Analyse économique d'une nouvelle technique d'élevage de crevettes en cages flottantes au Brésil Auteur(s) : Paquotte, Philippe Éditeur(s) : Actes de la 8ème Conférence de l'IIFET (Marrakech, juillet 1996) Résumé : In the framework of French-Brazil co-operation, the French Institute of research for exploitation of the sea (IFREMER), the State of Bahia and the company Littoral Sul Maricultura have developed a new technique for shrimp farming (Penaeus vannamei) using floating cages. After trials carried out during three years, this technique turns out very interesting from several viewpoints. The growth speed and the survival rate are very good, the negative effects on the environmental quality are weak and local fishermen have proved eager to adopt this technique as a part-time activity. In the same time, a technico-economic analysis, using a computerised simulation tool, as well as a market analysis in the State of Bahia have been carried out in order to assess the feasibility of such a project and to help research and development decisions. Despite low first investment and good biological results, the ex-farm production cost of shrimps weighing 15 g is as high as 4,5 US$ per kilo. This price is due to the low productivity of the labour and to the cost of nets which have a short life-time. It is possible to consider this activity as profitable on the market of Salvador de Bahia. Nevertheless, the question is how to face the international competition, which is particularly strong in the case of tropical shrimps. The conditions allowing enterprises to develop a strategy based on product quality and environmentally friendliness are reviewed. [NOT CONTROLLED OCR] Dans le cadre de la coopération Franco-Brésilienne, l'IFREMER, l'Etat de Bahia et la société Littoral Sul Maricultura ont développé une nouvelle technique d'élevage de crevettes pénéides (Penaeus vannamei) en utilisant des cages flottantes. Après trois années de mise au point, cette technique s'est avérée intéressante sur plusieurs aspects. La vitesse de croissance et le taux de survie sont très satisfaisants, les effets négatifs sur la qualité du milieu naturel sont faibles et les pêcheurs locaux ont accueilli favorablement cette nouvelle pratique en complément de leur activité traditionnelle. Parallèlement, une analyse technico-économique utilisant un outil de simulation informatique ainsi qu'une analyse du marché de la crevette dans l'état de Bahia ont été conduites afin d'évaluer la faisabilité d'un tel projet et d'orienter les actions de recherche et développement. En dépit d'investissements initiaux plus faibles que dans les élevages en bassins et de bonnes performances zootechniques, le coût de production de crevettes de 15 g atteint 4,7 US$ par kilo. Dans les conditions de marché de l'Etat de Bahia, la rentabilité d'un projet d'élevage de crevettes en cage serait assurée, mais la faible productivité du travail et le coût de remplacement des filets ne permettent pas d'avoir des coûts de production aussi bas que dans les élevages en bassins. C'est pourquoi la question se pose de la possibilité pour ce type de production innovante d'affronter une concurrence internationale très forte comme dans le cas des crevettes tropicales. "Summer syndrome" in Litopenaeus stylirostris grow out ponds in New Caledonia: zootechnical and environmental factors Auteur(s) : Lemonnier, Hugues Herbland, Alain Salery, Lucas Soulard, Benoit Éditeur(s) : Elsevier Résumé : The "Summer Syndrome", a septicemic vibriosis caused by Vibrio nigripulchritudo, is responsible for one of the two main seasonal mortalities which affect shrimp aquaculture in New Caledonia. It was identified for the first time in December 1997 in an intensive shrimp farm (called farm DF) and has been enzootic ever since. The "Summer syndrome" affects shrimp during warm-season growout. Although the geographic area concerned is limited, it is a potential threat for the industry in that the disease could spread. Analysis of grow out data from 1991 to 2002 in this farm was carried out in order to characterize the disease and the environmental conditions in relation to mortality. Results of farm DF were compared to data from farms not affected by the disease (called farms HC). The "Summer syndrome" occurs during the warm season which is characterized by an increase in water temperature and sunny duration. This disease is characterized by simultaneously highest densities and weakest drying duration between crops in the farms studied. Between 1991 and 2002, the increase of feed input and nitrogen rate in feed has led to a significant increase of the growth rate over the years in farm DF. At the same time, water management has not changed and has induced an increase in environmental waste production. In