INRA, CFCS Description : The importance of longevity as an economically trait gives a picture of the flock efficiency and adaptation in a particular environment. A study was conducted in the experimental herd of Creole goats at INRA in Guadeloupe in order to test environmental (year and season at first kidding, age at first kidding and weight at first mating as well as genetic (index of resistance, sire) factors that affect longevity of does. Lifetime data set of 387 Creole does, reared at pasture all year long, was recorded over a period of 11 years (2001-2012). Does were bred for reproduction at 11months of age. Three mating periods were organized per year, corresponding to 3 climatic seasons, using buck effect. Data were analyzed using survival models (Survival Kit 6.1). The average age for culling was 5.03 years. The culling rate was higher for goats between 2 and 3 years (17 and 24%) and then gradually decreased. Year and season at first kidding did not have a long term influence on does? longevity; neither does age at first kidding. In contrast, weight at first mating had a significant effect and it can be recommended to farmers to mate primiparous goats heavier than 17 kg. Heritability was estimated to 0.16 allowing some genetic progress. No significant correlation was shown with the genetic breeding value of resistance to gastrointestinal parasitism. This study gave indications to breeders to improve their female flock management and increase does? longevity. L'importance de la longévité comme trait économique donne une image de l'efficacité et de l'adaptation de troupeau dans un environnement particulier. Une étude a été entreprise dans le troupeau expérimental de chèvres créoles à l'AICN en Guadeloupe afin d'examiner ambiant (l'année et la saison d'abord badiner, âge d'abord badiner et poids d'abord joindre aussi bien que (index de résistance, de père) les facteurs génétiques affectez dont la longévité fait. L'ensemble de données de vie du Créole 387 fait, élevé au pâturage tout au long de l'année, a été enregistré pendant 11 ans (2001-2012). Fait ont été multipliés pour la reproduction à 11months d'âge. Trois périodes d?accouplement ont été organisées par an, correspondant à 3 saisons climatiques, utilisant l'effet de mâle. Des données ont été analysées utilisant des modèles de survie (trousse de survie 6,1). L'âge moyen pour cueillir était de 5,03 ans. Le taux de cueillage était plus haut pour des chèvres entre 2 et 3 ans (17 et 24%) et alors graduellement diminué. L'année et la saison à premier badiner n'ont pas eu une influence à long terme sur la longévité des does ; ni l'un ni l'autre ne vieillit d'abord badiner. En revanche, le poids au premier accouplement a eu un effet significatif et il peut recommander aux agriculteurs de joindre les chèvres primipares des que 17 kilogrammes plus lourds. L'héritabilité a été estimée à 0,16 permettant du progrès génétique. Aucune corrélation significative n'a été montrée avec la valeur d'élevage génétique de la résistance au parasitisme gastro-intestinal. Cette étude a donné des indications aux éleveurs afin d'améliorer la gestion féminine de troupeau et d'augmenter la longévité de ces dernières. Along with an analysis of eastern Caribbean trade networks through surviving shipping records, extensive government correspondence, court records, and accounts in historic newspapers from the region, this will subsequently permit insights into how the shifting bounds and tolerances of legal and illicit trade by colonial officials on these islands fostered a smuggling and laundering ring for ships, goods, and people engaged in privateering, piracy during the Latin American Wars of Independence and the later Cisplatine War, and the post-abolition slave trade. By understanding the processes involved in smuggling and laundering illegal prizes by privateers and pirates, direct evidence these activities can become visible in the archaeological record through a predictive model based upon local practices of intentionally sinking illegally captured ships. Ce système rejetait la graphie étymologique, trop imprévisible. La force du standard 1 était d'être basée sur l'API (ainsi, à un phonème correspond un unique graphème) et d'être abordable tant pour un créolophone francophone que pour un créolophone anglophone. Mais s'agissait-il à l'époque d'une proposition provisoire ? expérimentale ? Il semblerait que oui car le temps et la réflexion ont fait émerger les limites d'une telle graphie. Ces critiques ont émané de Jean Bernabé lui-même qui a vu dans cette proposition une limitation de l'aisance de la lecture. The present-day structural relationships and the kinematics of these two faults, especially at their junction in Lavasanat region, is still a matter of debate. In this paper, we present the results of a morphotectonic analysis (aerial photos and field investigations) within the central part of the Mosha and eastern part of the North Tehran faults between the Mosha valley and Tehran City. Our investigations show that, generally, the traces of activity do not follow the older traces corresponding to previous long-term dip-slip thrusting movements. The recent faulting mainly occurs on new traces trending E-W to ENE-WSW affecting Quaternary features (streams, ridges, risers, and young glacial markers) and cutting straight through the topography. Often defining en-echelon patterns (right- and left-stepping), these new traces correspond to steep faults with either north- or south-dipping directions, along which clear evidences for left-lateral strike-slip motion are found. At their junction zone, the two sinistral faults display a left-stepping en-echelon pattern defining a positive flower structure system clearly visible near Ira village. Further west, the left-lateral strike-slip motion is transferred along the ENE-WSW trending Niavaran fault and other faults. The cumulative offsets associated with this left-lateral deformation