The penaeidins, initially characterized from the shrimp Litopenaeus vannamei, are a family of antimicrobial peptides that appear to be expressed in all penaeid shrimps. As of recent, a large number of penaeid nucleotide sequences have been identified from a variety of penaeid shrimp species and these sequences currently reside in several databases under unique identifiers with no nomenclatural continuity. To facilitate research in this field and avoid potential confusion due to a diverse number of nomenclatural designations, we have made a systematic effort to collect, analyse, and classify all the penaeidin sequences available in every database. We have identified a common penaeidin signature and subsequently established a classification based on amino acid sequences. In order to clarify the naming process, we have introduced a 'penaeidin nomenclature' that can be applied to all extant and future penaeidins. A specialized database, PenBase, which is freely available at http://www.penbase.immunaqua.com, has been developed for the penaeidin family of antimicrobial peptides, to provide comprehensive information about their properties, diversity and nomenclature. Feasibility of polyculture of blue shrimp Litopenaeus stylirostris and goldlined rabbitfish Siganus lineatus in a mesocosm system Auteur(s) : Luong, Trung Cong Hochard, Sebastien Royer, Florence Lemonnier, Hugues Letourneur, Yves Éditeur(s) : Elsevier Science Bv Résumé : The study was conducted to (1) estimate the effects of polyculture of blue shrimp Litopenaeus stylirostris with goldlined rabbitfish Siganus lineatus on production, environmental quality and water and sediment metabolism and (2) to determine if blue shrimp and goldlined rabbitfish polyculture is possible. The experiment was carried out for 12 weeks in a mesocosm system that consisted of 12 circular outdoor mesocosm fiberglass tanks (1.7 m2, 1275 L water volume). Shrimp (2.9 g) were randomly stocked at density of 15 shrimp.m− 2 without rabbitfish (control), with rabbitfish (25.5 g) at low density (1.2 fish.m− 2) (LDRB) and high density (2.4 fish.m− 2) (HDRB). Results indicated that goldlined rabbitfish S. lineatus did not affect shrimp growth performance. At the end of the experiment, the combined shrimp and rabbitfish yields in the LDRB (213 g.m− 2) treatment and the HDRB (295 g.m− 2) treatment were significantly greater (P < 0.05) than the shrimp yield in the control (143 g.m− 2). Most of the water and sediment parameters were similar among treatments. There was no significant difference (P > 0.05) in gross primary productivity and respiration between the HDRB treatment and the control. It was concluded that the polyculture of blue shrimp with goldlined rabbitfish is technically possible without degradation of the environment in the culture system. Trade has involved, in particular, the following species: - Penaeus japonicus from Japan to Europe, the Pacific Islands and South America - P. monodon from South-East Asia to almost all tropical countries - P. vannamei and P. stylirostris from countries along the Pacific coast of South and Central America to the United States of America, the islands of the South Pacific, countries along the Atlantic coast of the Americas and certain countries in Africa. In the 1980s, research conducted by American and French teams enabled the breeding of stocks of spawners in captivity. This resulted in the establishment of lines of captive broodstock outside natural areas of distribution, and the development of new breeding farms in previously unproductive regions. Trade related to movements of these species is facilitated by the absence of legislation in most countries; even when such legislation does exist, it is not always applied These practices have led to the rapid development of world shrimp production, but have also contributed to the dissemination of pathogens. In recent years, the occurrence of serious epizootics - occasioning heavy losses world-wide - has led to a more cautious approach involving trade of post-larvae obtained from captive broodstock in which thorough control measures have been implemented for known diseases. Trade in wild shrimp will probably be abandoned progressively, to avoid the risk of introducing new pathogens. The main challenges for research in the near future are the development of diagnostic tools, to enable continuous control of captive broodstock, and the selection of strains which are resistant to the principal pathogens affecting these species. Evaluation du probiotique bactérien Pediococcus acidilactici MA18/5M chez la crevette