consequence, an early eutrophication of water is observed, which is concomitant with the beginning of the mortality outbreaks. This could play a role by inducing directly or indirectly a stress for shrimp and/or a growth and/or virulence factors of the pathogen. Searching for genetic markers of virulence in Vibrio nigripulchritudo, a Litopenaeus stylirostris shrimp pathogen Auteur(s) : Reynaud, Yann Éditeur(s) : Colloque IMBC - International Marine Biotechnology Conference Résumé : Since 1997, a new pathology seasonally occurs in new caledonian shrimp farms during the warm season. Diseased Litopenaeus stylirostris shrimp suffer from a systemic vibriosis which was attributed to highly pathogenic Vibrio nigripulchritudo. At the present time, only two farms among 17 are affected by the so called "summer syndrome". In such a context it appears urgent to develop reliable diagnostic tools to detect V. nigripulchritudo strains and to differentiate highly virulent strains from non virulent ones in order to understand the mechanisms of infection and overcome disease spreading. Multilocus Sequencing Typing (MLST) analysis on 24 bacterial strains isolated in geographically distinct shrimp farms highlighted the existence of a cluster A containing only strains displaying moderate to high virulence. Since experimental infection procedure for virulence status determination is material intensive and time consuming, and MLST method not accurate enough to inform on the virulence status of strains, it appeared of great interest to develop other diagnostic tools. Two phylogenetically closed strains, one highly virulent SFn1 and the other non-virulent SFn118 were selected in order to characterize genes specific of the virulent strain by Substractive Suppression Hybridization (SSH). Firstly, a total of 622 DNA fragments specific of SFn1 were sequenced, characterized and further used as probes in a DNA macroarray. Testing 24 strains, hybridization profiles were found to be strongly correlated with both MLST genotypes and bacterial virulence status. A total of 96 representative probes were secondly selected to validate macroarray results on 34 more additionnal strains, allowing identification of genetic markers of virulence. Most of DNA fragments specific of highly virulent strains associated to summer syndrome were found to be bore by a plasmid successfully isolated in the laboratory. In next future, these results would permit the development of relevant diagnostic tools useful for accurate epidemiological survey of the summer syndrome. Using sequence polymorphisms evidenced in this previous MultiLocus Sequence Typing study, we developed two new quantitative PCR assays permitting the detection and quantification of V. nigripulchritudo at the genospecies level using SYBR Green I chemistry and at the emerging cluster level using Fluorescence Resonance Energy Transfer technology with hybridization probes. The use of this molecular diagnostic tool evidenced the colonization of the shrimp pond ecosystem by the pathogenic cluster at least at the onset of the disease. March 2 - 6, 2009 Tahiti, French Polynesia Résumé : New Caledonian shrimp farming feasibility studies started in the mid-seventies. Today ca. 2000 tons of Litopenaeus stylirostris, introduced in 1978, are semi-intensively produced each year. Since fifteen years, production has been impacted by two seasonal vibriosis, the “Summer” syndrome caused by Vibrio nigripulchritudo in relation to pond bottom and water column quality, and the “Winter Syndrome” caused by Vibrio penaeicida in relation to drastic temperature drops . With the ambition of elucidating processes possibly leading to mortality outbreaks in ponds, Ifremer has been conducting a research program since 2003 under the DESANS project (2003-2006) and the present DEDUCTION project, equally funded by the South and North Provinces and by the Government of New Caledonia. This program has been designed in a multidisciplinary approach integrating the suspected multifactorial origin of shrimp mortalities. Environment quality in ponds (either sediment or water column) plays a major role in the shrimp ecophysiological status, influencing sensitivity to pathogens . DEDUCTION focuses on a better knowledge of phytoplankton composition and biogeochemical fluxes at the sediment-water interface in ponds. Impact of farm discharges is also investigated. In addition, a more fundamental work is conducted to understand the pathogenicity mechanisms of these two Vibrio species but also to prevent the occurrence of viral infections through a RNA interference approach. Markers of the ecophysiological status of broodstock, larvae, post-larvae and reared animals, such as enzymes testifying from an oxidative stress, are developed. Gain