SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. Éditeur(s) : HAL CCSD Ferrata Storti Foundation Résumé : International audience BACKGROUND: Abnormal interactions between red blood cells, leukocytes and endothelial cells play a critical role in the occurrence of the painful vaso-occlusive crises associated with sickle cell disease. We investigated the interaction between circulating leukocytes and red blood cells which could lead to aggregate formation, enhancing the incidence of vaso-occlusive crises. DESIGN AND METHODS: Blood samples from patients with sickle cell disease (n=25) and healthy subjects (n=5) were analyzed by imaging and classical flow cytometry after density gradient separation. The identity of the cells in the peripheral blood mononuclear cell layer was determined using antibodies directed specifically against white (anti-CD45) or red (anti-glycophorin A) blood cells. RESULTS: Aggregates between red blood cells and peripheral blood mononuclear cells were visualized in whole blood from patients with sickle cell disease. The aggregation rate was 10-fold higher in these patients than in control subjects. Both mature red blood cells and reticulocytes were involved in these aggregates through their interaction with mononuclear cells, mainly with monocytes. The size of the aggregates was variable, with one mononuclear cell binding to one, two or several red blood cells. Erythroid Lu/basal cell adhesion molecule and α(4)β(1) integrin were involved in aggregate formation. The aggregation rate was lower in patients treated with hydroxycarbamide than in untreated patients. CONCLUSIONS: Our study gives visual evidence of the existence of circulating red blood cell-peripheral blood mononuclear cell aggregates in patients with sickle cell disease and shows that these aggregates are decreased during hydroxycarbamide treatment. Our results strongly suggest that erythroid Lu/basal cell adhesion molecule proteins are implicated in these aggregates through their interaction with α(4)β(1) integrin on peripheral blood mononuclear cells. Les pêcheries mixtes de langoustine et de merlu du golfe de Gascogne. Description, préparation à une modélisation et à une simulation des procédures de gestion Auteur(s) : Charuau, Anatole Ifremer Résumé : 1 - Study's goals review The management of Norway Lobster and Hake fisheries can be considered as the most important issue in the Gascogne gulf. Calculation methods used to evaluate prediction variations under various efforts and mesh constraints all pinpoint to the crucial role narrow mesh fishing plays in Hakes' Northern stock status. The involvement of Gascogne gulf Norway Lobster fisheries regarding Hakes over exploitation seems over dimensioned when comparing the size of both of these stocks. Hake stocks spread from Northern Great Britain to Northern Spain, while Norway Lobster stocks are located in the Gascogne gulf's mud flats. The biological solution is well known and consists in globally increasing mesh nets to 80mm which would bring up Hakes yield to its most. It would be unreal to pretend all parties involved in Hake stocks will come to use them. For now, it is mostly an economical issue since many vessels continue on with an exploitation scheme dating from the 60's and fish on coastal mud flats which are the most exploited zones and where their catch is detrimental to juvenile class Hakes and Norway Lobsters. Considering our limited time, this study will cover bio- economical modelisation preparation. You will therefore find a description of fisheries and an exploitation assessment based on 1986's available data.
2 - Accomplished work
2. 1 Fishery description
Presenting the problems and preparing ulterior analyses was first and foremost. Norway Lobster vessels supposedly, according to evaluation models used, test immature Hakes in such a manner that all stock exploitation is jeopardized. Norway Lobster distribution is closely linked to the nature of the ocean floor since it can only build its habitat on fine sediment. Hakes' habitat is not as restrictive and is linked to food supply, especially euphausiid shrimps, so they can be found on sandy and sandy-sedimental floors. Norway Lobsters are sought by a fleet of 400 to 450 ships which characteristics widely vary according to their home port. On the northern part, fisheries are close to exploitation ports where Norway Lobsters and miscellaneous catch yield is sufficient to support an older fleet. On the southern part, the structure is diversified and flexibility usually rules, more modern vessels can easily switch their target on a seasonal or daily basis. Globally, Hakes' northern stocks are slightly on the rise. This rise is probably due to global mesh size increase, even if slight and a decrease on fishing effort where immature species are caught. Norway Lobster stocks unloading is stable or slightly lower and yields, as far as northern fishery are concerned, vary and oscillate between ±15 to ±20 % at about an average of 100 kg/day. Biological parameters used in the evaluations for each species are those used in CIEM and CEE work cells. Among those parameters, those describing growth are the greatest. For Hakes, an intermediary growth equation between males and females has been used. For Norway Lobsters, both sexes are treated separately. Simulations were carried out according to 3 types of data: Norway Lobster vessels fishing effort variations. Norway Lobster vessels mesh increase. Norway Lobster vessels fishing effort decrease or mesh increasing go along in the same direction for Hake and Norway Lobster stock production (chart A and B). Lastly, a brief study on results evaluation significance was done to modify: - the size of Hake rejects by Norway Lobster vessels. The range of sizes stays the same but the amount in each class varies according to the same percentage. - Norway Lobsters growth parameters, both values used (L» = 76 mm and k = 0.11 for males, L°° = 56 mm and k = 0.14 for females) representing the "high" hypothesis. In the first case, a 20% error rate for rejects evaluation with 60 mm mesh would only bring a 3% modification rate on global stock production. Those first results are contradicting usual evaluation conclusions and it will be necessary to recalculate by modifying the size class step. In the second case, to decrease L» and increase growth speed, in time production variations remain within usual inter annual fluctuations. In all figure cases, calculations confirming greater yield for in time production through hypothetical mesh size increase to 60 mm, are definitely well founded.