pénéide Litopenaeus stylirostris en Nouvelle- Calédonie Auteur(s) : Castex, Mathieu Éditeur(s) : Agro Paris Tech Résumé : P. acidilactici MA 18/5M is a dietary probiotic (BACTOCELL®) which has shown its efficiency on several terrestrial and aquatic species. The present thesis aimed to investigate more fully the effects of this probiotic on cultured penaeid shrimps. For this study, we chose a global approach by replacing the problem studied in a model of interactions between three compartments: the shrimp, its intestinal microflora and the probiotic. Having first defined how the probiotic should be administrated, we studied its effects under controlled laboratory conditions at several levels of Litopenaeus stylirostris biology: (i) Nutritional status and growth, (ii) associated bacterial microflora, and (iii) health status with respect to oxidative stress. Then the probiotic improves the growth of the shrimps and the feed conversion ratio. Our study shows that these effects are partially due to a better use of the feed, particularly of dietary carbohydrates, but also suggest an action on the metabolism, andor on the growth via a contribution of essential nutrients. On the other hand, although the intestinal bacterial communities seems little affected, we show an antagonism effect of the probiotic toward total and vibrionacea intestinal flora, as well as a decrease in the level of infection of shrimps during challenge with pathogenic V. nigripulchritudo. Beside, our works highlight that infection by this pathogen, led to a reduction in antioxidant defences, to a rise in oxidative damages, and to a release of mortalities within 48 hours. This oxidative stress following bacterial infection was better controlled by animals receiving the probiotic diet, and resistance of the shrimps to infection was significant improved. A nutritional hypothesis is proposed to explain such result, hypothesis based on a link between carbohydrate metabolism and antioxidant status. As a conclusion of these works, we can propose a first explanatory scenario for the effects of P. acidilactici on a penaeid shrimp. Finally probiotic's effects were assessed under a pilot scale in a commercial farm from New Caledonia currently affected by the "summer syndrome ". We confirm the improvements brought in term of zootechnical results, with in particular, a better resistance to this syndrome. This work was made possible thanks to a breeding method in floating cages, developed during this thesis, and therefore transferred, as an experimental tool, to the shrimp industry of New Caledonia. Pediococcus acidilacticii MA18/5M est un probiotique bactérien (BACTOCELL®) qui a démontré son efficacité chez plusieurs espèces d'animaux terrestres et aquatiques. Cette thèse avait pour objectif d'investiguer plus en détails les effets de ce probiotique chez les crevettes pénéides d'élevage. Pour cette étude, nous avons choisi une approche globale replaçant la problématique dans un modèle d'interactions entre trois compartiments : la crevette, sa microflore intestinale et le probiotique. Après avoir défini les modalités d'administration, l'effet de P. acicilactici a été étudié, en conditions de laboratoire, chez la crevette Litopenaeus stylirostris, à différents niveaux: (i) statut nutritionnel et croissance, (ii) microflore bactérienne associée, et (iii) santé de l'animal à travers son statut antioxydant et son niveau de stress oxydant. Ainsi, le probiotique améliore la croissance des crevettes et l'indice de conversion de l'aliment. Notre étude montre que ces effets sont en partie liés à une meilleure utilisation de l'aliment, notamment des carbohydrates alimentaires, mais suggèrent aussi une action sur le métabolisme, et/ou sur la croissance via un apport de nutriments essentiels. D'autre part, bien que les communautés bactériennes intestinales semblent peu affectées, un effet d'antagonisme du probiotique vis-à-vis de la flore intestinale totale et vibrionacée a été montré, ainsi qu'une réduction du niveau d'infection des crevettes au cours de challenge avec le pathogène V. nigripulchritudo. Par ailleurs, nous mettons en évidence que l'infection par ce pathogène, conduit à un abaissement des défenses antioxydantes, à une élévation des dégâts oxydatifs, et au déclenchement de mortalités dans les 48 heures. Le probiotique atténue le niveau de ce stress oxydant, et améliore significativement la résistance des crevettes à l'infection. Une hypothèse nutritionnelle est proposée afin d'expliquer ce résultat, hypothèse reposant sur un lien entre le métabolisme des carbohydrates