is small compared with the topography associated with the previous Late Tertiary thrusting motion, showing that it corresponds to a recent change of kinematics. Sampling efforts in such environments may be constrained and many field studies only rely on subjective and/or qualitative approaches to design collection strategy. The region of Cacao, in French Guiana, provides an excellent study site to understand the presence and abundance of Anopheles mosquitoes, their species dynamics and the transmission risk of malaria across various environments. We propose an objective methodology to define a stratified sampling design. Following thorough environmental characterization, a factorial analysis of mixed groups allows the data to be reduced and non-collinear principal components to be identified while balancing the influences of the different environmental factors. Such components defined new variables which could then be used in a robust k-means clustering procedure. Then, we identified five clusters that corresponded to our sampling strata and selected sampling sites in each stratum. RESULTS: We validated our method by comparing the species overlap of entomological collections from selected sites and the environmental similarities of the same sites. The Morisita index was significantly correlated (Pearson linear correlation) with environmental similarity based on i) the balanced environmental variable groups considered jointly (p = 0.001) and ii) land cover/use (p-value<0.001). The Jaccard index was significantly correlated with land cover/use-based environmental similarity (p-value = 0.001). CONCLUSIONS: The results validate our sampling approach. Land cover/use maps (based on high spatial resolution satellite images) were shown to be particularly useful when studying the presence, density and diversity of Anopheles mosquitoes at local scales and in very heterogeneous landscapes. SUMMARY ANSWER: IVM strongly modified apoptosis- and autophagy-related relative protein levels in mice testicular tissue whereas the impact of cryopreservation procedures was minimal at the end of the culture. WHAT IS KNOWN ALREADY: In vitro spermatogenesis remains a challenging technical issue as it imposes to find a very close balance between survival and death of germ cell natural precursors (i.e. gonocytes and spermatogonia), which will eventually undergo a complete spermatogenesis close to in vivo conditions. The establishment of efficient culture conditions coupled with suitable cryopreservation procedures (e.g. controlled slow freezing [CSF] and solid surface vitrification [SSV]) of pre-pubertal testicular tissue is a crucial step in the fields of fertility preservation and restoration to improve the spermatic yield obtained in vitro. STUDY DESIGN, SIZE, DURATION: Here, we study cryopreservation procedures (i.e. CSF or SSV) and the impact of culture media compositions. A first set of 66 mouse pre-pubertal testes were directly cultured during 30, 36, 38 and 60 days (D) from 2.5 to 6.5-day-old CD-1 mice to evaluate the impact of time-aspect of culture and to endorse the reverse phase protein microarrays (RPPM) technique as an adapted experimental tool for the field of in vitro spermatogenesis. Ninety others fresh, slow-frozen and vitrified pre-pubertal testes were cultured during 30 days for the principal study to evaluate the impact of cryopreservation procedures before and after culture. Thirty-four testes dissected from 2.5, 6.5, 36.5, 40.5, 42.5 and 62.5 days postpartum (dpp) mice, corresponding to the time frames of spermatogenesis orchestrated in vitro, were used as in vivo controls. PARTICIPANTS/MATERIALS, SETTING, METHODS: After in vitro culture, testicular tissue samples originated from 2.5 or 6.5-day-old CD-1 male mice were analyzed using RPPM. This targeted proteomic technique allowed us to assess the expression level of 29 apoptosis- and autophagy-related factors by normalizing blank-corrected signal values. In addition, morphological analyses (e.g. HES, PAS, TRA98 and CREM) and DNA fragmentation in intra-tubular cells (i.e. terminal deoxynucleotidyl transferase dUTP nick end labeling; TUNEL) were assessed for the distinct experimental conditions tested as well as for in vivo control mouse testes. MAIN RESULTS AND THE ROLE OF CHANCE: A validation of the RPPM procedure in the field of in vitro spermatogenesis was completed with assay and array robustness before a principal study concerning the evaluation of the impact of in vitro culture and cryopreservation procedures. The proportion of elongated spermatids and the total cell number per seminiferous tubule tended to be very different between the in vivo and in vitro conditions (P < 0.05), suggesting the presence of a beneficial regulation on the first spermatogenesis wave by intrinsic apoptosis (Caspase₉) and autophagy (Atg5) factors (P < 0.0003 and r2 = 0.74). Concerning the impact of culture media compositions, a basic medium (BM) composed of αMEM plus 10% KnockOut™ serum replacement and gentamicin supplemented with retinol (Rol) and vitamin E (Vit. E) was selected as the best culture medium for fresh 6.5 dpp tissue cultured during 30D with 27.7 ± 8.10% of seminiferous tubules containing elongated spermatids. Concerning the impact of cryopreservation procedures, SSV did not have any impact on the morphological parameters evaluated after culture in comparison to fresh tissue (FT) controls. The proportion of tubules with elongated spermatids on testicular explants cultured with BMRol+Vit. E was not different between SSV (6.6 ± 1.6%) and CSF (5.3 ± 1.9%); however, round spermatids were observed more frequently for SSV (19 ± 6.2%) than CSF (3.3 ± 1.9%, P = 0.0317). Even if the proportion of TUNEL-positive cells for BMRol+Vit. E was higher at D30 after SSV (4.12 ± 0.26%) than CSF (1.86 ± 0.12%, P = 0.0022) and FT (2.69 ± 0.33%, P = 0.0108), the DNA damages observed at the end of the culture (i.e. D30) were similar to