction) et les 15 sites d'échantillonnage comprennent à la fois des sites de ponte, d'alimentation et de développement pour les immatures. Deux types de marqueurs ont été utilisés : la région contrôle de l'ADN mitochondrial et 6 loci microsatellites, afin d'appréhender au mieux l'apport des lignées maternelles et paternelles. Nous avons pu mettre en évidence la présence dans le sud-ouest de l'océan Indien de 29 haplotypes distincts, appartenant à trois clades fortement divergents dont l'un constitué d'haplotypes originaires de l'océan Atlantique. Parmi ces haplotypes, 7 ont été détectés pour la première fois dans la zone d'étude, et 15 autres n'ont jamais été précédemment décrits chez cette espèce. Ils sont présents dans chacun des 3 clades d'haplotypes. Ces nouveaux haplotypes semblent spécifiques à la région, et en font une zone originale. On observe par ailleurs une grande richesse allélique dans les effectifs analysés. Ces résultats montrent que le sud-ouest de l'océan Indien est une zone riche et très diversifiée. Cette région joue un rôle important dans la diversité génétique globale de l'espèce. Le sud-ouest de l'océan Indien constitue l'une des deux seules zones connues à l'heure actuelle de contact entre les deux métapopulations de tortues vertes (Atlantique-Méditerranée et Indo-Pacifique). Ce contact a entraîné la formation d'un cline génétique portant principalement sur les fréquences relatives des haplotypes CM8 (Atlantique) et C3 (Indo-Pacifique). Les résultats obtenus lors de l'analyse microsatellite de la différenciation entre les individus originaires des deux métapopulations montrent que le sud-ouest de l'océan Indien constitue une zone d'échanges génétiques entre les deux métapopulations, participant au brasage génétique de l'espèce. L'étude de facteurs, intrinsèques et extrinsèques, pouvant influencer la structuration des populations apportent de nombreuses informations qui pourraient s'avérer utiles lors de l'élaboration de plans de gestion. La structure des femelles et des mâles en alimentation ne diffère pas, contrairement à celle des immatures qui semble s'organiser en 'pools régionaux' qui seraient le fruit de l'interaction d'un comportement de philopatrie et d'une influence des courants océaniques. La forte différenciation mitochondriale des femelles en ponte et la très faible différenciation microsatellite observée à l'échelle de la région, indiquent l'existence de flux de gènes via les mâles. La composition génétique d'un site ne varie pas de manière significative au cours de l'année. Par contre, elle peut varier d'une année à l'autre, signifiant l'alternance dans certains sites de ponte de plusieurs populations distinctes. L'évolution de la composition génétique d'un groupe, au cours de 10 ou 20 ans, diffère selon le site considéré. La distance ne semble pas influencer de manière significative la structuration des populations au niveau microsatellite. Les femelles en ponte sur la plage de Saziley (Mayotte) diffèrent génétiquement de celles pondant sur les deux autres plages de l'île. La structure observée des populations est en accord avec l'organisation des courants océanique dans la région. La confrontation des résultats obtenus à partir des deux marqueurs génétiques utilisés, permet la détermination de 8 ensembles génétiquement différenciés dans la zone d'étude et l'identification d'au moins 6 populations distinctes. Ces ensembles constituent autant d'unités de gestion (MUs) potentielles qui pourront servir de base à l'élaboration de plans de gestion et de conservation.

