Experimental evaluation of co-culture of juvenile sea cucumbers, Holothuria scabra (Jaeger), with juvenile blue shrimp, Litopenaeus stylirostris (Stimpson) Auteur(s) : Purcell, S Patrois, Jacques Fraisse, N Éditeur(s) : Blackwell science Résumé : The co-culture of juvenile sea cucumber Holothuria scabra (Jaeger), or 'sandfish', with juvenile blue shrimp Litopenaeus stylirostris (Stimpson) was tested by growing groups in co-culture and monoculture for 3 weeks in tanks with enriched sand substratum. Feed was supplied on trays, accessible only to shrimp. Survival of shrimp and sandfish was high in all treatments (73-100%). Growth of shrimp did not differ between monoculture and co-culture, but sandfish grew significantly slower in co-culture (P=0.03), although their sand burying and surface foraging were apparently unaffected by shrimp (P=0.76). However, shrimp increased the levels of total ammonia-N in tanks, which related inversely with sandfish growth (P=0.04). Conversely, sandfish did not appear to lower the water quality for shrimp culture. While sandfish bioturbate sediments and eat organic deposits, the juveniles did not significantly reduce the organic content of sand in tanks. Co-culturing juveniles of the two species in earthen ponds appears feasible, with no detriment to shrimp production, presenting a cost-effective method for growing sandfish to larger sizes for restocking. These findings underpin further studies to test the viability of commercial co-culture of sandfish with blue shrimp at later stages in the production cycle of shrimp. Probiotic P-acidilactici application in shrimp Litopenaeus stylirostris culture subject to vibriosis in New Caledonia Auteur(s) : Castex, Mathieu Chim, Liet Pham, Dominique Lemaire, Pierrette Wabete, Nelly Nicolas, Jean-louis Schmidely, P Mariojouls, C Éditeur(s) : Elsevier Résumé : We studied the effects of a lactic acid bacterium, Pediococcus acidilactici (strain MA 18/5M, CNCM), as a dietary probiotic on growth performance and some nutritional and microbiological aspects of the shrimp Litopenaeus stylirostris. Experimental shrimp culture was carried out over 10 weeks, using floating cages of 14 m(2) each set in earthen ponds, in a New Caledonia farm affected by "Summer syndrome", a septicaemic vibriosis caused by Vibrio nigripulchritudo. The experiment design testing probiotic-coated pellets against control was replicated in two earthen ponds. High mortalities, characteristic of Summer syndrome, were observed during the first 2 weeks of the trial. The probiotic improved production in the treated cages from both ponds with increases in the survival rate (7% and 15% respectively) and final biomass (8% and 12% respectively). No differences were recorded among treatments on growth performances, but significant lower food conversion ratios were obtained with probiotic treatment. After 5 weeks of rearing, the Hepatosomatic Index and the adjusted dry weight of the digestive gland were significantly increased by 10% and by 9% respectively in shrimps treated with probiotic. In the meantime, the specific activities of alpha amylase and trypsin in the digestive gland showed a significant effect of the probiotic treatment with respective increases by 35% and 55%. The rise in total trypsin activity following morning feeding was also enhanced by the probiotic treatment (P < 0.001). The highest concentration of probiotic (between 10(4)-10(5) CFU g(-1) of fresh gut) in the shrimp gut was obtained 2 h after feeding. The concentration remained high for 4 h after feeding before decreasing until the next meal. Total bacterial counts on Marine agar and TCBS in the gut were significantly lowered by the probiotic treatment over the 10 weeks of the experiment. Additionally, during the first 2 weeks, prevalence and load of V nigripulchritudo strains in haemolymph was lower in animals fed with the probiotic diet. This study demonstrated, under pond conditions, that feeding shrimp with live terrestrial lactic acid bacteria can be an effective treatment for improving shrimp culture affected by vibriosis. Digestive enzyme activity and food ingesta in juvenile shrimp Litopenaeus vannamei (Boone, 1931) as a function of body weight Auteur(s) : Gamboa Delgado, Julián Molina Poveda, César Cahu, Chantal Éditeur(s) : Blackwell science Résumé : A study was conducted to evaluate variations of digestive enzyme activities in Litopenaeus vannamei (Boone) reared in commercial ponds under semi-intensive conditions. Shrimp were collected at each body weight