1 - Rappel des buts de l'étude La gestion des pêcheries de merlu et de langoustine peut être considérée comme le problème le plus important du Golfe de Gascogne. Les méthodes de calcul employées pour évaluer les variations de la production sous diverses contraintes d'effort et de maillage mettent toutes l'accent sur le rôle capital des pêcheries à petits maillages sur l'état du stock nord de merlu. Le niveau de responsabilité des pêcheries de langoustine du Golfe de Gascogne dans la surexploitation du merlu semble démesuré eu égard aux dimensions respectives des deux stocks. Le stock de merlu s'étend du Nord des Iles Britanniques au Nord de l'Espagne alors que le stock de langoustine est localisé aux vasièrés du Golfe de Gascogne. La solution biologique du problème est connue et tient à une augmentation généralisée des maillages à 80 mm qui amènerait le rendement du stock de merlu à son maximum. Il serait illusoire de prétendre que tous les acteurs opérant sur le stock de merlu arriveront jamais à l'utiliser. Pour le moment, le problème se pose essentiellement en termes économiques car beaucoup de navires poursuivent un schéma d'exploitation hérité des années 1960 et pèchent sur des vasièrés côtières qui sont les zones les plus exploitées et où ils effectuent leurs captures au détriment des jeunes classes d'âge de merlu et de langoustine. En raison des délais impartis, cette étude constitue la préparation à une modélisation bioéconomique. On y trouvera donc une description des pêcheries et un bilan de l'exploitation sur les données disponibles en 1986. 2 - Travaux effectués 2 . 1 Description de la pêcherie Il s'agissait avant tout de poser les problèmes et de préparer les analyses ultérieures. Les langoustiniers sont présumés, d'après les modèles d'évaluation employés, effectuer sur les immatures de merlu des prélèvements tels qu'ils mettent en danger toute l'exploitation du stock. La distribution de la langoustine est très liée à la nature du fond car elle ne peut construire son terrier que sur des fonds de vase fine. Celle du merlu est beaucoup plus large et inféodée à la recherche de nourriture, crevettes euphausiacées en particulier, on le trouve donc sur les fonds sableux à sablo-vaseux. La langoustine est recherchée par une flottille de 400 à 450 bateaux dont les caractéristiques sont très variables suivant leur appartenance géographique. Au nord, la pêcherie est proche des ports exploitants et les rendements en langoustine et en prise accessoire sont suffisants pour faire vivre une flottille ancienne. Vers le sud, la structure est très diversifiée et la flexibilité est généralement la règle et les navires de construction récente peuvent changer de cible aisément de façon saisonnière ou au cours de la même journée de pêche. Globalement, les débarquements de merlu du stock nord sont en légère hausse. Ce redressement est dû probablement aux augmentations de maillage, même légères, généralisées et à une baisse de l'effort sur les pêcheries où s'effectuent des captures d'immatures. Les débarquements de langoustine sont stables ou en légère baisse et les rendements, au moins sur la pêcherie du nord, varient avec des oscillations de ±15 à ±20 % autour d'une moyenne de 100 kg/jour. Les paramètres biologiques utilisés pour chaque espèce dans les évaluations sont ceux en usage dans les groupes de travail du CIEM et de la CEE. Parmi ces paramètres, ceux décrivant la croissance sont les plus importants. Pour le merlu une équation de croissance intermédiaire entre mâles et femelles a été utilisée. Pour la langoustine les deux sexes sont traitées séparément. Les simulations ont porté sur trois types de données : Variations de l'effort des langoustiniers Augmentation du maillage des langoustiniers La diminution de l'effort des langoustiniers ou L'augmentation de maillage agisse très exactement dans le même sens sur la production des stocks de merlu et de langoustine. Enfin une étude succincte de la sensibilité des résultats des évaluations a été effectuée pour des modifications : - de la composition en taille des rejets de merlu par les langoustiniers. L'étendue des tailles reste la même mais le nombre dans chaque classe varie selon le même pourcentage. des paramètres de croissance de la langoustine, le couple de valeurs utilisées (L» = 76 mm et k = 0.11 pour les mâles, L°° = 56 mm et k = 0.14 pour les femelles) constituant l'hypothèse "haute" Dans le premier cas, une erreur de 20 % sur l'évaluation des rejets pour un cnaillage de 60 mm n'apporterait qu'une modification de 3 % dans la production globale du stock. Ces premiers résultats sont en contradiction avec les conclusions habituelles des évaluations et il sera nécessaire de reprendre les calculs en modifiant le pas des classes de taille. Dans le deuxième cas, pour une diminution de L» et une augmentation de la vitesse de croissance, les variations de la production à terme restent à l'intérieur des fluctuations interannuelles habituelles. Dans tous les cas de figure, les calculs confirmant l'augmentation de la production à terme dans l'hypothèse d'une augmentation de maillage à 60 mm sont d'une robustesse à toute épreuve. Le merlu du golfe de Gascogne et de la mer Celtique : Croissance, Répartition spatiale et bathymétrique, Ecologie alimentaire et Assemblages Auteur(s) : Kacher, Mohamed Éditeur(s) : Université du Littoral Côte d'Opale (Dunkerque) Résumé : In the Bay of Biscay and Celtic Sea waters, hake is considered as genetically homogeneous and form a unique stock with identical meristic characteristics. The vertebral mean vary from 50,68 (vertebrae) during the first year of life to 51,11 (vertebrae) for adults. 0-group Merluccius display a rapid growth with significant interannual variability (0.71 mm.day-1 in 2001 versus 0.74 mm.day-1 in 2002). The spawning period occurs in April and the average length on the firth January following the hatching is of 17.3 cm (first seasonal check / increment on the sagittal otolith is located about 0.143 cm from the nucleus). At the end of the first year of life the average length is about 24 cm. The longevity of the species is important and growth parameters (L = 138,24 cm ; K = 0,132) explain natural mortality coefficient of M = 0.21. No difference for growth between male and female has been observed for this stock. The ratio total weight / eviscerated weight is equal to Fc = 1,086. Both male and female have same spatial and bathymetric distribution. Juveniles are found in deep waters (< 17 cm length) and move to coastal waters where they display an average length of (33 cm length). Longer individuals are found in nursery areas where they feed and grow then move to deeper areas to spawn. Nursery areas are numerous and located in the Celtic Sea and in the bay of Biscay. In the bay of Biscay, disturbances resulted from hydrondynamics affect the settlement on the nursery grounds particularly in the southern part of the bay. Diet composition varies with age. Juveniles feed mainly on crustaceans (Euphausia kroni). At the length of 23 cm, individuals feed strictly on fishes. Sizes of preys are proportional with sizes of predators and preys are composed mainly of species that feed and reproduce close to the bottom. Cannibalism has been observed for this specie, and increases with age notably in the northern part of the bay of Biscay and in the Celtic Sea. Main preys are 0-group juveniles. This specie is assumed to be a passive predator catching its preys when they come to feed in the bottom. Although they are punctual, these results have been obtained after studies on the evolution of sizes during the first year of life (daily increments measurements) and on the spawning period parameters (back-calculating). Average length on the firth January following hatching and the position of the first seasonal check on the otolith are based on observations of the sizes on the firth January following hatching (obtained with growth rates and spawning period parameters). The results are similar to those described in previous studies, nevertheless they have to be confirmed with growth and diet composition analyses. A long term study appear essential to determine biological and ecological