Résumé : Environmental conditions experienced by aquatic organisms are archived in biogenic carbonates such as fish otoliths, bivalve shells and coral skeletons. These calcified structures present an accretionary growth and variations in optical properties - color or opacity - that are used to reconstruct time. Full and reliable exploitation of the information extracted from these structures is, however, often limited as the metabolic processes that control their growth and their optical properties are poorly understood. Here, we propose a new modeling framework that couples both the growth of a biogenic carbonate and its optical properties with the metabolism of the organism. The model relies on well-tested properties of Dynamic Energy Budget (DEB) theory. It is applied to otoliths of the Bay of Biscay anchovy (Engraulis encrasicolus) for which a DEB model has been previously developed. The model reproduces well-known otolith patterns and thus provides us with mechanisms for the metabolic control of otolith size and opacity at the scale of an individual lifespan. Two original contributions using this framework are demonstrated. First, the model can be used to reconstruct the temporal variations in the food assimilated by an individual fish. Reconstructing food conditions of past and present aquatic species in their natural environment is key ecological information to better understand population dynamics. Second, we show that non-seasonal checks can be discriminated from seasonal checks, which is a well-recognized problem when interpreting fish otoliths. We discuss further developments of the model and the experimental settings required to test this new promising framework.

Résumé : The Algerian margin has originated from the opening of the Algerian basin about 25-30 Ma ago. The central margin provides evidence for large-scale normal faults of Oligo-Miocene age, whereas transcurrent tectonics characterizes the western margin. A set of NW-SE oriented dextral transform faults was active during basin opening and divided the 600 km long central margin into segments of similar to 120-150 km. The upper Miocene, Plio-Quaternary, and present-day tectonic setting is, however, compressional and supports the occurrence of a margin inversion, a process still poorly documented worldwide. We show that the central Algerian margin represents a rare example of inverted margin, where the process of subduction inception is particularly well expressed and helps understand how extensional and transtensive structures are involved in margin shortening. Using multibeam bathymetry and multichannel seismic reflection sections from the MAR-ADJA 2003 and 2005 cruises, we evidence Pliocene-Pleistocene shortening with contrasting styles along the margin between west (Khayr Al Din bank) and east (Boumerdes-Dellys margin) of Algiers. Pre-Miocene structures such as basement highs and transform faults appear to control changes of the deformation pattern along this part of the margin, resulting in different widths, geometries, and relative positions of folds and faults. Plio-Quaternary and active blind thrust faults do not reuse Oligo-Miocene normal and transform faults during inversion, but instead grow within the continental margin (as testified for instance by the 21 May 2003 M-w 6.8 Boumerdes-Zemmouri earthquake), at the foot of the continental slope and at the northern sides of basement highs interpreted as stretched continental blocks of the rifted margin. The inherited structures of the margin appear, therefore, to determine this deformation pattern and ultimately the earthquake and tsunami sizes offshore. The complex geometry of the fault system along the Algerian margin suggests a process of initiation of subduction in its central and eastern parts.

A reactive percolation experiment was conducted by injecting seawater into a permeable olivine aggregate at 190 °C and 19 MPa to explore the relationships between hydration reactions and hydrodynamic properties during the onset of serpentinization in the ultramafic lithosphere exposed at mid-ocean ridges. The experiment was stopped after 23 days when the sample became impermeable. The initial flow rate was 0.2 mL/h and then was decreased down to 0.06 mL/h after 8 days. Permeability decreased continuously throughout the experiment. The analyses of fluid chemistry time series and of the mineralogy and structure of the reacted sample showed olivine dissolution and precipitation of proto-serpentine, brucite and Fe-oxides. These reactions are controlled by coupled hydrodynamic and chemical processes interacting at different time and spatial scales. Their first characteristic is the production of silica rich outlet fluids in disequilibrium with the observed reacted mineral assemblages. These compositions are interpreted as resulting from a suite of coupled dissolution-precipitation reactions, controlled at the pore scale by surface kinetic processes, that rapidly reaches steady-state (constant fluid composition independent of fluid flux). Hence, the effective rate of serpentinization was controlled, to the first order, by the transport of reactants during the experiment. Mass balance calculations show that the rate of olivine conversion was fast (0.2-0.5 wt%/day), yet only ∼8 wt% of the olivine sample reacted, because the permeability drop limited fluid circulation. Porosity varied little during the experiment compared to permeability changes: the decrease of permeability was explained by the structure of the newly formed serpentine favouring the clogging of fluid paths. The rate at which permeability decreased was the fastest at low flux conditions. This suggests that permeability changes were not controlled simply by the kinetics of the serpentinization reactions, but mainly reflected the development of low Peclet pore scale zones forming micro-environments that control the serpentinization reaction-paths. These results outline the need to take into account the coupling of hydrodynamic and chemical processes when modelling the effects of reactive transport within the hydrating oceanic lithosphere.