respective 6.5 dpp controls. In addition, the relative protein level expression ratio of an apoptotic factor, the phosphorylated FADD on Fas, was reduced by 64-fold in vitrified testes cultured with BMRol+Vit. E. Furthermore, we found in this study that the StemPro®-34 SFM culture medium supplemented with growth factors (e.g. EGF, bFGF, GDNF and LIF) prevented the differentiation of spermatogonial stem cells in favor of a significant proliferation with a better architectural pattern than in vivo 6.5 dpp controls with an increase of seminiferous tubules area for FT (P = 0.0357) and CSF (P = 0.0317). LIMITATIONS REASONS FOR CAUTION: Despite our promising results, the evaluation of apoptotic- and autophagic-related proteins was studied for a limited amount of proteins and on global testicular tissue. WIDER IMPLICATIONS OF THE FINDINGS: The data presented herein will help to improve apoptotic and autophagic understanding during the first spermatogenic wave. Moreover, our findings illustrate for the first time that, using finely-tuned experimental conditions, a testicular in vitro culture combined with proteomic technologies may significantly facilitate the study of cryopreservation procedures and in vitro culture evaluations. This study may also contribute to improve work on testicular tissues from pre-pubertal and adolescent cancer survivors. In order to characterize Basralocus clump organisation, many approaches were used : the study of different forest variables (total height, diameter at breast height, .... j, a spatial analysis studied by population splitting in 3 classes according to their diameters at breast height and an architectural analysis. To evaluate the pertinence of different approach, 2 clump structures, located on control and logged plots,are compared. The trees of the logged clump have a smaller height and diameter. a slender habit, a taller crown and a better growth than the control. These 2 clumps stand out by a different number of mature trees and different types of spatial distribution of juvenile trees around them are possible. The opening of the environment by forestry provokes a Jump rejuvenation by the presence of numerous unbranched young trees. The architectural analysis helps to detect a large expression variability in growth strategies which can be reflected by different architectural vigours. The combination between forest variables, spatial and architectural analysis results could suggest a dynamic evolution of these 2 clumps. L'Angélique (Dicorynia guianensis Amshoff, Caesalpiniaceae), première espèce exploitée Pour le bois d'oeuvre en Guyane française, a une répartition spatiale de type agrégatif. Afin de caractériser la structure d’un agrégat d'Angélique, plusieurs approches sont utilisées: une étude des différents paramètres dendrométriques (hauteur totale, diamètre à 1.30 m....j. une analyse spatiale réalisée en découpant la population des individus en 3 classes de diamètre bien distinctes et une analyse architecturale. Afin de valider la pertinence de ces approches, 2 agrégats, l'un en parcelle témoin et l'autre en parcelle exploitée pour le bois d'oeuvre, sont comparés. Ainsi, les arbres de l'agrégat exploité ont une hauteur et un diamètre plus petit, un port plus élancé, un houppier plus grand et une meilleure croissance. Ces 2 agrégats se distinguent par un nombre variable de semenciers et différents types de répartition spatiale des juvéniles autour de ceux-ci sont possibles. L'ouverture du milieu par l'exploitation forestière provoque un rajeunissement de l'agrégat souligné par la présence de nombreux individus jeunes non ramifiés. L'analyse architecturale permet de déceler une grande variabilité d'expression dans le développement qui se traduit par différentes vigueurs architecturales. Construction et étude d'un modèle de réseau trophique de la vasière de Brouage (bassin de marennes Oléron, France). Prise en compte de la saisonnalité et des échanges physiques pour la synthèse constructive des connaissances sur une zone intertidale d'une région tempérée. Auteur(s) : Leguerrier, Dephine Éditeur(s) : Université de la Rochelle Résumé : In order to better understand the functioning of the Brouage intertidal mudflat (Marennes- Oléron Basin, France), its carbon-based trophic web has been modelled and analyzed. The foodweb building is based on the 4 step method of Inverse Analysis: 1) conceive an a priori model as the graph of compartments (nodes) between which exist fluxes of material (vertices). These vertices are the unknowns for the problem; 2) gather all the existing knowledge about the ecosystem and translate it into linear equations and inequalities involving the fluxes; 3) complete this set of data by common knowledge on the behaviour of the compartments and translate it into inequalities, 4) solve the obtained linear system under the parsimony principle to find a unique solution vector. The food-web analysis is based on. The first purpose of this work was a methodological one: inverse analysis method has been adapted to consider the seasonal and spatial variability of the temperate ecosystem: two seasons have been coupled in the computation and the area has been divided into three geographical zones along a cross-shore gradient. Hence, two types of physical exchanges are involved: the benthicpelagic ones and the advection ones. Statistical results can also be obtained by coupling Monte Carlo methods to Inverse Analysis. To study the obtained networks, new indices based on Markov Chains take delay into account in describing integrative transfers of material between compartments. This lets emerge the systems' properties that are not obvious at first sight and emphasizes the role of peculiar components. The second purpose was to apply such methods to the Brouage mudflat ecosystem in order to better understand its functioning. As the knowledge and methods evolve, various food webs were computed and analyzed with the help