et le statut antioxydant. A l'issue de ces travaux, nous pouvons alors proposer un premier scénario explicatif des effets de P. acidilactici chez une crevette pénéide. Finalement le probiotique a été éprouvé à l'échelle pilote sur une ferme commerciale de Nouvelle-Calédonie affectée par le « syndrome d'été ». Nous confirmons les améliorations apportées en termes de résultats zootechniques, avec notamment une meilleure résistance au syndrome. Physiological response of the blue shrimp Litopenaeus stylirostris to short-term confinement on a pond bottom Auteur(s) : Mugnier, Chantal Lemonnier, Hugues Legrand, Alexandre Éditeur(s) : Elsevier Résumé : The blue shrimp Litopenaeus stylirostris was exposed to pond sediment for 48 h and its physiological response was studied in relation to molt stage. Shrimp were maintained in cages within a semi-intensive pond after 4 months of rearing. Hypoosmoregulatory capacity (hypo-OC), magnesium ions (Mg ions), glucose, total proteins and oxyhaemocyanin were assayed in haemolymph. An increase of 370% to 500% in Mg ions concentration and 200% to 266% in glucose concentration, and a decrease in hypo-OC in shrimp maintained close to the centre of the pond indicated a short-term stress response in all molt stages. Indicators of water and sediment pore water quality were within normal operating limits, however this area was characterized by high water content and relatively high total ammonia nitrogen in pore water of sediment. To a lesser extent, shrimp maintained in two other stations also presented a stress response. Physiological response of shrimp in molt stage D, compared to C/D-0 and D-2 seems to indicate that shrimp were more sensitive to environment at this stage. The use and choice of physiological parameters to detect shrimp stress in a pond are discussed. In this study, we report the characterization in the white leg shrimp Litopenaeus vannamei of RNAi pathway associated proteins Lv-Ago 1 and Lv-Ago 2, two members of the Argonaute family of proteins, as well as Lv-sid 1, the first shrimp homologue of Sid-1, a membrane channel-forming protein implicated in the cellular import of dsRNA. To decipher their functional implication in RNAi-related phenomena, we monitored their relative expression following stimulation by specific and non-specific RNA duplexes of diverse length. The findings show that the length of small RNA duplexes plays a critical role in the activation of both RNAi-related and innate antiviral responses. To understand the mechanisms affecting shrimp resistance and leading to significant mortality, a shrimp ecophysiology and immunology survey was conducted on two farms, the first considered as a "control" farm (HC), the second affected by the disease (DF). Mortality observed during the survey at DF started 50 days after stocking and was typical of this disease. The main observations regarding shrimp were: (a) growth was not affected by the disease and was faster in the DF than in the HC pond; (b) disease did not affect one sex more than the other, or a specific part of the population in terms of weight; (c) the physical condition of shrimp did not specifically allow us to foresee disease outbreak; (d) shrimp at late premolt stage D-2 and early postmolt stage A appeared to be at some points of the mortalities - but not continuously - the most sensitive to disease; (e) physiological, immunological and nutritional parameters of uninfected shrimp in the DF pond were altered, suggesting that environmental stress occurred just before the first mortalities; (f) data suggest that Vn-infected shrimp are more stressed than the presumed healthy shrimp. Combined with pathological and environmental knowledge gained in parallel during this survey, a conceptual model is proposed. Results suggest that an unstable environment induced conditions (i) stressful for the shrimp, increasing their susceptibility to bacterial infections and (ii) favoring the proliferation of the pathogen in the pond. Marine crustacean farming: present status and perspectives Auteur(s) : Laubier, Annie Laubier, Lucien Éditeur(s) : EDP Sciences Résumé : For centuries, several species of prawns and crabs have been raised from wild-caught juveniles in coastal brackish-water fish ponds in various countries of south east Asia. The Indonesian "tambaks" are well known examples of such traditional practices. In western countries, since the turn of the century, advances of marine biology and fast increase of marine fisheries enabled the development of large-scale production and release of larval stages of American and European