parameters of this species for sustainable management of the stock. Les alertes quant à la surexploitation du stock de merlu européen sont nombreuses et récurrentes. Seulement l'estimation du niveau réel de l'état du stock a toujours posé des problèmes du fait de certaines lacunes dans la connaissance de la biologie et de l'écologie de cette espèce. Dans les eaux du golfe de Gascogne et de la mer Celtique, le merlu européen fait partie de la même population génétique et possède les mêmes caractéristiques meristiques. Sa moyenne vertébrale est de 50,68 durant sa première année de vie et évolue pour se stabiliser à 51,11 chez l'adulte. La croissance du merlu durant sa première année de vie et très rapide mais présente une variabilité interannuelle significative (0,71 mm .J-1 en 2001 et 0,74 mm .J-1 en 2002). Sa période de ponte maximale se déroule au mois d'avril et sa longueur au premier janvier suivant sa naissance est de 17,3 cm (la première marque hivernale est positionnée à 0,143 cm du nucléus de la sagittae). Au terme de sa première année de vie le merlu atteint une longueur de 24 cm. Sa longévité est assez longue (23 ans environ) et ses paramètres de croissance (L = 138,24 cm ; K = 0,132) permettent d'estimer son coefficient de mortalité naturelle à M = 0,21. Il n'a pas été établi de croissance différentielle entre sexe chez le merlu dans ces eaux. Le rapport poids total / poids éviscéré a été estimé à Fc = 1,086. Mâles ou femelles, les merlus ont une même répartition spatiale et bathymétrique. En général, ils sont très profonds à leurs stades juvéniles (< 17 cm) et ils se dirigent vers les eaux côtières pour les atteindre à 33 cm de longueur environ. Au-delà de cette taille, les merlus se concentrent au niveau des zones de nourricerie pour s'y alimenter avant de rejoindre les eaux plus profondes pour y pondre. Les zones de nourricerie du merlu sont très nombreuses et sont localisées en mer Celtique et dans le golfe de Gascogne aussi bien dans sa partie sud que dans sa partie nord. Les perturbations importantes dans l'hydrodynamisme du golfe de Gascogne (upwelling) semble influencer les niveaux de colonisation des zones de nourricerie notamment celles situées dans la partie sud du golfe. Le régime alimentaire du merlu évolue avec l'âge. Juvénile, il se nourrit principalement de crustacés (Euphausia krohni) et devient ichtyophage exclusif dès 23 cm de longueur. Le cannibalisme est une réalité chez le merlu et les merlus-proies sont les juvéniles du groupe d'âge G-0. Il s'intensifie avec l'âge notamment dans la partie nord du golfe de Gascogne et en mer Celtique ; Il est très faible dans le sud du golfe de Gascogne. La taille des proies évolue avec la taille du merlu et les poissons proies sont en général des espèces qui vont se nourrir ou se reproduire sur le fond. Ce comportement alimentaire fait que le merlu est un prédateur peu actif et qui attaque ses proies lorsqu'elles viennent sur le fond. Ces résultats, bien qu'ils ne soient que ponctuels, ont été obtenus après avoir déterminé le schéma d'évolution de la longueur du merlu durant sa première année de vie (dénombrement des accroissement journaliers) et des paramètres de sa période de ponte (rétro-calcul).La taille au premier hiver et la position de la première marque hivernale ont été déterminées en relativisant nos observations à l'estimation de la longueur du merlu au premier janvier suivant sa naissance : celle-ci étant obtenue en utilisant le taux de croissance et les paramètres de la période de ponte. Bien que l'ensemble des résultats obtenus, en ce qui concerne la répartition spatiale et les données sur la période de ponte, soient généralement conformes à ceux décrits dans la littérature, il est nécessaire de confirmer ceux concernant la croissance et le régime alimentaire. Pour cela, une étude à plus long terme semble indispensable pour parvenir à bien maîtriser l'ensemble des paramètres de biologie et d'écologie du merlu permettant ainsi une meilleure gestion de son stock. Its incidence is not known in the Afro-Caribbean population.Objective: To assess the incidence of SCC in Guadeloupe, the largest island of the Lesser Antilles (405 000 inhabitants, mostly black people of African and European descent). The second objective was to characterize clinical and histological patterns of SCC occurring in the Afro-Caribbean community.Methods: This retrospective study was conducted over an 11-year period (2000-2010). Data regarding 723 histological confirmed cases of SCC identified using the three Guadeloupean pathology laboratories computerized databases were retrieved from the records of 551 patients. Private practice dermatologists and general practitioners were contacted to obtain any missing data. Results: The annual age-adjusted incidence of SCC was 15 per 100.000 residents in Guadeloupe. In the Afro-Caribbean community, SCC had a greater size (i.e.: 2.8±2.8 cm versus 1.5±1.0 cm, p<0.001), was more often located on the anogenital area (i.e. :48/79-60.8% versus 14/320-4.4%, p<0.001) in association with an underlying dermatosis due to HPV infection (15/71- 21.1% versus 3/366, 0.8%, p<0.001) and led more frequently to metastasis (13/84-15.5% versus 10/366-2.7%, p<0.001) and/or fatal evolution (11/83-13.3% versus 7/365-1.9%, p<0.001). Conclusions: The results of this original study, which first estimated the incidence of SCC in West Indies, suggest that anogenital examination should be routinely performed in skin cancer screening of Afro-Caribbean people in order to detect the presence of SCC at an early stage. Intriguing diversity among diazotrophic picoplankton along a Mediterranean transect: a dominance of rhizobia Auteur(s) : Le Moal, Marc Collin, H. Biegala, Isabelle C. Éditeur(s) : Copernicus Gesellschaft Mbh Résumé : The Mediterranean Sea is one of the most oligotrophic marine areas on earth where nitrogen fixation has formally believed to play an important role in carbon and nitrogen fluxes. Although this view is under debate, the diazotrophs responsible for this activity have still not been investigated in the open sea. In this study, we characterised the surface distribution and species richness of unicellular and filamentous diazotrophs across the Mediterranean Sea by combining microscopic counts with size fractionated in situ hybridization (TSA-FISH), and 16S rDNA and nifH genes phylogenies. These genetic analyses were possible owing to the development of a new PCR protocol adapted to scarce microorganisms that can detect as few as 1 cell ml(-1) in cultures. Low concentrations of diazotrophic cyanobacteria were detected and this community was dominated at 99.9% by picoplankton hybridized to the Nitro821 probe, specific for unicellular diazotrophic cyanobacteria (UCYN). Among filamentous cyanobacteria only 0.02 filament ml(-1) of Richelia were detected in the eastern basin, while small (0.7-1.5 mu m) and large (2.5-3.2 mu m) Nitro821-targeted cells were recovered at all stations with a mean concentration of 3.5 cell ml(-1). The affiliation of the small Nitro821-targeted cells to UCYN-A was confirmed by 16S and nifH phylogenies in the western Mediterranean Sea. In the central and the eastern Mediterranean Sea no 16S rDNA and nifH sequence from UCYN was obtained as cells concentration were close to, or below PCR detection limit. Bradyrhizobium sequences dominated nifH clone libraries from picoplanktonic size fractions. A few sequences of gamma-proteobacteria were also detected in the central Mediterranean Sea. While low phosphate and iron concentrations could explain the absence of Trichodesmium sp., the factors that prevent the development of UCYN-B and C remain unknown. We also propose that the dominating picoplankters probably developed specific strategies, such as associations with protists or particles, and/or photosynthetic activity, to acquire carbon for sustaining diazotrophy. The objective of the present study was to assess the potential involvement of adipose tissue in these two aspects. Adipose tissue is composed of adipocytes and the stromal vascular fraction (SVF); the latter comprises immune cells such as CD4+ T cells and macrophages (both of which are important target