increase of 2 g. As the shrimp grew (2-12 g), significant increases in the activities of lipase and chymotrypsin were observed. The total protease activity decreased from 6 g onwards. Trypsin activity showed a peak at 6 g and amylase activity increased two-fold after 2 g. Additionally, the stomach contents were analysed microscopically for shrimp between 2 and 10 g. Plant matter contributed above 30% of the total stomach content in 6-, 8- and 10-g shrimp. Detritus represented 58% and 62% of the total stomach content in 2- and 4-g shrimp, respectively, decreasing to 33-43% at greater shrimp weights. Artificial feed showed a maximum contribution of 20% in 6-g shrimp. The present results show changes in the enzyme activity after the shrimp reach 6 g in body weight, evidenced by a decrease in total protease and an increase in lipase and amylase activities. The amylase/protease ratio was 2.6 in 2-g shrimp and increased steadily to 9.6 in 12-g shrimp. These findings suggest an adaptation of the enzymatic activity to diets with lower protein content as body weight increases, and may be related to the variation of the different items found in the stomach. Our experiments in hapas within shrimp ponds were designed to detect any deleterious effects of sandfish on shrimp, and vice versa. These experiments showed that a high stocking density of juvenile sandfish had no significant effects on growth and survival of shrimp. However, survival and growth of sandfish reared with shrimp for 3 weeks were significantly lower than for sandfish reared alone. Increased stocking density of shrimp also had a significant negative effect on survival and/or growth of sandfish. A grow-out trial of juvenile sandfish in 0.2-ha earthen ponds stocked with 20 shrimp post-larvae m(-2), and densities of sandfish between 0.8 and 1.6 individuals m(-2), confirmed that co-culture is not viable. All sandfish reared in co-culture were dead or moribund after a month. However, sandfish stocked alone into 0.2-ha earthen ponds survived well and grew to mean weights of similar to 400 g within 12 months without addition of food. The grow-out trial demonstrated that there is potential for profitable pond farming of sandfish in monoculture. Survival rate in FLOC and clear water were recorded and a probiotic was tested under both conditions. Daily growth rate (DGR) was higher in FLOC (p < 0.05) than in CW. Survival in each system increased significantly when a probiotic was included in the diet (p < 0.05). The Vibrionaceae community from the hepatopancreas (HP) and the culture medium did not differ between the two culture media. Nevertheless, a novel group of Vibrio strains was found to be unique to FLOC. No high level of lesions was observed in shrimp tissues from the FLOC + probiotic treatment; it suggests that the probiotic contributed to homeostasis and prevented outbreak of opportunistic pathogenic species. The size of the farming earthen ponds (510 ha) and the subsequent production constraints make it difficult to use them as experimental units for random experiments. Moreover, since every pond has its own characteristics, ponds' effects cannot be ruled out, thus making it hard to define true replicates. In order to design future experiments under conditions resembling those used in production, we evaluated the possible use of floating cages as experimental units with the aim of assessing treatment effects with a reasonable statistical power. To this end, two sets of floating cages were placed respectively in two different ponds in a commercial farm. In both cases the zootechnical conditions in the cages were similar in post-larvae origin, management, and diet and feeding regime. The aim of the study was to evaluate on a technical basis the possibility of rearing shrimps in floating cages set up in earthen ponds and to assess this method from a statistical standpoint. Shrimps reared in and outside the cages showed comparable growth and survival rates. The variability in the zootechnical parameters between cages, expressed as the estimated coefficient of variation (CV) for survival rate, total final biomass (g), final body weight (g), daily increment in body weight (g day− 1) and FCR were 11.0, 13.7, 4.4, 5.4, and 7.0%, respectively. On the basis of these figures, we calculated for a given statistical power (80%) the number of replicates (cages) that would be required to reveal significant differences between two treatments, at a 5% level of significance. We found that for expected differences of 