lobsters in a fruitless attempt to restock natural populations. After the Second World War, the increasing demand for crustaceans in United States and Japan was satisfied by opening new prawn fisheries all over the world. A major breakthrough was achieved with the development of hatchery technologies for the penaeid prawn Penaeus japonicus (Hudinaga, 1942 in Japan) and the caridean prawn Macrobrachium rosenbergii (Ling, 1969 in Malaysia), which occurred during the first decades of the second half of this century. Together with the increasing market demand in developed countries for sea food, this led to a considerable interest of both public agencies and private investors in marine shrimp and prawn culture. In western countries, a large number of pioneering commercial ventures, often based on assumptions not scientifically founded, failed. Nevertheless, the aquaculture production of prawns mainly based on wild-caught juveniles increased in South-East Asia and Central America during the 1980s. This overall positive trend should not hide important failures which occurred at a local scale, such as the Taiwanese crisis of 1988 due principally to environmental degradation, resulting in severe disease problems and a near collapse of the farming activity. Following the early period of hatchery technology development, the major scientific achievements were related to food requirements and formulation of compound diets for larvae, juveniles and adults and to a better knowledge of diseases caused by bacteria and several viruses which have been identified from hatcheries and intensive farming ponds. Additional new technological advances have emerged from recent research in the fields of physiology (endocrinology) and genetics. By far, the major part of the world production of marine crustaceans relies on penaeid prawns and, to a lesser extent, on Macrobrachium species. However, some other species of marine crustaceans have potential for aquaculture. The economic aspects of marine crustacean aquaculture should be considered together with those of the fishing industry: market prices are rather similar, depending on the quality of the product. The balance between market demand and production is an important constraint which, in turn, establishes the success of prawn farming. Since the early 1980s, crustacean aquaculture has increased tremendously in both Asia and America: the world production for 1991 approximated 700,000 tons, with more than 600,000 tons from penaeid prawn culture. Après la Seconde Guerre mondiale, la demande croissante des Etats-Unis et du Japon a été satisfaite par l'ouverture de nouvelles pêcheries de crevettes dans l'océan mondial. Un progrès décisif a été réalisé avec la mise au point des techniques d'écloserie pour la crevette Pénaeidé Penaeus juponicus (Hudinaga, 1942 au Japon) et la crevette Caridé Macrobrachium rosenbergii (Ling, 1969 en Malaisie), qui sont apparues au cours des premières décennies de la seconde moitié de ce siècle. Parallèlement à la demande croissante du marché des pays développés pour les produits de la mer, ce résultat s'est traduit par un intérêt considérable porté à l'élevage des crevettes marines de la part des institutions de recherche et des investisseurs. Dans les pays occidentaux, un grand nombre de tentatives d'élevage commerciales, fondées sur des considérations non scientifiquement établies, ont échoué. Néanmoins, la production aquacole de crevettes reposant sur la capture d'animaux juvéniles dans la nature, s'est développée en Asie du Sud-Est et en Amérique Centrale au cours des années 1980. Cette tendance globalement positive ne doit pas dissimuler les faillites importantes qui se sont produites à l'échelle locale, comme la crise des élevages de Taiwan en 1988, causée principalement par une dégradation de l'environnement, entraînant l'apparition de problèmes pathologiques sévères et une disparition presque totale de l'activité d'élevage. Après la période pionnière du développement de la technologie d'écloserie, les principaux acquis scientifiques concernent les besoins alimentaires et la formulation d'aliments composés pour les larves, les jeunes et les adultes, et une meilleure connaissance des maladies bactériennes et virales qui interviennent dans les écloseries et les fermes d'élevage intensif. De nouveaux progrès techniques complémentaires ont vu le jour à la suite de recherches dans les domaines de la physiologie (endocrinologie) et de la génétique. De loin, la majeure partie de la production mondiale de Crustacés marins repose sur les crevettes Pénaeidés et, dans une