cells for HIV). The inflammatory potential of adipose tissue has been extensively described in the context of obesity. During HIV infection, the inflammatory profile of adipose tissue has been revealed by the occurrence of lipodystrophies (primarily related to ART). Data on the impact of HIV on the SVF (especially in individuals not receiving ART) are scarce. We first analyzed the impact of simian immunodeficiency virus (SIV) infection on abdominal subcutaneous and visceral adipose tissues in SIVmac251 infected macaques and found that both adipocytes and adipose tissue immune cells were affected. The adipocyte density was elevated, and adipose tissue immune cells presented enhanced immune activation and/or inflammatory profiles. We detected cell-associated SIV DNA and RNA in the SVF and in sorted CD4+ T cells and macrophages from adipose tissue. We demonstrated that SVF cells (including CD4+ T cells) are infected in ART-controlled HIV-infected patients. Importantly, the production of HIV RNA was detected by in situ hybridization, and after the in vitro reactivation of sorted CD4+ T cells from adipose tissue. We thus identified adipose tissue as a crucial cofactor in both viral persistence and chronic immune activation/inflammation during HIV infection. Biometrie et Croissance des Jeunes Crevettes Trachypenaeus de la Guadeloupe (Antilles Francaises). Éditeur(s) : ISTPM Résumé : The principal biometric relations of juvenile Trachypenaeus similis similis and T. constrictus in Guadeloupe were calculated from 1976 to 1978. The examination of these relations yields few differences between the two species. A growth curve was established according to the sequences of modal values; these were determined according to weekly size frequency distributions over three different periods of time. In the zone studied, the growth of young Trachypenaeus is more rapid than that of the young Penaeus. Various PKC-independent effects of Ro 31-8220 have however been demonstrated, including inhibition of the ATP-binding cassette drug transporter breast cancer resistance protein. In the present study, we reported that the BIM also blocks activity of the solute carrier organic cation transporter (OCT) 1, involved in uptake of marketed drugs in the liver, in a PKC-independent manner. Ro 31-8220, in contrast to other pan-PKC inhibitors such as staurosporine and chelerythrine, was thus shown to cis-inhibit uptake of the reference OCT1 substrate tetraethylammonium in OCT1-transfected HEK293 cells in a concentration-dependent manner (IC50 = 0.18 μM) and without altering membrane expression of OCT1. This blockage of OCT1 was also observed in human hepatic HepaRG cells that constitutionally express OCT1. It likely occurred through a mixed mechanism of inhibition. Ro 31-8220 additionally trans-inhibited TEA uptake in OCT1-transfected HEK293 cells, which likely discards a transport of Ro 31-8220 by OCT1. Besides Ro 31-8220, 7 additional BIMs, including the PKC inhibitor LY 333531, inhibited OCT1 activity, whereas 4 other BIMs were without effect. In silico analysis of structure-activity relationships next revealed that various molecular descriptors, especially 3D-WHIM descriptors related to total size, correspond to key physico-chemical parameters for inhibition of OCT1 activity by BIMs. In addition to activity of OCT1, Ro 31-8220 inhibited those of other organic cation transporters such as multidrug and toxin extrusion protein (MATE) 1 and MATE2-K, whereas, by contrast, it stimulated that of OCT2. Effets de l'exploitation par la pêche sur la dynamique de diversité des peuplements de poissons démersaux : analyse comparée du rôle des interactions entre espèces dans le golfe de Gascogne et dans le golfe du Lion Auteur(s) : Blanchard, Fabian Éditeur(s) : Université Bretagne Ouest Résumé : This work deals with the fishing effects on the diversity dynamics of demersal fish communities, focusing on the biotic interactions. Multispecies indices are used in order to characterize the demersal fish community dynamics of the Bay of Biscay and of the Gulf of Lions. These are the diversity indices, the temporal variability of the total biomass and of the slope of the number size spectra and the slope of the species-area curves (spatial segregation). The yearly bottom trawl surveys EVHOE, CHALIST and MEDITS, carried out in these two areas since 1983 by IFREMER for the stocks assessment provide the data. The differences between areas are related to a fishing index (mean value of the annual landings of demersal fish per surface area). Other areas are included in the analysis when the index values are available from the published literature (North Sea, Scotian Shelf, Kenya, French Guyana). It is argued from the diversity indices analysis that the Gulf of Lions community is more diverse and less disturbed than the Bay of Biscay one. This result is not consistent with a fishing effect but with biogeography. The total biomass variability and the slope variability of size spectra are higher in the most harvested areas than in the least ones. On the contrary, the species-area slope is the lowest in the most harvested areas. Theoretically, biotic interactions decrease the total biomass variability and the spectra slope variability because of compensations and increase the species area slope because of competitive exclusion. It is concluded that harvesting alters biotic interactions and favour local coexistence by decreasing competitive exclusion. The species diversity maintenance depends on competitive exclusion. Hence, biodiversity is threatened not only by a direct fishing effect but also by a dynamics alteration. Ce mémoire aborde le problème de l'impact de l'exploitation par la pêche sur la dynamique de diversité des peuplements de poissons démersaux. Les effets de la pêche sur le rôle des interactions biotiques entre espèces dans la dynamique des peuplements sont plus particulièrement analysés à l'aide de descripteurs multispécifiques. Les descripteurs utilisés sont les indices de diversité, la variabilité temporelle de la biomasse totale et de la pente multispécifique du spectre de poids et la pente des droites d'accumulation d'espèces mesurant la ségrégation spatiale. Les valeurs de ces indices sont calculées à partir des données de campagnes annuelles de chalutage de fond EVHOE, CHALIST et MEDITS, menées par l'IFREMER depuis 1983 pour l'évaluation des ressources halieutiques dans le golfe de Gascogne et dans le golfe du Lion. Les différences de caractéristiques de dynamique entre les deux peuplements révélées par ces descripteurs sont mises en relation avec un indice d'exploitation (valeur moyenne des débarquements annuels, divisée par la surface du plateau de la région concernée). D'autres régions ont été intégrées dans cette analyse lorsque les valeurs des indicateurs étaient disponibles dans la littérature (Mer du Nord, plateau de Nouvelle-Ecosse, Kenya et Guyane Française). Les indices de diversité utilisés permettent de mettre en évidence que le peuplement du golfe du Lion est plus diversifié et moins soumis à perturbations que celui du golfe de Gascogne. Ces caractéristiques semblent dues à l'histoire biogéographique de ces deux régions et non à un effet de la pêche. La variabilité de la biomasse totale et celle de la pente du spectre multispécifique augmentent depuis les régions les moins exploitées vers les plus exploitées. A l'inverse, la ségrégation spatiale diminue. En théorie, les interactions entre espèces diminuent la variabilité temporelle de la biomasse totale et de la pente du spectre multispécifique, à cause de compensations entre espèces, et augmentent la ségrégation spatiale par exclusion compétitive. L'interprétation donnée des résultats obtenus est ainsi une altération des interactions entre les espèces par l'exploitation. En prélevant de la biomasse, l'exploitation augmente la disponibilité des ressources pour les individus non pêchés. En conséquence, l'exclusion compétitive diminue et la coexistence locale des individus est facilitée. A long terme, la diversité est donc menacée, non seulement par raréfaction des espèces prélevées par l'exploitation mais aussi parce que les interactions