20% from the control mean, 3 and 6 floating cages per treatment would be reasonable to determine statistical differences for growth parameters and survival rate, respectively. Moreover, we showed a significant pond effect in regard to survival and growth between the two sets of cages. These results illustrated the within-farm variability among the ponds, and confirmed that the specific characteristics of each pond from the same farm make it difficult to use the ponds themselves as experimental units. The study demonstrates that rearing in floating cages is an economical, powerful and sensitive experimental tool for shrimp culture studies specifically carried out under conditions close to semi-intensive production. 'Syndrome 93' is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Litopenaeus stylirostris in grow-out ponds in New Caledonia. This study assessed the toxic activities of extracellular products (ECPs) from V: penaeicida, V. alginolyticus and V, nigripulchritudo using in vivo injections in healthy juvenile L. stylirostris (= Penaeus stylirostris) and in vitro assays on shrimp primary cell cultures and the fish cell line epithelioma papulosum cyprini (EPC). Toxic effects of ECPs were demonstrated for all pathogenic Vibrio strains tested both in vivo and in vitro, but for shrimp only; no effect was observed on the fish cell line. ECP toxicity for New Caledonian V. penaeicida was found only after cultivation at low temperature (20 degrees C) and not at higher temperature (30 degrees C). This points to the fact that 'Syndrome 93' episodes are triggered by temperature drops. The assays used here demonstrate the usefulness of primary shrimp cell cultures to study virulence mechanisms of shrimp pathogenic bacteria. Assessment of tropical shrimp aquaculture impact on the environment in tropical countries, using hydrobiology, ecology and remote sensing as helping tools for diagnosis Auteur(s) : Fuchs, Jacques Martin, Jean-louis M. Populus, Jacques Résumé : After a period of rapid development of the sites (an increase in both reared surface area and production), tropical shrimp aquaculture is currently being faced with critical problems due to economical and ecological constraints. In many countries with favourable conditions for shrimp aquaculture such as indonesia or Vietnam, sites are often badly selected and/or over-exploited. This conducts to sorne extent to decreases or collapses in the production due to the difficulty in predicting the maximum production capacity of the sites. Furthermore, it appears that production sustainability depends on many factors among which socio-economy and ecolo gy are of prime importance.
The STD3 project titled « Assessment of tropical shrimp aquaculture impact on the environment in tropical countries using hydrobiology, ecology and remote sensing as helping tools for diagnosis )) was initiated in 1994, with the aim to study the sustainability of marine shrimp aquaculture in tropical areas, main! y Indonesia and Vietnam, through the following tasks: (i) improve site selection and study the impact of aquaculture on marine environments presenting a variable sensitivity to organic sewage, (ii) analyse the socio-economical aspects and profitability of the aquaculture industry and of common resources (iii) use remote sensing and geographical data bases for diagnosis and monitoring of site degradation (iv) reinforce the capacity of Asian
scientists through training.
Severa! aquaculture sites have been investigated in the Lampung region (South Sumatra, Indonesia), the Mekong delta (Vietnam) and on the West coast of New-Caledonia, providing a large range of typical ecosystems encountered in Asia and in the Pacifie, from coralline sites to coastal plains with mangrove and deltaic areas. The study concerned the spatial structure and seasonal influence of these ecosystems, the temporal variation during the course of the project (3 years) and the relation between the ecological structure and farm productions.
Remote sensing provides a synoptic vision over large land expanses. Severa! scenes were processed for landuse mapping using conventional classification techniques. Concerning water quality assessment, a general relation for the Java sea was found between image and field data in terms of total suspended matter. Applying this relation to a new site in Sumatra has provided an initial approach to water type and, together with land use mapping, a preliminary assessment of the suitability of the area to shrimp aquaculture development.