Résumé : The development of large pelagic fishing in the French Antilles is an integral part of a difficult economic situation of the fishing sector in Martinique where this development can seem like a solution to increasingly acute problems. Indeed, for the regulatory authorities as well as for many participants in the sector, it is imperative to find solutions to the lack of profitability of a part of the fishing ships and to confront the regular increase of the share of imports in supplying Martinique with seafood. This question about the possibilities of developing large pelagic fishing is not new since it is already mentioned in the Notice from the Economic and Social Committee of the Martinique Region in 1982. We will not show in this document a detailed analysis of small-scale fishing in Martinique. Since the latter has been abundantly studied by the team of the Caribbean Oceanological and Halieutic Research Centre (PROHC) between 1985 and 1987, we will take up these principal conclusions (with which we are in full agreement) and bring them up to date based on data collected during our mission from 11 to 24 April 1988. The first section devoted to the supply structure (local fishing and imports) will attempt to define under what conditions large pelagic fishing can be developed. The second section will address marketing and demand. Indeed, at the same time as the problems of developing large pelagic fishing, we are faced with the question of the organisation of seafood markets. This organisation is a fundamental issue for the small-scale fishing sector in the French Antilles since its long-term future depends upon it by setting different prices and amounts exchanged according to the structure it is given. We will come to a conclusion on the economic conditions of developing a large pelagic fishery and organising a market, this based on research and study work already done. We will indicate which investigations must be conducted to complete the set of factors for decision support in this area. To perform this assessment, we reconstructed from the long series 1970-1986 (sometimes with significant inadequacies): these are presented as an appendix.
(OCR non controlé) Le développement de la pêche de grands pélagiques dans les Antilles Françaises s'inscrit dans un contexte économique difficile du secteur de la pêche en Martinique où ce développement peut apparaître comme une solution à des problèmes de plus en plus aigus. En effet, pour les autorités de tutelle ainsi que pour de nombreux intervenants dans le secteur, il est impératif de trouver des solutions au manque de rentabilité d'une partie des unités de pêche et de faire face à l'augmentation régulière de la part des importations dans l'approvisionnement de la Martinique en produits de la mer. Cette question sur les possibilités d'un développement de la pêche de grands pélagiques n'est pas nouvelle puisqu'elle est déjà évoquée dans l'Avis du Comité Economique et Social de la Région Martinique en 1982. On ne présentera pas dans ce document une analyse détaillée de la pêche artisanale en Martinique. Celle-ci ayant été abondamment étudiée par l'équipe du Pôle de Recherche Océanologique et Halieutique Caraïbe (PROHC) entre 1985 et 1987, on reprendra ces principales conclusions (avec lesquelles nous sommes en plein accord) en les actualisant à partir des informations recueillies lors de notre mission du 11 au 24 avril 1988. La première section consacrée à la structure d'offre (pêche locale et importations) s'attachera à définir dans quelles conditions peut être développée une pêche de grands pélagiques. Là seconde section traitera de la commercialisation et de la demande. En effet parallèlement aux problèmes de développement d'une pêche de grands pélagiques, se pose la question de l'organisation du marché des produits de la mer. Cette organisation est un enjeu primordial pour le secteur de la pêche artisanale dans les Antilles Françaises puisqu'elle conditionne son avenir à long terme en définissant des prix et des quantités échangées différents suivant la structure qu'on lui donne. On conclura sur les conditions économiques du développement d'une pêche de grands pélagiques et de l'organisation d'un marché, ceci à partir des travaux de recherche et des études déjà réalisés. On indiquera quelles investigations doivent être menées pour compléter l'ensemble des éléments d'aide à la décision dans ce domaine. Pour effectuer cette évaluation, on a reconstituer des séries longues 1970-1986 (parfois avec des carences importantes) : celles-ci sont présentées en annexe.

