Il s’appuie sur les résultats des études menées à l’échelle de la Martinique en 2010 dans le cadre de l’évaluation de la valeur économique et sociale des récifs coralliens et écosystèmes associés et à celle de la commune du Prêcheur en 2011 dans le cadre de la mise en place de la réserve marine régionale. Il met en exergue les éléments qui composent les disparités de perception et de souhaits quant aux mesures de gestion des espaces littoraux. Il montre toutefois que la création d’une aire marine protégée dans la commune du Prêcheur emporte l’adhésion de tous. The article presents the differences in perception between the tourists and residents regarding the protection of the marine and coastal environment. It is based on the results of studies conducted across Martinique in 2010, in the framework of the evaluation of the economic and social value of coral reefs and associated ecosystems, and at the level of the rural community of le Prêcheur in 2011, in the context of the implementation of the regional marine reserve. It highlights the elements that compose the disparities of perception and wishes with respect to coastal areas management measures. Cette faiblesse économique est liée à la faible productivité agricole, à l’absence de matières premières et d’industries et à l’instabilité des différents régimes politiques. La société repose sur une majorité de pauvres. Dans ces conditions le rapport à l’environnement est très différent de celui que l’on trouve dans le reste des Antilles à niveau de vie plus élevé.Pour le Haïtien, le milieu est un moyen de production qui peut être utilisé dans l’immédiat. C’est ce milieu dans sa globalité qui est alors exploité; le sous-sol pour la construction, le sol quand il existe pour l’agriculture, les arbres pour le charbon de bois ou les poteaux de construction et quand la faim persiste : les eaux littorales. Il n’existe aucune limite à l’exploitation des écosystèmes. La misère se vit à l’instant. L’idée de protection durable n’a pas de sens en période de crise. Les pressions exercées par les populations littorales sont diverses et d’intensité variée. Ces atteintes aux milieux littoraux et marins augmentent et couvrent des étendues croissantes.Les littoraux supportent ainsi un grand nombre d’activités ; les unes sont liées à la mer (la pêche, le cabotage), les autres reposent sur l’exploitation de la bordure terrestre (l’extraction de matériaux, l’exploitation des mangroves et l’agriculture, le tourisme). ![]() | Trans-Atlantic distribution of a mangrove oyster species revealed by 16S mtDNA and karyological analyses Auteur(s) : Lapegue, Sylvie Boutet, Isabelle Leitao, Alexandra Heurtebise, Serge Garcia, P Thiriot Quievreux, C Boudry, Pierre Résumé : Three species of man.-rove oysters, Crassostrea rhizophorae, C. brasiliana, and C. gasar, have been described along the Atlantic shores of South America and Africa. Because the distribution of these molluscs is of great biological and commercial interest, their taxonomy and distribution deserve further clarification. Therefore, 15 populations were sampled from both continents. Their 16S mitochondrial polymorphism was studied by sequencing and PCR-RFLP analysis. Two haplotypes were identified. Haplotype a was the only one observed in Africa, but it was also observed in South America together with haplotype b. Because C. gasar is the only mangrove oyster identified on the west coast of Africa, haplotype a was attributed to this species, which has thus been shown to occur in South America. Haplotype b is attributed to C. rhizophorae. The karyotypes of specimens of C. gasar, from Africa and from South America, were very similar, and both species were observed at the same location in Brazil. The occurrence of C. gasar in South America adds a third species-in addition to C rhizophorae and C. brasiliana-to the list of species present along these coasts. The predominant surface circulation patterns in this part of the Atlantic Ocean favor the hypothesis that C. gasar was transported from Africa to America. Finally, a phylogenetic tree built with seven 16S sequences from Crassostrea and Saccostrea species showed that C. gasar is intermediate between the American Crassostrea species (C virginica and C. rhizophorae) and the Asian species (C. gigas and C ariakensis). Ce texte aborde deux types de réserves de conservation environnementale: une « réserve extractiviste » conquise par des populations rurales riveraines au confluent des fleuves Xingu et Amazone; et une « réserve marine» de populations rurales côtières – les deux à l’État du Pará. Au Brésil comme en Europe, ces initiatives ont visé un meilleur mode de vie et le développement des populations locales et, éventuellement, par surplus, régionales ou nationales. Au Brésil, ces objectifs ont été liés au droit à l’usage collectif des