Socio-economic research has been focused on the identification ofneeds for collective action, including public policy, in the perspective of shrimp farming sustainability. The main concern is in the regulation of shrimp farming intensification and extensification at the scale of coastal ecosystemic entities in a common property resource management perspective. The common considered here is coastal water quality. Comparing the local development profiles and the institutional grounds for the design and implementation of collective management rules shows that variables such as land tenure system, farm owner socio-economic profile, social homogeneity or heterogeneity of the farmers are key factors to analyse the potential for sustainability. The defmition of
water quality and the possible means to ensure its collective management are discussed. The main conclusion is that the economie incentives to farming development are strong but there is no significant difference among traditional, semi-intensive and intensive systems in terms of economie efficiency or wealth distribution.
The description of the functioning of the different kinds of ecosystems make it possible to give an advice concerning the positioning of the activity inside the ecosystem, and furthermore to have a reference state in order to determine the impact of the activity on the coastal environnement. This impact can be summarized as an increase in the concentration of total suspended matt€?r, of particulate organic matter and of sulfate reducing bacterias. This organic ma~.er is either issued from rearing activities or from telluric origine, due to mangrove eradication when building the ponds.
An evaluation of the relationship existing between productivity and characteristics of each ecosystem bas been conducted by correlating ecological indicators data (concentration of total suspended matter, particulate organic matter, chlorophyll, percentage of pheopigments in total pigments and cyanobacteria and sulfatereducing bacteria in water and sediments) with the average production of the farms. This comparison lead to the compilation of a scale of observed production integrating the range of these environmental parameters with shrimp production levels, bound to facilitate the positioning of shrimp farming in relation with the confmement leve! of each ecosystem.
Lastly, ali geographie data originated from the various compartments of the study have been geo-referenced and loaded into a geographical information system. This allows to display any query made on spatial variables and their related statistical data, including the ir variations over the last few years and to reveal patterns and phenomena otherwise not obvious. Le programme mis en oeuvre par la DRV (Paris, CREMA-L'Houmeau, DELIAO Brest, COP Tahiti, GIE/RA Nouvelle Calédonie), dans le cadre d'un projet européen STD3 «(Sciences et Technique au Service du Développement», en collaboration avec l'Université de Montpellier II, l'Ecole Nationale d'Agronomie de Rennes, l'ITC (Hollande), l'Institut Océanographique de Nha Trang (Vietnam), le BADC de Jepara et le BPPT de Jakarta (Indonésie), avait pour but la mise au point de méthodes permettant d'améliorer la sélection des sites ainsi que le suivi du développement et de l'impact de l'aquaculture des crevettes Péneides en milieu tropical pour éviter la surexploitation. Feasibility of polyculture of blue shrimp Litopenaeus stylirostris and goldlined rabbitfish Siganus lineatus in a mesocosm system Auteur(s) : Luong, Trung Cong Hochard, Sebastien Royer, Florence Lemonnier, Hugues Letourneur, Yves Éditeur(s) : Elsevier Science Bv Résumé : The study was conducted to (1) estimate the effects of polyculture of blue shrimp Litopenaeus stylirostris with goldlined rabbitfish Siganus lineatus on production, environmental quality and water and sediment metabolism and (2) to determine if blue shrimp and goldlined rabbitfish polyculture is possible. The experiment was carried out for 12 weeks in a mesocosm system that consisted of 12 circular outdoor mesocosm fiberglass tanks (1.7 m2, 1275 L water volume). Shrimp (2.9 g) were randomly stocked at density of 15 shrimp.m− 2 without rabbitfish (control), with rabbitfish (25.5 g) at low density (1.2 fish.m− 2) (LDRB) and high density (2.4 fish.m− 2) (HDRB). Results indicated that goldlined rabbitfish S. lineatus did not affect shrimp growth performance. At the end of the experiment, the combined shrimp and rabbitfish yields in the LDRB (213 g.m− 2) treatment and the HDRB (295 g.m− 2) treatment were