such indices and existing ones. Sensitivity analyses allow to point at the most crucial lacks of knowledge, which can orientate future field research. New data would improve the quality of the model in two ways: one the one hand, they would validate or invalidate the outcomes and on the other hand, they would give useful information to better constrain some of the black boxes and most uncertain fluxes, especially the advection and vertical flows. The main results of the study show a strongly seasonal system in which benthic and pelagic components are linked with intensity. The system is driven by its high local benthic primary production, but also depends on imports from the basin. Each of the 3 geographic zones has a peculiar functioning. The central zone corresponds to the classical description of European intertidal mudflats: very productive, it can feed the other regions. It is a production zone. The upper part is less productive but is fed by the middle one via the water column and can be exploited by shorebirds. It is a degradation zone (material is transformed for less qualitative). The lower part strongly depends on the basin imports for its functioning, as the cultivated bivalves act as a pump for pelagic material of which a great part is deposited under the cultivation structures (oyster racks or mussels "bouchots"). It is a transformation zone. Un double objectif (méthodologique et appliqué) a été visé par une modélisation du réseau trophique de la vasière de Brouage (Bassin de Marennes Oléron, France) à l'aide de l'analyse inverse, et son étude à l'aide de l'analyse des réseaux. L'Analyse Inverse permet en effet de reconstruire l'ensemble du réseau à partir de données éparses, le problème du manque de données étant récurrent en Ecologie. Au plan méthodologique, l'analyse inverse a été adaptée à la prise en compte des caractères saisonnier et spatialement hétérogène d'un écosystème tempéré de zone intertidale : deux saisons ont ainsi été couplées et la vasière a été considérée le long d'un transect perpendiculaire au rivage et divisé en trois zones spatiales. Deux types de flux physiques interviennent donc : les flux verticaux de dépôt et remise en suspension, et les flux d'advection via la colonne d'eau. Des pistes d'amélioration et de nouvelles méthodes sont proposées. Un résultat sous forme statistique peut être obtenu en couplant l'utilisation de méthodes de Monte Carlo à l'analyse inverse pour la construction des réseaux trophiques. Les moyens d'étude des modèles ont été enrichis par la mise au point d'indices issus de la construction de chaînes de Markov et prenant en compte le caractère temporel des transferts de matière entre compartiments. Enfin, la transformation mathématique du modèle statique pour une étude dynamique de la stabilité de l'équilibre décrit est abordée. Plusieurs modèles ont été construits au cours de l'évolution des méthodes et de l'acquisition de connaissances sur le site. Des méthodes d'analyses existantes et d'autres mises au point dans le cadre de cet ouvrage ont été appliquées aux réseaux obtenus afin de faire émerger les propriétés du fonctionnement de la vasière et de souligner le rôle de certains compartiments. Des analyses de sensibilité permettent d'évaluer la robustesse des résultats et d'orienter les recherches futures en mettant en évidence les manques les plus cruciaux. Des données supplémentaires, notamment concernant les foraminifères benthiques, le zooplancton ou les mouvements verticaux du microphytobenthos permettront de fournir de meilleures contraintes et de valider (ou non) les résultats obtenus. Les principaux résultats montrent un système au fort caractère saisonnier dont les compartiments benthiques et pélagiques sont très liés. La production primaire benthique locale est un moteur important, mais qui ne suffirait pas à suppléer l'indispensable importation de matériel détritique. Le découpage de la vasière en trois zones fait apparaître les particularités de chacune. La zone centrale correspond à la description classique des « vasières intertidales européennes » : très productive, elle peut alimenter les autres régions. C'est une zone de production et d'exportation. La partie supérieure est moins productive et reçoit de la matière de la zone de milieu d'estran via la colonne d'eau. Sa production secondaire peut alors être exploitée par les limicoles, nombreux en hiver. C'est une zone de dégradation et d'exportation. Le bas d'estran dépend fortement des importations extérieures, « pompées » par les bivalves cultivés qui sont ainsi directement responsables d'une forte sédimentation sous les structures mytilicoles et ostréicoles. Stability of spatial structures of demersal assemblages: a multitable approach Auteur(s) : Gaertner, Jean-claude Chessel, Daniel Bertrand, Jacques Éditeur(s) : Elsevier Résumé : The present work is based on a set of bottom trawl surveys performed in the Gulf of Lions between 1983 and 1992. A new method which couples the STATIS multitable approach with the logic of correspondence analysis (CoA) is proposed to study the stability of spatial organization of demersal assemblages in terms of species composition. This CoA version of STATIS has provided a clear representation of the stable part of the spatial structure of the assemblages. In addition, it gave an insight into the interannual variations of the distribution of each population around the reference structure. Our results showed that the demersal assemblages of the Gulf of Lions exhibited strong spatial structuring, mainly orientated along a bathymetric gradient. This gradient is structured around three major regions: the coast, the continental shelf and the continental slope. Despite sampling variations that occurred during the course of the study, the analysis showed a high degree of reproducibility for this spatial pattern. With the exception of Mullus surmuletus, the species considered in this investigation exhibit