Résumé : We investigated the influence of bioturbation by macrofauna on the vertical distribution of living (stained) benthic foraminifera in marine intertidal sediments. We investigated the links between macrofaunal bioturbation and foraminiferal distribution, by sampling from stations situated on a gradient of perturbation by oyster-farming, which has a major effect on benthic faunal assemblages. Sediment cores were collected on the French Atlantic coast, from three intertidal stations: an oyster farm, an area without oysters but affected by oyster biodeposits, and a control station. Axial tomodensitometry (CT-scan) was used for three-dimensional visualization and two-dimensional analysis of the cores. Biogenic structure volumes were quantified and compared between cores. We collected the macrofauna, living foraminifera, shells and gravel from the cores after scanning, to validate image analysis. We did not investigate differences in the biogenic structure volume between cores. However, biogenic structure volume is not necessarily proportional to the extent of bioturbation in a core, given that many biodiffusive activities cannot be detected on CT-scans. Biodiffusors and larger gallery-diffusors were abundant in macrofaunal assemblage at the control station. By contrast, macrofaunal assemblages consisted principally of downward-conveyors at the two stations affected by oyster farming. At the control station, the vertical distribution of biogenic structures mainly built by the biodiffusor Scorbicularia plana and the large gallery-diffusor Hediste diversicolor was significantly correlated with the vertical profiles of living foraminifera in the sediment, whereas vertical distributions of foraminifera and downward-conveyors were not correlated at the station affected by oyster farming. This relationship was probably responsible for the collection of foraminifera in deep sediment layers (> 6 cm below the sediment surface) at the control station. As previously suggested for other species, oxygen diffusion may occur via the burrows built by S. plana and H. diversicolor, potentially increasing oxygen penetration and providing a favorable microhabitat for foraminifera in terms of oxygen levels. By contrast, the absence of living foraminifera below 6 cm at the stations affected by oyster farming was probably associated with a lack of biodiffusor and large gallery-diffusor bioturbation. Our findings suggest that the effect of macrofaunal bioturbation on the vertical distribution of foraminiferal assemblages in sediments depends on the effects of the macrofauna on bioirrigation and sediment oxidation, as deduced by Eh values, rather than on the biogenic structure volume produced by macrofauna. The loss of bioturbator functional diversity due to oyster farming may thus indirectly affect infaunal communities by suppressing favorable microhabitats produced by bioturbation.

Résumé : Estuarine systems are complex environments where seasonal and spatial variations occur in concentrations of suspended particulate matter, in primary constituents, and in organic matter content. This study investigated in the laboratory the flocculation potential of estuarine-suspended particulate matter throughout the year in order to better identify the controlling factors and their hierarchy. Kinetic experiments were performed in the lab with a "video in lab" device, based on a jar test technique, using suspended sediments sampled every 2 months over a 14-month period at three stations in the Seine estuary (France). These sampling stations are representative of (1) the upper estuary, dominated by freshwater, and (2) the middle estuary, characterized by a strong salinity gradient and the presence of an estuarine turbidity maximum. Experiments were performed at a constant low turbulent shear stress characteristic of slack water periods (i.e., a Kolmogorov microscale > 1,000 A mu m). Flocculation processes were estimated using three parameters: flocculation efficiency, flocculation speed, and flocculation time. Results showed that the flocculation that occurred at the three stations was mainly influenced by the concentration of the suspended particulate matter: maximum floc size was observed for concentrations above 0.1 g l(-1) while no flocculation was observed for concentrations below 0.004 g l(-1). Diatom blooms strongly enhanced flocculation speed and, to a lesser extent, flocculation efficiency. During this period, the maximum flocculation speed of 6 A mu m min(-1) corresponded to a flocculation time of less than 20 min. Salinity did not appear to automatically enhance flocculation, which depended on the constituents of suspended sediments and on the content and concentration of organic matter. Examination of the variability of 2D fractal dimension during flocculation experiments revealed restructuring of flocs during aggregation. This was observed as a rapid decrease in the floc fractal dimension from 2 to 1.4 during the first minutes of the flocculation stage, followed by a slight increase up to 1.8. Deflocculation experiments enabled determination of the influence of turbulent structures on flocculation processes and confirmed that turbulent intensity is one of the main determining factors of maximum floc size.