significantly greater (P < 0.05) than the shrimp yield in the control (143 g.m− 2). Most of the water and sediment parameters were similar among treatments. There was no significant difference (P > 0.05) in gross primary productivity and respiration between the HDRB treatment and the control. It was concluded that the polyculture of blue shrimp with goldlined rabbitfish is technically possible without degradation of the environment in the culture system. Moreover, obtained results also demonstrated that the “summer syndrome”was caused by a single, possibly emerging, cluster of virulent strains. Consequently, it was hypothesized that pathogenic strains of V. nigripulchritudo may persist from one year to the next in the shrimp farm environment and re‐develop inside the grow‐out system at the following rearing cycle (3). This study was therefore aimed at determining whether V. nigripulchritudo isolates may survive, or not, in a shrimp pond bottom soil during a 18‐week drying period. Pathogenicity of Vibrio penaeicida or white shrimp Litopenaeus vannamei: a cysteine protease-like exotoxin as a virulence factor Auteur(s) : Aguirre Guzman, G Ascencio, F Saulnier, Denis Éditeur(s) : Inter-Research Résumé : The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. Toxicity and median lethal doses (LD50) of Fast Protein Liquid Chromatography (FPLC) products were evaluated on L. vannamei. The first FPLC fraction sample (A) from PFs40 of the AM101 strain displayed LD50 values of 1.68 and 5.61 mu g protein ind.(-1), respectively. The second FPLC process from Fraction A showed a peak (A1) also with toxic effects to shrimp. PFs40, Fraction A, and Peak At showed a 38.5 kDa molecular band (SDS-PAGE), with activity on a gelatin protease zymogram. The lethal effect of PFs40 and Fraction A was inhibited by Proteinase K, CuCl2, E-64, and heat (60 and 100 degrees C) treatments, but was not inhibited by EDTA-Na-2, aprotinin, and soy trypsin treatments. These results and the zymogram inhibition test suggest the presence of a cysteine protease-like proteinaceous exotoxin as a dominant protease, secreted by V. penaeicida Strain AM101. Shrimps reared in and outside the cages presented comparable growths and survival rates. Then it was concluded that shrimp culture in floating cages did not present zootechnical biases comparing to pond culture. The second objective of this study was to assess the present method as a statistical point of view. We found that for expected differences of 20% to the control mean, 3 and 6 floating cages per treatment will be reasonable to determined statistical differences for growth parameters and survival. Furthermore, we showed a significant pond effect as regard as survival and growth between both sets of cages. This results illustrated the within farm variability among the ponds, and confirmed that specific characteristics of each ponds from a same farm make difficult to use them as experimental unit. The study demonstrates that floating cages rearing is an economical, powerful, and sensitive experimental tool for shrimp culture studies specifically undertaken under close to semi-intensive production conditions. Rapid and sensitive PCR detection of Vibrio penaeicida, the putative etiological agent of syndrome 93 in New Caledonia Auteur(s) : Saulnier, Denis Avarre, Jean-christophe Le Moullac, Gilles Ansquer, Dominique Levy, Peva Vonau, Vincent Éditeur(s) : Inter-Research Résumé : Experimental infections of Penaeus (Litopenaeus) stylirostris were performed with a Vibrio penaeicida strain (AM101) isolated in New Caledonia from Syndrome 93 diseased shrimp. Cumulative mortalities resulting from intramuscular injection or immersion of shrimp in bacterial suspensions demonstrated high virulence for this bacterial strain and suggested that V. penaeicida could be the etiological agent of Syndrome 93. The median lethal dose (LD50) for AM101 was 1.3 x 104 CFU (colony forming units) ml-1 by immersion and less than 5 CFU shrimp-1 by intramuscular challenge, with mortality outbreaks at 48 and 22 h after challenge, respectively. A polymerase chain reaction (PCR) detection assay using a primer set designed from the 16S ribosomal RNA gene of V. penaeicida was developed. It gave an expected amplicon of approximately 310 bp in ethidium bromide-stained agarose gels. The specificity of these primers was assessed with different Vibrio species. Furthermore, DNA extracted by the ChelexTM method could be used to detect fewer than 20 cultured Vibrio cells in seawater or shrimp hemolymph by this assay. It appears to be a reliable screening method for detecting V. penaeicida in shrimp and from the aquatic environment. Then a spotlight is offered toward 3 projects: DEDUCTION 2 which aims at optimizing the performances of the shrimp aquaculture sector (T. Laugier), AMICAL which focuses on micro-algae aquaculture (L. Chim) and AMBIO centred on the monitoring and the definition of performance indicators for management of Marine Protected Areas and zones Registered on the World heritage (D. Pelletier). Observation sur la maturation et la reproduction en captivité des crevettes peneides en milieu tropical Auteur(s) : Michel, A Éditeur(s) : Publ. CNEXO (France) (Actes Colloq.)