sont altérées et que le maintien de la diversité en dépend pour partie. The importance and distinctiveness of small-sized phytoplankton in the Magellan Straits Auteur(s) : Zingone, Adriana Sarno, Diana Siano, Raffaele Marino, Donato Éditeur(s) : Springer Résumé : The distribution of summer phytoplankton across the Straits of Magellan (SOM) was studied with the aims of tracing differences among the distinct subregions of the area and contributing to the knowledge of its biodiversity. Samples collected at 25 stations were observed and counted in light microscopy. Selected samples were observed with transmission electron microscopy. The main unifying feature of the phytoplankton in the SOM was the high abundance and numerical dominance of small-sized (<10 μm) eukaryotic species, among which coccoid cells of <3 μm size were predominant (56.2 ± 30.6 of the total phytoplankton abundance). They mostly belonged to the prasinophyte Pycnococcus provasolii, which was abundant (0.8–6,834 cells × 103 ml−1) at all stations with the exception of those in proximity to the Atlantic entrances, where it was not recorded. Small-sized (<3 and 3–5 μm) diatoms (Minidiscus trioculatus, Lennoxia faveolata and other undetermined centric species) attained high densities (<3,757 cells 103 ml−1) especially at stations of the Patagonian sectors, whereas microplanktonic diatoms were only found at the two entrances of the Straits. Dinoflagellates were constituted mainly by >10 μm forms in the Andean subregion and <10 μm naked species in the Patagonian subregion, contributing up to 75.9 and 41.8% of the total carbon in these two areas, respectively. In the Patagonian subregion, flagellates mainly constituted by <5 μm forms and by cryptomonads <10 μm comprised up to 53.9% of the total biomass. Several species identified in this study have never been reported in other investigations in the SOM, while others, including Pycnococcus provasolii and Lennoxia faveolata, have rarely been recorded elsewhere. Overall, the summer phytoplankton of the Straits does not resemble that of any other region of the world’s seas. Although some of the predominant species might have been overlooked elsewhere, their abundance and relative importance apparently constitute a distinctive feature of the SOM. They are receptors of laminin-10/11 and are expressed in red blood cells (RBCs), epithelial cells of multiple tissues and vascular endothelial cells. To gain more insights into the biological function of Lu/BCAM gps, we looked for potential partners of their cytoplasmic tail. We isolated Ubc9 (Ubiquitin-conjugating enzyme 9) protein by screening a human kidney library using the yeast two-hybrid system. Lu/Ubc9 interaction was validated by GST pull-down and co-immunoprecipitation experiments. Endogenous Ubc9 formed a complex with endogenous or recombinant Lu gp in A498 and MDCK epithelial cells, respectively. Substitution of lysine 585 by alanine in the Lu gp, abolished in vitro and ex vivo interaction of Lu gp with Ubc9 protein. Lu K585A mutant transfected in MDCK cells exhibited a normal basolateral membrane expression but was overexpressed at the surface of polarized MDCK cells as compared to wild-type Lu. Pulse-chase experiments showed extended half-life of Lu K585A gp at the plasma membrane, suggesting an impaired endocytosis of this mutant leading to protein accumulation at the membrane. Furthermore, we showed that the ability of MDCK-Lu K585A cells to spread on immobilized laminin was dramatically decreased. Our data supports a physiological role for the direct interaction between Lu gp and Ubc9 protein, and reveals a role for this enzyme in regulating the stability of Lu gp at the cell membrane. Special attention was paid to describing the heterogeneity of the population by using single-cell approaches including flow cytometry (FCM) and different microscopic techniques and by analyzing a cell size fractionation experiment. Up to seven different subpopulations were distinguished by FCM based on nucleic acid content and light side scattering of the cells. The cell size analysis of symbionts showed that the symbiotic population was very heterogeneous in size, i.e., ranging from 0.5 to 5 _m in length, with variable amounts of intracellular sulfur. The side-scatter signal analyzed by FCM, which is often taken as a proxy of cell size, was greatly influenced by the sulfur content of the symbionts. FCM revealed an important heterogeneity in the relative nucleic acid content among the subclasses. The larger cells contained exceptionally high levels of nucleic acids, suggesting that these cells contained multiple copies of their genome, i.e., ranging from one copy for the smaller cells to more than four copies for the larger cells. The proportion of respiring symbionts (5-cyano-2,3-ditolyl-terazolium chloride positive) in the bacteriocytes of Codakia revealed that around 80% of the symbionts hosted by Codakia maintain respiratory activity throughout the year. These data allowed us to gain insight into the functioning of the symbionts within the host and to propose some hypotheses on how the growth of the symbionts is controlled by the host. Seasonal variations in planktonic community structure and production in an atlantic coastal pond: The importance of nanoflagellates Auteur(s) : Dupuy, Christine Ryckaert, Mireille Le Gall, Solange Hartmann, Hans Éditeur(s) : Springer Résumé : The structure and summertime production of planktonic communities and the role of nondiatom planktonic cells were studied in coastal ponds, which are areas traditionally used for fattening and greening table-sized oysters. The abundance and biomass of nano-microplanktonic protists were determined at weekly intervals between February 1998 and February 1999 in a coastal pond without oysters in the French Atlantic coast near La Rochelle. The production of these microbiotas was determined in the summer period. The structure of plankton communities revealed the following observations: (1) microphytoplanktonic cells were mostly diatoms and dinoflagellates, (2) microzooplanktonic cells were mainly ciliates, and (3) nanoplanktonic cells were represented by pigmented (80-90% of the nanoplankton biomass) and colorless nanoflagellates. Diatoms were dominated by Naviculiineae. Dinoflagellates were dominated by Peridiniales. Oligotrichida were predominant in the ciliate community. Protist biomass levels were nine times higher from April to August (summer period 1033 mu g C L-1) than from September to March (winter period 114 mu g C L-1). Whatever the season, nanoflagellates were dominant in the water column (66 and 53% of the entire protist biomass in the summer and winter periods, respectively). Nanoflagellates represented the highest production of nano-microplanktonic communities (76% of carbon protist production) in the coastal pond in summer and showed the shortest generation time (7.1 h). Dinoflagellates came after nanoflagellates in production (19.5% of carbon protist production). Diatoms represented only a supplementary carbon resource available for higher trophic levels, whereas, until now, they were considered as the principal food of oysters in coastal ponds. Ciliates were a small source of carbon, but their growth rate was high. We suggest, first, that nanoflagellates represented the primary resource available in the pond and could constitute an important food resource for higher trophic levels, such as oysters, farmed in this type of pond. Overall, the system appeared to be more autotrophic than heterotrophic. Because inorganic nutrients are quickly exhausted in a semiclosed pond, pigmented flagellates dominated the carbon biomass, production and biomass of bacteria were high (thus, the microbial food web appeared to be active in this pond), and mixotrophy seemed to be an important trophic mode there. La dynamique des pêches côtières du pays bigouden Auteur(s) : Charuau, Anatole Résumé : In the beginning, the coastal fleet of the Bigouden region fished by trawling and was completely focused on exploiting lobster and hake. It is a traditional business that expanded between the last war and the 70s, reaching its peak between 1975 and 1980.