moindre mesure, sur des espèces de Macrobrachium. Cependant, quelques autres espèces de Crustacés marins intéressent potentiellement l'aquaculture. Les aspects économiques de l'aquaculture de crustacés marins doivent être considérés avec ceux de l'industrie des pêches maritimes : les prix du marché sont à peu près les mêmes, et dépendent de la qualité du produit. Le rapport entre la demande du marché et la production est une contrainte importante qui à son tour, détermine les conditions du succès de l'élevage de crevettes. Les capacités osmorégulatrices chez la crevette bleue Litopenaeus stylirostris, au cours de l’ontogenèse Auteur(s) : Pham, Dominique Éditeur(s) : Université de la Polynésie Française Résumé : The ontogeny of osmoregulation was studied in the blue shrimp Litopenaeus stylirostris in New Caledonia. The implication of branchial organs in ionic regulation was first demonstrated in juveniles by immunolocalization of three ion transporting proteins ( namely NKA, co-transporter NKCC1 and CFTR ) and ionocyte detection was done by means of electron microscopy. The role of the main ion transporting enzyme, NKA, was subsequently investigated during post-embryonic development using an integrative approach associating physiological, histological, immunological and biomolecular studies. Results obtained allowed us to propose a model of the osmoregulatory pattern in L. stylirostris based on weak regulation in the early stages, then progressively this species acquires the adult pattern of hyper-hypo-regulation after metamorphosis with the concurrent development of epipodites and gills. Strong NKA immunofluorescence and abundance of NKA transcripts in these two tissues compared to others organs suggest their functional implication in osmoregulatory processes. The results of this study allow us to increase our knowledge of the penaeid shrimp osmoregulation and to propose zootechnical measures to improve the animal physiological comfort. These propositions have been already tested during larval rearing trials, leading a two-fold-growth in 19 days-old postlarvae. L’étude de l’ontogenèse de l’osmorégulation a été entreprise chez la crevette bleue Litopenaeus stylirostris en Nouvelle-Calédonie. L’implication des tissus de la cavité branchiale dans la régulation ionique a été déterminée au préalable chez des juvéniles par immunolocalisation de trois transporteurs protéiques d’ions, ( la NKA, le co-transporteur NKCC1 et le CFTR ) et par la détection de ionocytes en microscopie électronique. Le rôle de la NKA a été particulièrement étudié et son implication a été examinée au cours du développement post-embryonnaire par une approche intégrative associant des études physiologiques, histologiques, immunologiques et moléculaires. Cette démarche nous a permis de proposer un modèle d’osmorégulation chez L. stylirostris: de faible régulatrice dans les premiers jours de sa vie, cette espèce évolue vers l’hyper-hypo-régulation après la métamorphose avec le développement progressif des épipodites et des branchies. L’immunofluorescence prononcée de la NKA ainsi que l’abondance des transcrits du gène codant la NKA dans les tissus de ces deux organes suggèrent leur implication fonctionnelle dans l’ionorégulation. Les résultats de ce travail nous ont permis d’accroître nos connaissances sur l’osmorégulation chez les pénéidés et de proposer des améliorations zootechniques visant à augmenter leur confort physiologique. Selection and characterization of potential probiotic bacteria for Litopenaeus stylirostris shrimp hatcheries in New Caledonia Auteur(s) : Pham, Dominique Ansquer, Dominique Chevalier, Anne Dauga, Clement Peyramale, Aude Wabete, Nelly Labreuche, Yannick Éditeur(s) : Elsevier Science Bv Résumé : In New Caledonia, shrimp hatcheries are confronted with mass mortality in the larval stages, a phenomenon poorly understood as no specific causative agent has been identified. This has resulted in an excessive use of prophylactic antibiotics, although their adverse effects in aquaculture are notorious. The present work was thus aimed at selecting potential probiotic strains for penaeid hatcheries. From a pool of more than 400 marine bacterial isolates sampled from the local marine environment seven strains exhibited in vitro antagonistic activity towards Vibrio harveyi. These isolates were characterized both phenotypically and genotypically using a biochemical approach and 16S rDNA sequencing. Six out of these seven strains were found to belong to the genus Pseudoalteromonas, the last one belonging to the Vibrionaceae family and related