limited variations in their spatial distribution. Finally, in the absence of log-books covering the activities of commercial fishing vessels, these findings constitute a base of reference for the analysis of the dynamics of fishing fleets exploiting the demersal resources of the Gulf of Lions. Ce travail porte sur une série de campagnes de chalutage de fond réalisé dans le golfe du Lion entre 1983 et 1992. Une nouvelle méthode fondée sur le couplage de la technique multitableaux STATIS et de l'analyse factorielle des correspondances (AFC) est proposée afin d'étudier l'organisation spatio-temporelle des assemblages démersaux, poissons et quelques céphalopodes, en terme de composition spécifique. Cette version AFC de STATIS a fourni une représentation claire de la part stable de l'organisation spatiale des assemblages. De plus, elle nous a permis d'appréhender les variations interannuelles de la distribution de chaque population autour de la structure de référence. Nos résultats montrent que les assemblages démersaux du golfe du Lion présentent une forte structuration spatiale, principalement orientée selon un gradient bathymétrique. Ce dernier est structuré en trois grandes régions: la côte, le plateau continental et le talus continental. En dépit des variations d'échantillonnage survenues au cours de la période d'étude, l'analyse a mis en évidence une forte reproductibilité de ce schéma d'organisation. A l'exception de Mullus surmuletus, les espèces considérées dans ce travail présentent des variations limitées de leur distribution spatiale. We argue that P, B, and Taxes provide a reasonable estimate of principal stress directions when faulting is close to Andersonian conditions, and that this can be estimated from the location on a triangular diagram. We analyze the geometrical relationship between the plunges of P, B, and Taxes on one hand, and the rake of slip and dip of nodal planes on the other hand. We show that the rake and dip level curves correspond to trajectories of the vertical direction along great and small circles with respect to the frame of the P, B. and Taxes. This shows that dip-slip faulting is compatible with vertical P or Taxes, but does not require it, and instead requires horizontal B axes. It also shows that strike-slip faulting does not require vertical B axes, but P and Taxes with equal plunges. This also reveals that focal mechanisms where P, B, and Taxes all have moderate plunge correspond to two very different types of nodal planes: a steeply dipping one with oblique slip and a moderately dipping one with strike-slip. Seismically active and moderately dipping strike-slip faults are to be found among these events. Seismic hazard assessment for Tehran, Tabriz and Zandjan cities (NW Iran) based on morphotectonical and paleoseismological approaches. ; Evaluation de l'aléa sismique pour les villes de Téhéran, Tabriz et Zandjan dans le NW de l'Iran. Within the Tehran capital, we focused our analysis on the Mosha-North Tehran (M-NT) active fault system. Our investigations show that this fault system is mainly dominated by left-lateral faulting. Along the eastern part of the M-NT fault system, we found evidences of at least seven strong (M>7) paleoearthquakes during the past 10 ka (recurrence interval : 1100-1400 yrs). Our study along the central portion of the fault system (in Abali region) suggests that it corresponds probably to the source of the 1830 AD historical earthquake (M=7.1, Io= IX). Within Tabriz region, we studied the SE segment of the North Tabriz fault. We found evidences of at least three strong events, which occurred during the past 33.5 ka. The North Tabriz dextral fault corresponds to the central portion of a fault system (TFS), and contains two fault segments (NW and SE). Our paleoseismological analysis shows a variation of the slip rate, which decreases from NW to SE along the strike-slip fault. The western (Mishu) and eastern (Bozghush) terminations of the fault system correspond to oblique-compression zones. TFS belongs to the eastern portion of a regional fault system (GSKT), linking the active extension within the central part of the collision and the active compression to the East. Finally, we focus on the Zandjan area, where no important historical seismicitiy is recorded. There, based on morphotectonics analysis we discovered a large active fault network, whose southeastern part affects the Zandjan city. To conclude this work, we replace our observations within the general geodynamic framework of the Arabia- Eurasia collision. One of the striking pictures is the occurrence of opposite kinematics along the M-NT (and other range parallel faults within the south Central Alborz) and the North Tabriz fault systems that have the same trend. This shows the rapid variation of the regional stress field in the Northern Iran. L'objectif de ce mémoire de thèse est l'étude de la tectonique récente et l'évaluation de l'aléa sismique pour trois villes importante, Téhéran, Tabriz et Zandjan, situées dans le nord-ouest de l'Iran. Dans la capitale de l'Iran, Téhéran, nous nous sommes concentrés sur le système de faille active de Mosha-Nord Téhéran. Nos travaux démontrent que ce système est essentiellement décrochant sénestre. Nos analyses paléosismologiques sur la portion est de ce système (dans la vallée de Tar) montrent au minimum les évidences de sept grands événements sismiques (M>7) pendant les derniers 10000 ans (l'intervalle de récurrence est de 1100 à 1400 ans). Les études que nous avons mené dans la partie centrale de ce système (région d'Abali) révèlent qu'il pourrait être la source du dernier événement sismique ayant affecté la ville de Téhéran en 1830 AD. Dans la région de Tabriz, nous nous sommes intéressés au segment SE de la faille décrochante dextre de Nord Tabriz où nous avons trouvé les évidences de trois grands séismes pendant les derniers 33500 ans. Nos études montrent que la faille Nord Tabriz qui est la