of performance (resistance to pathogens and growth) were obtained on hybrids of the New Caledonian and introduced Hawaiian strains. Finally the database Stylog is exploited in view of discriminating “good” from “bad” rearings on the basis of relevant indicators. Les études de faisabilité de l'aquaculture de crevettes ont débuté en Nouvelle-Calédonie dans les années soixante dix. Aujourd'hui environ 2000 tonnes de Litopenaeus stylirostris, introduite en 1978, sont produites chaque année de façon semi-intensive. Depuis quinze ans la production a été impactée par deux vibrioses : le syndrome d'été provoqué par Vibrio nigripulchritudo en relation avec la qualité de la tranche d'eau et des fonds de bassins et le syndrome d'hiver provoqué par Vibrio penaeicida en relation avec des chutes rapides de température. Dans le but d'élucider les processus entrainant des mortalités dans les élevages, Ifremer a conduit un programme de recherche depuis 2003 dans le cadre du projet DESANS (2003-2006) puis du présent projet DEDUCTION co-financé par les provinces Nord et Sud et le Gouvernement de la Nouvelle-Calédonie. Evaluation du probiotique bactérien Pediococcus acidilactici MA18/5M chez la crevette pénéide Litopenaeus stylirostris en Nouvelle- Calédonie Auteur(s) : Castex, Mathieu Éditeur(s) : Agro Paris Tech Résumé : P. acidilactici MA 18/5M is a dietary probiotic (BACTOCELL®) which has shown its efficiency on several terrestrial and aquatic species. The present thesis aimed to investigate more fully the effects of this probiotic on cultured penaeid shrimps. For this study, we chose a global approach by replacing the problem studied in a model of interactions between three compartments: the shrimp, its intestinal microflora and the probiotic. Having first defined how the probiotic should be administrated, we studied its effects under controlled laboratory conditions at several levels of Litopenaeus stylirostris biology: (i) Nutritional status and growth, (ii) associated bacterial microflora, and (iii) health status with respect to oxidative stress. Then the probiotic improves the growth of the shrimps and the feed conversion ratio. Our study shows that these effects are partially due to a better use of the feed, particularly of dietary carbohydrates, but also suggest an action on the metabolism, andor on the growth via a contribution of essential nutrients. On the other hand, although the intestinal bacterial communities seems little affected, we show an antagonism effect of the probiotic toward total and vibrionacea intestinal flora, as well as a decrease in the level of infection of shrimps during challenge with pathogenic V. nigripulchritudo. Beside, our works highlight that infection by this pathogen, led to a reduction in antioxidant defences, to a rise in oxidative damages, and to a release of mortalities within 48 hours. This oxidative stress following bacterial infection was better controlled by animals receiving the probiotic diet, and resistance of the shrimps to infection was significant improved. A nutritional hypothesis is proposed to explain such result, hypothesis based on a link between carbohydrate metabolism and antioxidant status. As a conclusion of these works, we can propose a first explanatory scenario for the effects of P. acidilactici on a penaeid shrimp. Finally probiotic's effects were assessed under a pilot scale in a commercial farm from New Caledonia currently affected by the "summer syndrome ". We confirm the improvements brought in term of zootechnical results, with in particular, a better resistance to this syndrome. This work was made possible thanks to a breeding method in floating cages, developed during this thesis, and therefore transferred, as an experimental tool, to the shrimp industry of New Caledonia. Pediococcus acidilacticii MA18/5M est un probiotique bactérien (BACTOCELL®) qui a démontré son efficacité chez plusieurs espèces d'animaux terrestres et aquatiques. Cette thèse avait pour objectif d'investiguer plus en détails les effets de ce probiotique chez les crevettes pénéides d'élevage. Pour cette étude, nous avons choisi une approche globale replaçant la problématique dans un modèle d'interactions entre trois compartiments : la crevette, sa microflore intestinale et le probiotique. Après avoir défini les modalités d'administration, l'effet de P. acicilactici a été étudié, en conditions de laboratoire, chez la crevette Litopenaeus stylirostris, à différents niveaux: (i) statut nutritionnel et croissance, (ii) microflore bactérienne associée, et (iii) santé de l'animal à travers son statut antioxydant et son niveau de stress oxydant. Ainsi, le probiotique améliore la croissance des crevettes et l'indice de conversion de l'aliment. Notre étude montre que ces effets sont