Pituitary adenomas are currently classified by histological, immunocytochemical and numerous ultrastructural characteristics lacking unequivocal prognostic correlations. We investigated the prognostic value of a new clinicopathological classification with grades based on invasion and proliferation. This retrospective multicentric case-control study comprised 410 patients who had surgery for a pituitary tumour with long-term follow-up. Using pituitary magnetic resonance imaging for diagnosis of cavernous or sphenoid sinus invasion, immunocytochemistry, markers of the cell cycle (Ki-67, mitoses) and p53, tumours were classified according to size (micro, macro and giant), type (PRL, GH, FSH/LH, ACTH and TSH) and grade (grade 1a: non-invasive, 1b: non-invasive and proliferative, 2a: invasive, 2b: invasive and proliferative, and 3: metastatic). The association between patient status at 8-year follow-up and age, sex, and classification was evaluated by two multivariate analyses assessing disease- or recurrence/progression-free status. At 8 years after surgery, 195 patients were disease-free (controls) and 215 patients were not (cases). In 125 of the cases the tumours had recurred or progressed. Analyses of disease-free and recurrence/progression-free status revealed the significant prognostic value (p < 0.001; p < 0.05) of age, tumour type, and grade across all tumour types and for each tumour type. Invasive and proliferative tumours (grade 2b) had a poor prognosis with an increased probability of tumour persistence or progression of 25- or 12-fold, respectively, as compared to non-invasive tumours (grade 1a). This new, easy to use clinicopathological classification of pituitary endocrine tumours has demonstrated its prognostic worth by strongly predicting the probability of post-operative complete remission or tumour progression and so could help clinicians choose the best post-operative therapy.

Long-term exposure to persistent pollutants with hormonal properties (endocrine-disrupting chemicals, EDCs) may contribute to the risk of prostate cancer (PCa). However, epidemiological evidence remains limited. We investigated the relationship between PCa and plasma concentrations of universally widespread pollutants, in particular p,p'-dichlorodiphenyl dichloroethene (DDE) and the non-dioxin like polychlorinated biphenyl congener 153 (PCB153). We evaluated before treatment 576 men with newly diagnosed PCa and 655 controls in Guadeloupe (French West Indies). Exposure was analyzed according to case-control status. Associations were assessed by unconditional logistic regression analysis, controlling for confounding factors. Missing data were handled by multiple imputation. We estimated a significant positive association between DDE and PCa (adjusted odds ratio [OR] 1.53; 95% CI 1.02, 2.30 for the highest versus lowest quintile of exposure; PTrend = 0.01). PCB153 was inversely associated with PCa (OR 0.30; 95% CI 0.19, 0.47 for the highest versus lowest quintile of exposure values; PTrend < 0.001). Also, PCB153 was more strongly associated to low-grade than high grade PCa. Associations of PCa with DDE and PCB153 were in opposite directions. This may reflect differences in the mechanisms of action of these EDCs, and although our findings need to be replicated in other populations, they are consistent with complex effects of EDCs on human health.