portion centrale d'un système de faille (TFS) est constituée par deux segments NW et SE dextres. Notre étude paléosismologique suggère une variation de la vitesse qui diminue du NW vers SE le long de ces segments. Les extrémités ouest (Mishu) et est (Bozghush) de ce système correspondent à des zones compressives qui absorbent les déplacements horizontaux sur les segments NW et SE. D'un point de vue général la TFS est une partie d'un système de failles régionales (GSKT) qui relie une zone d'extension active dans la partie centrale de la collision Arabie-Eurasie et une zone de compression active à l'Est. Enfin, à Zandjan qui est une ville située dans une région de lacune sismique, nous avons découvert un important réseau de failles actives. Le tracé sud-est de ce réseau de failles passe par la ville de Zandjan. En conclusion, les localisations et les cinématiques des systèmes de faille étudiés sont discutées dans le cadre géodynamique général de la collision Arabie-Eurasie. On remarquera que la faille majeure de Nord Tabriz qui correspond essentiellement à un décrochement dextre a la même direction NW-SE que les décrochements sénestres présents dans la chaîne de l'Alborz (les failles de : Mosha, Talaghan et Manjil). Combined effect of exposure to ammonia and hypoxia on the blue shrimp Litopenaeus stylirostris survival and physiological response in relation to molt stage Auteur(s) : Mugnier, Chantal Zipper, Etienne Goarant, Cyrille Lemonnier, Hugues Éditeur(s) : Elsevier Résumé : The effect of ambient ammonia, hypoxia and combination of both on survival and the physiological and immunological response of the blue shrimp Litopenaeus stylirostris in relation to molt stage was studied. Shrimp were submitted to 44.0-71.5 mg 1(-1) total ammonia-N corresponding to 2.0 mg 1(-1) unionized ammonia NH3-N and/or to 1.5 mg O-2 1(-1) (4.3 kPa) for 24 hours. Survival was recorded and the molt stages of both dead and surviving shrimp determined. Only shrimp in intermolt and premolt stages were sampled for analysis of haemolymph. Haemolymph was assayed for osmoregulatory capacity (OC), magnesium ion (Mg2+), calcium ion (Ca2+), total proteins, oxyhaemocyanin, lactate, glucose and total haemocyte count (THC). Low mortalities were recorded for shrimp submitted independently to ammonia or hypoxia. Seventy five percent of dead shrimp were in early post molt (stage A) in ammonia treatment, while hypoxia affected mainly late premolt animals (stage D-2). A synergic effect of ammonia and hypoxia combination (A+ O-2 treatment) on mortality was observed, affecting nearly exclusively shrimp in late premolt stage D-2. Analysis of molt stage repartition at the end of the experiment suggests that ammonia treatment may have accelerated molting. The common physiological response of shrimp to the different treatments was characterized by a reduced OC and an increase in Ca2+. Increase in Mg2+ could not be validated by the statistical analysis, as well as glycaemia variations. Plasmatic lactate level increased and THC decreased in shrimp submitted to hypoxia and the combination of hypoxia and ammonia. Total proteins concentration was reduced in ammonia and A+O-2 treatments. The effect was more pronounced in late premolt shrimp than in intermolt shrimp. Combination of ammonia and hypoxia led to a physiological response stronger than this observed for ammonia-alone and/or hypoxia-alone treatments, except for oxyhaemocyanin. Caractérisation chimique des composés humiques et de leurs diverses classes de poids moléculaires dans les dépôts du delta du Rhône Auteur(s) : Gadel, F Charriere, B Serve, L Comellas, L Éditeur(s) : Gauthier-Villars Résumé : The analysis of humic compounds by elemental analysis, PY-GC-MS and HPLC techniques permitted determination of the origin of organic inputs, essentially terrestrial, and the effects of environmental conditions in the deposits along a north-south transect through the Rhone delta, from the river mouth to the open sea. In the prodelta the sedimentation of fine particles enriches the sediments in organic matter. The humic compound contents were relatively low in sediments, perhaps due to a rapid burying of organic matter which leads to anoxic conditions. In sediments fulvic acids are more abundant than humic acids. Elemental analysis indicated that fulvic acids were enriched in hydrogen and nitrogen as compared to humic acids. Furthermore, H/C and N/C ratios increased in the fulvic and humic acids towards the open sea, related to a more marine character. Pyrolysis analysis showed some differences between the chemical composition of fulvic acids which correspond to low molecular weights enriched in carbohydrates, and that of humic acids, high molecular weight compounds which are enriched in phenols and aromatic hydrocarbons. The pyrolysis-derived phenols were abundant near the river mouth, decreasing towards the open sea as nitrogen-containing-compounds increased. Near the river mouth, the abundance of phenolic aldehydes demonstrated by HPLC emphasized the existence of fresh organic matter. On the other hand, off the mouth, an increase in phenolic acids could indicate a more degraded organic material. The fractionation of humic acids in various classes of molecular weight species using ultrafiltration (Amicon membranes) and their analysis showed that, close to the river mouth, the large proportion of the high molecular weight humic compounds observed was enriched in phenols and carbohydrates, probably inherited from the terrestrial ligno-cellulosic complex. This process seemed to be superimposed on the increase of molecular weights which is usually observed as the environment becomes purely marine. These humic compounds would derived either from organic matter from terrestrial soils swept along by the river flow, after deposition on its bed or, more probably, from high molecular weight compounds which, associated with the mineral phase, flocculated when reaching the