en partie liés à une meilleure utilisation de l'aliment, notamment des carbohydrates alimentaires, mais suggèrent aussi une action sur le métabolisme, et/ou sur la croissance via un apport de nutriments essentiels. D'autre part, bien que les communautés bactériennes intestinales semblent peu affectées, un effet d'antagonisme du probiotique vis-à-vis de la flore intestinale totale et vibrionacée a été montré, ainsi qu'une réduction du niveau d'infection des crevettes au cours de challenge avec le pathogène V. nigripulchritudo. Par ailleurs, nous mettons en évidence que l'infection par ce pathogène, conduit à un abaissement des défenses antioxydantes, à une élévation des dégâts oxydatifs, et au déclenchement de mortalités dans les 48 heures. Le probiotique atténue le niveau de ce stress oxydant, et améliore significativement la résistance des crevettes à l'infection. Une hypothèse nutritionnelle est proposée afin d'expliquer ce résultat, hypothèse reposant sur un lien entre le métabolisme des carbohydrates et le statut antioxydant. A l'issue de ces travaux, nous pouvons alors proposer un premier scénario explicatif des effets de P. acidilactici chez une crevette pénéide. Finalement le probiotique a été éprouvé à l'échelle pilote sur une ferme commerciale de Nouvelle-Calédonie affectée par le « syndrome d'été ». Nous confirmons les améliorations apportées en termes de résultats zootechniques, avec notamment une meilleure résistance au syndrome. Ce travail a été réalisé grâce une méthode d'élevage en cages flottantes, mis au point au cours de cette thèse, et qui a dés lors été transférée, comme outil expérimental, à la filière crevetticole Calédonienne. Les échanges internationaux de crevettes pénéidés vivantes entre les pays du Pacifique, de l'Atlantique et de l'Europe Auteur(s) : Michel, Alain Éditeur(s) : Office Int Epizooties Résumé : The tremendous development of penaeid shrimp culture across the world over the past twenty years has led to international trade in eggs, larvae and spawners of the best shrimp species for aquaculture. Trade has involved, in particular, the following species: - Penaeus japonicus from Japan to Europe, the Pacific Islands and South America - P. monodon from South-East Asia to almost all tropical countries - P. vannamei and P. stylirostris from countries along the Pacific coast of South and Central America to the United States of America, the islands of the South Pacific, countries along the Atlantic coast of the Americas and certain countries in Africa. In the 1980s, research conducted by American and French teams enabled the breeding of stocks of spawners in captivity. This resulted in the establishment of lines of captive broodstock outside natural areas of distribution, and the development of new breeding farms in previously unproductive regions. Trade related to movements of these species is facilitated by the absence of legislation in most countries; even when such legislation does exist, it is not always applied These practices have led to the rapid development of world shrimp production, but have also contributed to the dissemination of pathogens. In recent years, the occurrence of serious epizootics - occasioning heavy losses world-wide - has led to a more cautious approach involving trade of post-larvae obtained from captive broodstock in which thorough control measures have been implemented for known diseases. Trade in wild shrimp will probably be abandoned progressively, to avoid the risk of introducing new pathogens. The main challenges for research in the near future are the development of diagnostic tools, to enable continuous control of captive broodstock, and the selection of strains which are resistant to the principal pathogens affecting these species. The "Syndrome 93" is a cold season disease due to Vibrio penaeicida affecting all shrimp farms, while the "Summer Syndrome" is a geographically restricted vibriosis caused by a virulent lineage of Vibrio nigripulchritudo. Microbiological procedures for diagnosis of these diseases are time-consuming and do not have the ability to discriminate the range of virulence potentials of V. nigripulchritudo. In this study, we developed a multiplex PCR method to simultaneously detect these two bacterial species and allow for pathotype discrimination. The detection limits of this assay, that includes an internal amplification control to eliminate any false-negative results, were determined at 10 pg purified DNA and 200 cfu/ml. After confirming the effectiveness of our method using experimentally infected animals, its accuracy was compared to standard biochemical methods during a field survey using 94 samples collected over 3 years from shrimp farms encountering mortality events. The multiplex PCR showed very high specificity