forêts, des bois, des cours d’eau et des mangroves. À des degrés différents, ces territoires sont issus de luttes pour la défense des ressources qui étaient la source de la subsistance de leurs habitants, de leur mode de vie et culture, en un mot, leur patrimoine – contre la pénétration d’« étrangers » venus d’ailleurs en dépit de la loi et de la légitimité: entreprises du bois, de grands élevages et de pêche. Ce texte cherche à confronter, au niveau de l’analyse sociologique, des intérêts de groupes, des acteurs, des stratégies des uns et des autres et de leurs perspectives pour les actuelles et futures générations. This article refers to what some writers call, in Brazil, "new territories". Since the debates during the Federal Constitution elaboration, in 1988, they have been implemented, wholly or partly, within the jurisdiction of a state, a municipality, or a district. This paper addresses two types of environmental conservation reserves: an "extractive reserve" conquered by rural people at the confluence of the rivers Xingu and Amazon; and a "marine reserve" of rural populations on the northern coast of Brazil - both in the federal state of Pará. In Brazil as in Europe, these initiatives aim at better living standards and collective development of local populations. In Brazil, these have been linked to the respect of their collective rights to use the forests, wood, water streams and mangroves. To varying degrees, these territories result from struggles in defense of natural resources that were source of livelihood, lifestyle and culture of the inhabitants, in short, their heritage – against the invasion of "foreigners", timber companies, large farms and fishers. This text contrasts, through sociological analysis, the interest groups, the stakeholders, the challenges and their development prospects vis-à-vis the present and future generations. Cette découverte est à l’origine d’un nouveau dispositif énergétique : la pile à combustible microbienne (PACM). Depuis les années 2000, la littérature scientifique sur les biofilms EA et sur les PACM explose, notamment grâce à la découverte de bactéries capables de transférer par voie directe – par des pili ou protéines transmembranaires – des électrons vers les électrodes. Plusieurs sources de bactéries EA ont été à ce jour découvertes, allant de cultures pures à des communautés bactériennes plus complexes. Ces dernières sont issues de milieux aqueux naturels (milieux marins ou d’eau douce), industriels ou urbains (effluents d’industrie, eaux usées domestiques). La plupart de ces sources bactériennes proviennent d’environnements de climat tempéré. Dans ce travail de thèse, plusieurs sols de milieux humides de la Guyane ont été identifiés comme étant de sources de bactéries EA. Les expériences menées sous potentiel d’électrode imposé et constant ont permis d’étudier l’adhésion à la surface d’électrode des biofilms EA issus de la flore endogène des milieux sélectionnés. La formation de bioanodes et de biocathodes a été possible en présence respectivement d’acétate et d’oxygène dans les milieux. Une étude par voltammétrie cyclique a mis en évidence les pics d’oxydo-réduction en lien avec les échanges électroniques du biofilm EA et de l’électrode. En optimisant la procédure de formation des biofilms EA par chronoampérométrie (surface d’électrode plus importante, apport en continu et progressif du substrat), une densité de courant maximale de 12 A/m2 et un rendement faradique de 24 % ont été obtenus. Une autre méthode pour former des biofilms EA à partir d’un milieu choisi, la mangrove, a consisté à utiliser deux prototypes de PACM : une pile à compartiment unique et à cathode à air, et une pile benthique. Dans les deux cas, les biofilms EA ont pu être formés et étudiés, complétant certaines observations faites sous potentiel imposé. La PACM benthique s’est avérée être la plus proche d’une application à grande échelle puisqu’elle a été complètement autonome : anode et cathode utilisant uniquement le milieu pour fonctionner. Elle a pu être étudiée en laboratoire comme sur le terrain. Electroactive biofilms (EA) are mono or multi-species bacterial consortia, which have the ability to exchange electrons from their metabolism with solid surfaces of conductive electrodes. This discovery is the basis for a device of energy production: microbial fuel cell (MFC). Since the 2000s, the scientific literature on EA biofilms and MFC explodes, thanks to the discovery of bacteria that are able to transfer directly – by pili or trans-membrane proteins – electrons to electrodes. Several sources of EA bacteria were discovered to date, ranging from pure cultures to more complex bacterial communities. Those last are from natural (marine and freshwater), industrial or urban (industrial effluents, domestic wastewater) aqueous environments. However, the