fluvio-marine front. Generally, the low molecular weight fractions were enriched in hydrogen and nitrogen. Near the river mouth, nitrogen-containing compounds were concentrated in the low molecular weight classes which correspond to organic molecules regarded as humic precursors. Off the mouth, they were incorporated into the high molecular weight complexes, certainly of marine origin. Analyses of the sedimentary filling of paleokarsts help completing the geological record of regions that have been submitted to post-depositional, long-term, continental evolution. Jurassic carbonate platform of Languedoc (South of France) has undergone several karstification phases from Cretaceous to Neogene. Later incision of canyons through the carbonate massifs allows to observe paleokarsts over 400m depth, within the massifs. Paleokarsts are partly filled with sediments. Some have yielded marine bioclasts (echinoderms, radiolars), foraminifera and nannofossils; others are composed of polygenic detrital sediments, including sources from the upstream Paleozoic basement (Cevennes). The age of the filling of successive paleokarsts can be constrained by structural relationships and by biostratigraphy. These findings suggests 1) the marine elements of the karstic filling relate to a Late Cretaceous to Early Paleocene interval, while 2) the Paleozoic basement-sourced-sediments were trapped in the karst during Miocene to Present. Karstic sediment containing Early Paleocene foraminifera and nannofossils are found in paleokarsts cavities distributed across the entire thickness of the carbonate massif (≥350m). This requires base-level lowering and associated karstification, followed by base-level rise and karst filling of at least 350 m amplitude, respectively. The time interval corresponding to the occurrence of foraminifera and nannofossils in karsts covers 10 Myrs ; surprisingly, no equivalent marine sediments are preserved on the surface. In addition, analyses of the different forams species suggests several (up to 3) distinct karstification and marine filling cycles. Finally, sedimentological facies analysis of the karst filling reveals the following succession of processes: low energy settling of mudstone, high energy reworking, transport and deposition of silts and sandstones within the karst system. Integration of geological, paleontological and sedimentological data, leads to a polyphase scenario in response to repeated base-level variations, more than 350m amplitude. Such an amplitude excludes eustacy, and the improbable repeated sequence of uplift and subsidence rules out tectonics, as driving forces for base-level change, respectively. We propose that the high-amplitude base-level changes results from a succession of desiccation-flooding events of an endorheic, silled, basin during Early Paleogene. The later detrital assemblage sourced in the Cevennes occurs on perched paleosurfaces and in karst cavities across the whole 350m-deep canyon walls. When found on paleosurfaces, they correspond to the south-flowing Early Miocene fluvial drainage, and can be correlated downstream with the marine, well dated, Early Miocene, sediments. When found within the karst cavities, they correspond to successive base-level surfaces connected to the progressive incision of the canyon. If the seminiferous epithelium is well described in terms of organization and cellular morphology of cells that compose it, the processes by which undifferentiated diploid germ cells enter meiosis and give haploid cells that undergo many morphological transformations, are not fully decrypted. These processes rely on the coordinated and sequential expression of genes, including specific products for each stage of germ cell development These gene products are essential at each key stage of spermatogenesis. Transcriptomics since the 1990s, and proteomics since the 2000s have contributed to the improved. understanding of these mechanisms. A long term proteomic study aiming at characterizing the proteomes of Sertoli cells and germ cells, and a recent study that characterized and quantified the transcriptome of isolated rat testicular cells at high resolution using de novo sequencing of transcripts (RNA-Seq), have been the basis of my thesis work. The latter study showed the accumulation of long non-Coding RNAs (lncRNAs) and testicular unannotated transcripts (TUTs) at meiotic and post-Meiotic stages of spermatogenesis in the rat. In this context, my thesis work aimed at validating the coding potential of many genes expressed in germ cells using RNA-Seq combined with shotgun proteomics, a so-Called PIT (Proteomics Informed by transcriptomics) approach. In this approach, the protein sequences translated from the transcripts assembled by RNA-Seq in the different testicular cell types are integrated into a custom database of protein sequences used to query mass spectrometry data obtained from proteins of meiotic and post-Meiotic cells. The PIT approach showed that 69 TUTs or lncRNA (corresponding to 44 loci) code for proteins in meiotic cells and post meiotic cells, and we confirmed experimentally the meiotic and post-Meiotic expression for two new transcripts encoding for VAMP9, a protein of the SNARE family, and a new testicular enolase T-ENOL. The post-Meiotic expression of T-ENOL protein was confirmed by immunohistochemistry using a polyclonal antibody raised against the recombinant protein. This approach also allowed us to identify new isoforms of known proteins, specific to each stage of spermatogenesis. Germ cells and Sertoli cells maintain a dialogue which is necessary to the success of spermatogenesis and spermiogenesis. Another part of my work aimed at identifying membrane proteins, in germ cells and residual bodies, that may be involved in the dialogue between Sertoli cells and germ cells, using a ICPL relative quantification proteomic approach. The ICPL analysis enabled us to establish a list of 166 proteins