Résumé : In Taiwan , the subduction of the Chinese continental margin under the Luzon arc results in the progressive growth of an active orogenic wedge. It is one of the best places to study the complex relationships that occurs between the tectonic processes controlling deformation (plate rheology and kinematics, metamorphism and magmatism) and surface processes (erosion and sedimentation). In order to study the kinematics of the present day convergence of Philippine Sea plate to the Asian continent around Taiwan, we perform 2D and 3D sandbox modeling and we analyzed the models with PIV (Particle Image Velocimetry) software. In 2D experiments, a model Coulomb wedge is submitted to erosion and sedimentation under topography and flux steady state conditions. The 3D experiments are mainly used to study the escape tectonics occurring in the middle west and southwest Taiwan. A wedge shaped indenter and a basement relief were used to simulate the mechanical plate boundary between Philippine Sea plate and Eurasia plate and the basement topography to the west of Taiwan respectively. We examine the exhumation patterns, the mode of fault propagation and displacement patterns by strain partitioning of vertical vs. horizontal displacement and convergent parallel vs. orogen parallel components. The preliminary conclusions are: 1. The interactions between tectonics, erosion and sedimentation control geometry and kinematics of the doubly vergent transpressional orogen. 2. Exhumation processes are controlled by the balance between erosion, frontal accretion and underplating in different structural domains of the wedge. 3. Retrowedge erosion favorites backfolding and backthrusting, induces flower structures and fan shaped cleavage distribution. 4. Sedimentation promotes underplating and out-of sequences thrusting. 5. Continental subduction or subduction of topographic highs will enhance underthrusting/underplating, thick-skin tectonics and inversion of sedimentary basin.

BACKGROUND: This study examines gene-environment interaction between the MTHFR C667T polymorphism and folic acid in the etiology of orofacial clefts (OFC). We used a pooled-analytical approach on four studies that used similar methods. METHODS: We used logistic regression to analyze the pooled sample of 1149 isolated cases and 1161 controls. Fetal and maternal MTHFR C677T genotypes, and maternal periconceptional exposure to smoking, alcohol, vitamin containing folic acid and folic acid supplements were contrasted between the cleft types [non-syndromic clefts lip or without cleft palate (CL(P)) and non-syndromic cleft palate (CP)] and control groups. RESULTS: There was a reduced risk of CL(P) with maternal folic acid use (p = 0.008; OR = 0.70, 95% CI: 0.65-0.94) and with supplements containing folic acid (p = 0.028, OR = 0.80, 95% CI: 0.65-0.94). Maternal smoking increased the risk of both CL(P) (p < 10 e-3; OR = 1.62, 95% CI: 1.35-1.95) and CP (p = 0.028; OR = 1.38, 95% CI: 1.04-1.83). No significant risk was observed with either maternal or fetal MTHFR C677T genotypes. CONCLUSION: This individual participant data (IPD) meta-analysis affords greater statistical power and can help alleviate the problems associated with aggregate-level data-sharing. The result of this IPD meta-analysis is consistent with previous reports suggesting that folic acid and smoking influence OFC outcomes.

Résumé : Genetic variation was surveyed at 11 microsatellite loci and at 517 bp of the mitochondrial control region to investigate the presence of genetic stock structure in swordfish (Xiphias gladius) in four proximal localities of the southwest Indian Ocean. One aim of this study was to serve as a preliminary examination for congruency of structure detected by these two genetic markers, prior to conducting a more comprehensive basin-wide survey of the Indian Ocean and nearby surrounding areas. Analyses of multilocus microsatellite genotypes and mitochondrial control region sequences both revealed a great homogeneity between samples. Genetic diversity detected at the regional scale was not significantly higher than detected at the local scale. Results suggest that the southwest Indian Ocean globally functions as a unique panmictic population. However, some discrete genetic differences appeared that could possibly indicate influence from a second genetic pool in the northern part of the Indian Ocean. This structure appeared to be sex-dependent with genetic differences higher among female than among male samples. This result may indicate a higher level of spawning area fidelity for females with a subsequent sampling bias tending to homogenise male genotypic distributions. (c) 2009 Elsevier B.V. Résumé : French Guiana is a region located on the Guiana Shield in South America. Is presents at the same time a high originality in biodiversity, a good state of conservation and a strong correlation to