vast majority of these are from temperate environments.In this thesis, several wetland soils of French Guiana have been identified as sources of EA bacteria. Experiments under poised and constant electrode potential were used to examine adherence to the electrode surface of EA biofilms from the endogenous flora of selected environments. Formation of bioanodes and biocathodes was possible in the presence respectively of acetate and oxygen in the media. A study by cyclic voltammetry showed the redox peaks related to electronic exchanges between EA biofilm and electrode. By optimizing the process of EA biofilm formation by chronoamperometry (larger electrode surface, providing continuous and progressive substrate), a maximum current density of 12 A/m2 and a coulombic efficiency of 24% were obtained.Another method to form EA biofilms from a chosen medium (mangrove) was to use two MFC prototypes: a single compartment and air cathode one, and a benthic one. In both cases, the EA biofilms have been trained and studied supplementing certain observations made under poised polarization. MFC benthic proved to be the closest to a wide application since it was completely autonomous, anode and cathode only using the medium to function. Instituée pour développer la pêche maritime et garantir un revenu aux pêcheurs, la politique commune de la pêche (PCP) s’est orientée progressivement vers la protection des espèces et de l’environnement marin. Pour cela, elle a sacrifié une part importante du secteur productif, mais a échoué et, avec le déclin de la pêche en Europe, les régions ultrapériphériques de la Caraïbe (RUP-C) ont gagné en importance relative dans ce domaine. Elles représentent aujourd’hui près de 30 % de la flotte nationale et les fondements malthusiens de la PCP ne sont plus adaptés à leurs caractéristiques : ressources halieutiques, développement économique, emplois, sécurité des approvisionnements alimentaires, etc. La réforme actuellement en cours, qui devrait aboutir avant 2012, offre peut-être une alternative aux RUP-C permettant la mise en œuvre des mesures spécifiques en application de l’article 349 du Traité sur le Fonctionnement de l’Union européenne. The European Union currently operates the second reform of its Common Fisheries Policy since its creation in 1970. Established to develop the fisheries and provide incomes for fishermen, the Common Fisheries Policy (CFP) was directed gradually towards the protection of the species and the marine environment. To do this, it sacrificed a large part of the productive sector, but failed, and with the decline of fisheries in Europe, the outermost regions of the Caribbean gained relative importance in this field. They now account for nearly 30% of the national fleet and the Malthusians bases of the CFP are not suited any more to their characteristics: fisheries resources, economic development, jobs, security of food supply, etc. The ongoing reform, which should be in force before 2012, may offer an alternative to the outermost regions of the Caribbean for the implementation of specific measures under Article 349 of the Treaty on the Functioning of the European Union. Dans la partie est du plateau continental guyanais, les mérous géants se localisent autour de tous les sites rocheux marins où ils se répartissent en fonction de la profondeur et du faciès (éboulis ou tombant). Les analyses des isotopes stables du carbone et de l’azote suggèrent qu’environ 30 % des mérous géants < 115 cm présents sur ces sites rocheux, proviendraient des mangroves alentours. Les analyses de contenus stomacaux révèlent un changement ontogénétique du régime alimentaire. Les petits individus (< 90 cm) s’alimentent principalement de Crustacés puis intègrent progressivement les poissons à leur alimentation jusqu’à devenir essentiellement piscivores au-delà de 140 cm. La population de Guyane est principalement composée d’individus juvéniles : seulement 8,5 % des individus capturés en Guyane a plus de 6 ans. Ces jeunes mérous sont très territoriaux durant les 4-5 premières années de leur vie puis ne sont plus retrouvés sur les sites rocheux. Le devenir de ces juvéniles est incertain, soit ils sont pêchés, soit ils migrent vers d’autres sites ou vers les agrégations de reproduction. Cependant, les mérous géants ne semblent pas se reproduire en Guyane. Il est possible que les conditions environnementales (forte turbidité et température) ne favorisent pas le développement gonadique, ce qui engendrerait une omission totale de reproduction. Le suivi par tags satellite suggère que les mérous géants migrent dans le sens des courants marins vers Trinidad-et-Tobago, potentiellement pour la reproduction. Aucun mouvement vers le Brésil n’a été mis en évidence bien que les stocks de mérous géants du Brésil et de Guyane semblent liés. En effet, ces deux