(no. 7) Résumé : The present development of Peneid shrimp culture is limited by the problem set by the routine obtainment of reproduction in captivity. In Polynesia where no species of commercial interest is found naturally, this obtainment is a necessary prerequisite. The observations at the Centre Océanologique du Pacifique of CNEXO were made on 6 species of the genus Penaeus (P. merguiensis, P. aztecus, P. japonicus, P. monodon, P. vannamei and P. stylirostris) raised and maintained in 12 m² outdoortanks. At temperatures, between 25 and 32°, a salinity of 35 and a pH of 8,2, maturations and spawnings are seen throughout the year and several generations were obtained : P. merguiensis, F7 ; P. aztecus et P. japonicus, F3 ;P. monodon, F2 ; P. stylirostris and P. vannamei F1. For P. aztecus et P. mondon maturation is induced by unilateral eyestalk ablation. The animals behavior copulation and external signs of ovarian, development are described. The factors which seem essential to the proper progress of maturation are the temperature, the light intensity, the food and the state of health of the animals. Le développement des élevages de crevettes Pénéidés est actuellement limité par les problèmes que pose l'obtention en routine de la reproduction en captivité. En Polynésie où aucune espèce d'intérêt commercial ne se trouve à l'état naturel, cette obtention était un préalable nécessaire. Les observations effectuées au Centre Océanologique du Pacifique du CNEXO ont porté sUr 6 espèces du genre Penaeus (P. merguiensis, p. aztecus, P. japonicus, P. monodon, P. vannamei et P. stylirostris) élevées et maintenues dans des bacs extérieurs de 12 m² en circulation ouverte. Evaluation du probiotique bactérien Pediococcus acidilactici MA18/5M chez la crevette pénéide Litopenaeus stylirostris en Nouvelle- Calédonie Auteur(s) : Castex, Mathieu Éditeur(s) : Agro Paris Tech Résumé : P. acidilactici MA 18/5M is a dietary probiotic (BACTOCELL®) which has shown its efficiency on several terrestrial and aquatic species. The present thesis aimed to investigate more fully the effects of this probiotic on cultured penaeid shrimps. For this study, we chose a global approach by replacing the problem studied in a model of interactions between three compartments: the shrimp, its intestinal microflora and the probiotic. Having first defined how the probiotic should be administrated, we studied its effects under controlled laboratory conditions at several levels of Litopenaeus stylirostris biology: (i) Nutritional status and growth, (ii) associated bacterial microflora, and (iii) health status with respect to oxidative stress. Then the probiotic improves the growth of the shrimps and the feed conversion ratio. Our study shows that these effects are partially due to a better use of the feed, particularly of dietary carbohydrates, but also suggest an action on the metabolism, andor on the growth via a contribution of essential nutrients. On the other hand, although the intestinal bacterial communities seems little affected, we show an antagonism effect of the probiotic toward total and vibrionacea intestinal flora, as well as a decrease in the level of infection of shrimps during challenge with pathogenic V. nigripulchritudo. Beside, our works highlight that infection by this pathogen, led to a reduction in antioxidant defences, to a rise in oxidative damages, and to a release of mortalities within 48 hours. This oxidative stress following bacterial infection was better controlled by animals receiving the probiotic diet, and resistance of the shrimps to infection was significant improved. A nutritional hypothesis is proposed to explain such result, hypothesis based on a link between carbohydrate metabolism and antioxidant status. As a conclusion of these works, we can propose a first explanatory scenario for the effects of P. acidilactici on a penaeid shrimp. Finally