Amazonian carbon stocks. Knowledge on evolution of the forest structure of French Guiana is however still insufficient to bring an optimal management at territory scale. Availability as never seen before of remote sensing data at high spatial resolution such as LiDAR, on large coverage, offers the opportunity to assess the forest resource in French Guiana. The work done during this thesis aim mostly to study the structure and the variations of the canopy height, the factors determining its variability and the consequences of forest logging activities. The data used during this thesis come from an airborne LiDAR and field data from Office National des Forêts (ONF). This thesis manuscript is divided in three parts. The first part introduces the problem and sets the objectives of the thesis. The second part takes the so-called "article" form, that is, the different chapters that compose it are scientific articles published or in the course of publication, it is composed of three studies (I) A study on the gaps detection and environmental determinants on the gap sizes distribution in natural forest: How to characterize the gap size distribution and are there environmental determinants that influence the statistical distribution of gap size? This article has been submitted in Biogeoscience. (ii) A study of the control of the environment on the canopy height structure and variations at different spatial resolutions. Which spatial scales are relevant for analyzing the environmental determinants of canopy height changes in tropical forests from remote sensing data? How can the environment help or not map the height of the canopy in the rainforest? This article was accepted for publication in Remote Sensing. (iii) a third study where we quantified the effects of Reduced Impact Logging (RIL) on the structure and variation of canopy height. What are the effects of logging on canopy height variations and gap size distribution? This article will be submitted for publication in a Forest Ecology and Management journal. In the last part, after a synthesis of the main results, we propose a conclusion.
La Guyane française est une région située sur le Plateau des Guyanes en Amérique du sud. Elle présente à la fois une grande originalité en termes de biodiversité, un bon état de conservation et une forte contribution au stock de carbone Amazonien. Les connaissances sur l’évolution de la structure forestière de la Guyane française restent cependant insuffisantes pour offrir une gestion optimale à l’échelle du territoire. La disponibilité sans précédent de données de télédétection à très haute résolution spatiale telles que le LiDAR , sur de larges couvertures nous offre l'opportunité d’évaluer la ressource forestière de la Guyane Française. Les travaux réalisés dans le cadre de cette thèse visent prioritairement à étudier la structure et les variations de la hauteur de canopée, les facteurs déterminant sa variabilité ainsi que les conséquences des activités d’exploitation forestière. Les données utilisées au cours de la thèse proviennent d’un LiDAR aéroporté et des données de terrains de l'Office National des Forêts (ONF). Ce manuscrit de thèse est divisé en trois grandes parties. La première partie introduit la problématique et fixe les objectifs de la thèse. La seconde partie prend la forme dite « sur article », c'est-à-dire que les différents chapitres qui la compose sont des articles scientifiques publiés ou en cours de publication, elle est composée de trois études. (i) Une étude concernant la détection des trouées et des déterminants environnementaux sur la distribution de tailles des trouées en forêt non exploitée : Comment caractériser la distribution de taille des trouées et est-ce qu’il y a des déterminants environnementaux qui influencent la distribution statistique de taille de trouées? Cet article à été soumis dans Biogeoscience. (ii) Une étude du contrôle de l'environnement sur la structure et les variations de la hauteur de canopée à différentes résolutions spatiales. Quelles échelles spatiales sont pertinentes pour analyser les déterminants environnementaux des variations de hauteur de la canopée en forêt tropicale à partir des données de télédétection ? Comment l’environnement peut aider, ou pas, à cartographier la hauteur de la canopée en forêt tropicale ? Cet article à été accepté pour publication dans Remote Sensing. (iii) Une troisième étude où nous avons quantifié les effets de l’exploitation à faible impact (EFI) sur la structure et la variation de la hauteur de canopée. Quelles sont les effets de l’exploitation forestière sur les variations de hauteur de canopée et sur la distribution de taille des trouées ? Cet article sera soumis pour publication dans une revue scientifique (Forest Ecology and Management). Dans la dernière partie, après une synthèse des principaux résultats, nous proposons des pistes de développement.