to the Harveyi clade. Selected probiotic candidates were individually tested for antagonistic activities in vitro using a green fluorescent protein (GFP)-labelled transconjugant of V. harveyi and for inherent pathogenicity towards cultured shrimp larvae at two different developmental stages. This approach enabled rapid processing and selection of candidates to be tested in our experimental hatchery. Four different experiments were conducted to test candidate strains either alone or in combination. Repeated trials showed that postlarval survival was significantly improved by adding the strain NC201 individually to the rearing water compared to unchallenged controls. The analysis of immune-related gene expressions showed that the Litsty PEN3 transcript abundance of larvae was significantly increased after being reared in probiotic-containing water; however no significant difference in lysozyme gene expression was recorded in this study. Together, these results open new insights into the use of these strains as potential substitutes to antibiotherapy in shrimp larval rearing in New Caledonia. Five species have been investigated. Wi1d-caught juveni1es from the New Ca1edonia 1agoon (f. merguiensis, f. semisulcatus, ~. ensis) and post1arvae from the Ga1veston NMFS 1aboratory (P. aztecus) and Fujinaga Institute (f. japonicus) have been used to-obtain adu1t stock. Shrimps are reared in circu1ar tanks of 12 m3 with a continuous water circulation through a coral sand bed. Rate of exchange is from 1 to 3 times a day. Throughout the year water temperature is between 25 and 29 C, sa1inity around 34.5 ppt, and pH 8.2. Solar energy is reduced by shade covers. Artificial pellets of different composition are given as food. In these conditions we have obtained: 1) about 1,000 spawnings and two generations in captivity from September 1973 to September 1974 with f. merguiensis; 2) spawnings of ~. ensis in 1973; 3) four spawnings of f. japonicus in September 1974; 4) about 20 spawnings of P. aztecus, but only aftcr removing one eyestalk; 5) maturation signs in P. semizulcatus but no spawning. In this study, we have specifically analysed the expression in circulating hemocytes of antimicrobial peptides (AMPs) encoding genes, such as PEN2 and PEN3, ALF, crustin, lysozyme and a putative cysteine-rich peptide. We evidenced a relationship between the level of expression of some AMPs and the successful response of the shrimp, Litopenaeus stylirostris, to circumvent a pathogenic Vibrio penaeicida infection. Additionally, significant differences in some AMP transcript amounts are evidenced between control, non-selected shrimp line and the third generation breeding of shrimp selected for their survival to natural V. penaeicida infections. On the basis of these results, it will now be of great interest to determine if these AMPs are directly involved in the resistance of shrimp to infection or if they only reflect other acquired defence mechanisms which can confer a resistance. Response of the blue shrimp Litopenaeus stylirostris to temperature decrease and hypoxia in relation to molt stage Auteur(s) : Mugnier, Chantal Soyez, Claude Éditeur(s) : Elsevier Résumé : The effect of different levels of dissolved oxygen (DO) and a decreased temperature combined with severe hypoxia were studied in the blue shrimp Litopenaeus stylirostris in relation to molt stage. In the first experiment, shrimps were submitted to DO concentrations of 5 to 1 mg l(-1). Osmoregulatory capacity (OC) was measured in shrimps at molt stage C after 6, 24, or 48 h of exposure. No mortality was recorded but a significant negative effect of DO concentration on hypo-OC was observed for DO concentrations below 3 mg l(-1). Osmoregulation improved after 48 h exposure, compared to 6 and 24 h exposure. In a second experiment, temperature was decreased from 28 to 22 degreesC in 24 h, before a severe hypoxia of 0.5+/-0.1 mg O-2 l(-1) was applied. Mortality and OC were studied in relation to molt stages. Half of the shrimps died after respectively 100 min at 0.5 mg O-2 l(-1) at 28 degreesC and 153 min at 0.4 mg O-2 l(-1) at 22degreesC. Mortality was significantly higher in stages D-2 and B compared to stages C and Do. Control shrimps in stage D2 had a lower hypo-OC than shrimps in stages C and Do both at 28 and 22 degreesC. Temperature decrease had no effect on hypo-OC. Hypoxia reduced hypo-OC for all stages, whatever the temperature, but the effect was greater in stage C at 27 degreesC (68% decrease) compared to 22 degreesC (49%) and stage D-2 at both temperatures (respectively, 43% and 58% at 