for the detection of V. penaeicida and V. nigripulchritudo (inclusivity and exclusivity 100%) and allowed us to detect the spreading of highly pathogenic isolates of V. nigripulchritudo to a farm adjoining the "Summer Syndrome area." This assay represents a simple, rapid, and cost-effective diagnostic tool for implementing timely risk management decisions but also understanding the seasonal and geographical distribution of these pathogens. In the laboratory, the bioaccumulation kinetics of Ag, Cd, Co, Cr, and Zn have been investigated in shrimp exposed via seawater and food, using the corresponding gamma-emitting radiotracers (Ag-110m, Cd-109, Co-57, Cr-51, and Zn-65) and highly sensitive nuclear detection techniques. Results showed that hepatopancreas and intestine concentrated the metals to the highest extent among the blue shrimp organs and tissues. Moulting was found to play a non negligible detoxification role for Co, Cr and, to a lesser extent, Zn. Metal retention by L stylirostris widely varied (from a few days to several months), according to the element and exposure pathway considered (a given metal was usually less strongly retained when ingested with food than when it was taken up from the dissolved phase). In the field study, Ag, As, Cd, Co, Cr, Cu, Mn, Ni, and Zn were analysed in shrimp collected from a New Caledonian aquaculture pond. Metal concentrations in the shrimp muscles were generally relatively low and results confirmed the role played by the digestive organs and tissues in the bioaccumulation/storage/detoxification of metals in the Pacific blue shrimp. Colonization of coastal environments by foraminifera: insight from shrimp ponds in New Caledonia (sw Pacific) Auteur(s) : Debenay, J. -p. Della Patrona, Luc Goguenheim, H. Éditeur(s) : Cushman Foundation for Foraminiferal Research Résumé : The objectives of this study were to observe foraminiferal colonization patterns and behavior in shrimp ponds in New Caledonia during a shrimp-growing (farming) cycle. Weekly collecting at 10 stations in 8 shrimp ponds yielded a total of 170 samples accompanied by environmental data. Seawater pumped from the nearby ocean filled the ponds at the beginning of the growth cycle and its daily renewal maintained salinity at 32-39, and supplied the ponds with influxes of smaller, mostly juvenile, foraminifera. A few days after initial filling, the pioneering species Ammonia tepida and Quinqueloculina seminula appeared, with A. tepida dominant. Their high reproduction rates increased both living and dead assemblage densities during the first 10 weeks. Populations of these two species then stabilized with higher oxygen demand (drop of redox) and consumption of living foraminifera by shrimp. Only a few colonizers subsequently appeared, which was attributed to the isolation of the pond, despite the high rate of water renewal. Only one pond had notably higher species richness, but it could not be distinguished from the others by its physicochemical parameters. The species that appeared in ponds initially barren of foraminifera also survived where there was water seepage between growing cycles, suggesting that the assemblages had already reached equilibrium with the environment. Despite the number of environmental parameters measured, only oxygen and reactive organic matter correlated with the microfauna on a weekly timescale. We assume that other parameters do not significantly affect foraminifera until they reach critical threshold. Consistent with previous studies, A. tepida was the species most tolerant of organic influx, but its relative abundance dropped once the organic matter flocculated and settled, leading to disoxic conditions in the sediment. Conversely, Q. seminula was able to climb through the floc and reach the oxygenated layer, where its relative abundance increased. Effect of water exchange on effluent and sediment characteristics and on partial nitrogen budget in semi-intensive shrimp ponds in New Caledonia Auteur(s) : Lemonnier, Hugues Faninoz, Sebastien Éditeur(s) : Blackwell science Résumé : An experiment was conducted in six earthen ponds with 20 shrimps m(-2) Litopenaeus stylirostris (Stimpson) during the warm season in New Caledonia to study the dynamics of wastes in relation with water exchange rate (WER). The nitrogen budget was established, taking into account the different forms of nitrogen in the water, sediment, feed and shrimp. Data from a wide range of treatments applied in unreplicated ponds were treated using regression analysis to establish the relationship between WER and partial nitrogen budget, sediment characteristics and shrimp performance. To compare effluent quality between treatments during the season, data were analysed using the non-parametric sign test. The