The best image to give an idea of this fleet is that of a "bucket waterwheel" endlessly discharging its catch in the four ports of the Guilvinec district. Where there is a seafaring business in Guilvinec, Loctudy and Saint-Guénolé, there is a complementarity that is expressed in two ways:
- coastal fishing constitutes a very noticeable and regular supplement to production, since sales happen daily and only bad winter weather causes a relative decrease in the supplement. - coastal fishing gives a flattering image of the port based on the display of high-quality leading market products: daily live lobster, hake and angler.
In this study, we have above all tried to evaluate the situation of the Bigouden coastal system as an entity, while including in the same analysis the species, fleets and the exploitation of the resource. For the main species, a balance sheet is proposed. It covers the lobster, hake and angler separately. Lobster, because of its small amount of movement, can be considered as being an isolated stock, while fish belong to much more wide-ranging groups.
The lobster has gained from various increases in meshing that have been imposed since 1971, even if these have not always been strictly applied. One of the results of this management by means of meshing is a "depreciation" of the variations of daily yields around an average value of 100kg.
For the hake, the dominant trait is an increase in yields in the last few years in accordance with an improvement of the situation in the Gulf of Gascony due to a generalised decrease of effort and a modification of the exploitation profile since the appearance of boulters and straight nets. Whereas the situation for lobster fisheries, as regards the hake's marketable size, did not improve, since the demand for small hake still exists, we have nonetheless been witnessing for about ten years a slow diversification of the fleets' business activities, which is much more in line with sequential hake fisheries' logical outline of exploitation. When lobster yields decrease, one part of the fleet focuses on the fish at the edges of the lobster fisheries.
For the angler, we see rather similar phenomena and a significant increase in unloading of the two species by lobster fishermen due also to a widening of their fishing areas. On the lobster mudflats themselves, there is no notable change in exploitation in the historical records. All studies of coastal fishing always come up against the problem of acquiring statistical data and the Bigouden region is no exception to the rule.
To improve this situation, in 1987 and 1988 two items were chosen:
- a better awareness of the activity and of the areas frequented, by survey during unloading in Guilvinec and in Loctudy.
- a detailed description of the haul by species, by sampling, in Loctudy, on two-thirds of coastal sales, between December 1987 and November 1988.
For this reason, analysis of fleets by type is still cursory and covers only one year. The appearance of new trends towards fish can only be described qualitatively, since there is no historical record. The classification of boats was done by analysing data furnished by the auctions. This first assessment is not necessarily reliable, above all because of the auctions' inaccurate description of the hauls, a deficiency that we have tried to overcome by surveying in Loctudy.
In conclusion, we are witnessing in the Bigouden region, as in other entities, a rebalancing between traditional business activities and modern trends. Lobster fishing, almost exclusive before the 1980s, is tending to decrease for two main reasons:
- a generalised decrease in activity in the lobster fisheries due to non-renewal of coastal fish fleets,
- The transfer of part of the lobster fishermen's activities to fish, since the lobster and its accompanying catch no longer seemed able to insure the economic viability of new ships.
Other maritime districts resolved their lobster problem in a different way. The Lorient fishermen improved their yields by using more capable trawlers, the fishermen of Sable by exploiting the hake fish run to the fullest and limiting their lobster fishing to the best season. The Bigouden region is slowly turning to the second solution, as it renews its fleet.