probiotic's effects were assessed under a pilot scale in a commercial farm from New Caledonia currently affected by the "summer syndrome ". We confirm the improvements brought in term of zootechnical results, with in particular, a better resistance to this syndrome. This work was made possible thanks to a breeding method in floating cages, developed during this thesis, and therefore transferred, as an experimental tool, to the shrimp industry of New Caledonia. Pediococcus acidilacticii MA18/5M est un probiotique bactérien (BACTOCELL®) qui a démontré son efficacité chez plusieurs espèces d'animaux terrestres et aquatiques. Cette thèse avait pour objectif d'investiguer plus en détails les effets de ce probiotique chez les crevettes pénéides d'élevage. Pour cette étude, nous avons choisi une approche globale replaçant la problématique dans un modèle d'interactions entre trois compartiments : la crevette, sa microflore intestinale et le probiotique. Après avoir défini les modalités d'administration, l'effet de P. acicilactici a été étudié, en conditions de laboratoire, chez la crevette Litopenaeus stylirostris, à différents niveaux: (i) statut nutritionnel et croissance, (ii) microflore bactérienne associée, et (iii) santé de l'animal à travers son statut antioxydant et son niveau de stress oxydant. Ainsi, le probiotique améliore la croissance des crevettes et l'indice de conversion de l'aliment. Notre étude montre que ces effets sont en partie liés à une meilleure utilisation de l'aliment, notamment des carbohydrates alimentaires, mais suggèrent aussi une action sur le métabolisme, et/ou sur la croissance via un apport de nutriments essentiels. D'autre part, bien que les communautés bactériennes intestinales semblent peu affectées, un effet d'antagonisme du probiotique vis-à-vis de la flore intestinale totale et vibrionacée a été montré, ainsi qu'une réduction du niveau d'infection des crevettes au cours de challenge avec le pathogène V. nigripulchritudo. Par ailleurs, nous mettons en évidence que l'infection par ce pathogène, conduit à un abaissement des défenses antioxydantes, à une élévation des dégâts oxydatifs, et au déclenchement de mortalités dans les 48 heures. Le probiotique atténue le niveau de ce stress oxydant, et améliore significativement la résistance des crevettes à l'infection. Une hypothèse nutritionnelle est proposée afin d'expliquer ce résultat, hypothèse reposant sur un lien entre le métabolisme des carbohydrates et le statut antioxydant. A l'issue de ces travaux, nous pouvons alors proposer un premier scénario explicatif des effets de P. acidilactici chez une crevette pénéide. Finalement le probiotique a été éprouvé à l'échelle pilote sur une ferme commerciale de Nouvelle-Calédonie affectée par le « syndrome d'été ». Nous confirmons les améliorations apportées en termes de résultats zootechniques, avec notamment une meilleure résistance au syndrome. Eutrophication in a tropical pond: Understanding the bacterioplankton and phytoplankton dynamics during a vibriosis outbreak using flow cytometric analyses Auteur(s) : Lucas, Ronan Courties, C. Although difficult to maintain, its stability is required to guarantee the success of the culture. A 110-day period of phytoplankton and bacterioplankton stock and dynamics in an earthen pond (1.2 ha area, 1 m depth) was monitored using flow cytometry to provide baseline information on community characteristics and ecosystem instability. Seven autotrophic cell types were identified over the whole sampling period. Prokaryotic cells included Synechococcus sp., a group named UNK which presented an atypical new flow cytometric signature and picoeukaryotes (PEUK). Nanophytoplankton cells were represented by 4 groups: NAN1, NAN2, NAN3 and Cryptophytes. During the first part of the survey, picophytoplankton dominated the phytoplanktonic assemblage. The mean abundance of total cells (up to 8 x 10(6) cells mL(-1)) was among the highest recorded in marine and brackish waters. Bacterial abundance and production ranged from 0.8 to 5.1 x 10(7) cells mL(-1) and from 30 to 110 mu g C L-1 h(-1). A shift from pico to nanophytoplankton abundance was observed for a few days from d 96. During this period, heterotrophic bacteria production and abundance suddenly dropped, implying a change in the functioning of the microbial loop. This shift was concomitant with a significant shrimp mortality outbreak due to Vibrio penaeicida, the etiological agent of a disease known as Syndrome 93, which affects the shrimp industry in New Caledonia. This survey suggests that