stocks évoluent conjointement avec un déclin des populations au début des années 90. Le maintien de la population de mérou géant en Guyane se ferait grâce à l’apport de larves des sites d’agrégation du Brésil par le courant marin. La mortalité totale des mérous géants (0,65) est aussi élevée au sein de la réserve marine ainsi que sur les sites de pêche. Cela suggère que l’interdiction de pêche à la réserve n’est pas respectée et que les grands individus quittent la réserve pour des sites plus spacieux. En supposant que la population de mérous géants adultes a été réduite à cause de la pêche, une protection régionale des individus > 150 cm doit être instaurée afin de régénérer le stock d’individus matures. Il semble nécessaire de développer une politique de gestion internationale de l’espèce, surtout entre la Guyane et le Brésil, afin de conserver les populations de mérous géants d’Amérique du Sud. The critically endangered Goliath Grouper, Epinephelus itajara, is found in the turbid and brackish waters of French Guiana, where the species is subject to a coastal fishery. Goliath Grouper populations inhabit all rocky habitats and exhibit increasing abundance with depth. Around 30% of individuals less than 115 cm may derive from the abundant mangrove habitat along the french Guiana coast. There is a ontogenetic shift of the diet, primarily crustacivorous, individuals >140 cm become piscivorous. The Goliath Grouper population in French Guiana is composed primarily og juvelines. The fate of the juvelines is unknonw, some may be caught in the fishery, others may move to other habitat. No evidence of spawning or gonadal readiness was found in french Guiana. it is possible that environmental conditions (high turbidity and temperature) are not conducive to reproductive activity. Goliath Grouper may migrate down current from French Guiana to Trinidad and Tobago or further. No movement of Goliath Grouper was found against the current toward Brazil. French Guiana juveniles may be derived from spawns occuring off Brazil. Calculated total mortality of Goliath Grouper off French Guiana is high (0.65) in both fished and protected areas suggesting that illegal fishing is occuring within the protected area, or that emigration rates are high. The recently proposed phylum Thaumarchaeota may represent the deepest branching lineage in the archaeal phylogeny emerging before the divergence between Euryarchaeota and Crenarchaeota. Here we report the first characterization of two marine thaumarchaeal species from shallow waters that consist of multiple giant cells. One species is coated with sulfur-oxidizing g-Proteobacteria. These new uncultured thaumarchaeal species are able to live in the sulfide-rich environments of a tropical mangrove swamp, either on living tissues such as roots or on various kinds of materials such as stones, sunken woods, etc. These archaea and archaea/bacteria associations have been studied using light microscopy, transmission electron microscopy and scanning electron microscopy. Species identification of archaeons and the putative bacterial symbiont have been assessed by 16S small subunit ribosomal RNA analysis. The sulfur-oxidizing ability of the bacteria has been assessed by genetic investigation on alpha-subunit of the adenosine-5_- phosphosulfate reductase/oxidase's (AprA). Species identifications have been confirmed by fluorescence in situ hybridization using specific probes designed in this study. In this article, we describe two new giant archaeal species that form the biggest archaeal filaments ever observed. One of these species is covered by a specific biofilm of sulfur-oxidizing g-Proteobacteria. This study highlights an unexpected morphological and genetic diversity of the phylum Thaumarchaeota. Physicochemical variables (depth, temperature, salinity, dissolved oxygen, pH) and population descriptors (species and family richness, density and biomass) were measured. The Whitefield and the well-being indexes were calculated to appreciate species equilibrium. Canonical correspondence analysis and generalized linear models were used to link species assemblages to physicochemical variables and to search for a special link with salinity. A total of 30,733 individuals belonging to 34 species and 23 families were collected. The Gerreidae, Clupeidae, Engraulidae, Scianidae, and Sparidae represented almost 98% of the total number of individuals. Our results show that organization of the fish assemblages in the lagoon is poorly dependent on salinity, whereas dissolved oxygen, temperature, and pH could have a stronger role. Looking at response curves to salinity change, most fishes species appeared to be visitors, either temporarily present or regular migrators. Only a few species compose the year-round resident population. They have a high patrimonial value