water outflow was characterized by a decrease in the concentrations of N-mineral forms (TAN, NO2--NO3-), an increase in the concentration of organic soluble and sestonic organic forms (expressed in terms of particulate nitrogen, particulate organic carbon, chlorophyll a) compared with the water inflow. Increasing WER increased the amount of exported wastes and mainly in the organic forms and TAN can be considered as negligible. The nitrogen budget showed that 19-46% of nitrogen input (feed+water) was exported into the coastal environment. The results showed that the quality of the sediment decreased as WER decreased. The potential negative impact of the developing industry in New Caledonia on the costal environment could be partially reduced in a first step by decreasing WER. However, if applied in the farms, this practice should be linked to a close survey of the evolution of sediment quality. Vibrio nigripulchritudo is an emerging pathogen of the cultured shrimp Litopenaeus stylirostris in New Caledonia and other regions in the Indo-Pacific. The molecular determinants of V. nigripulchritudo pathogenicity are unknown; however, molecular epidemiological studies have revealed that recent pathogenic V. nigripulchritudo isolates from New Caledonia all cluster into a monophyletic clade and contain a small plasmid, pB1067. Here, we report that a large plasmid, pA1066 (247 kb), can also serve as a marker for virulent V. nigripulchritudo, and that an ancestral version of this plasmid was likely acquired prior to other virulence-linked markers. Additionally, we demonstrate that pA1066 is critical for the full virulence of V. nigripulchritudo in several newly developed experimental models of infection. Plasmid pB1067 also contributes to virulence; only strains containing both plasmids induced the highest level of shrimp mortality. Thus, it appears that these plasmids, which are absent from non-pathogenic isolates, may be driving forces, as well as markers, for the emergence of a pathogenic lineage of V. nigripulchritudo. Influence of sediment characteristics on shrimp physiology: pH as principal effect Auteur(s) : Lemonnier, Hugues Bernard, E Boglio, E Goarant, Cyrille Cochard, Jean-claude Éditeur(s) : Elsevier Résumé : Penaeid shrimp reared in earthen ponds are exposed to sediment, which can, in some instances, induce a stress. In seawater, the osmoregulatory capacity (hyporegulation) is a useful tool to compare the physiological condition of shrimp exposed to various stressors. By keeping some shrimp in cages at different locations of a single pond heterogeneous in terms of sediment quality, it was possible, using osmotic pressure (OP), to identify some locations where the stress was maximum and some others where it was minimum. Simultaneously, sediment samples were taken and analysed in order to evaluate some physico-chemical parameters that could be related to the stress observed in the shrimps kept in the cages. This approach allowed to show a significant positive correlation between the pH of the sediment surface and the shrimp osmotic pressure. This result was confirmed in a study carried out in experimental 70-1 tanks, where osmotic pressure decreased significantly as water pH decreased from 7.0 to 6.5. Impact de la crevetticulture calédonienne sur l'environnement : état des lieux, recherche de traceurs des effluents et définition d'un programme de recherche pluriannuel Auteur(s) : Lemonnier, Hugues Thomas, Yoann Legrand, Alexandre Martin, Jean-louis Herbland, Alain Résumé : In the booming context of shrimp culture in French Caledonia, some research actions need to be considered in order (i) to make sure this production will not impact the quality and biodiversity of the coastal environment it is contingent upon (ii) for the field to highlight, based on objective bases, its good "environmental image", for an efficient commercialization of its products. In the framework of the MOM project, a number of actions have been carried out: Action 1: A bibliographical synthesis of the impact of shrimp culture on coastal environment has been conducted, taking into account the works carried out throughout the world and in French Caledonia in order to draw the main issues the Territory will have to deal with. It seems necessary to start with an assessment of the future and the impact of the effluents on the coastal environment. Action 2: With this aim in mind, one or more indicators, among the lagoon's environmental variables, of the influence of the farms' rejects on the coastal environment, have been searched for. The first results are presented in this report. Action 3: In parallel, a long-term program taking into account the different aspects of the problem in its entirety and complexity has been defined in