A ses origines, la flottille côtière du pays bigouden péchant au chalut, est tout entière orientée vers l'exploitation de la langoustine et du merlu. Il s'agit d'une activité traditionnelle qui s' amplifie entre la dernière guerre et les années 70 pour atteindre son apogée entre 1975 et 1980. La meilleure image que l'on puisse donner de cette flottille est celle d'une "noria" débarquant inlassablement sa capture dans les quatre ports du quartier du Guilvinec. Là où existe une activité hauturière à Guilvinec, Loctudy et Saint-Guénolé, il y a une complémentarité qui s'exprime de deux manières : - la pêche côtière constitue un appoint très appréciable et régulier de production, puisque la vente est quotidienne et que seules les intempéries hivernales entraînent une baisse relative des apports. - la pêche côtière donne du port une image flatteuse basée sur la présentation de produits d'appel de grande qualité : langoustine vivante, merlu et baudroies quotidiens. Dans cette étude, nous avons surtout tenté de faire le point sur le système côtier bigouden en tant qu'entité, en englobant dans la même analyse des espèces, les flottilles et l'exploitation de la ressource. Pour les espèces principales, un bilan est proposé. Il couvre séparément la langoustine, le merlu et les baudroies. La langoustine, en raison de ses faibles déplacements, peut être considérée comme constituant un stock isolé, alors que les poissons appartiennent à des ensembles beaucoup plus vastes. La langoustine a beaucoup gagné des diverses augmentations de maillage qui ont été imposées depuis 1971, même si elles n'ont pas toujours été appliquées avec rigueur. Un des résultats de cette gestion par les maillages est un "amortissement" des variations des rendements journaliers autour d'une valeur moyenne de 100 kg. Pour le merlu, le trait dominant est une augmentation des rendements dans les dernières années en accord avec une amélioration de la situation dans le Golfe de Gascogne due à une baisse généralisée de l'effort et à une modification du profil d'exploitation depuis l'apparition des palangriers et des filets droits. Si la situation sur les pêcheries de langoustine, au regard de la taille marchande du merlu, ne s'est pas améliorée puisque la demande de petits merluchons existe toujours, on assiste par contre depuis une dizaine d'années à une diversification lente des activités des flottilles entrant beaucoup mieux dans le schéma logique d'exploitation des pêcheries séquentielles de merlu. Quand les rendements en langoustine baissent, une partie de la flottille s'oriente vers les poisson sur les marges des pêcheries de langoustine. Pour les baudroies on observe des phénomènes assez analogues et une augmentation importante des débarquements des deux espèces par les langoustiniers due également à un élargissement de leurs zones de pêche. Sur les vasières à langoustine elles-mêmes, il n'y a pas de modification notable de l'exploitation dans la série historique. Toute étude des pêches côtières se heurte toujours au problème d'acquisition des données statistiques et le pays bigouden n'échappe pas à la règle. Pour améliorer cette situation, en 1987 et 1988, deux points ont été retenus : - une meilleure connaissance de l'effort et des zones fréquentées, par enquête au moment des débarquements à Guilvinec et à Loctudy, - une description détaillée des apports par espèce, par échantillonnage, à Loctudy, sur les 2/3 des ventes côtières, entre décembre 1987 et novembre 1988. Pour cette raison, l'analyse de flottilles en types est encore sommaire et n'a porté que sur une année. L'apparition des tendances nouvelles vers le poisson ne peut être décrite que qualitativement puisque la série historique n'existe pas. La classification des bateaux a été faite par l'analyse des données fournies par les criées. Ce premier diagnostic ne présente pas toutes les garanties en raison surtout d'une description peu fidèle des apports par les criées, déficience que l'on a essayé de pallier en enquêtant à Loctudy. En conclusion, on assiste dans le pays bigouden comme dans d'autres entités à un rééquilibrage entre les activités traditionnelles et des orientations modernes. La pêche de langoustine, quasi exclusive avant les années 1980, tend à décroître pour deux raisons principales : - une baisse généralisée de l'effort sur les pêcheries de langoustine due au nonrenouvellement des flottilles de pêche côtière, - le report d'une partie de l'effort des langoustiniers vers les pêcheries de poissons, la langoustine et sa capture accessoire ne semblant plus pouvoir assurer la rentabilité économique des navires neufs. D'autres quartiers maritimes ont résolu de façon différente leur problème langoustine. Les Lorientais ont amélioré leurs rendements en adoptant des chaluts plus performants, les Sablais en exploitant à fond la séquence des pêcheries de merlu et en limitant leur intervention sur la langoustine à la saison la plus favorable. Le pays bigouden s'engage lentement dans la seconde solution, au rythme du renouvellement de sa flottille. Despite their ecological importance, knowledge on seasonal dynamics, community composition and abundance at the species level is limited due to their small cell size and few morphological features visible under the light microscope. Here, we present unique data on haptophyte seasonal diversity and dynamics from two annual cycles, with the taxonomic resolution and sampling depth obtained with high-throughput sequencing. From outer Oslofjorden, S Norway, nano- and picoplanktonic samples were collected monthly for 2years, and the haptophytes targeted by amplification of RNA/cDNA with Haptophyta-specific 18S rDNA V4 primers. We obtained 156 operational taxonomic units (OTUs), from c. 400.000 454 pyrosequencing reads, after rigorous bioinformatic filtering and clustering at 99.5%. Most OTUs represented uncultured and/or not yet 18S rDNA-sequenced species. Haptophyte OTU richness and community composition exhibited high temporal variation and significant yearly periodicity. Richness was highest in September-October (autumn) and lowest in April-May (spring). Some taxa were detected all year, such as Chrysochromulina simplex, Emiliania huxleyi and Phaeocystis cordata, whereas most calcifying coccolithophores only appeared from summer to early winter. We also revealed the seasonal dynamics of OTUs representing putative novel classes (clades HAP-3-5) or orders (clades D, E, F). Season, light and temperature accounted for 29% of the variation in OTU composition. Residual variation may be related to biotic factors, such as competition and viral infection. This study provides new, in-depth knowledge on seasonal diversity and dynamics of haptophytes in North Atlantic coastal waters. Are algae relevant to the detritus-based food web in tank-bromeliads? The distributions of both algal abundance and biomass were examined based on physical parameters, the morphological characteristics of bromeliad species and with regard to the structure of other aquatic microbial communities held in the tanks. Algae were retrieved in all of the bromeliad species with mean densities ranging from ∼10(2) to 10(4) cells/mL. Their biomass was positively correlated to light exposure and bacterial biomass. Algae represented a tiny component of the detrital food web in shaded bromeliads but accounted for up to 30 percent of the living microbial carbon in the tanks of Catopsis berteroniana, located in a highly exposed area. Thus, while nutrient supplies are believed to originate from wind-borne particles and trapped insects (i.e., allochtonous organic matter), our results indicate that primary producers (i.e., autochtonous organic matter) are present in this insectivorous bromeliad. Using a 24-h incubation of size-fractionated and manipulated samples from this plant, we evaluated the impact of mosquito foraging on algae, other microorganisms and rotifers. The prey assemblages were greatly altered by the predation of mosquito larvae. Grazing losses indicated that the dominant algal taxon, Bumilleriopsis sp., like protozoa and rotifers, is a significant part of the diet of mosquito larvae. We conclude that algae are a relevant functional community of the aquatic food web in C. berteroniana and might form the basis of a complementary non-detrital food web. The effects of starvation were observed using transmission electron microscopy, fluorescence in situ hybridization and catalyzed reporter deposition (CARD-FISH), and flow cytometry to monitor the anatomical and physiological modifications in the gill organization of the host and in the symbiotic population housed in bacteriocytes. The abundance of the symbiotic population decreased through starvation, with a loss of one-third of the bacterial population