and could be considered bioindicator species for long-term studies (e.g., of global change and natural or anthropogenic disturbances). The Euclidean distance and biomass variables were not useful descriptors of confinement and biological zonation in the lagoon, whereas the negative gradient of specific richness, number of individuals, Whitefield index, and index of well-being better described confinement. On the basis of these last parameters, biological zonation in the Manche-à- Eau lagoon was established and is provided. Poor in species, the mangrove of Benin belongs to the tropical Atlantic type tending to be humid, considering the "drought belt" which affects this region. It undergoes profound hydrological changes in the course of the different seasons in relation to the Mono and Couffo Rivers. The granulometric analysis and the study of the facies makes clear a lenticular-type sedimentation in relation to numerous and rapid hydrodynamic variations. The clayish quartzose sediment also contains a chemical phase (pyrite is transformed through oxidation into jarosite, gypsum and halite in relation to the micromilieux of evaporation) and an important organic phase - with a pH level ranging from 5 to 6. Kaolinite, from the nearby "Continental terminal", is deposited during calm periods. The smectites seem linked to high energy, sandier deposits and their origin should probably be sought in a far away back country (Eocene, basement). Des conditions climatiques et océanographiques homogènes permettent à l’ensemble du bassin Caraïbe de constituer une unité sur les plans floristique et faunistique. Cette unité n’exclut pas une diversité biologique exceptionnelle avec notamment des espèces endémiques à la zone ; ce qui confère aux côtes des pays caribéens un intérêt écologique, paysager et scientifique certain. La reconnaissance sur le plan international et régional de l’importance de la Mer des Caraïbes mais aussi de sa fragilité a permis au milieu marin caribéen d’acquérir son statut de patrimoine naturel.En plus des services écosystémiques qu’elle rend (ressources halieutiques, protection des littoraux…), la Mer des Caraïbes constitue un atout indéniable dans le cadre du développement touristique de la zone aussi bien par le balnéaire que, plus récemment, par l’écotourisme. Ce dernier s’appuie généralement sur des aires marines protégées et doit permettre un développement durable des îles.A partir d’une étude de cas, le parc marin de Saba, l’objectif de cette étude est d’analyser les rapports entre patrimoine naturel marin et l’écotourisme, ainsi que les impacts économiques, sociaux et environnementaux de la mise en valeur de ces ressources. The Caribbean Sea is home to a diversity of coastal habitats that support the installation of three rich and complex ecosystems - coral reefs, mangroves and seagrasses -. There are also other unique formations such as stromatolites or blue holes. Oceanographic and climatic conditions allow homogeneous throughout the Caribbean basin to form a unit in terms of flora and fauna. This unit does not exclude an exceptional biological diversity, including species endemic to the area, which give the coasts of the Caribbean countries an ecological, landscape and some science. Recognition on the international and regional importance of the Caribbean Sea but also its fragile marine environment has allowed the Caribbean to gain its status as a natural heritage.In addition to the ecosystem services it render (fisheries, coastal protection ...), the Caribbean Sea is a definite asset in the development of tourism in the area by the seaside as well as, more recently, ecotourism. The latter is generally based on marine protected areas and should facilitate the sustainable development of islands.From a case study, the Saba Marine Park, the objective of this study is to analyze the relationship between natural heritage and marine ecotourism, as well as economic, social and environmental development of these resources. However our knowledge of oyster phylogeny and systematics is fragmentary. This is principally due to the plastic growth patterns of these animals, which result in a wide range of overlapping, ecophenotypic variants that greatly reduce the value of analysis based on shell morphology. Besides that, many intentional or accidental anthropogenic transfers have emphasised this situation. In several cases of misclassification or misidentification of oysters, DNA molecular data, have provided valuable new insights on the tracing of introductions or more generally on the phylogeographic relationships between oyster species. We will illustrate this issue with recent case studies from our lab on several flat and cupped oysters: (1) the Crassostrea gigas Crassostrea angulata couple of Asian species and their successive introductions in Europe, (2) the