whose expression is differential between pachytene spermatocytes, round spermatids and residual bodies. Their differential expression suggests that these proteins may play a role in spermiogenesis. Thanks to the Gene Ontology annotations, a list of 8 proteins with a putative role in signal transduction, cell recognition or differentiation, thus potentially involved in the dialogue between Sertoli and germ cells was drawn. In addition, I provided a first proteome of rat Sertoli cells, germ cells and residual bodies obtained by shotgun proteomics. La spermatogenèse chez les mammifères est une fonction biologique complexe incluant des processus de prolifération cellulaire, de méiose et de différenciation uniques visant à la production des gamètes mâles au sein du testicule. Si l’épithélium séminifère est bien décrit sur le plan de son organisation et de la morphologie des cellules qui le composent, les processus par lesquels les cellules germinales diploïdes indifférenciées entrent en méiose pour donner ensuite des cellules haploïdes subissant par la suite de nombreuses transformations morphologiques, ne sont pas totalement décryptés. Ils reposent sur l’expression coordonnée et séquentielle de gènes dont les produits spécifiques de chaque stade de développement des cellules germinales sont essentiels aux étapes clés de la spermatogenèse. La transcriptomique depuis les années 1990 et la protéomique depuis les années 2000 ont contribué à l’amélioration de la connaissance de ces mécanismes. Une étude protéomique visant à caractériser par des approches systématiques et différentielles les protéomes des cellules de Sertoli et de la lignée germinale, et d’autre part une étude récente, réalisée dans notre unité, qui a permis de caractériser et de quantifier le transcriptome des cellules testiculaires isolées de rat en utilisant le séquençage de novo des transcrits (RNA-Seq), ont été à la base de mes travaux de thèse. Cette dernière étude a mis en évidence l’accumulation de longs ARNs non codants (lncRNAs) et de transcrits testiculaires non annotés (TUTs) aux stades méiotique et post- méiotique de la spermatogenèse chez le rat. Dans ce contexte, mon travail a consisté à valider le potentiel codant de nombreux gènes exprimés dans les cellules germinales par une approche dite PIT (Proteomics Informed by Transcriptomics) couplant protéomique Shotgun et RNA-Seq. Dans ce type d’approche, les séquences protéiques déduites des transcrits des différents types cellulaires, assemblés par RNA-Seq, sont intégrées dans une base personnalisée de séquences protéiques utilisée pour interroger les données de spectrométrie de masse obtenues à partir de protéines de cellules méiotiques et post-Méiotiques. L’approche PIT a permis de montrer que 69 TUTs ou lncRNA (correspondant à 44 loci) codent pour des protéines dans les cellules méiotiques et post méiotiques. L’expression post-Méiotique de deux nouveaux transcrits, l’un codant pour la protéine VAMP9, une protéine de la famille SNARE, et l’autre pour une nouvelle énolase T-ENOL a pu être confirmée. L’expression post-Méiotique de T-ENOL a été confirmée par immunohistochimie à l’aide d’un anticorps polyclonal produit contre la protéine recombinante. Cette approche nous a également permis d’identifier de nouvelles isoformes de protéines connues spécifiques de chaque stade de la spermatogenèse. Les cellules germinales et les cellules de Sertoli entretiennent le dialogue nécessaire au bon déroulement de la spermatogenèse. Une autre partie de mon travail a consisté à identifier des protéines membranaires des cellules germinales et des corps résiduels, susceptibles d’intervenir dans le dialogue entre les cellules de Sertoli et les cellules germinales, par une approche protéomique de quantification relative ICPL. Cette approche a permis d’établir une liste de 166 protéines différentiellement exprimées entre les spermatocytes pachytène, les spermatides rondes et les corps résiduels, qui sont susceptibles de jouer un rôle dans la spermiogénèse. Grâce aux annotations de le Gene Ontology, j’ai pu établir une liste de 8 protéines ayant un rôle supposé dans la transduction du signal, la reconnaissance cellulaire ou bien la différenciation. Ici, nous avons analysé l'effet de cette perturbation sur les caractéristiques démo-génétiques d’une espèce forestière héliophile Jacaranda copaia. Nous avons travaillé sur quatre parcelles (chacune de 6,25 ha) du dispositif de Paracou, dont une parcelle témoin et trois parcelles à degré d'exploitation croissant. Un échantillonnage exhaustif de tous les arbres appartenant à l'espèce a été réalisé sur les quatre parcelles, La population se répartit en deux groupes : les adultes ayant potentiellement participé à la régénération (DBH > 10 cm ; N = 133) et les juvéniles issus de la régénération (DBH < 10 cm). Ces derniers sont séparés en deux sous-groupes selon leur position : dans les trouées d'exploitation (N = 564) ou hors des trouées d'exploitation (N = 127). A l’aide de quatre marqueurs microsatellites SSR. Nous avons donc comparé la diversité génétique, le coefficient de consanguinité, la différenciation, le déséquilibre génotypique et la proximité génétique des individus en fonction de la distance spatiale (auto corrélation génétique spatiale). Les résultats ainsi obtenus ne montrent pas d'effet de l’exploitation sur la diversité génétique globale. Cependant celle-ci est plus structurée dans l'espace et se traduit par une consanguinité plus forte. La présence de déséquilibres génotypiques et une répartition en agrégats des juvéniles présents dans les trouées d’exploitation. This IntuiScript project aims at developing a digital workbook to help with teaching children how to handwrite. To do so, we must be able to analyse their handwriting, to evaluate if the letters are correctly written, and to detail what aspects of the child symbols – letters, numbers, and geometric forms-do not correspond to the teacher models.