27 and 22 degreesC). Combination of temperature and hypoxia had a significant effect on hypo-OC. Low temperature reduced the effect of hypoxia presumably by slowing down the metabolism. Influence of sediment characteristics on shrimp physiology: pH as principal effect Auteur(s) : Lemonnier, Hugues Bernard, E Boglio, E Goarant, Cyrille Cochard, Jean-claude Éditeur(s) : Elsevier Résumé : Penaeid shrimp reared in earthen ponds are exposed to sediment, which can, in some instances, induce a stress. In seawater, the osmoregulatory capacity (hyporegulation) is a useful tool to compare the physiological condition of shrimp exposed to various stressors. By keeping some shrimp in cages at different locations of a single pond heterogeneous in terms of sediment quality, it was possible, using osmotic pressure (OP), to identify some locations where the stress was maximum and some others where it was minimum. Simultaneously, sediment samples were taken and analysed in order to evaluate some physico-chemical parameters that could be related to the stress observed in the shrimps kept in the cages. This approach allowed to show a significant positive correlation between the pH of the sediment surface and the shrimp osmotic pressure. This result was confirmed in a study carried out in experimental 70-1 tanks, where osmotic pressure decreased significantly as water pH decreased from 7.0 to 6.5. Arbitrarily primed PCR to type Vibrio spp. Pathogenic for shrimp Auteur(s) : Goarant, Cyrille Merien, F Berthe, Franck Mermoud, I Perolat, P Éditeur(s) : Amer Soc Microbiology Résumé : A molecular typing study on Vibrio strains implicated in shrimp disease outbreaks in New Caledonia and Japan was conducted by using AP-PCR (arbitrarily primed PCR). It allowed rapid identification of isolates at the genospecies level and studies of infraspecific population structures of epidemiological interest. Clusters identified within the species Vibrio penaeicida were related to their area of origin, allowing discrimination between Japanese and New Caledonian isolates, as well as between those from two different bays in New Caledonia separated by only 50 km. Other subclusters of New Caledonian V. penaeicida isolates could be identified, but it was not possible to link those differences to accurate epidemiological features. This contribution of AP-PCR to the study of vibriosis in penaeid shrimps demonstrates its high discriminating power and the relevance of the epidemiological information provided. This approach would contribute to better knowledge of the ecology of Vibrio spp. and their implication in shrimp disease in aquaculture. Shrimp at different larval and postlarval stages were exposed to various salinities and survival was monitored for 24 h. Survival rates exceeded 80% at salinity over 25 ppt (750 mOsm.kg(-1)) at all the stages. At salinities below to 25 ppt, salinity tolerance was higher in nauplii and zoeae than in mysis larvae. Postlarval stages were able to withstand lower salinities, e.g. 6.0 ppt (176 mOsm.kg(-1)) at PL9 stage, but they were more sensitive than larvae to salinities over 35 ppt (1035 mOsm.kg(-1)). Zoea and mysis larvae slightly hyper-regulated at all tested salinities. After metamorphosis, postlarvae progressively acquired the adult pattern of hyper-hypo-osmoregulation: At PL9, the estimated isosmotic salinity was 24.5 ppt (720 mOsm.kg(-1)); below and over this salinity, animals hyper-regulated and hypo-regulated, respectively. Finally, we determine the effects of lowering salinity at different animal development stages. We conclude that seawater salinity (35 ppt) is optimum during larval stages; but for postlarval stages, lowering salinity at 27 ppt leads to a better growth in 19 days compared to those maintained at 35 ppt (1.07 mg vs 0.47 mg). These results are in agreement with penaeid natural life cycle during which larvae are released in oceanic water while juveniles live in coastal areas where salinity is more fluctuant. Although this gene silencing process has been reported to be relatively well conserved in species of different phyla, there are important discrepancies between plants, invertebrates and mammals. In penaeid shrimp, the existence of an intact and functional RNAi machinery is supported by a rapidly growing body of evidence. However, the extent to which this process participates to the host immune responses remains poorly defined in this non-model organism. This review summarizes our current knowledge of RNAi mechanisms in shrimp and focuses on their implication in antiviral activities and shrimp immune defences.