new species of cupped oysters, Crassostrea honkongensis, described in Hong-Kong and very divergent from the species Crassostrea gigas it was initially supposed to be, (3) the mangrove oyster Crassostrea gasar that was shown not only to be present along the coasts of Western Africa but also along the Atlantic coasts of South America, and (4) the geographic disjunction between Ostreola stentina, described in the Mediterranean Sea, and the genetically closely related species Ostrea aupouria (from New Zealand) and Ostreola equestris (from Mexico Gulf/Atlantic). At the intra specific level, we will focus on one of the main documented case in Europe, the flat oyster, Ostrea edulis. It is a marine bivalve whose natural geographical distribution ranges along the European Atlantic coast from Norway to Morocco, in addition to the Mediterranean and Black Sea. Studies of allozymes, microsatellites and mitochondrial differentiation over the whole range concluded that a significant divergence existed between Mediterranean and Atlantic populations, together with an isolation-by-distance pattern. However, the average mitochondrial haplotypic diversity displayed a high among populations variance, reflecting smaller effective population size in some locations. Additionally, a ten-fold quantitative difference was observed in the same study in Fst between the mitochondrial and the nuclear genomes, which could be due to sex biased differential reproductive success between males and females. In order to further document this hypothesis, two experiments at the population level were conducted. First, brooding females were sampled in a wild population and the number of males fertilizing a given female estimated. Then, parentage analyses were achieved under experimental conditions: successive mass spawnings were collected from a population of potential genitors kept in hatchery, whose genotypes were known, in order to infer a posteriori the relative contribution of each genitor. Moreover, we aimed to better understand the reproduction dynamics of this species. Several patterns of spawning could be distinguished: unique, successive or extended in time. The different parental contributions and reproductive behaviors observed in these experiments are discussed in the context of the hypothesis of a variance in the reproductive success of males and females and consequences in local and temporal reduced effective population sizes. Moreover, the effluent shows hydrobiological characteristics (minerals, phytoplankton biomass and organic matter) significantly higher than that of the receiving environment. Separate surveys were carried out in a bay (CH Bay) with a medium-size intensive farm (30 ha) (PO) and in a mangrove-lined creek (TE Creek) near a larger semi-intensive farm (133 ha) (SO). Net loads of nitrogen exported from the semi-intensive farm and the intensive farm amounted to 0.68 and 1.36 kg ha(-1) day(-1), respectively. At CH Bay, discharge effects were spatially limited and clearly restricted to periods of effluent release. The high residence time at site TE favoured the installation of a feedback system in which organic matter was not exported. Mineralization of organic matter led to the release of nutrients, which in turn, caused in an increased eutrophication of this ecosystem. The study of the pico- and nanophytoplankton assemblages showed (i) a shift in composition from picophytoplankton to nanophytoplankton from offshore towards the coast and (ii) a shift within the picophytoplankton with the disappearance of Prochlorococcus and the increase of picoeucaryotes towards the shoreline. These community changes may partially be related to a nitrogen enrichment of the environment by shrimp farm discharges. Thus, in view of the recent addition of the New Caledonian lagoon to the UNESCO World Heritage list, the data presented here could be a first approach to quantify farm discharges and evaluate their impact on the lagoon. En effet, les écosystèmes marins sont essentiels, d’autant plus, qu’ils présentent en Nouvelle-Calédonie une grande richesse, notamment en termes de patrimoine culturel. Ce sont majoritairement les Kanak qui habitent sur la Côte Est de la Province Nord, où la Collectivité et le WWF collaborent à la mise en place de plusieurs aires marines protégées (AMP). Jusqu’à présent, la gestion coutumière Kanak a permis une relative préservation de ces écosystèmes dans un secteur littoral non soumis aux pressions de l’urbanisation, du tourisme ou de la navigation. L’étude s’attache à souligner les enjeux autour de la mise en place des AMP et des perspectives de développement local, sur le milieu marin et les populations locales. Il s’agit de voir comment cette expérimentation est envisagée par les différents acteurs (État, Province Nord, associations, coutumiers etc.) et de