relation with our partners in French Caledonia. With this aim in mind, a symposium entitled "Ecosystems and shrimp culture", organised by the Ifremer in French Caledonia, with the support of the IRD, was held in Nouméa from June 22nd to June 24th, 2005. This symposium enabled the drawing out of the research perspective on the benthos, on the functioning of the lagoon ecosystem, and on the evolution of the mangrove swamps subjected to organic matter inflows from farms. Dans un contexte d'essor de la filière crevette de Nouvelle Calédonie, des actions de recherche doivent être envisagées (i) pour assurer que cette production n'hypothéquera pas la qualité de l'environnement littoral et sa biodiversité dont elle dépend et (ii) pour que la filière puisse mettre en avant, sur des bases objectives, son image «environnementalement correcte» garante de la bonne commercialisation de son produit. Dans le cadre de ce projet MOM, plusieurs actions ont été menées : Action 1 : Une synthèse bibliographique de l'impact de l'aquaculture de crevettes sur l'environnement littoral a été réalisée en prenant en compte les travaux conduits à travers le monde et en Nouvelle-Calédonie afin de dégager les principales problématiques concernant le Territoire. Il apparaît nécessaire comme première priorité d'être capable d'évaluer le devenir et l'impact des effluents sur l'environnement littoral. Action 2 : A ce titre, un travail de recherche d'un ou de plusieurs indicateurs, parmi les variables environnementales du lagon, de l'influence des rejets aquacoles sur l'environnement littoral a été mené. Les premiers résultats sont présentés dans ce rapport. Action 3 : Parallèlement, il a été défini en relation avec nos partenaires présents en Nouvelle-Calédonie un programme pluriannuel prenant en compte les différents aspects du problème dans sa globalité et sa complexité. Pour cela, un séminaire intitulé « Écosystèmes et crevetticulture » organisé par l'Ifremer en Nouvelle-Calédonie avec le soutien de l'IRD s'est tenu à Nouméa du 22 au 24 juin 2005. The molecular determinants of V. nigripulchritudo pathogenicity are unknown; however, molecular epidemiological studies have suggested that pathogenicity is linked to particular lineages. Here, we performed high-throughput sequencing-based comparative genome analysis of 16 V. nigripulchritudo strains to explore the genomic diversity and evolutionary history of pathogen-containing lineages and to identify pathogen-specific genetic elements. Our phylogenetic analysis revealed three pathogen-containing V. nigripulchritudo clades, including two clades previously identified from New Caledonia and one novel clade comprising putatively pathogenic isolates from septicemic shrimp in Madagascar. The similar genetic distance between the three clades indicates that they have diverged from an ancestral population roughly at the same time and recombination analysis indicates that these genomes have, in the past, shared a common gene pool and exchanged genes. As each contemporary lineage is comprised of nearly identical strains, comparative genomics allowed differentiation of genetic elements specific to shrimp pathogenesis of varying severity. Notably, only a large plasmid present in all highly pathogenic (HP) strains encodes a toxin. Although less/non-pathogenic strains contain related plasmids, these are differentiated by a putative toxin locus. Expression of this gene by a non-pathogenic V. nigripulchritudo strain resulted in production of toxic culture supernatant, normally an exclusive feature of HP strains. Thus, this protein, here termed 'nigritoxin', is implicated to an extent that remains to be precisely determined in the toxicity of V. nigripulchritudo. Salinité et confort physiologique - Application pratique en élevage larvaire Auteur(s) : Pham, Dominique Mailliez, Jean-rene Peignon, Jean-marie Broutoi, Francis Marteau, Anne-laure Wabete, Nelly Résumé : The caledonian blue shrimp Litopenaeus stylirostris is the second largest export commodity in New Caledonia. This industry is based on 19 production farms but also four hatcheries, two feed production units and two packing plants. The maximum production was reached in 2005 with 2400 tons. Supply of juveniles by hatchery is the bottle neck of the production chain because this species is not present in the natural environment. Recent problems of larval survival led some scientists to work on eco-physiology to propose adaptations of larval rearing protocol. Salinity of the rearing environment is taken as an example to study the influence of abiotic parameters on the animal physiology according to its development stage.