each month, as shown by CARD-FISH. At the same time, flow cytometry revealed significant changes in the physiology of symbiotic cells, with a decrease in cell size and modifications to the nucleic acid content, while most of the symbionts maintained a high respiratory activity (measured using the 5-cyano-2,3-ditolyl tetrazolium chloride method). Progressively, the number of symbiont subpopulations was reduced, and the subsequent multigenomic state, characteristic of this symbiont in freshly collected clams, turned into one and five equivalent genome copies for the two remaining subpopulations after 3 months. Concomitant structural modifications appeared in the gill organization. Lysosymes became visible in the bacteriocytes, while large symbionts disappeared, and bacteriocytes were gradually replaced by granule cells throughout the entire lateral zone. Those data suggested that host survival under these starvation conditions was linked to symbiont digestion as the main nutritional source. Reproductive dynamics of swordfish (Xiphias gladius) in the southwestern Indian Ocean (Reunion Island). Part 2: fecundity and spawning pattern Auteur(s) : Poisson, Francois Fauvel, Christian Éditeur(s) : EDP Sciences Résumé : Batch fecundity and relative fecundity of swordfish (Xiphias gladius) in the southwestern Indian Ocean were estimated from seven gravid swordfish females (size range 127-225 cm lower jaw-to-fork length, LJFL) with unovulated, hydrated oocytes collected onboard Reunion-based (France) longline swordfish fishing vessels between December 1999 to January 2001. To investigate the spawning pattern of swordfish, we used data collected through a combination of two at sea sampling regimes. A total of 17 007 geo-located size data of swordfish were recorded during 8 years (1993-2001) and a total of 1727 (size range 75-289 cm, LJFL) swordfish gonads (1107 females and 620 males) were sampled from May 1998 to January 2001. The estimated batch fecundity ranged from 995 000 hydrated oocytes for the smallest ripe female to 4.3 millions for the largest female sampled measuring respectively 127 to 225 cm in curved length (LJFL). The relative fecundity ranged from 25 to 72 hydrated oocytes per gram of body weight. We found that batch fecundity was positively correlated with fish length and that the older/larger females have earlier and longer spawning seasons than younger/smaller females. These findings suggested that older/larger females which are seasonally migrating in this spawning ground seem to play a major role in reproductive success of the species in producing significantly more offspring than younger females during an extended spawning season. Examination of the length-frequency date from the fishery indicated that the young fish are resident around Reunion and around the seamounts off Reunion Island. Our results highlight the important role of the older/larger females in the reproductive capacity of southwestern Indian Ocean stock. We discuss the potential implications of fishing the older/larger females for this stock in terms of reproduction and recruitment. La fécondité par acte de ponte et la fécondité relative chez l'espadon (Xiphias gladius) dans le sud-ouest de l'océan Indien sont estimées d'après sept femelles de taille comprise entre 127 et 225 cm (de l'extrémité de la mâchoire inférieure à la fourche caudale) ; celles-ci ayant atteint la maturité sexuelle, les ovaires contenant des ovocytes hydratés. Ces femelles ont été pêchées entre décembre 1999 et janvier 2001, par des palangriers ciblant l'espadon et basés à La Réunion. La stratégie de la reproduction chez l'espadon a été étudiée en utilisant des données collectées au cours de deux campagnes d'échantillonnages en mer : soit 17 007 mensurations géo-référencées d'espadons, de 75 à 289 cm, qui ont été enregistrées durant les 8 années d'étude (1993-2001) et 1727 gonades d'espadon (1107 femelles et 620 mâles) prélevées entre mai 1998 et juin 2001.La fécondité par acte de ponte estimée varie de 995 000 ovocytes hydratés, pour la plus petite femelle (127 cm), à 4,3 millions pour la plus grande femelle (225 cm). La fécondité par acte de ponte est corrélée positivement à la taille du poisson. La fécondité relative s'étend de 25 à 72 ovocytes hydratés par gramme de poids de corps. Les femelles les plus âgées/grandes pondent plus tôt et sur une période plus grande que les plus jeunes/petites femelles. Ainsi, les femelles plus âgées/grandes qui migrent de façon saisonnière vers cette aire de ponte, contribueraient plus largement au renouvellement de l'espèce en produisant de façon significative davantage de recrues que les femelles plus jeunes et ceci pendant une saison de ponte plus étendue. Par ailleurs, les jeunes individus semblent séjourner aux abords de l'île de La Réunion et des monts sous-marins situés au large. Nos résultats soulignent l'importance du rôle des femelles âgées/grandes dans la capacité reproductrice du stock d'espadon du sud-ouest de l'océan Indien. Les implications potentielles de la pêche : des femelles âgées et de grandes tailles sont discutées pour ce stock, du point de vue de la reproduction et du recrutement. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and, consequently, for the study of bivalve larvae genetics. Culling, by selective sieving of the smallest larvae is an advantageous practice at a phenotypic scale as it reduced variance in larval size, variance of developmental rate and time to settlement. Culling of 50% of the larval population only led to 15% less spat, showing a positive phenotypic correlation between larval growth and settlement success. However, culling represents a substantial risk for diversity loss, because it increases the variance of reproductive success among parental oysters. The effective population sizes of early settling cohorts of settlement were lower than those of later ones. Our results show that the settlement of slow growing larvae significantly contributes to minimizing the variability of reproductive success and therefore to maximizing genetic diversity. These results corroborate the low estimations of variability of broodstocks sampled in several French commercial hatcheries, relative to natural populations. The genetic composition of the larval population and the resulting spat was significantly different between the two tested temperatures, revealing genotype x environment interaction for survival. Similarly, genotype x environment interaction was also observed for larval growth as a higher temperature exerted a positive influence on the expression of genetic variability for this trait. Consequently, we can conclude that a temperature of 26°C coupled with culling, to common practice in oyster hatcheries, is likely to amplify the selection pressure for fast growing larvae. To test for this hypothesis, we compared larval developmental traits in the progeny of a hatchery broodstock closed for 7 generations, with the progeny of wild oysters and the two possible hybrids. Our results show that selection of fast growing larvae can counteract presumed inbreeding depression, due to higher mean relatedness among hatchery broodstock than in the wild. Genetic effects of intensive rearing conditions at larval stage are significant and should be taken into account in hatchery practices, especially in terms of genetic diversity management. Afin d'étudier les conséquences génétiques des pratiques de production en écloserie d'huître creuse, deux facteurs ont été examinés : l'élimination des petites larves et la température. Nos résultats montrent que l'assignation de parenté par marqueurs microsatellites est un outil performant pour les études génétiques en phase larvaire de familles élevées en mélange. Bien qu'avantageux d'un point de vue phénotypique, le tamisage sélectif représente un risque de perte de diversité. La fixation des larves à croissance lente permet en effet de minimiser la variabilité du succès reproducteur et de fait, de maximiser la variabilité génétique. Ces résultats corroborent les estimations de variabilité sur les stocks d'écloseries françaises où l'on constate une diversité allélique inférieure à celle de populations issues du milieu naturel. La température exerce également une influence sur la précocité de l'expression de la variabilité génétique pour la croissance larvaire. Ainsi une température élevée associée à une procédure de tamisage peut amplifier l'effet sélectif. Enfin, la sélection de larves à croissance rapide semble démontrée, s'opposant à la dépression de consanguinité présumée. Les conditions d'élevage peuvent donc avoir un effet génétique significatif qui devrait être pris en considération dans les pratiques d'écloserie, notamment dans la gestion de la diversité génétique.