s’interroger sur les mécanismes de gouvernance. This study allows a distinct approach to thinking about the marine resources in a protected area. Indeed, marine ecosystems are essential, especially as they are in New Caledonia, particularly in terms of cultural heritage. It is mainly Kanak who live on the East Coast of the North Province, where the Community and WWF collaborate on the installation of several marine protected areas (MPAs). So far, the Kanak customary management allowed a relative preservation of these ecosystems in a coastal area not subject to the pressures of urbanization, tourism and navigation. The study aims to highlight the issues surrounding the establishment of MPAs and opportunities for local development on the marine environment and local populations. ![]() | Etude de génétique des populations sur l'huître creuse de mangrove, Crassostrea gasar (Adanson, 1757), par l'apport du marqueur ITS2 Auteur(s) : Moreau, Dimitri Résumé : Le genre Crassostrea regroupe de nombreuses espèces dont la répartition mondiale s'étend sur une grande partie des régions côtières des basses et moyennes latitudes. La classification des différentes espèces décrites à partir des critères morphologiques et de répartition géographique est aujourd'hui remise en cause dans de nombreux cas par les nouvelles études utilisent les techniques récentes de biologie moléculaire. C'est en particulier grâce à l'apport des séquences génomiques variables, qu'il a été possible de déterminer différentes espèces présentes sur les côtes africaines et américaines (Lapègue & al.). La présence de l'espèce C. gasar, d'abord décrite en Afrique, a ainsi été montré en Amérique. La répartition de cette espèce sur les deux continents laisse supposer que les barrières géographiques on induit une limitation des flux géniques et donc permis une divergence génétique des populations. Pour confirmer cette hypothèse une étude génétique des populations sur 3 populations de chaque continent a donc été réalisée. Elle s'est basée sur le marqueur nucléaire ITS2, séquence non transcrite et variable du génome nucléaire. Il a donc été possible, grâce aux techniques de RFLP (restriction fragments length polymorphisme) de démontrer une forte différenciation génétique entre les deux continents. Cependant le séquençage montre une divergence nucléotidique faible (3%). Ceci laisse supposer qu'il n'y a pas de flux génique entre les deux continents, et que les populations ont divergé récemment. De plus, le polymorphisme intracontinental observé uniquement en Afrique nous laisse supposer les populations américaines seraient originaires d'Afrique. La mise au point du séquençage et la lecture de deux séquences pour des individus de chaque continent a de plus permis de sélectionner de nouvelles enzymes de restriction utilisable en RFLP pour poursuivre l'étude du polymorphisme entre les populations. In this study, the spatial and temporal variability of porewater silicic acid (Si(OH)4) profiles e that integrate benthic transport and reaction processes e was investigated at different spatial (metre, longitudinal and crosssection, intra-estuary) and temporal (tidal, seasonal) scales in two macrotidal estuaries, very close geographically but essentially differing in their shape. Studying the spatial and temporal variability of Si(OH)4 concentrations in porewaters provided evidence for the importance of transport processes, e.g. bio-irrigation, tidal pumping, resuspension and any combination of these processes, in affecting Si(OH)4 concentrations and fluxes and therefore temporary or permanent retention along the landeocean continuum. We confirm that aSiO2 (amorphous silicate) transported by rivers and estuaries clearly needs to be better characterized as it provides an important source of reactive aSiO2 to sediments. This study allows us to: (1) interrogate spatial and temporal scales, although both are most often in complete interaction; (2) design the most appropriate sampling schemes to be representative of any given system and to extrapolate at the scale of the whole estuary; (3) quantify uncertainty associated to the estimations of Si(OH)4 stocks and fluxes in this type of ecosystem, essential for budget calculations. We showed that two adjacent small macrotidal estuaries, may exhibit different behaviours regarding Si retention. Temporary retention has been observed in the meanders of the Aulne Estuary and not along the more linear Elorn Estuary, demonstrating the importance of the morphology and hydrodynamic components of the estuarine filter. Research is needed in other systems and climatic zones, but our study suggests that the typology should not only account for the different types of landeocean continuum (fjord, delta, mangrove.), but also incorporate the physical or biological attributes of the estuarine filter.