flow cytometric analysis could be used for the monitoring of aquaculture systems to improve our understanding of the complex phytoplankton and bacterial dynamics of these systems and its potential influence on disease development. Les échanges internationaux de crevettes pénéidés vivantes entre les pays du Pacifique, de l'Atlantique et de l'Europe Auteur(s) : Michel, Alain Éditeur(s) : Office Int Epizooties Résumé : The tremendous development of penaeid shrimp culture across the world over the past twenty years has led to international trade in eggs, larvae and spawners of the best shrimp species for aquaculture. Trade has involved, in particular, the following species: - Penaeus japonicus from Japan to Europe, the Pacific Islands and South America - P. monodon from South-East Asia to almost all tropical countries - P. vannamei and P. stylirostris from countries along the Pacific coast of South and Central America to the United States of America, the islands of the South Pacific, countries along the Atlantic coast of the Americas and certain countries in Africa. In the 1980s, research conducted by American and French teams enabled the breeding of stocks of spawners in captivity. This resulted in the establishment of lines of captive broodstock outside natural areas of distribution, and the development of new breeding farms in previously unproductive regions. Trade related to movements of these species is facilitated by the absence of legislation in most countries; even when such legislation does exist, it is not always applied These practices have led to the rapid development of world shrimp production, but have also contributed to the dissemination of pathogens. In recent years, the occurrence of serious epizootics - occasioning heavy losses world-wide - has led to a more cautious approach involving trade of post-larvae obtained from captive broodstock in which thorough control measures have been implemented for known diseases. Trade in wild shrimp will probably be abandoned progressively, to avoid the risk of introducing new pathogens. The main challenges for research in the near future are the development of diagnostic tools, to enable continuous control of captive broodstock, and the selection of strains which are resistant to the principal pathogens affecting these species. Selection and characterization of potential probiotic bacteria for Litopenaeus stylirostris shrimp hatcheries in New Caledonia Auteur(s) : Pham, Dominique Ansquer, Dominique Chevalier, Anne Dauga, Clement Peyramale, Aude Wabete, Nelly Labreuche, Yannick Éditeur(s) : Elsevier Science Bv Résumé : In New Caledonia, shrimp hatcheries are confronted with mass mortality in the larval stages, a phenomenon poorly understood as no specific causative agent has been identified. This has resulted in an excessive use of prophylactic antibiotics, although their adverse effects in aquaculture are notorious. The present work was thus aimed at selecting potential probiotic strains for penaeid hatcheries. From a pool of more than 400 marine bacterial isolates sampled from the local marine environment seven strains exhibited in vitro antagonistic activity towards Vibrio harveyi. These isolates were characterized both phenotypically and genotypically using a biochemical approach and 16S rDNA sequencing. Six out of these seven strains were found to belong to the genus Pseudoalteromonas, the last one belonging to the Vibrionaceae family and related to the Harveyi clade. Selected probiotic candidates were individually tested for antagonistic activities in vitro using a green fluorescent protein (GFP)-labelled transconjugant of V. harveyi and for inherent pathogenicity towards cultured shrimp larvae at two different developmental stages. This approach enabled rapid processing and selection of candidates to be tested in our experimental hatchery. Four different experiments were conducted to test candidate strains either alone or in combination. Repeated trials showed that postlarval survival was significantly improved by adding the strain NC201 individually to the rearing water compared to unchallenged controls. The analysis of immune-related gene expressions showed that the Litsty PEN3 transcript abundance of larvae was significantly increased after being reared in probiotic-containing water; however no significant difference in lysozyme gene expression was recorded in this study. Together, these results open new insights into the use of these strains as potential substitutes to antibiotherapy in shrimp larval rearing in New Caledonia.