The influence of salinity on survival and growth was evaluated at different development stages in Litopenaeus stylirostris. The study showed that ocean salinity (33-35 ppt) should be maintained during the larval stage, but after the post-larval metamorphosis, better growth could be achieved by lowering the salinity gradually to 27 ppt. This measure is now applied routinely in the Saint Vincent's hatchery and is the result of research in eco-physiology showing that the blue shrimp iso-osmotic point is around 27 ppt. La crevette bleue calédonienne Litopenaeus stylirostris est au deuxième rang des produits exportés en Nouvelle-Calédonie. L’industrie crevetticole calédonienne est basée sur 19 fermes de production mais également 4 écloseries, 2 provendiers et 2 usines de conditionnement. La production maximale a été atteinte en 2005 avec 2400 tonnes. La fourniture de juvéniles par les écloseries est le point crucial de la chaîne de production car cette espèce n’est pas présente dans le milieu naturel. Les récents problèmes de survie larvaire ont orienté une partie de la recherche vers l’éco-physiologie en vue de proposer des adaptations du protocole d’élevage. La salinité du milieu d’élevage est prise comme exemple pour étudier l’influence d’un paramètre abiotique sur la physiologie de l’animal en fonction de son stade de développement. L’influence de la salinité sur la survie et la croissance a été évaluée aux différents stades de développement chez Litopenaeus stylirostris. L'étude a permis de montrer que la salinité océanique (33-35 ppt) devait être maintenue durant la phase larvaire mais qu'après la métamorphose en post-larve, une meilleure croissance pouvait être obtenue en abaissant la salinité progressivement à 27 ppt. Cette mesure est maintenant appliquée en routine en élevage larvaire à l'écloserie de Saint-Vincent et découle des recherches menées en éco-physiologie qui situe le point iso-osmotique de la crevette bleue autour de 27 ppt. The weakness of farmed shrimp, Litopenaeus stylirostris, during winter is a major problem in New Caledonia, because of seasonal climate (tropical-sub-temperate). The transfer of broodstock in winter from earthen outdoor ponds to indoor maturation tanks in the hatchery (T=20 degrees C, Salinity=35 parts per thousand, fed shrimp) usually leads, after 48 h, to high mortality (up to 70%). Eyestalk ablation to induce ovarian maturation in females leads to further mortality. Actes Colloq. 38, 75-84.), we developed aprotocol using a soft technology, based on modifications of water salinity, temperature and feeding regime. The aim was to minimize problems of osmoregulatory imbalance and associated mortalities. The protocol we developed, called the LSD OT protocol (Low Salinity and Diet, Optimal Temperature), was first evaluated on sub-adult shrimp (20-25 g) and then applied to broodstock. Survival after transfer and following eyestalk ablation, as well as reproductive achievement (spawning rate, nauplii number) was considerably improved when shrimps were transferred under "physiological comfort" i.e. warmed isosmotic water (26 degrees C and 26 parts per thousand) and unfed for 3 d. This new handling protocol, based on a better control of salinity, temperature and feeding conditions, has been transferred successfully to private hatcheries and already contributes to an increased profitability of New-Caledonian shrimp industry. Effluents from land based marine farms: nature, treatment, valorisation, modelisation. Three thematic of research on aquaculture ponds are carried out in CREMA-L’Houmeau, in collaboration with producers. The aim of the first one is to determine the optimal rearing conditions (stocking density, water renewal rate, input of oxygen, rate of feeding...) which allow to minimise the quantity of wastes for a similar final biomass. The second part aims to reduce the nutrient loading in the effluent, through open-air treatments: sedimentation ponds, foam fractionation, mass production of phytoplankton, and rearing of molluscs on the produced phytoplankton. The third part concerns research aiming to model the “ production - treatment - valorisation ” relationships. Closed cycle quarantine facilities using recycled artificial seawater are an adequate answer to this challenge by allowing secure rearings and additional pathogen testing. L’importation de sang neuf dans les meilleures conditions de biosécurité impose que de multiples précautions soient prises afin d’éviter que la présence d’un pathogène, non identifié lors des tests préliminaires à l’importation, puisse se propager à l’ensemble de la filière. L’utilisation d’installations de quarantaine fonctionnant en circuit fermé permet de répondre à ce souci en réalisant l’élevage des crevettes importées, sur une période de plusieurs mois, dans des conditions d’isolement tout en permettant la réalisation de tests de détection de pathogènes.
